Antimicrobial Properties on Non-Antibiotic Drugs in the Era of Increased Bacterial Resistance.

In recent years, due to the dramatic increase in and global spread of bacterial resistance to a number of commonly used antibacterial agents, many studies have been directed at investigating drugs whose primary therapeutic purpose is not antimicrobial action. In an era where it is becoming increasingly difficult to find new antimicrobial drugs, it is important to understand these antimicrobial effects and their potential clinical implications. Numerous studies report the antibacterial activity of non-steroidal anti-inflammatory drugs, local anaesthetics, phenothiazines such as chlorpromazine,

Combating Bacterial Drug Resistance via Dual-Mechanism Drive Penetration and Triple-Approach Metabolic Disruption.

The emergence of multidrug-resistant (MDR) bacteria poses a serious threat to global public health. Enhancing the permeability across the physical barriers, including lipopolysaccharide (LPS) and extracellular polymeric substances (EPS), while simultaneously preventing drug resistance, remains a major challenge. To address this issue, we developed a novel biotin-conjugated cationic photosensitizer (PS), ACR-DM-Bio. It not only effectively binds to the bacterial outer membrane and EPS via electrostatic interactions but also facilitates active uptake through the biotin transport system. This dua

Combating bacterial biofilms and related drug resistance: Role of phyto-derived adjuvant and nanomaterials.

The emergence of antimicrobial resistance (AMR) in clinical microbes has led to a search for novel antibiotics for combating bacterial infections. The treatment of bacterial infections becomes more challenging with the onset of biofilm formation. AMR is further accelerated by biofilm physiology and differential gene expression in bacteria with an inherent resistance to conventional antibiotics. In the search for innovative strategies to control the spread of AMR in clinical isolates, plant-derived therapeutic metabolites can be repurposed to control biofilm-associated drug resistance. Unlike a

Spontaneous bacterial peritonitis - therapeutic challenges in the era of increasing drug resistance of bacteria.

Spontaneous bacterial peritonitis (SBP) is one of the most common bacterial infections in patients with liver cirrhosis and it significantly contributes to the deterioration of the prognosis and increased risk of mortality. Previous data suggested that the most common pathogens causing SBP are G-negative aerobic bacteria and treatment recommended by the international guidelines (EASL, AASLD) is highly effective. In recent years, due to the widespread use of antibiotic prophylaxis and the increased frequency of hospitalization along with the use of invasive procedures in patients with cirrhosis

Survey of drug resistance associated gene mutations in Mycobacterium tuberculosis, ESKAPE and other bacterial species.

Tuberculosis treatment includes broad-spectrum antibiotics such as rifampicin, streptomycin and fluoroquinolones, which are also used against other pathogenic bacteria. We developed Drug Resistance Associated Genes database (DRAGdb), a manually curated repository of mutational data of drug resistance associated genes (DRAGs) across ESKAPE (i.e. Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacter spp.) pathogens, and other bacteria with a special focus on Mycobacterium tuberculosis (MTB). Bacterial quorum sensing inhibitors: attractive alternatives for control of infectious pathogens showing multiple drug resistance.

Quorum sensing (QS) is a bacterial communication process that depends on the bacterial population density. It involves small diffusible signaling molecules which activate the expression of myriad genes that control diverse array of functions like bioluminescence, virulence, biofilm formation, sporulation, to name a few. Since QS is responsible for virulence in the clinically relevant bacteria, inhibition of QS appears to be a promising strategy to control these pathogenic bacteria. With indiscriminate use of antibiotics, there has been an alarming increase in the number of antibiotic resistant

Antimicrobial peptides RI8 and RW8 target bacterial membranes and genomic DNA to overcome drug resistance.

The global rise of multidrug-resistant (MDR) pathogens invalidates conventional antibiotics, while natural antimicrobial peptides (AMPs) lack proteolytic stability and precise targeting. This study reports a protease-resistant symmetric (RRYY)2P(YYRR)2 scaffold (Y = isoleucine/tryptophan) with alternating cationic-hydrophobic repeats, a central proline hinge, and palindromic topology for dual bacterial membrane-genomic DNA targeting. Two derived peptides, RI8 and RW8, were characterized: RI8 showed broad-spectrum activity against MDR pathogens (MIC: 2-16 μg/mL), high therapeutic index (39.4),

Bacterial internalins exploit E-cadherin to promote head and neck tumor metastasis and drug resistance.

