Indoleamine 2,3-dioxygenase (IDO1) is a heme enzyme that catalyzes the oxygenation of the indole ring of tryptophan to afford N-formylkynurenine. This activity significantly suppresses the immune response, mediating inflammation and autoimmune reactions. These consequential effects are regulated through redox changes in the heme cofactor of IDO1, which autoxidizes to the inactive ferric state during turnover. This change in redox status increases the lability of the heme cofactor leading to further suppression of activity. The cell can thus regulate IDO1 activity through the supply of heme and

The mechanism of maternal immunotolerance of the semiallogeneic fetus has been a matter of intense investigation. The tryptophan-degrading enzyme indoleamine 2,3-dioxygenase (IDO) is reported to be critically implicated. This article discusses findings pertaining to the role of IDO in pregnancy, its location at the feto-maternal interface, systemic induction of IDO in pregnancy and the association of IDO to spontaneous abortion and preeclampsia. Whereas there is a large body of evidence supporting the relevance of IDO as a key immunoregulatory factor in feto-maternal tolerance, open questions

Guanidine-responsive riboswitches control genes that enable either detoxification or assimilation of guanidino compounds. In Vreelandella boliviensis and other halophilic bacteria, genes encoding the guanidine carboxylase pathway are found in a guanidine riboswitch-regulated operon, along with two uncharacterized genes annotated as 2-oxoglutarate (2-OG/Fe(II))-dependent dioxygenase family protein and hypothetical protein, respectively. Here we show that the 2-OG/Fe(II)-dependent dioxygenase efficiently hydroxylates methylguanidine. The resulting N-(hydroxymethyl)guanidine constitutes an unexpe

AIMS: Heart failure (HF) is one of the most devastating consequences of cardiovascular diseases. Regardless of etiology, heart failure is often preceded by cardiac hypertrophy. In the present study we investigated the effect of cardiomyocyte-specific deletion of ten-and-eleven translocation 2 (TET2) on cardiac hypertrophy.
METHODS AND MATERIALS: Cardiac hypertrophy was induced in vitro by exposing cardiomyocytes to angiotensin II (Ang II), isoproterenol (ISO), or phenyleprine (PE) and in vivo by subjecting the mice to transverse aortic constriction (TAC).
KEY FINDINGS: TET2 expression was el

IDO1 (indoleamine 2,3-dioxygenase 1) is a member of a unique class of mammalian haem dioxygenases that catalyse the oxidative catabolism of the least-abundant essential amino acid, L-Trp (L-tryptophan), along the kynurenine pathway. Significant increases in knowledge have been recently gained with respect to understanding the fundamental biochemistry of IDO1 including its catalytic reaction mechanism, the scope of enzyme reactions it catalyses, the biochemical mechanisms controlling IDO1 expression and enzyme activity, and the discovery of enzyme inhibitors. Major advances in understanding the

BACKGROUND: Pleural fluid is a frequent manifestation in pulmonary diseases, such as lung cancer and infectious diseases, including pulmonary tuberculosis (TB). The enzyme indoleamine 2,3-dioxygenase (IDO) catalyses tryptophan through the kynurenine pathway, and is considered a crucial immunoregulatory molecule mediating immune tolerance. Recent studies have shown IDO activity to be a novel prognostic factor not only in cancer patients but also in those with infectious diseases, including pneumonia and pulmonary TB. However, no studies have measured and determined the clinical significance of

Persulfides (RSSH) have been proposed as key players in biochemical transformations that often involve iron, including iron-sulfur cluster assembly, H2S regulation, post translational modifications, and mitochondrial sulfur oxidation. An example of the latter is found in the O2-mediated oxidation of glutathione persulfide to sulfite dianion (SO32-) catalyzed by ETHE1, a nonheme iron persulfide dioxygenase (PDO). The iron-mediated mechanism of persulfide oxidation by PDOs remains poorly understood, and there are no synthetic analogues to date. Herein, we report the synthesis, characterization,

Owing to their diverse coordination patterns and catalytic mechanisms, non-heme iron-dependent dioxygenases catalyze a variety of biochemical reactions involved in the synthesis of numerous natural products and valuable compounds. Recently, we discovered a novel and atypical non-heme iron-dependent dioxygenase, BTG13, that features a unique coordination center consisting of four histidines and a carboxylated lysine (Kcx). This enzyme catalyzes the C-C bond cleavage of anthraquinone through two unconventional steps, with modified Kcx playing a key role in facilitating these processes, as reveal

Non-heme iron oxygenases constitute a versatile enzyme family that is crucial for incorporating molecular oxygen into diverse biomolecules. Despite their importance, only a limited number of these enzymes have been structurally and functionally characterized. Surprisingly, there remains a significant gap in understanding how these enzymes utilize a typical architecture and reaction mechanism to catalyze a wide range of reactions. Improving our understanding of these catalysts holds promise for advancing both fundamental enzymology and practical applications. This chapter aims to outline method

