Acute lymphoblastic leukemia and developmental biology: a crucial interrelationship.

The latest scientific findings in the field of cancer research are redefining our understanding of the molecular and cellular basis of the disease, moving the emphasis toward the study of the mechanisms underlying the alteration of the normal processes of cellular differentiation. The concepts best exemplifying this new vision are those of cancer stem cells and tumoral reprogramming. The study of the biology of acute lymphoblastic leukemias (ALLs) has provided seminal experimental evidence supporting these new points of view. Furthermore, in the case of B cells, it has been shown that all the

Human retinal organoids: from developmental biology to translational applications.

Human retinal organoids (ROs), derived from pluripotent stem cells, have emerged as versatile human-relevant systems for studying retinal development, modeling disease, and evaluating therapeutic strategies. This review summarizes the evolution of human RO technology, from early stem-cell-based differentiation protocols to advanced culture platforms that improve scalability, reproducibility, and functional maturation. We compare human ROs with native human retinas regarding developmental trajectory, cellular composition, and photoreceptor function, while also highlighting persistent gaps in va

Human neural tube defects: developmental biology, epidemiology, and genetics.

Birth defects (congenital anomalies) are the leading cause of death in babies under 1 year of age. Neural tube defects (NTD), with a birth incidence of approximately 1/1000 in American Caucasians, are the second most common type of birth defect after congenital heart defects. The most common presentations of NTD are spina bifida and anencephaly. The etiologies of NTDs are complex, with both genetic and environmental factors implicated. In this manuscript, we review the evidence for genetic etiology and for environmental influences, and we present current views on the developmental processes in

Developmental and functional landscape of theca cells in ovarian biology.

Ovarian theca cells constitute essential components of the follicular microenvironment and play central roles in follicular development, steroidogenesis, and endocrine regulation. Despite their significance, the developmental origins, differentiation processes, and functional dynamics of theca cells remain incompletely defined, particularly in humans. This review provides an updated synthesis of current knowledge on the ontogeny, molecular signaling pathways, and intercellular interactions of theca cells. It also presents recent findings on the role of theca stem or progenitor cells and their

The organizer: What it meant, and still means, to developmental biology.

This article is about how the famous organizer experiment has been perceived since it was first published in 1924. The experiment involves the production of a secondary embryo under the influence of a graft of a dorsal lip from an amphibian gastrula to a host embryo. The early experiments of Spemann and his school gave rise to a view that the whole early amphibian embryo was "indifferent" in terms of determination, except for a special region called "the organizer". This was viewed mainly as an agent of neural induction, also having the ability to generate an anteroposterior body pattern. Earl

The evolution of venom-conducting fangs: insights from developmental biology.

The present study of the origin of the various types of fang represented among colubroid snakes (i.e., tubular, grooved, and ungrooved) attempts to reconcile the morphology of adult fangs with current phylogenetic hypotheses. Observations of growth series of developing tubular fangs were hypothesised to shed light on the evolutionary origin of fangs in snakes. While molecular phylogenies and evolutionary studies of venom proteins and of other anatomical components of the venom-delivery system reconstruct a consistent evolutionary scenario, the character of a tubular venom-conducting fang does

Behavioral inhibition: linking biology and behavior within a developmental framework.

Behavioral inhibition refers to a temperament or style of reacting that some infants and young children exhibit when confronted with novel situations or unfamiliar adults or peers. Research on behavioral inhibition has examined the link between this set of behaviors to the neural systems involved in the experience and expression of fear. There are strong parallels between the physiology of behaviorally inhibited children and the activation of physiological systems associated with conditioned and unconditioned fear. Research has examined which caregiving behaviors support the frequency of behav

Gyrodactylid developmental biology: historical review, current status and future trends.

In the viviparous gyrodactylids, embryos develop one inside another within the parental uterus, a phenomenon with major implications for the biology of this species-rich group. Development occurs via two routes: first-born daughters develop at the centre of an embryo cluster in utero, whereas all other daughters develop from oocytes. The resulting offspring are, however, morphologically indistinguishable. We review here the history of gyrodactylid embryology in the context of current knowledge and, present additional cytogenetic and ultrastructural observations of embryonic development. These

Tendons and Ligaments: Connecting Developmental Biology to Musculoskeletal Disease Pathogenesis.

Tendons and ligaments provide connections between muscle and bone or bone and bone to enable locomotion. Damage to tendons and ligaments caused by acute or chronic injury or associated with aging and arthritis is a prevalent cause of disability. Improvements in approaches for the treatment of these conditions depend on a better understanding of tendon and ligament development, cell biology, and pathophysiology. This review focuses on recent advances in the discovery of transcription factors that control ligament and tendon cell differentiation, how cell and extracellular matrix homeostasis are

Centering Biodiversity: Integrating the Complexity of Sex Into a Developmental Biology Course.

In biology, variation is the norm, not the exception. This particularly holds true for the concepts of sex and sex determination, which we refer to here as biology of sex. Unfortunately, biology of sex topics are often taught using oversimplified content, situating sex as a simple, genetically determined binary, and that mammals are the predominant model for biology of sex across organisms. Teaching using these oversimplifications leads students to develop a limited understanding of biology of sex topics, which limits their capacity as future researchers and also harmfully and erroneously supp

Bridging maternal effects and epitranscriptomics: A novel perspective in developmental biology.

Maternal effects, encompassing both genetic (maternally expressed gene products) and non-genetic (maternal state) influences, are powerful determinants of offspring phenotype, yet their RNA-level mechanisms remain incompletely resolved. In parallel, epitranscriptomics, an emerging field centered on chemical modifications to RNA, has revealed new layers of gene regulation with implications for cell fate, plasticity, and response to environmental cues. In this perspective article, a conceptual link is proposed between maternal effects and epitranscriptomic mechanisms, focusing on how maternal en

Developmental biology and classification of congenital anomalies of the hand and upper extremity.

Recent investigations into the mechanism of limb development have clarified the roles of several molecules, their pathways, and interactions. Characterization of the molecular pathways that orchestrate limb development has provided insight into the etiology of many limb malformations. In this review, we describe how the insights from developmental biology are related to clinically relevant anomalies and the current classification schemes used to define, categorize, and communicate patterns of upper limb malformations. We advocate an updated classification scheme for upper limb anomalies that i

Nail patella syndrome: a review of the phenotype aided by developmental biology.

Nail patella syndrome (NPS) is an autosomal dominant condition affecting the nails, skeletal system, kidneys, and eyes. Skeletal features include absent or hypoplastic patellae, patella dislocations, elbow abnormalities, talipes, and iliac horns on x ray. Kidney involvement may lead to renal failure and there is also a risk of glaucoma. There is marked inter- and intrafamilial variability. The results of a British study involving 123 NPS patients are compared with previously published studies and it is suggested that neurological and vasomotor symptoms are also part of the NPS phenotype.
