Microemulsion electrokinetic chromatography for separation and analysis of curcuminoids in turmeric samples.

Microemulsion EKC (MEEKC) was developed for quantitative analysis of curcuminoids, such as curcumin (C), demethoxycurcumin (D), and bis-demethoxycurcumin (B). MEEKC separation of curcuminoids was optimized, and a change in resolution was explained using a modified equation for resolution in MEEKC without electroosmosis. The suitable MEEKC conditions for separation of curcuminoids were obtained to be the microemulsion buffer containing 50 mM phosphate buffer at pH 2.5, 1.1% v/v n-octane as oil droplets, 180 mM SDS as surfactant, 890 mM 1-butanol as cosurfactant, and 25% v/v 2-propanol as organi

Electrokinetically based approach for single-nucleotide polymorphism discrimination using a microfluidic device.

In this work, we describe and implement an electrokinetic approach for single-nucleotide polymorphism (SNP) discrimination using a PDMS/glass-based microfluidic chip. The technique takes advantage of precise control of the coupled thermal (Joule heating), shear (electroosmosis), and electrical (electrophoresis) energies present at an array of probes afforded by the application of external electrical potentials. Temperature controllers and embedded thermal devices are not required. The chips can be easily and inexpensively fabricated using standard microarray printing methods combined with soft

Effect of pH and sodium dodecyl sulfate concentration on the analytical window in the direct-injection analysis of plasma samples by capillary electrophoresis.

Plasma samples can be analyzed with direct injection onto a capillary electrophoresis system if interactions between the plasma proteins and capillary walls are minimized. This can be achieved using micellar electrokinetic capillary chromatography with a surfactant such as sodium dodecyl sulfate. The surfactant complexes with the proteins, giving them a net negative charge, and thus causes them to be repelled from the negatively-charged fused-silica capillary walls. Migrating as anions, the complexed proteins appear late in the electropherogram. In such analyses, it is important to manipulate

Electromotive Enhanced Drug Administration in Oncology: Principles, Evidence, Current and Emerging Applications.

Local-regional administration of cytotoxic drugs is an important adjunct to systemic chemotherapy amongst cancer patients. It allows for targeted delivery of agents at high concentration to target sites while minimizing systemic side effects. Despite the pharmacokinetic advantages of the local-regional approach, drug transport into tumor nodules remains limited due to the biophysical properties of these tissues. Electromotive enhanced drug administration (EMDA) represents a potential solution to overcome challenges in local drug transport by applying electric currents. Through electrokinetic p

Capillary zone electrophoresis for the estimation of transdermal iontophoretic mobility.

The objective of the study was to investigate the relationship between transdermal iontophoretic flux--specifically, the electromigratory component--and electrophoretic mobility as determined by capillary zone electrophoresis (CZE). First, the steady-state iontophoretic transport rates of a series of dipeptides across porcine skin were determined in vitro. Co-iontophoresis of acetaminophen was used to quantify the respective contributions of electroosmosis (EO) and electromigration (EM). Second, the electrophoretic mobilities of the dipeptides and three other cationic drugs (lidocaine, propran

Validation of CE modeling with a contactless conductivity array detector.

Dynamic computer simulation data are compared for the first time with CE data obtained with a laboratory made system comprising an array of 8 contactless conductivity detectors (C(4) Ds). The experimental setup featured a 50 μm id linear polyacrylamide (LPA) coated fused-silica capillary of 70 cm length and a purpose built sequential injection analysis manifold for fluid handling of continuous or discontinuous buffer configurations and sample injection. The LPA coated capillary exhibits a low EOF and the manifold allows the placement of the first detector at about 2.7 cm from the sample inlet.

A mathematical model of dielectrophoretic data to connect measurements with cell properties.

Dielectrophoresis (DEP) brings about the high-resolution separations of cells and other bioparticles arising from very subtle differences in their properties. However, an unanticipated limitation has arisen: difficulty in assignment of specific biological features which vary between two cell populations. This hampers the ability to interpret the significance of the variations. To realize the opportunities made possible by dielectrophoresis, the data and the diversity of structures found in cells and bioparticles must be linked. While the crossover frequency in DEP has been studied in-depth and

Semi-permanent cationic coating for protein separations.

Capillary electrophoresis has been used extensively for protein separations, but interactions of proteins with the negative charge on the surface of fused silica capillary create band broadening and diminish the separation efficiency. Coatings developed to mask the negative charge of the capillary affect the electroosmotic flow. The method presented in this work addresses these concerns through the use of a two-layer coating of a semi-permanent phospholipid substrate and cetyltrimethylammonium bromide (CTAB). When used alone, phospholipid coating suppresses the electroosmotic flow but cannot b

Iontophoretic drug delivery across the nail.

INTRODUCTION: Topical drug delivery to treat nail diseases such as onychomycosis and psoriasis is receiving increasing attention. Topical nail delivery is challenged by the complicated structure of the nail and the low permeability of most drugs across the nail plate. Considerable effort has been directed at developing methods to promote drug permeation across the nail plate. Iontophoresis efficiently enhances molecular transport across the skin and the eye and is now being tested for its potential in ungual delivery. AREAS COVERED: This review covers the basic mechanisms of transport (electr

The Role of pH, Electrodes, Surfactants, and Electrolytes in Electrokinetic Remediation of Contaminated Soil.

