The selective dopamine D2 receptor antagonist raclopride discriminates between dopamine-mediated motor functions.

The actions on central dopamine (DA) mechanisms of raclopride, a new substituted benzamide, were studied by means of behavioural and biochemical methods in the rat. Raclopride blocked the in vitro binding of the dopamine D2 antagonist 3H-spiperone (IC50 = 32 nM), but not of the unselective D1 antagonist 3H-flupenthixol (IC50 greater than 100,000 nM) in rat striatum, and failed to inhibit striatal DA-sensitive adenylate cyclase in vitro (IC50 greater than 100,000 nM). Raclopride caused a dose-dependent increase in the DA metabolites HVA and DOPAC in the striatum and olfactory tubercle. Behaviou

Mesolimbic dopamine neurotransmission is increased by administration of mu-opioid receptor antagonists.

Microdialysis and high pressure liquid chromatography were used to assess the effects of ventral tegmental area microinjections of the mu-opioid receptor antagonists D-Pen-Cys-Tyr-D-Trp-Orn-Thr-Pen-Thr-NH2 (CTOP) and beta-funaltrexamine (beta-FNA) on extracellular ventral striatal dopamine and metabolite concentrations. While CTOP is known to antagonize the increases in extracellular ventral striatal dopamine and dopamine metabolite concentrations induced by ventral tegmental area microinjections of a mu-opioid receptor agonist, it produced dose-orderly increases in ventral striatal dopamine a

Effects of D1 and D2 dopamine antagonists on behavior of polydipsic rats.

The behavioral and neurochemical effects of SCH3390 (SCH), a dopamine (DA) D1 antagonist, and haloperidol (HAL), a DA D2 receptor antagonist, on schedule-induced polydipsia (SIP) were examined. Once animals were made polydipsic, a vehicle or one of three doses of SCH or HAL were administered to seven groups of rats in a series of three five-session blocks in a drug condition, no-drug condition, drug condition design. Detailed behavioral measures and brain regional levels of monoamine neurotransmitters and their major acidic metabolites were analyzed. The volume of water consumed and the percen

Effects of muscarinic receptor agonists and antagonists on dopamine-mediated behavioural paradigms.

The effects of some muscarinic M1 and M2 receptor agonists and antagonists on two dopamine-mediated behavioural paradigms, namely post-swim grooming response and apormorphine-induced climbing behaviour, was investigated in mice. The M1 agonists and M2 receptor antagonists attenuated both paradigms, whereas the M1 receptor antagonists and M2 receptor agonists facilitated both behaviours. However, carbachol and physostigmine, exhibited a similar dose-dependent dual effect with the lower doses augmenting and the higher doses inhibiting both the behaviours, suggesting that the M2 agonist receptor

Effects of different dopamine agonists and antagonists on post-menopausal hot flushes.

The dopaminergic system seems to be involved in both pulsatile luteinizing hormone (LH) secretion and hot flushes in post-menopausal women. With the aim of further clarifying its role, the effectiveness of dopaminergic and antidopaminergic drugs in the treatment of hot flushes was studied. Self-assessed scores for vasomotor symptoms were evaluated in 5 groups of 15 patients treated for 20 days with one of the following agents: placebo; the dopamine receptor agonist, bromocriptine; the indirect dopaminergic agent, Liposom; the antidopaminergic drug, veralipride or the peripheral antidopaminergi

Effects of dopamine analogues and antagonists on carotid body chemosensors in situ.

1. The effects of dopamine, its analogues and antagonists on the chemosensory discharges originating from carotid bodies in situ were studied in anaesthetized cats. 2. Intracarotid (I.C.) injections of 100 ng or more of dopamine produced transient depression of the frequency of carotid nerve chemosensory discharges. Short term (1-5 sec) complete inhibition was usually elicited by 2 microgram dopamine. 3. I.V. injections of dopamine also produced inhibition of chemosensory discharges, an effect observed with doses which were still too low to produce changes in systemic arterial pressure. Half-m

Review of Dopamine Antagonists for Nausea and Vomiting in Palliative Care Patients.

Symptoms of nausea and vomiting are common in palliative care and hospice patients. One of the many classes of medications used for the treatment of nausea and vomiting is dopamine receptor antagonists which are particularly helpful for treating nausea mediated by the chemoreceptor trigger zone (CTZ) and impaired gastrointestinal function. While dopamine antagonists can be very effective treatments for nausea they should be used with caution as they carry the risk of QTc prolongation, have a FDA black box warning for tardive dyskinesia (TD), and increased risk of precipitating psychosis and de

Preferential adsorption of dopamine antagonist binding sites by fluphenazine-agarose.

