This study examined the presence of Treponema in lesions using conventional PCR detection methods and investigated the microbiome by performing high-throughput DNA sequencing. Twenty-nine bovine digital dermatitis (BDD) lesions were collected from 25 dairy farms in South Korea that were tested by PCR amplification using sets of one universal, one genus-specific, and three species specific Treponema PCR primers. Three BDD samples were randomly selected and normal tissue samples were submitted for 16S rRNA sequencing using the Illumina MiSeq platform. The dominant phylum present in all tested BD

In 2018, digital dermatitis (DD) associated with Treponema spp. was detected in a herd of European bison (EB, Bison bonasus) in Switzerland. This follow-up study was carried out in 2021-2022 to evaluate the DD prevalence in the Swiss EB population (n = 49) by taking photographs and biopsies of the interdigital cleft of 10 anesthetized and eight deceased animals from five out of the six Swiss EB herds. Further samples of captive animals were available from Germany (n = 4), France (n = 1), and Poland (n = 1), and of free-living individuals from Germany (n = 4) and Poland (n = 11). Except for the

This nonrandomized clinical intervention study was designed as a prospective, multicenter group comparison to evaluate the efficacy of a risk assessment and mitigation program to control bovine digital dermatitis (BDD). The program was implemented over a 1-year period on 9 intervention (INT) farms and 10 control (CTR) farms. Mainstays of the program derived from results of a previous BDD risk factor analysis. All farms were visited monthly to assess within-herd BDD prevalences to perform risk assessments and to treat BDD lesions with salicylic acid paste. Bulk milk samples were collected every

OBJECTIVE: The use of antibiotics is a proven approach for local and systemic treatment of digital dermatitis (DD). Even if they are administered only locally and at low dosage, their use should be restricted to an absolute minimum. The efficacy of a commercial polyurethane wound dressing in treating acute digital dermatitis was compared with topical treatment using a commercially available tetracycline spray and a salicylic acid paste.
MATERIALS AND METHODS: A total of 105 cows in a dairy farm (with about 160 dairy cows) were included in the study, with 101 cows needing topical treatment for

The increased frequency of diseases, especially those of the hoof, cause economic losses, such as premature culling of affected animals, decreased milk production, weight loss, reduced fertility, and the high costs of treatment. A great variety of hoof conditions may affect cattle, one of them is digital dermatitis. These conditions are probably due to multiple factorial diseases and present with similar clinical signs. Bovine lameness is typically treated by foot trimming and debridment of the lesions, coupled when necessary with systemic antibiotics and therapeutic footbaths, which results i

Three Treponema sp.-T. medium, T. pedis, and T. phagedenis-are associated with bovine digital dermatitis (BDD). This painful infectious disease has been reported worldwide, causing economic and animal welfare problems. Although detection by real-time PCR (rtPCR) is the method of choice, published assays differ in specificity and sensitivity. We multiplexed target regions of 2 published rtPCR assays and slightly optimized another one to compare their performance. We then used the 2 novel TaqMan rtPCR assays to determine the prevalence of the 3 Treponema sp. in a random sample of hind feet from

Digital dermatitis (DD) is a skin infection of cattle's feet with multiple bacteria suspected to be involved, yet its precise etiopathogenesis remains unclear. This longitudinal study explored the temporal changes of seven DD-associated bacteria in feet developing lesions or remaining healthy, while simultaneously investigating their persistence in potential reservoirs as sources of infection. Weekly swabs were collected from feet skin and saliva of 53 Holstein cows without DD lesions sequentially enrolled at calving in a commercial dairy herd. At the end of the study, samples from all cases a

BACKGROUND: Bovine digital dermatitis (BDD) is an infectious foot disease found commonly in dairy herds. Foot-trimming is an important husbandry procedure for reducing the ensuing lameness; however, epidemiological, and microbiological studies have identified this as a risk activity for transmitting BDD. Three disinfectants have previously been identified in laboratory work as effective for removing viable BDD-associated Treponema spp., from hoof knife blades. The present study enrolled 133 dairy cattle with BDD lesions, and swabbed hoof knife blades before and after foot-trimming, and after k

Digital dermatitis is a bacterial infection that causes lesions above the heel bulbs on cattle hooves, and several bacterial species from the genus Treponema are suspected to be causative agents of this polymicrobial condition. Transmission of the bacteria to healthy cows is understudied, particularly with regard to potential insect vectors. Therefore, the objective of this research was to determine if flies captured from a dairy farm known to have digital dermatitis are contaminated with Treponema bacteria. The DNA-based assays did not detect any Treponema phagedenis from stable flies and hou

The aim of this study was to determine the prevalence of the three Treponema species as well as D. nodosus in Digital dermatitis (DD) and slurry of Swiss cattle using PCR. A total of 86 specimens from 24 farms were enrolled in the study. Slurry samples from 21 DD-affected and one unaffected farm were collected to assess the potential of environmental transmission. Nested and real-time PCR were performed from the specimens to detect Treponema species and D. nodosus, respectively. The DD-stages were positive for at least one or more of the DD-associated Treponema species in 50 of 61 cases (82.0%

