Neurogenetics of courtship and mating in Drosophila.

The reproductive biology of Drosophila melanogaster is described and critically discussed, primarily with regard to genetic studies of sex-specific behavior and its neural underpinnings. The investigatory history of this system includes, in addition to a host of recent neurobiological analyses of reproductive phenotypes, studies of mating as well as the behaviors leading up to that event. Courtship and mating have been delved into mostly with regard to male-specific behavior and biology, although a small number of studies has also pointed to the neural substrates of female reproduction. A Drosophila model of Barth syndrome.

Barth syndrome is an X-linked disease presenting with cardiomyopathy and skeletal muscle weakness. It is caused by mutations in tafazzin, a putative acyl transferase that has been associated with altered metabolism of the mitochondrial phospholipid cardiolipin. To investigate the molecular basis of Barth syndrome, we created Drosophila melanogaster mutants, resulting from imprecise excision of a P element inserted upstream of the coding region of the tafazzin gene. Homozygous flies for that mutation were unable to express the full-length isoform of tafazzin, as documented by RNA and Western bl

Genetics of sexual behaviour in Drosophila.

The analysis of genetics of behaviour within and between species provides important clues about the forces shaping the evolution of behavioural genes. In Drosophila, a number of key processes such as emergence from the pupal case, locomotor activity, feeding, olfaction and aspects of mating behaviour are under circadian regulation. Genes controlling sexual behaviour are likely to control species specific differences in courtship that are involved in reproductive isolation of closely related species. Courtship in Drosophila is characterized by a series of stereotyped behaviours that lead to cop

Taste representations in the Drosophila brain.

Drosophila taste compounds with gustatory neurons on many parts of the body, suggesting that a fly detects both the location and quality of a food source. For example, activation of taste neurons on the legs causes proboscis extension or retraction, whereas activation of proboscis taste neurons causes food ingestion or rejection. We examined whether the features of taste location and taste quality are mapped in the fly brain using molecular, genetic, and behavioral approaches. We find that projections are segregated by the category of tastes that they recognize: neurons that recognize sugars p

Development of the genitalia in Drosophila melanogaster.

The imaginal discs of Drosophila melanogaster are an excellent material with which to analyze how signaling pathways and Hox genes control growth and pattern formation. The study of one of these discs, the genital disc, offers, in addition, the possibility of integrating the sex determination pathway into this analysis. This disc, whose growth and shape are sexually dimorphic, gives rise to the genitalia and analia, the more posterior structures of the fruit fly. Male genitalia, which develop from the ninth abdominal segment, and female genitalia, which develop mostly from the eighth one, disp

Chemical induction of nondisjunction in drosophila.

Tests for chemically induced nondisjunction and loss of the sex chromosomes in Drosophila were performed. Of 31 compounds tested four gave rise only to an increase of XO exceptions, indicating the induction of chromosome loss. Six compounds, all known spindle inhibitors (colchicine, organic mercury, lead, and tin compounds) gave rise to an increase both of XXY and XO or of only XXY. The effect by metalloorganic compounds of which methylmercury was studied particularly closely, follows a peculiar pattern. In females with structurally normal X chromosomes only an increase of XX gametes is obtain

Elements of olfactory intelligence in Drosophila.

The ability to make the world of odorants intelligible is a key capability of the Drosophila olfactory system that we shall call olfactory intelligence. Characterizing the functional logic of the Drosophila early olfactory system that makes the natural world of odorants intelligible is a major challenge in neuroscience. Starting by modeling the space of odorants using constructs of both semantic and syntactic information, we establish that the Antenna Lobe and Mushroom Body Calyx first decompose the confounding representation of the Antenna into a concentration independent odorant semantics in

Effects of lithium on aggression in Drosophila.

Lithium is a common medication used to treat mania and bipolar disorder, but the mechanisms by which lithium stabilizes mood and modifies aggression are still not fully understood. Here we found that acute but not chronic lithium significantly suppresses aggression without affecting locomotion in Drosophila melanogaster. Male flies treated with acute lithium are also less competitive than control males in establishing dominance. We also provided evidence that glycogen synthase kinase-3 (GSK-3), a well-known target of lithium, plays an important role in the anti-aggressive effect of lithium in

Modulation of Huntington's Disease in Drosophila.

Huntington's disease (HD) is a progressive neurodegenerative disorder which deteriorates the physical and mental abilities of the patients. It is an autosomal dominant disorder and is mainly caused by the expansion of a repeating CAG triplet. A number of animal models ranging from worms, fruit flies, mice and rat, pig, sheep and monkeys are available, which have been helpful in understanding various pathways involved during the progression of the disease. Drosophila is one of the most commonly used model organisms for biomedical science, due to low cost maintenance, short life span and easy im

Drosophila Model in Cancer: An Introduction.