Head and neck squamous cell carcinoma (HNSCC) is an aggressive malignancy characterized by local invasion, lymph node metastasis, and therapeutic resistance. Chronic periodontal disease has been linked to HNSCC progression, yet the responsible pathogens and underlying molecular mechanisms remain unclear. Here, we show that the keystone periodontal pathogen Porphyromonas gingivalis promotes HNSCC metastasis and chemoresistance through two internalin proteins that are secreted via the type IX secretion system (T9SS). These internalin proteins specifically bind the EC1 domain of E-cadherin throug

Bacterial profile, drug resistance pattern, clinical and laboratory predictors of ascites infection in cirrhosis patients.

Ascites is a pathological collection of free fluid in the peritoneal cavity, which is a common complication in patients with cirrhosis, an advanced liver disease. Bacterial infection increases the mortality rate of hospitalized patients with cirrhosis, irrespective of the severity of the liver disease. Around 60% of patients with compensated cirrhosis developed ascites within 10 years during the course of their disease. The in-hospital mortality rate due to spontaneous bacterial peritonitis (SBP) could exceed 90%, but with early diagnosis and prompt antibiotic therapy, this rate has been shown

Characterization and in vitro Analysis of Probiotic-Derived Peptides Against Multi Drug Resistance Bacterial Infections.

An inexorable switch from antibiotics has become a major desideratum to overcome antibiotic resistance. Bacteriocin from Lactobacillus casei, a cardinal probiotic was used to design novel antibacterial peptides named as Probiotic Bacteriocin Derived and Modified (PBDM) peptides (PBDM1: YKWFAHLIKGLC and PBDM2: YKWFRHLIKKLC). The loop-shaped 3D structure of peptides was characterized in silico via molecular dynamics simulation as well as biophysically via spectroscopic methods. Thereafter, in vitro results against multidrug resistant bacterial strains and hospital samples demonstrated the strong

Engineering Nano-Antibiotics for Accelerating Wound Healing in Drug-Resistant Bacterial Infections.

Plenty of nano-antimicrobial materials have been developed successively, aiming to address severe clinical challenges such as wound healing disorders and high postoperative mortality rates caused by drug-resistant bacterial infections. However, their reliance on external stimuli (light, thermal energy, or exogenous H2O2 addition) for bactericidal activation severely hampers clinical translation from bench to bedside. Herein, we report an engineered Cu/CeO2 nanoplatelet (NP) system that functions as a stimulus-independent, time-dependent nano-antibiotic against methicillin-resistant Staphylococ

Biocompatible Gallium Nanodots against Drug-Resistant Bacterial Pneumonia and Liver Abscess.

Resistant bacterial infection remains a severe public health threat, and conventional antibiotic drugs work poorly in effectively treating infectious diseases. Here, we developed gallium-based nanodots (Ga NDs), consisting of specific disruption of bacterial iron ability, to treat multidrug-resistant (MDR) Gram-negative bacteria-infected diseases. The Ga NDs significantly suppress the proliferation of two typical MDR bacteria strains (P. aeruginosa and ESBL E. coli) compared with clinically used antibacterial drugs, including penicillin and levofloxacin. Ga NDs could also disrupt the biofilms

DNA replication proteins as potential targets for antimicrobials in drug-resistant bacterial pathogens.

With the impending crisis of antimicrobial resistance, there is an urgent need to develop novel antimicrobials to combat difficult infections and MDR pathogenic microorganisms. DNA replication is essential for cell viability and is therefore an attractive target for antimicrobials. Although several antimicrobials targeting DNA replication proteins have been developed to date, gyrase/topoisomerase inhibitors are the only class widely used in the clinic. Given the numerous essential proteins in the bacterial replisome that may serve as a potential target for inhibitors and the relative paucity o
