The effect of restricted medication with lasalocid sodium on the development of acquired immunity against Eimeria tenella was evaluated. The medication was allowed for all or part of 6-day test period (one day before until 4 days after infection). The parameters used for such evaluations were lesion score, caecal, bursal and splenic weights. The optimum treatment time for the drug was clearly indicated by lesion score which was very low when the medication was initiated 1 day before until 1 day after inoculation, but only partly effective if given on Day 2 post-inoculation. The challenge with

BACKGROUND: Coccidia are a group of intracellular protozoal parasites within the phylum Apicomplexa. Eimeria tenella, one of the species that cause intestinal coccidiosis in poultry, can cause significant mortality and morbidity. Diploid oocysts of Eimeria species are shed in the feces of an infected host and must sporulate to achieve infectivity. This process results in eight haploid infectious units, called sporozoites, held within a single oocyst. Each Eimeria spp. parasite possesses a single apicoplast and a single mitochondrion, both of which carry multiple copies of their respective orga

The purpose of this study was to characterize Eimeria maxima immune-mapped protein 1 (IMP1) that is hypothesized to play a role in eliciting protective immunity against E. maxima infection in chickens. RT-PCR analysis of RNA from unsporulated and sporulating E. maxima oocysts revealed highest transcription levels at 6-12h of sporulation with a considerable downregulation thereafter. Alignment of IMP1 coding sequence from Houghton, Weybridge, and APU-1 strains of E. maxima revealed single nucleotide polymorphisms that in some instances led to amino acid changes in the encoded protein sequence.

Validation of transgenic Eimeria parasites is essential for confirming stable genomic integration and expression of exogenous genes. This protocol describes a comprehensive workflow for verifying transgene insertion and protein expression in Eimeria tenella. The procedure begins with genomic DNA extraction from sporulated oocysts, followed by PCR amplification to preliminarily verify the presence of the target fragment. To further confirm genomic integration, whole-genome resequencing is performed to identify insertion sites and evaluate the stability of the integrated construct in the parasit

This study aims to explore the coding sequence (CDS) of the putative DUS gene in Eimeria media and assess its potential biological functions during the parasite's lifecycle. Initially, oocysts were isolated from fecal samples of rabbits infected with E. media, from which DNA and RNA were extracted. These extractions were used as templates for PCR to successfully amplify the CDS of the DUS gene, confirming its presence within the E. media genome. Further analysis using quantitative PCR (qPCR) demonstrated significantly higher expression of the DUS gene in the precocious line (PL) compared to th

Coccidiosis is an intestinal parasitic disease that causes huge economic losses in the poultry industry globally. Henan and Hubei, as important poultry production provinces in China, have great pressure for the prevention and control of chicken coccidiosis. In order to obtain information on the local prevalence of Eimeria species, we used an internal transcribed spacer 1 (ITS1) sequence of ribosomal DNA to identify the species from 318 fresh fecal samples. The fecal samples and the data relating to farm information were collected from 137 farms in Hubei and Henan provinces. As shown by genus-s

Toxoplasma gondii and Eimeria spp. are widely prevalent Coccidian parasites that undergo sexual reproduction during their life cycle. T. gondii can infect any warm-blooded animal in its asexual cycle; however, its sexual cycle is restricted to felines. Eimeria spp. are usually restricted to one host species, and their whole life cycle is completed within this same host. The literature reviewed in this article comprises the recent findings regarding the unique biology of the sexual development of T. gondii and Eimeria spp. The molecular basis of sex in these pathogens has been significantly unr

The present study aimed to compare the effect of different Artemisia annua extracts on sporulation rate of mixed oocysts of Eimeria acervulina, Eimeria necatrix, and Eimeria tenella. Three types of A. annua extracts including petroleum ether (PE), ethanol 96° (E), and water (W) extracts were prepared. Artemisinin, a sesquiterpene lactone endoperoxide derived from the A. annua analysis of each extract was done by high-performance liquid chromatography with ultraviolet detection (HPLC-UV). Fresh fecal samples containing three Eimeria species were floated and counted, and the oocysts were transfe

When sporozoites of Eimeria tenella were inoculated into an isolated horn of the bicornuate caecum of the domestic fowl, replicating infections, culminating in the production of oocysts, were found, almost exclusively, in the inoculated horn. In contrast, sporozoites of E. tenella introduced into the bloodstream via a superficial vein induced infections of equivalent intensity in both horns of the caecum. These findings are discussed with reference to (1) the detection (by tissue transfer) of infective sporozoites in samples of cardiac blood and vascular tissues taken within 3 to 18 h of the o

Eimeria gruis and Eimeria reichenowi are common coccidial parasites of a number of species of cranes. Until recently, little was known about either the site for invasion or the dynamics of early development of the crane coccidia because of the difficulty of identifying sporozoites and early developmental stages of these parasites by conventional staining methods. In the present study, monoclonal antibodies (MAbs) elicited against Eimeria spp. of chickens and turkeys were found to cross-react with sporozoites and developmental stages of E. reichenowi in the tissues of Florida sandhill cranes (G

