First report of human infection by Christensenella minuta, a gram-negative, strickly anaerobic rod that inhabits the human intestine.

Christensenella minuta is a Gram-negative strictly anaerobic short rod that inhabits the human gut. This bacterium was isolated in a mixed infection with Desulfovibrio desulfuricansfrom the blood of a patient with a diagnosis of acute appendicitis. The strain was identified by 16S rRNA sequence analysis. As far as we know, this is the first time C.minuta has been isolated from a human clinical specimen.

Impact of Mycoplasma hyopneumoniae and Lawsonia intracellularis on the performance of pigs divergently selected for feed efficiency.

Feed efficiency (FE) is a valuable trait, yet how genetic selection for enhanced FE affects other processes such as response to disease is unknown. Disease from endemic respiratory and enteric pathogens such as Mycoplasma hyopneumoniae (Mh) and Lawsonia intracellularis (LI) are common in swine production. Therefore, the aim of this study was to examine if pigs selected for high vs. low FE based on residual feed intake (RFI) respond differently to a dual respiratory and enteric challenge. Pigs selected for low RFI (LRFI, high FE) are considered more FE compared to their high RFI (HRFI, low FE)

Occurrence of diarrhoea and intestinal pathogens in non-medicated nursery pigs.

BACKGROUND: Intestinal disease in nursery pigs is the most common cause of antibiotic usage in pigs in Denmark. The decision to initiate batch medication of intestinal diseases in nursery pigs is typically made by the stock personnel based on clinical assessments of pigs and counting of diarrhoeic faecal pools on the pen floor. The target population of this study was batches of nursery pigs (10-66 days after weaning) where the stock personnel assessed the pigs to be without signs of intestinal disease and therefore did not needed treatment. The objective was to determine the within-herd preval

Evaluation of immunogenicity and protection mediated by Lawsonia intracellularis subunit vaccines.

Lawsonia intracellularis is an economically important bacterium that causes ileitis in pigs. Current vaccines for L. intracellularis do not allow for differentiation between infected and vaccinated animals (DIVA), which is beneficial for disease tracking and surveillance. Previously, we identified five putative surface L. intracellularis proteins that were targeted by antibodies from pigs infected with L. intracellularis which could serve as antigens in a subunit vaccine. We conducted two trials to determine whether these antigens were immunogenic and provided protection against infectious cha

Rapid and visual detection of Lawsonia intracellularis via an RPA-CRISPR/Cas12a assay.

The swine industry faces a substantial economic threat from Lawsonia intracellularis (LI), the bacterium responsible for porcine proliferative enteropathy. Infection commonly presents with diarrhea and increased enterocyte proliferation in the ileum and colon, ultimately resulting in impaired growth performance in affected pigs. Effective management of porcine proliferative enteropathy requires timely and accurate diagnosis. The method provides significant advantages over existing detection techniques, offering enhanced sensitivity and high accuracy. The assay achieves detection within one hou

[A case of Desulfovibrio desulfuricans cultured from blood in Japan].

We report a case of Desulfovibrio desulfuricans bacteremia in a 60-year-old-man. In our case, anaerobic blood culture bottle turned out positive after five days' incubation. Gram stain showed the presence of slightly-curved Gram negative rod. Suspecting Campylobacter and Helicobacter, we added microaerobic culture while tentatively reporting Campylobacter to the physician. We then added anaerobic culturing with Brucella HK (RS) Agar because microaerobic culture proved the absence of microaerophile. We found small colonies on the third day, then we started anaerobic culture and eventually ident

Evaluation of porcine ileum models of enterocyte infection by Lawsonia intracellularis.

The early interaction of Lawsonia intracellularis with host cells was examined with the use of porcine ileum models. Two conventional swine were anesthetized, and ligated ileum loops were prepared during abdominal surgery. The loops were inoculated with 108 L. intracellularis or saline. After 60 min, samples of each loop were processed for routine histologic and electron microscopic study. Histologic and ultrathin sections of all the loops appeared normal, with no apposition of bacteria and host cells or bacterial entry events in any loop. Portions of ileum from a single gnotobiotic piglet wer

A low-endotoxic Salmonella vector with dual bacterial-host promoter expression of Lawsonia intracellularis antigens elicits protective immunity in a murine model.

Lawsonia intracellularis, the etiological agent for proliferative enteropathy, remains a major cause of productivity loss and economic burden in the global swine industry. Currently available vaccines provide suboptimal protection and raise safety concerns, highlighting the need for improved immunization strategies. In this study, we developed Salmonella Typhimurium-based vaccine candidates against L. intracellularis in a murine model, incorporating immunoprotective epitopes from the surface autotransporter protein LatA, the flagellar protein FliC, and the heat shock protein 60 (HSP60). Alteration of the gut microbiota following SARS-CoV-2 infection correlates with disease severity in hamsters.

Mounting evidence suggests that the gut-to-lung axis is critical during respiratory viral infections. We herein hypothesized that disruption of gut homeostasis during severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection may associate with early disease outcomes. To address this question, we took advantage of the Syrian hamster model. Our data confirmed that this model recapitulates some hallmark features of the human disease in the lungs. We further showed that SARS-CoV-2 infection associated with mild intestinal inflammation, relative alteration in intestinal barrier propert

Development and application of a triplex real-time PCR method for the detection of Lawsonia intracellularis, Brachyspira hyodysenteriae, and Clostridium perfringens.