Digital dermatitis (DD) is the leading cause of lameness in cattle, posing major animal welfare and economic concerns. Effective prevention strategies are increasingly important given emerging antimicrobial resistance associated with common DD treatments. Supplementation with Saccharomyces cerevisiae fermentation postbiotics (SCFP) has been shown to enhance innate immunity and reduce DD lesion development. This study evaluated the effect of a commercial SCFP supplement on the microbial composition of DD lesions using shotgun metagenomic sequencing to characterize microbial communities and asso

The source of infection of digital dermatitis (DD), an infectious lameness condition, is still uncertain. In this cross-sectional study, we aimed to identify potential reservoirs of DD bacteria in dairy cattle body sites with different stages of the disease and farm environments. We collected skin swabs from 85 dairy cows from 5 herds, 3 with and 2 without DD, from foot, hock, and udder cleft skin (with lesions or not), saliva, urine, and feces. We also obtained environmental samples. Real-time quantitative PCR targeted Treponema phagedenis, Treponema medium, Treponema pedis, Porphyromonas lev

CASE HISTORY: Thirty adult Marlborough green geckos (Naultinus manukanus) were collected from Stephens Island and held over winter, prior to their translocation. Five adult geckos developed skin lesions after husbandry changes affected the humidity of their enclosures. Two geckos underwent ecdysis and recovered. One animal died and two others progressively worsened and were presented for treatment.
CLINICAL AND PATHOLOGICAL FINDINGS: The geckos were in poor body condition and had multiple black powdery lesions and solitary raised white nodules on their skin. Both geckos died despite topical a

Identification of reservoirs and transmission routes of digital dermatitis (DD)-associated Treponema spp. is considered an effective means for controlling DD infection in dairy cows. The objective of this study is to identify and characterize the potential reservoir niches for DD-associated Treponema spp. from healthy udder cleft skin and foremilk in lactating dairy cows. A large dairy farm was visited weekly from March to July 2015. Clinical investigation revealed that a total of 25 lame cows had DD lesions located at the plantar aspect of the interdigital cleft. A total of 75 samples, three

Digital dermatitis (DD) is one of the main causes of lameness in dairy cattle worldwide, and it is frequently reported in high-yielding, free stall dairy herds from regions with a temperate climate. However, DD is also observed with high prevalence in grazing cattle with a low milk yield in tropical regions. To clarify whether these differences have an impact on the etiology of the disease, we studied DD lesions from all year round grazing cattle of mixed breed in Brazil using high-throughput 16S rRNA gene sequencing and fluorescent in situ hybridization. The study included samples from 66 ski

This study aimed to determine whether there was a difference in skin permeability to methylene blue dye or skin morphology between dairy cows that differed in their susceptibility to digital dermatitis (DD) and to assess the effect of contact with slurry on skin permeability. Twenty nine dairy cows were monitored for DD during the winter housing period and classed as DD+ (previous DD infection, n = 17), or DD- (no recorded infection, n = 12). The animals were culled and a skin sample was taken from above the heel of each hind foot and frozen. Samples were later defrosted and one sample from ea

BACKGROUND: Atopic dermatitis (AD) is one of the most common chronic inflammatory skin diseases, associated with itching, sleep disturbance, and impaired quality of life. Structured patient education can improve disease outcomes, but in-person programs are often unavailable in rural or underserved areas. Digital interventions may help overcome these barriers; however, their efficacy compared with routine care has not been evaluated.
OBJECTIVE: This study aims to assess whether a digitally delivered, interdisciplinary care concept (ADCompanion) is noninferior to routine, real-world care, as av

Bovine digital dermatitis (BDD) is an infectious hoof disease associated with multiple Treponema species, causing significant economic and welfare concerns in the cattle industry. Studies regarding T-cell mediated immune responses are limited in BDD. Using a mouse model, this study analyzed different T-cell subsets involved in immune responses against BDD-associated treponemes. Additionally, this study measured Treponema-specific IgG antibody titers to elucidate humoral immune responses. Mice were experimentally infected with 2 × 109 cells/mL of live T. phagedenis HNW1, T. pedis GNW45, T. pedi

Bovine ischaemic teat necrosis (ITN) is a disease affecting the skin of the teats of dairy cows with an unknown aetiopathogenesis. Digital dermatitis (DD)-associated treponemes have previously been suggested as a potential aetiological agent in ITN, although the sample size was small. The current study, using established PCR techniques, aimed to examine the association with the presence of DD-associated treponemes in a large number of ITN samples from a wider geographical area, and surveyed the potential of milk as an infection reservoir. From 95 ITN lesions, 35.8% (n = 34) were positive for a
