Mutational background influences P. aeruginosa ciprofloxacin resistance evolution but preserves collateral sensitivity robustness.

Collateral sensitivity is an evolutionary trade-off whereby acquisition of the adaptive phenotype of resistance to an antibiotic leads to the nonadaptive increased susceptibility to another. The feasibility of harnessing such a trade-off to design evolutionary-based approaches for treating bacterial infections has been studied using model strains. However, clinical application of collateral sensitivity requires its conservation among strains presenting different mutational backgrounds. Particularly relevant is studying collateral sensitivity robustness of already-antibiotic-resistant mutants w

Optimization of sequential therapies to maximize extinction of resistant bacteria through collateral sensitivity.

Antimicrobial resistance (AMR) threatens global health. A promising and underexplored strategy to tackle this problem is sequential therapies exploiting collateral sensitivity (CS), whereby resistance to one drug increases sensitivity to another. Here, we develop a four-genotype stochastic birth-death model with two bacteriostatic antibiotics to identify switching periods that maximize bacterial extinction under subinhibitory concentrations. We show that extinction probability depends nonlinearly on switching period, with stepwise increases aligned to discrete switch events: fast sequential th

Collateral sensitivity of antibiotic-resistant microbes.

Understanding how evolution of microbial resistance towards a given antibiotic influences susceptibility to other drugs is a challenge of profound importance. By combining laboratory evolution, genome sequencing, and functional analyses, recent works have charted the map of evolutionary trade-offs between antibiotics and have explored the underlying molecular mechanisms. Strikingly, mutations that caused multidrug resistance in bacteria simultaneously enhanced sensitivity to many other unrelated drugs (collateral sensitivity). Here, we explore how this emerging research sheds new light on resi

Transcriptomic correlates of ceftazidime-avibactam collateral sensitivity in tigecycline-resistant NDM-producing enterobacter cloacae complex.

PURPOSE: Carbapenem-resistant Enterobacter cloacae complex (CRECC) has become an increasingly important pathogen in healthcare-associated infections, with limited treatment options and a high mortality rate. As reports of antimicrobial resistance continue to rise, tigecycline (TGC) and ceftazidime-avibactam (CZA) have emerged as the last-line therapies for CRECC infections. The aim of this study was to investigate collateral sensitivity to ceftazidime-avibactam following the acquisition of tigecycline resistance in NDM-producing CRECC, and to explore the transcriptional changes associated with

Large scale laboratory evolution uncovers clinically relevant collateral antibiotic sensitivity.

The increasing prevalence of antibiotic resistance is a critical challenge, necessitating the development of strategies to mitigate the evolution of resistance. Collateral sensitivity (CS)-based sequential therapies have been proposed to mitigate resistance evolution. However, the evolutionary repeatability of CS across different experimental conditions and its clinical relevance remain underexplored, hindering its potential for translation into clinical practice. Here, we evolve 20-24 lineages of E. coli against tigecycline (TIG) and piperacillin (PIP), antibiotics suggested to produce CS, th

First Diagonal to Left Ventricle Fistula With LAD Collateral Flow: An Unusual Coronary Detour.

BACKGROUND: Coronary artery fistulas are rare vascular anomalies connecting a coronary artery to a cardiac chamber or vessel. While typically associated with ischemia via coronary steal, they can paradoxically serve a protective role. CASE SUMMARY: We present a 57-year-old Asian male with 2 weeks of effort angina who was found to have an elevated high-sensitivity troponin. Urgent coronary angiography revealed a 100% mid left anterior descending artery (LAD) occlusion with collateral flow supplied predominantly by a fistulizing first diagonal artery (D1) communicating with the left ventricle.

Phenotype and genotype alteration during adaptive evolution of Enterococcus faecalis to antimicrobials.