UNLABELLED: Avian coccidiosis (AC) is a parasitic disease caused by the protozoan parasite Eimeria tenella (E.tenella), which primarily parasitizes the cecum of chickens, resulting in a reduction of their production capacity. Areca alkaloids(AAl), derived from the plant Areca catechu L, have been repeatedly reported to exhibit anti-parasitic effects against protozoa, this suggests its potential efficacy in combating E.tenella.
THE PURPOSE: of this study is to explore the mechanism underlying the anti E.tenella effects of AAl.
METHODS: Screening potential active ingredients of AAl by establis

Apicomplexan parasites have divergent biogenesis machinery for small RNA generation. Analysis has shown that parasites in Plasmodium and Cryptosporidium as well as many species in Leishmania or Trypanosoma do not have a complete machinery in small RNA biogenesis. Recently, the miRNA-generating system of Toxoplasma has been identified as plant/fungal-like and its miRNAome has been elucidated. However, the microRNA (miRNA) expression profiles and their potential regulatory functions in different stages of Eimeria tenella remain largely unknown. In this study, we characterized the RNA silencing m

Molecular approaches supporting identification of Eimeria parasites infecting chickens have been available for more than 20 years, although they have largely failed to replace traditional measures such as microscopy and pathology. Limitations of microscopy-led diagnostics, including a requirement for specialist parasitological expertise and low sample throughput, are yet to be outweighed by the difficulties associated with accessing genomic DNA from environmental Eimeria samples. A key step towards the use of Eimeria species-specific PCR as a sensitive and reproducible discriminatory tool for

Freshly defecated unsporulated oocysts of Eimeria piriformis from rabbit were treated with various concentrations (1%, 2.5%, 5%, and 10%) of chromium compounds, potassium dichromate, potassium chromate, chromium oxide and chromium nitrate, to examine their effect on sporulation. The sporulation time of oocysts treated with 1 to 10% K(2)Cr(2)O(7) was 28 h. However, much longer sporulation times of about 60 h were required for oocysts treated with 2.5% CrO(3) and Cr(NO(3))(3). Moreover, for oocysts treated with distilled water, 1% K(2)CrO(4) and 10% K(2)CrO(4), the sporulation times required wer

Coccidiosis, caused by Eimeria species, is a significant disease affecting the poultry industry worldwide, leading to substantial economic losses due to reduced flock performance. Effective vaccination strategies require the precise quantification of the dosage of viable Eimeria oocysts to induce immunity in young chicks without causing disease. However, current methods for determining oocyst viability rely on sophisticated equipment and are not effective for routine monitoring. Recently, we documented the presence of granular structures exclusively in dead oocysts using high-resolution micros

Avian coccidiosis significantly impairs intestinal health in chickens and remains a major threat to the poultry industry worldwide. Frequent co-infections with three Eimeria species, i.e., Eimeria tenella, E. necatrix, and E. maxima, present challenges for effective vaccine development. Here, we first used two-dimensional gel electrophoresis and silver staining to identify 650 E. tenella sporozoite proteins and then detected 18 cross-reactive immunogens based on Western blotting and proteomic analysis. These immunogens were consistently recognized by hyperimmune sera raised against three Eimer

Eimeria tenella infections are known to cause severe caecal damage and death of the infected chicken. Gamogony is an essential stage in E. tenella life cycle and in the establishment of coccidiosis. Prior research had extensively explored isolation and separation of the parasite gametes - microgamete (male) and macrogamete (female). However, there is little information on the efficient, highly purified and distinctly separated male and female gametes. In this study, we generated a genome editing line expressing mCherry fluorescent protein fused with GCS1 protein in E. tenella by using Toxoplas

The widespread apicomplexan parasites Toxoplasma gondii (T. gondii) and Eimeria tenella (E. tenella) are important pathogens with high prevalence in poultry. The aim of our study was the investigation of mutual influences in co-infected chickens, focusing on immune response and course of infection. Two separate trials were performed using in total 96 1-day-old chickens, divided into four study groups: group NC (negative control, uninfected), group PC-T (oral or intramuscular infection with T. gondii oocysts (trial 1) or tachyzoites (trial 2), respectively), group PC-E (oral infection with E. t

Eimeria tenella (E. tenella) causes severe coccidiosis in chickens, leading to bloody diarrhea, growth retardation, high mortality, and substantial economic losses. Rising drug resistance and residue concerns limit conventional chemical anticoccidials, driving interest in Traditional Chinese Medicine as a safe, cost-effective, and sustainable alternative. To investigate the effects of Sophora flavescens seed extract (SFSE) on E. tenella-infected chicken ceca, 12-day-old chickens were assigned to control, infection, and SFSE groups, with the latter two orally inoculated with sporulated E. tenel