Separation of dopamine (DA) agonist and antagonist receptors was attempted by means of a covalently-bound fluphenazine-agarose (Flu-agarose). Incubation of striatal membranes with Flu-agarose resulted in loss of 3H-spiroperidol (3H-SPI) binding sites, while incubation with non-coupled agarose did not cause any change. The loss of 3H-SPI binding to the Flu-agarose treated membranes was not attributed to the release of fluphenazine from Flu-agarose as justified by several criteria. Flu-agarose adsorbed more effectively striatal membranes with 3H-SPI binding sites than those with 3H-DA binding si

Ventilatory effects of domperidone, a new dopamine antagonist, in anaesthetized rabbits.

The ventilatory effects of domperidone (DP), a dopamine antagonist that does not cross the blood-brain barrier, were studied on rabbits anaesthetized with pentobarbitone. DP induced hyperventilation (+20%), which was not dose-dependent, without any marked change in arterial blood gases. This effect was abolished by previous transection of both carotid sinus nerves. On an average, steady state hypoxia (PaO2 approximately 45 mmHg for 5 min) doubled ventilation before, and increased itself threefold after DP (0.10 mg kg-1 i.v.). Prazosin (PZ, 0.1 mg kg-1 i.v.), an alpha 1-adrenoceptor antagonist,

Dopamine D1 receptor antagonist reduces stimulant-induced conditioned place preferences and dopamine receptor supersensitivity.

Repeated administration of stimulants induces conditioned place preference (CPP). Dopamine receptor supersensitivity is developed in stimulant-induced CPP animals; however, dopamine receptor subtypes associated with the development of supersensitivity in CPP animals are largely unknown. The present preclinical study aimed to examine whether dopamine D1 or D2 receptor antagonists exert inhibitory effects on stimulant-induced psychological behaviors. Additionally, the authors aimed to elucidate the role of dopamine receptor supersensitivity on the development of reward-related behavior. Sprague

Clozapine acts as an antagonist of human dopamine D1 receptors in the atrium.

Clozapine is one of the oldest antipsychotic drugs, the first atypical neuroleptic drug, and can be found in the World Health Organization's list of essential drugs. The precise mechanism of action of clozapine as an antipsychotic is not known. However, clozapine binds to many proteins in vitro. The objective of this study was to investigate the role of the dopamine D1 receptor as a possible explanation for the cardiac side effects of clozapine. To this end, the hypothesis was tested that clozapine reduces the positive inotropic or positive chronotropic effects mediated by human cardiac dopami

Effects of dopamine receptor antagonist antipsychotic therapy on blood pressure.

WHAT IS KNOWN AND OBJECTIVE: Hypertension, a major risk factor for adverse cardiovascular events, such as stroke and myocardial infarction, affects 80 million American adults. The aetiology of hypertension is multifaceted and difficult to identify. Dopamine receptors, especially those in the kidneys, play a role in blood pressure regulation, and alterations in their function can cause hypertension. The objective of this review was to investigate the association between the use of dopamine antagonists with hypertension focusing especially on second-generation antipsychotics, like clozapine that

Pharmacological characterization of a dopamine transporter ligand that functions as a cocaine antagonist.

An N-butyl analog of benztropine, JHW007 [N-(n-butyl)-3α-[bis(4'-fluorophenyl)methoxy]-tropane], binds to dopamine transporters (DAT) but has reduced cocaine-like behavioral effects and antagonizes various effects of cocaine. The present study further examined mechanisms underlying these effects. Cocaine dose-dependently increased locomotion, whereas JHW007 was minimally effective but increased activity 24 hours after injection. JHW007 (3-10 mg/kg) dose-dependently and fully antagonized the locomotor-stimulant effects of cocaine (5-60 mg/kg), whereas N-methyl and N-allyl analogs and the dopami

Effects of the D3 and autoreceptor-preferring dopamine antagonist (+)-UH232 in schizophrenia.

The aminotetralin derivative (+)-UH232 is a dopamine-receptor antagonist with complex pharmacological properties, including a 4:1 selectivity for the D3 vs. D2 receptor and a preference for the autoreceptors. Its behavioral profile differs markedly from that of other dopamine antagonists in exhibiting both stimulant and inhibitory features. In an effort to elucidate the role of different dopamine receptor subtypes in psychosis, we administered (+)-UH232 to drug-free schizophrenic patients. Six patients received single doses of (+)-UH232 over a dose range of 80 to 180 mg in a rising-dose, doubl