The current worldwide emergence of resistance to antibiotics in bacteria constitutes an important growing public health threat. The mechanisms of the emergence and dissemination of resistance remain to be elucidated. Adaptation laboratory evolution provide an approach to investigate the acquisition of de novo mutations that confers drug resistance. In our study, 3 Enterococcus faecalis clinical isolates and E. faecalis ATCC29212 were evolved resistant to 8 kinds of antimicrobials spanning five chemical classes, including ciprofloxacin, levofloxacin, gatifloxacin, penicillin, imipenem, vancomyc

Multidrug Resistance (MDR) and Collateral Sensitivity in Bacteria, with Special Attention to Genetic and Evolutionary Aspects and to the Perspectives of Antimicrobial Peptides-A Review.

Antibiotic poly-resistance (multidrug-, extreme-, and pan-drug resistance) is controlled by adaptive evolution. Darwinian and Lamarckian interpretations of resistance evolution are discussed. Arguments for, and against, pessimistic forecasts on a fatal "post-antibiotic era" are evaluated. In commensal niches, the appearance of a new antibiotic resistance often reduces fitness, but compensatory mutations may counteract this tendency. The appearance of new antibiotic resistance is frequently accompanied by a collateral sensitivity to other resistances. Organisms with an expanding open pan-genome

Association between clinical antibiotic resistance and susceptibility of Pseudomonas in the cystic fibrosis lung.

BACKGROUND AND OBJECTIVES: Cystic fibrosis patients suffer from chronic lung infections that require long-term antibiotic therapy. Pseudomonas readily evolve resistance, rendering antibiotics ineffective. In vitro experiments suggest that resistant bacteria may be treated by exploiting their collateral sensitivity to other antibiotics. Here, we investigate correlations of sensitivity and resistance profiles of Pseudomonas aeruginosa that naturally adapted to antibiotics in the cystic fibrosis lung. METHODOLOGY: Resistance profiles for 13 antibiotics were obtained using broth dilution, E-test

Collateral effects of antibiotic resistance in Campylobacter jejuni.

Campylobacter jejuni is a leading cause of bacterial gastroenteritis worldwide, and resistance to major antibiotic classes is increasing. Exploiting collateral sensitivity (CS) may be an effective approach to optimize antibiotic use and mitigate the impact of resistance. In this study, we investigated CS and collateral resistance (CR) profiles in experimentally evolved C. jejuni mutants derived from ATCC 29428 resistant to ciprofloxacin, erythromycin, gentamicin, and tetracycline. Minimum inhibitory concentrations of thirteen antibiotics were determined in the wild-type and mutant strains to c

In vitro evolution of caspofungin resistance in Candidozyma auris via FKS1 hotspot I mutations results in moderate fitness trade-offs but no reduction in virulence.

Resistance to echinocandins in the recently emerged pathogen Candidozyma auris (previously known as Candida auris) poses significant clinical challenges. This study reports the in vitro evolution of a caspofungin-sensitive C. auris clade II isolate to acquire caspofungin resistance. We sequenced the whole genome of the parental strain and its drug-resistant progeny. The resistant isolates harboured mutations in the FKS1 gene altering a conserved residue (S639) also mutated in caspofungin-resistant clinical isolates; demonstrating that FKS1-S639 mutations are sufficient to confer resistance to

Rapid Phenotypic Convergence towards Collateral Sensitivity in Clinical Isolates of Pseudomonas aeruginosa Presenting Different Genomic Backgrounds.

Collateral sensitivity (CS) is an evolutionary trade-off by which acquisition of resistance to an antibiotic leads to increased susceptibility to another. This Achilles' heel of antibiotic resistance could be exploited to design evolution-based strategies for treating bacterial infections. To date, most studies in the field have focused on the identification of CS patterns in model strains. However, one of the main requirements for the clinical application of this trade-off is that it must be robust and has to emerge in different genomic backgrounds, including preexisting drug-resistant isolat

Cytotoxicity of natural products and derivatives toward MCF-7 cell monolayers and cancer stem-like mammospheres.

UNLABELLED: Although cancer stem-like cells (CSCs) are rare, they can enter a non-proliferative or dormant state and resist therapy. Furthermore, quiescent CSCs are responsible for metastases that can appear after curative surgical treatment of a primary tumor. Because of drug resistance of CSCs, the development of novel therapies is urgently required that specifically target CSCs. PURPOSE: The aim of the present study was to investigate the potential of a panel of natural products and derivatives to inhibit CSC-enriched mammospheres of MCF-7 breast cancer cells. METHODS: CD44(high)/CD24(low

Molecular mechanisms of collateral sensitivity to the antibiotic nitrofurantoin.