Cancers express tryptophan catabolising enzymes indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan 2,3-dioxygenase (TDO2) to produce immunosuppressive tryptophan metabolites that undermine patients' immune systems, leading to poor disease outcomes. Both enzymes are validated targets for cancer immunotherapy but there is a paucity of potent TDO2 and dual IDO1/TDO2 inhibitors. To identify novel dual IDO1/TDO2 scaffolds, 3D shape similarity and pharmacophore in silico screening was conducted using TDO2 as a model for both systems. The obtained hits were tested in cancer cell lines expressing mai

UNLABELLED: The salicylate 1,2-dioxygenase (SDO) from the bacterium Pseudaminobacter salicylatoxidans BN12 is a versatile gentisate 1,2-dioxygenase (GDO) that converts both gentisate (2,5-dihydroxybenzoate) and various monohydroxylated substrates. Several variants of this enzyme were rationally designed based on the previously determined enzyme structure and sequence differences between the SDO and the 'conventional' GDO from Corynebacterium glutamicum. This was undertaken in order to define the structural elements that give the SDO its unique ability to dioxygenolytically cleave (substituted)

Evidence for an iron(III)-indolenylperoxo (OOIn-) complex, [FeIII(OOIn)(TPP)(OCH3)]- (TPP = 5,10,15,20-tetraphenylporphyrinato2-) trapped at -78 °C, was obtained. The formation of this species was initiated by reaction of either FeIII(TPP)(Cl) in the presence of excess O2, OCH3- as axial ligand and base, and 3-methylindole, or from reaction of preformed [FeIII(O2•-)(TPP)(OCH3)]- with 3-methylindole under the same basic conditions. The new species was characterized by UV-vis, X-band EPR, and Mössbauer spectroscopies. Interestingly, the EPR spectra of [FeIII(OOIn)(TPP)(OCH3)]- exhibited two over

Rieske dioxygenases catalyze the initial steps in the hydroxylation of aromatic compounds and are critical for the metabolism of xenobiotic substances. Because substrates do not bind to the mononuclear non-heme FeII center, elementary steps leading to O2 activation and substrate hydroxylation are difficult to delineate, thus making it challenging to rationalize divergent observations on enzyme mechanisms, reactivity, and substrate specificity. Here, we show for nitrobenzene dioxygenase, a Rieske dioxygenase capable of transforming nitroarenes to nitrite and substituted catechols, that unproduc

Rieske nonheme iron oxygenases (ROs) catalyze the oxidation of a wide variety of substrates and play important roles in aromatic compound degradation and polycyclic aromatic hydrocarbon degradation. Those Rieske dioxygenases that usually act on hydrophobic substrates have been extensively studied and structurally characterized. Here, we report the crystal structure of a novel Rieske monooxygenase, NagGH, the oxygenase component of a salicylate 5-monooxygenase from Ralstonia sp. strain U2 that catalyzes the hydroxylation of a hydrophilic substrate salicylate (2-hydroxybenzoate), forming gentisa

Tryptophan metabolism via the kynurenine pathway plays a critical role in immune regulation and neurobiology. Dysregulation of this pathway, particularly through overactivation of indoleamine 2,3-dioxygenases (IDOs) and tryptophan 2,3-dioxygenase (TDO), has been implicated in various pathological conditions, including cancer, depression, chronic pain, and neurodegenerative diseases. Here, we report the design and synthesis of a novel class of dual IDO1/TDO inhibitors featuring a 5,6-dihydroimidazothiazole core linked to a urea motif via an alkyl spacer. These compounds were evaluated for their

EPR-silent, chemically reduced protocatechuate 3,4-dioxygenase (Er) binds NO at the active site Fe2+ to yield an EPR-active, S = 3/2 species that blocks subsequent binding of all other exogenous ligands. In contrast, addition of NO to a preformed Er.CN- complex yields an EPR-active, S = 1/2 species [Er.(CN)x.NO] that exhibits resolved superhyperfine splitting from 13CN-, 15/14NO, and a protein-derived 14N. Simulations of the EPR spectra observed for the Er.(CN)x.NO complex formed with 12CN- and 13CN- (1:1) show that CN- binds in two iron ligand sites (x >/= 2). The two cyanides exhibit similar

Oxygen-containing heterocycles (O-heterocycles), including tetrahydrofuran (THF) and 1,4-dioxane, are emerging water contaminants of growing concern because of their persistence and recalcitrance to conventional treatment processes. Biodegradation of these compounds requires ring cleavage, yet the enzymes responsible for this critical step have remained unidentified. By integrating affinity predictions with in vivo validation, we demonstrated that classical aromatic ring-cleaving dioxygenases, including the intradiol dioxygenases CatA1/CatA2 from Cupriavidus metallidurans ZM02 and the extradio
