Growth-promoting activity of Sanyak (Dioscoreae rhizoma) extract on injured sciatic nerve in rats.

The present study evaluates the potential effects of Sanyak (Dioscoreae rhizome) on the regenerative capacity of the peripheral sciatic nerve after crush injury in rats. Focal application of Sanyak extract at the injury site increased GAP-43 and Cdc2 protein levels in the distal portion of the injured nerve. Immunohistochemical analysis showed that the signals of phospho-vimentin as Cdc2 substrate protein were almost colocalized with Cdc2. Retrograde DiI-tracing revealed enhancement in distal elongation of regenerating axons. Furthermore, the number of non-neuronal cells was higher in Sanyak-t

Sustainable pH sensing: repurposing the anthocyanin-rich extract from Dioscorea alata for green analytical chemistry.

Over the years, synthetic indicators have been the choice for acid-base titrations. However, due to environmental and safety concerns, the search for natural compounds that can act as acid-base indicators is important. This study aims to identify an eco-friendly natural indicator from the Dioscorea alata L. yam species. Dried D. alata yam was ground into a coarse powder and extracted with an acidified methanol-water mixture for 15 minutes. The resulting extract was tested for colour changes in the pH range from 1 to 12. An acid-base titration was carried out using the D. alata extract as the i

Haplotype-resolved autotetraploid genome of Dioscorea nipponica provides insights into recent polyploidization, metabolic rewiring, and environmental adaptation.

Whole-genome duplication (WGD, or polyploidization) is a major evolutionary force in plants, yet the genomic, transcriptional, and metabolic consequences of autopolyploidy remain less understood than those of allopolyploidy. Dioscorea nipponica, a medicinal species producing diosgenin-type steroidal saponins, harbors both diploid and autopolyploid populations, providing a unique system to investigate these processes. Here, we present a haplotype-resolved, chromosome-scale genome assembly of autotetraploid D. nipponica (2n = 4x = 40). Comparative and population genomic analyses indicate a singl

CYP2C19-mediated metabolic activation and OAT3/P-gp-associated transport contribute to Dioscorea bulbifera L.-induced hepatotoxicity.

ETHNOPHARMACOLOGICAL RELEVANCE: Dioscorea bulbifera L. (DBL) is a traditional medicinal herb, but its clinical use is often limited by its potential to cause herb-induced liver injury. 8-Epidiosbulbin E acetate (EEA) is recognized as the major hepatotoxic constituent in DBL; however, the mechanisms underlying the drug transport and metabolic activation of EEA leading to hepatotoxicity are unclear, which severely impedes the understanding of its toxicity risk and safety control measures. AIM OF THE STUDY: This study aimed to identify the key EEA transporters and P450 enzymes mediating EEA meta

Chinese yam (Dioscorea) polysaccharide ameliorates ulcerative colitis in mice via modulating disorders of intestinal microecology and metabolism.

Clinical research has demonstrated that non-starch polysaccharides from natural sources exhibit protective and therapeutic effects on ulcerative colitis (UC). In this study, a non-starch polysaccharide (CYP-A) with a molecular weight of 1.54 × 103 kDa was isolated from a speciality Chinese yam (Dioscorea) variety, the Ma yam, consisting mainly of mannose and glucose. The results indicated that CYP-A alleviated colitis symptoms induced by dextran sulfate sodium (DSS), repaired the mucus barrier, and protected the integrity of the intestinal mechanical barrier. Furthermore, CYP-A suppressed pro-

Therapeutic impact of polysaccharide from Chinese yam (Dioscorea opposita Thunb) on DSS-induced ulcerative colitis in mice.

While yam polysaccharide (YP) demonstrates diverse biological activities, its anti-inflammatory mechanisms remain insufficiently characterized. This study systematically investigates the therapeutic efficacy of YP in ulcerative colitis (UC) through integrated in vitro and in vivo approaches. YP was purified from Dioscorea opposita Thunb. via ethanol precipitation, followed by cytotoxicity profiling. In Lipopolysaccharide (LPS)-stimulated intestinal epithelial cells (IEC-6), YP significantly attenuated proinflammatory responses. For in vivo validation, a murine colitis model was established thr

Integrated Metabolomics and Proteomics Approach To Identify Metabolic Abnormalities in Rats with Dioscorea bulbifera Rhizome-Induced Hepatotoxicity.

Previous studies have shown that Dioscorea bulbifera rhizome (DBR) can induce hepatotoxicity in clinical practice. However, its underlying mechanisms remain largely unexplored. In the present study, we investigated the global effect of DBR exposure on the proteomic and metabolomic profiles in rats over a 12-week administration using an integrated proteomics and metabolomics approach. The abundance of 1366 proteins and 58 metabolites in the liver of rats after subchronic exposure to DBR was dose-dependently altered. The results indicated that DBR mainly damaged hepatic cells through the aberran

A potential plasmid-curing agent, 8-epidiosbulbin E acetate, from Dioscorea bulbifera L. against multidrug-resistant bacteria.

Bioassay-guided fractionation of an aqueous methanolic extract of Dioscorea bulbifera L. bulbs was performed using organic solvents. A novel plasmid-curing compound was identified as 8-epidiosbulbin E acetate (EEA) (norditerpene) on the basis of modern spectroscopic analysis and X-ray crystallography. EEA exhibited broad-spectrum plasmid-curing activity against multidrug-resistant (MDR) bacteria, including vancomycin-resistant enterococci. EEA cured antibiotic resistance plasmids (R-plasmids) from clinical isolates of Enterococcus faecalis, Escherichia coli, Shigella sonnei and Pseudomonas aer

Dioscorea tokoro Makino extract suppresses the growth of Saccharomyces cerevisiae INVSc1 by disrupting the endolysosomal pathway.

Exploring novel antifungal mechanisms is needed because of the side effects of existing treatments and the increasing prevalence of drug resistance. We recently reported that inhibition of the endocytic pathway via a lysosome inhibitor or Fab1/Vac14 suppression enhances the localization of emerald green fluorescent protein-bound β-1,3-glucanase (BGL2-EmGFP) at the tip in Saccharomyces cerevisiae INVSc1, used as a non-pathogenic model. Overexpression of wild-type BGL2-EmGFP, but not an inactive mutant, completely abolished cell proliferation, suggesting that impairing tip growth through dysregu

Response of the Symbiotic Microbial Community of Dioscorea opposita Cultivar Tiegun to Root-Knot Nematode Infection.

Yam is an important medicinal and edible dual-purpose plant with high economic value. However, nematode damage severely affects its yield and quality. One of the major effects of nematode infestations is the secondary infection of pathogenic bacteria or fungi through entry wounds made by the nematodes. Understanding the response of the symbiotic microbial community of yam plants to nematodes is crucial for controlling such a disease. In this study, we investigated the rhizosphere and how endophytic microbiomes shift after nematode infection during the tuber expansion stage in the Dioscorea opp
