Effect of trichloroethylene and its metabolites, dichloroacetic acid and trichloroacetic acid, on the methylation and expression of c-Jun and c-Myc protooncogenes in mouse liver: prevention by methionine.

Trichloroethylene (TCE), dichloroacetic acid (DCA), and trichloroacetic acid (TCA) are environmental contaminants that are carcinogenic in mouse liver. 5-Methylcytosine (5-MeC) in DNA is a mechanism that controls the transcription of mRNA, including the protooncogenes, c-jun and c-myc. We have previously reported that TCE decreased methylation of the c-jun and c-myc genes and increased the level of their mRNAs. Decreased methylation of the protooncogenes could be a result of a deficiency in S-adenosylmethionine (SAM), so that methionine, by increasing the level of SAM, would prevent hypomethyl

A 2-year dose-response study of lesion sequences during hepatocellular carcinogenesis in the male B6C3F(1) mouse given the drinking water chemical dichloroacetic acid.

Dichloroacetic acid (DCA) is carcinogenic to the B6C3F(1) mouse and the F344 rat. Given the carcinogenic potential of DCA in rodent liver and the known concentrations of this compound in drinking water, reliable biologically based models to reduce the uncertainty of risk assessment for human exposure to DCA are needed. Development of such models requires identification and quantification of premalignant hepatic lesions, identification of the doses at which these lesions occur, and determination of the likelihood that these lesions will progress to cancer. In this study we determined the dose r

Age-related differences in the effect of dichloroacetate on postischemic lactate and acid clearance measured in vivo using magnetic resonance spectroscopy and microdialysis.

Numerous studies using adult animal models suggest that dichloroacetate (DCA) may have neuroprotective properties by virtue of its ability to increase rates of metabolism and, therefore, clearance of brain lactic acidosis, which may accumulate during cerebral ischemia. We tested the hypothesis that postischemic DCA administration affects lactate and acid clearance to different extents in immature versus mature brain. 31P and 1H magnetic resonance spectroscopy were used to measure intracellular acid and lactate clearance rates in vivo in newborn and 1-month-old swine after a 14-min episode of t

The role of mouse intestinal microflora in the metabolism of trichloroethylene, an in vivo study.

1. Both trichloroethylene and its metabolite, dichloroacetic acid, produce liver tumors peroxisome proliferation and other adverse cellular alterations in rodents. 2. The hepatic mechanism by which dichloroacetic acid is formed is not conclusively demonstrated, but pharmacokinetic models have successfully associated its formation with trichloroacetic acid as immediate precursor. 3. Previous investigations have shown that dichloroacetic acid is formed from trichloroacetic acid by gut microflora isolated in vitro. 4. To determine the impact of gut microflora on dichloroacetic acid formation from

Cytochrome P-450-dependent covalent binding of 1,1,2,2-tetrachloroethane in vitro.

Incubation of 1,1,2,2,-tetrachloro[1,2-14C]ethane with a reconstituted mono-oxygenase system or with intact rat liver microsomes led to the formation of a metabolite capable of binding covalently to proteins and other nucleophiles. The enzyme system responsible for metabolizing tetrachloroethane to both soluble and covalently bound metabolites was induced by phenobarbital or xylene but not by beta-naphthoflavone treatment. The only soluble metabolite detected upon incubation of tetrachloroethane with a reconstituted system was dichloroacetic acid, whereas microsomes produced both dichloroaceti

Trichloroethylene transformation in aerobic pyrite suspension: pathways and kinetic modeling.

The pathways and kinetics of trichloroethylene (TCE) degradation in aerobic pyrite suspension were investigated. The detection of hydroxyl radical in aqueous pyrite suspension suggested that TCE was degraded by this strong oxidant The reaction pathways of TCE degradation were proposed in which the degradation of TCE to formic acid and finally to CO2 was the main route. Degradation of TCE to oxalic acid and to dichloroacetic acid were found as minor pathways. Degradation rates of TCE to formic acid, glyoxylic acid, and dichloroacetic acid were obtained using kinetic model at 1.2 x 10(-2), 9.8 x

Novel Metabolite of Trichloroethylene in a Methanotrophic Bacterium, Methylocystis sp. M, and Hypothetical Degradation Pathway.