Cancer is a cumulative manifestation of several complicated disease states that affect multiple organs. Over the last few decades, the fruit fly Drosophila melanogaster, has become a successful model for studying human cancers. The genetic simplicity and vast arsenal of genetic tools available in Drosophila provides a unique opportunity to address questions regarding cancer initiation and progression that would be extremely challenging in other model systems. In this chapter we provide a historical overview of Drosophila as a model organism for cancer research, summarize the multitude of genet

A Drosophila Model of Essential Tremor.

Essential Tremor (ET) is one of the most common neurological diseases, with an estimated 7 million affected individuals in the US; the pathophysiology of the disorder is poorly understood. Recently, we identified a mutation (KCNS2 (Kv9.2), c.1137 T > A, p.(D379E) in an electrically silent voltage-gated K+ channel α-subunit, Kv9.2, in a family with ET, that modulates the activity of Kv2 channels. We have produced transgenic Drosophila lines that express either the human wild type Kv9.2 (hKv9.2) or the ET causing mutant Kv9.2 (hKv9.2-D379E) subunit in all neurons. We show that the hKv9.2 subunit

A Drosophila model for Angelman syndrome.

Angelman syndrome is a neurological disorder whose symptoms include severe mental retardation, loss of motor coordination, and sleep disturbances. The disease is caused by a loss of function of UBE3A, which encodes a HECT-domain ubiquitin ligase. Here, we generate a Drosophila model for the disease. The results of several experiments show that the functions of human UBE3A and its fly counterpart, dube3a, are similar. First, expression of Dube3a is enriched in the Drosophila nervous system, including mushroom bodies, the seat of learning and memory. Second, we have generated dube3a null mutants

Expression of antimicrobial peptides associated with different susceptibilities to environmental chemicals in Drosophila suzukii and Drosophila melanogaster.

Drosophila suzukii is a serious agricultural pest. The evolved morphology of the female D. suzukii assists in penetrating the surface of fresh fruit and spawns eggs with its unique ovipositor. Conversely, Drosophila melanogaster, a taxonomically close species with D. suzukii, largely inhabits decaying and fermenting fruits and is consistently exposed to extensive environmental chemicals, such as 2-phenylethanol, ethanol, and acetic acid, produced by microorganisms. Considering the distinct habitats of the two flies, D. suzukii is thought to be more susceptible to environmental chemicals than D

Adaptations of Drosophila and yeasts: their interactions with the volatile 2-propanol in the cactus-microorganism-Drosophila model system.

The interactions of yeasts growing in decaying cactus tissue with and without 2-propanol were studied with respect to the costs and benefits provided to three cactophilic Drosophila species (D. mojavensis, D. arizonensis and D. buzzatii). Two common cactus yeasts, Candida sonorensis and Cryptococcus cereanus, which can tolerate and metabolize 2-propanol, provide benefits to the three Drosophila species in the presence of the alcohol, as compared with another common cactus yeast, Pichia cactophila, which has less tolerance and cannot metabolize 2-propanol. Because 2-propanol is commonly found i

Sex-biased Expression of Genes and Transposable Elements in Hybrids from Drosophila mojavensis and Drosophila arizonae.

Sex-biased gene expression is one of the main forces driving differences between sexes. Transposable elements (TEs) have also been shown to display sex-specific expression, mainly in the gonads. Despite this clear sexual dimorphism in the transcriptome, their contribution to postzygotic isolation remains poorly understood. Here, we tested whether the balance of gene and TE expression between ovaries and testes is disrupted in hybrids, potentially contributing to reproductive barriers. We analyzed Drosophila mojavensis and Drosophila arizonae, sibling species that produce sterile hybrids with d

The sibling species Drosophila melanogaster and Drosophila simulans differ in the expression profile of glutathione S-transferases.

Two major forms of glutathione S-transferase are known in Drosophila melanogaster: GST D and GST 2. In the present paper we report the existence of a third major form of glutathione S-transferase in Drosophila simulans. Induction with phenobarbital revealed a different regulation of GST between these species. Despite the fact that these two species are closely related, there was a difference in the expression profile of the enzyme implicated in the detoxification system, suggesting variations in capacity to suit their environment.

Polycomb Group Proteins in Drosophila melanogaster.

The development of multicellular organisms relies on the precise establishment and stable inheritance of cell type-specific transcriptional programs without changes to the underlying DNA sequence. Polycomb group (PcG) proteins are evolutionarily conserved epigenetic repressors that govern developmental gene expression by assembling into multiprotein complexes to establish robust and spatiotemporally restricted transcriptional silencing. Since their discovery through the classic Polycomb mutant phenotype in Drosophila melanogaster, PcG proteins have become a central paradigm for understanding e