Lawsonia intracellularis (LI), Brachyspira hyodysenteriae (Bhy), and Clostridium perfringens (Cp) are three common swine enteric pathogens widespread worldwide, leading to huge economic losses to the pig industry. LI, Bhy, and Cp are co-infected and should not be distinguished, and the detection of bacterial nucleic acids in feces allows rapid identification of whether a herd is infected with these three pathogens. In this study, we developed and optimized a TaqMan triple quantitative real-time PCR (qPCR) method for LI, Bhy, and Cp, which was validated and applied to the analysis of actual sam

Vaccination Against Lawsonia intracellularis Decreases Shedding of Salmonella enterica serovar Typhimurium in Co-Infected Pigs and Alters the Gut Microbiome.

Salmonella enterica serovar Typhimurium continues to be a major cause of foodborne illness worldwide and pork can serve as a source of infection. Co-infection of S. enterica with Lawsonia intracellularis, a common intestinal pathogen of swine, has been found as risk factor for increased S. enterica shedding. The objective of this study was to investigate if vaccination against L. intracellularis could lead to decreased S. Typhimurium shedding. To test this hypothesis, pigs were challenged with either S. Typhimurium or S. Typhimurium and L. intracellularis, with and without L. intracellularis v

The Role of Gut Microbiome in the Pathogenesis of Multiple Sclerosis and Related Disorders.

Multiple sclerosis (MS) is a chronic, progressive neuroinflammatory disease with a complex pathophysiological background. A variety of diverse factors have been attributed to the propagation of inflammation and neurodegeneration in MS, mainly genetic, immunological, and environmental factors such as vitamin D deficiency, infections, or hormonal disbalance. Recently, the importance of the gut-brain axis for the development of many neurological conditions, including stroke, movement disorders, and neuroinflammatory disorders, has been postulated. The purpose of our paper was to summarize current

Metagenomics and metaproteomics alterations are associated with kidney disease in opisthorchiasis hamsters fed a high-fat and high-fructose diet.

BACKGROUND: Opisthorchis viverrini (O. viverrini, Ov) infection and consumption of high-fat and high-fructose (HFF) diet exacerbate liver and kidney disease. Here, we investigated the effects of a combination of O. viverrini infection and HFF diet on kidney pathology via changes in the gut microbiome and host proteome in hamsters. METHODOLOGY/PRINCIPAL FINDINGS: Twenty animals were divided into four groups; 1) fed a normal diet not infected with O. viverrini (normal group), 2) fed an HFF diet and not infected with O. viverrini (HFF), 3) fed a normal diet and infected with O. viverrini (Ov), a

Assessment of oral ciprofloxacin impaired gut barrier integrity on gut bacteria in mice.

Gut bacteria and gut barrier plays important roles in body homeostasis. Ciprofloxacin (CPFX) is widely used to treat bacterial infections. However, whether high dosage of CPFX has side effects on gut barrier integrity is still unclear. Our results indicated that the High CPFX treatment (1 mg/ml) caused weight loss, nervousness, anorexia, and increased apoptosis cells in gut, but less influence was observed in the Low CPFX group (0.2 mg/ml). Meanwhile, the High CPFX treatment impaired tight junction molecules Ocln/ZO-1 level and down-regulated antibacterial genes expression (reg3γ, pla2g2α and

HIV Tat Expression and Cocaine Exposure Lead to Sex- and Age-Specific Changes of the Microbiota Composition in the Gut.

The balance of microbial communities in the gut is extremely important for normal physiological function. Disruption of the balance is often associated with various disorders and diseases. Both HIV infection and cocaine use are known to change the gut microbiota and the epithelial barrier integrity, which contribute to inflammation and immune activation. Our recent study shows that Tat expression and cocaine exposure result in changes of genome-wide DNA methylation and gene expression and lead to worsen the learning and memory impairments. In the current study, we extended the study to determi

Desulfovibrio desulfuricans bacteraemia and associated liver abscess: Case report and literature review.

We present a rare case of Desulfovibrio desulfuricans bacteraemia and associated liver abscess. A 72-year-old Caucasian woman presented with an acute history of fevers, rigors and abdominal pain. She was managed empirically with ceftriaxone and metronidazole and required 24 hours of inotropic support due to hypotension from sepsis. A computed tomography scan of her abdomen demonstrated a 6cm liver abscess in maximal diameter. On day seven of her admission, blood cultures flagged an anaerobic gram-negative bacillus, which was later identified by MALDI-TOF MS as Desulfovibrio desulfuricans. The

Antimicrobial Resistance in Leptospira, Brucella, and Other Rarely Investigated Veterinary and Zoonotic Pathogens.

Leptospira, Brucella, and Borrelia are major agents of zoonotic disease, causing high morbidity and, in some cases, significant mortality in humans. For all three genera, prompt diagnosis and appropriate antimicrobial therapy are required to prevent the development of chronic, debilitating illness. Leptospira spp. are intrinsically resistant to several antimicrobial classes; however, there is little evidence in the literature for development of acquired resistance to antimicrobial agents used for clinical treatment of acute leptospirosis. For Brucella infections, there are numerous reports of

Co-incubation with IL-18 potentiates antigen-specific IFN-γ response in a whole-blood stimulation assay for measurement of cell-mediated immune responses in pigs experimentally infected with Lawsonia intracellularis.

The whole-blood interferon-gamma (IFN-γ) assay is a quantitative in vitro assay for a direct read-out of Ag-specific cell-mediated immune (CMI) responses to infectious diseases. The IFN-γ assay is robust in severe intracellular infections like Brucella or mycobacteria, but more difficult to evaluate for less severe or immunocompromising infections. Here we investigated the performance of the assay when recombinant co-stimulatory cytokines IL-12 and/or IL-18 were added along with Ag or PBS to cultures of whole-blood from pigs infected with Lawsonia intracellularis.