Previously, a new type II methanotrophic bacterium, Methylocystis sp. M (strain M), was isolated in our laboratory [H. Uchiyama et al., Agric. Bioi. Chem., 53, 2903-2907 (1989)]. In this paper, metabolites resulting from the degradation of trichloroethylene (TCE) by strain M were studied with gas chromatography-mass spectrometry. Trichloroacetic acid, dichloroacetic acid, and a small amount of 2,2,2-trichloroethanol were detected in the water-soluble fraction of the reaction mixture. These results suggest that the conversion of TCE to trichloroacetaldehyde via a CI-shift reaction, followed by

Effect of haloacetic acid water disinfection by-products on intestinal barrier function in nematode Caenorhabditis elegans.

Halogenated disinfection by-products are contaminants in drinking water that pose a significant public health concern. The intestine is particularly susceptible to the effects of disinfection by-products through oral ingestion; however, the intestinal toxicity of these compounds remains largely unexplored. This study aimed to assess the impact of 10 haloacetic acids, a major class of drinking water disinfection by-products, on intestinal barrier function using Caenorhabditis elegans as a model animal. Additionally, changes in the expression of five genes related to intestinal barrier function

A review of in vivo 1H magnetic resonance spectroscopy of cerebral ischemia in rats.

A number of metabolic alterations are initiated by cerebral ischemia including dramatic increases in lactate concentration, decreases in N-acetylaspartate, choline, and creatine concentrations, as well as changes in amino acid levels. A review of proton nuclear magnetic resonance spectroscopy studies of focal and global cerebral ischemia in rats is presented here. In particular, studies in neonatal rats have shown that a continued elevation of lactate levels without recovery after hypoxia-ischemia or a decrease in N-acetylaspartate concentration at any time are indicative of deleterious outcom

Trichloroethylene transformation by natural mineral pyrite: the deciding role of oxygen.

The transformation of trichloroethylene (TCE) in natural mineral iron disulfide (pyrite) aqueous suspension under different oxygen conditions was investigated in laboratory batch experiments. TCE transformation was pursued by monitoring its disappearance and products released with time. The effect of oxygen was studied by varying the initial dissolved oxygen concentration (DO(i)) inside each reactor. Transformation rates depended strongly on DO(i) in the system. In anaerobic pyrite suspension, TCE did not transform as it did under aerobic conditions. The transformation rate increased with an i

Absorption, elimination and metabolism of trichloroethylene: a quantitative comparison between rats and mice.

The absorption, elimination and metabolism of 14C-trichloroethylene (Tri) was studied in adult female Wistar rats and NMRI mice after administration of 200, 20 and 2 mg/kg Tri. Dose-dependent biotransformation of Tri to metabolites was observed in both species. Induction of hepatic mono-oxygenases by phenobarbital or polychlorinated biphenyls resulted in a higher rate of biotransformation after a single oral dose of 200 mg/kg 14C-Tri to rats. An increase in radioactivity covalently bound to liver and kidney macromolecules of induced rats as compared to control rats parallels the toxic effects

Formation and control of disinfection by-products during the trichloroisocyanuric acid disinfection in swimming pool water.

The increasing demand for trichloroisocyanuric acid (TCCA) in swimming pool disinfection highlights the need to evaluate its applicability in terms of disinfection by-product (DBP) formation. Nevertheless, there is limited understanding of DBP formation and control during TCCA disinfection, particularly concerning the effects of various management parameters. This study aimed to fill this knowledge gap by comprehensively investigating DBP formation during TCCA chlorination, with a particular focus on assessing the contribution and interaction of influencing factors using Box-Behnken Design and