Antibiotic resistance increasingly limits the success of antibiotic treatments, and physicians require new ways to achieve efficient treatment despite resistance. Resistance mechanisms against a specific antibiotic class frequently confer increased susceptibility to other antibiotic classes, a phenomenon designated collateral sensitivity (CS). An informed switch of antibiotic may thus enable the efficient treatment of resistant strains. CS occurs in many pathogens, but the mechanisms that generate hypersusceptibility are largely unknown. We identified several molecular mechanisms of CS against

β-lactamase expression induces collateral sensitivity in Escherichia coli.

Major antibiotic groups are losing effectiveness due to the uncontrollable spread of antimicrobial resistance (AMR) genes. Among these, β-lactam resistance genes -encoding β-lactamases- stand as the most common resistance mechanism in Enterobacterales due to their frequent association with mobile genetic elements. In this context, novel approaches that counter mobile AMR are urgently needed. Collateral sensitivity (CS) occurs when the acquisition of resistance to one antibiotic increases susceptibility to another antibiotic and can be exploited to eliminate AMR selectively. However, most CS ne

Catalase activity deficiency sensitizes multidrug-resistant Mycobacterium tuberculosis to the ATP synthase inhibitor bedaquiline.

Multidrug-resistant tuberculosis (MDR-TB), defined as resistance to the first-line drugs isoniazid and rifampin, is a growing source of global mortality and threatens global control of tuberculosis disease. The diarylquinoline bedaquiline has recently emerged as a highly efficacious drug against MDR-TB and kills Mycobacterium tuberculosis by inhibiting mycobacterial ATP synthase. However, the mechanisms underlying bedaquiline's efficacy against MDR-TB remain unknown. Here we investigate bedaquiline hyper-susceptibility in drug-resistant Mycobacterium tuberculosis using systems biology approach

Evolutionary trajectory of bacterial resistance to antibiotics and antimicrobial peptides in Escherichia coli.

The global crisis of antimicrobial resistance poses a major threat to human health, underscoring the urgency of developing new antibacterial strategies. Antimicrobial peptides (AMPs) are promising alternatives to antibiotic therapy, yet potential microbial resistance is of great concern. Resistance is often accompanied by fitness costs, which may in turn influence the spread of drug-resistant bacteria and their susceptibility to other antimicrobial agents. Herein, we investigate the evolutionary trajectory of bacterial resistance to antibiotics and AMPs in Escherichia coli, and evaluate the fi

Structures revealing mechanisms of resistance and collateral sensitivity of Plasmodium falciparum to proteasome inhibitors.

The proteasome of the malaria parasite Plasmodium falciparum (Pf20S) is an advantageous drug target because its inhibition kills P. falciparum in multiple stages of its life cycle and synergizes with artemisinins. We recently developed a macrocyclic peptide, TDI-8304, that is highly selective for Pf20S over human proteasomes and is potent in vitro and in vivo against P. falciparum. A mutation in the Pf20S β6 subunit, A117D, confers resistance to TDI-8304, yet enhances both enzyme inhibition and anti-parasite activity of a tripeptide vinyl sulfone β2 inhibitor, WLW-vs. Here we present the high-

Collateral sensitivity as a strategy against cancer multidrug resistance.

While chemotherapy remains the most effective treatment for disseminated tumors, acquired or intrinsic drug resistance accounts for approximately 90% of treatment failure. Multidrug resistance (MDR), the simultaneous resistance to drugs that differ both structurally and mechanistically, often results from drug efflux pumps in the cell membrane that reduce intracellular drug levels to less than therapeutic concentrations. Expression of the MDR transporter P-glycoprotein (P-gp, MDR1, ABCB1) has been shown to correlate with overall poor chemotherapy response and prognosis. This review will focus