Electrokinetic remediation has, in recent years, shown great potential in remediating polluted environments. The technology can efficiently remove heavy metals, chlorophenols, polychlorinated biphenyls, phenols, trichloroethane, benzene, toluene, ethylbenzene, and xylene (BTEX) compounds and entire petroleum hydrocarbons. Electrokinetic remediation makes use of electrolysis, electroosmosis, electrophoresis, diffusion, and electromigration as the five fundamental processes in achieving decontamination of polluted environments. These five processes depend on pH swings, voltage, electrodes, and e

Application of UV radiation for in-situ Cr(VI) reduction from contaminated soil with electrokinetic remediation.

Restoring hexavalent chromium (Cr(VI)) to trivalent chromium (Cr(III)) from contaminated soil is a cost-effective alternative for attenuating Cr(VI) toxicity to the ecosystem. A new electrokinetic remediation (EKR) system with UV light was explored to overcome an energy barrier to catalyze Cr(VI) reduction from the surface soil near the anodic reservoir. Natural organic matters and minerals from the contaminated soil acted as electron donors and catalysts for Cr(VI) photo-reduction and no additional chemical reagent. There was almost no residual Cr(VI) in anolyte after UV/EKR compared with the

The effect of Fe(0) on electrokinetic remediation of clay contaminated with perchloroethylene.

The present paper was to investigate the effect of Fe(0) reaction barrier position and Fe(0) quantity on the remediation efficiency and electrokinetic performances of tetrachloroethylene (PCE) contaminated clay under potential gradient of 2 V/cm for 5 days. The Fe(0) reaction barrier was composed of 2 to approximately 16 g of Fe(0) mixed with Ottawa sand in a ratio of 1: 2. Its positions were respectively located at the anode, the middle, and the cathode end of the electrokinetic cell. Results showed that a relatively higher remediation of 66% of PCE was found as the Fe(0) reaction barrier loc

Unraveling the Fluoride-Induced Interface Reconstruction Across Lead-Based Hierarchical MnO2 Anode in Zinc Electrowinning.

Although the hierarchical manganese dioxide film electrode shows promise as a durable and catalytically active anode for zinc electrowinning, it often fails and deactivates when it is exposed to fluoride-rich environments. The lack of understanding regarding the mechanism behind fluoride-induced irreversible interface reconstruction hinders their practical application in large-scale energy-saving and pollution-reduction efforts. Here, we conducted multidimensional operando investigations to gain insights into the dynamic evolution across the film electrode interface with temporal and spatial r

Preparation and application of hydrophobic hybrid monolithic columns containing polyhedral oligomeric silsesquioxanes for capillary electrochromatography.

Hydrophobic organic-inorganic hybrid monolithic columns were synthesized via thermally initiated free radical polymerization with the confines of 75 μm id capillary using a polyhedral oligomeric silsesquioxane (POSS) reagent containing eight or more methacrylate groups as the crosslinker. Three organic functional monomers, butyl methacrylate (BuMA), lauryl methacrylate (LMA) and methacrylic acid (MAA), were selected and copolymerized with the POSS in the presence of 1-propanol and 1,4-butanediol to prepare the poly(POSS-co-BuMA), poly(POSS-co-LMA), and poly(POSS-co-MAA) monoliths, respectively

Ion Transport in Charged Membranes: Linking Electric-Field-Driven Mechanisms to Pore Size via Perturbation Analysis.

Ion-exchange membranes are a critical component in electrochemical systems. Nevertheless, the understanding and modeling of ion transport within these porous structures have been limited by particular complexity reductions, either ignoring the dimensionality of their porous network architecture or imposing geometric assumptions (i.e., overlapping double layers). Before addressing this morphology-transport gap, a framework that relates the driving forces of transport to the geometry of a single pore is required. In this work, our modeling domain consists of a two-dimensional single pore with ch

Electrically controlled release of anticancer drug 5-fluorouracil from carboxymethyl cellulose hydrogels.

Transdermal drug delivery is a technique used to introduce a drug through the skin and into the blood system. The drug-matrix hydrogels based on carboxymethyl cellulose (CMC) were prepared by solution casting method using citric acid as the crosslinking agent at various amounts. The 5-fluorouracil, an anticancer non-ionic drug, was released from the CMC hydrogels under applied electrical field. The diffusion coefficient and the release mechanism of the drug of the CMC hydrogels were investigated by the modified Franz-diffusion cell with the PBS buffer solution at the pH value of 7.4 and at 37

Direct current stimulation of endothelial monolayers induces a transient and reversible increase in transport due to the electroosmotic effect.

We investigated the effects of direct current stimulation (DCS) on fluid and solute transport across endothelial cell (EC) monolayers in vitro. Our motivation was transcranial direct current stimulation (tDCS) that has been investigated for treatment of neuropsychiatric disorders, to enhance neurorehabilitation, and to change cognition in healthy subjects. The mechanisms underlying this diversity of applications remain under investigation. To address the possible role of blood-brain barrier (BBB) changes during tDCS, we applied direct current to cultured EC monolayers in a specially designed c

Therapeutic drug monitoring by reverse iontophoresis.

Therapeutic molecules possessing distinct pharmacokinetic variation, narrow therapeutic index and concentration dependent therapeutic/adverse effects demand constant monitoring. The current methods for blood sampling are invasive and possess low patient compliance. Human skin, selective and effective membrane to chemical permeation, offers an alternative route for the extraction of endogenous molecules in the body. Significant attention has been received in the application of reverse iontophoresis in extracting drugs/biomaterials from the subdermal region. This technique involves transiting of

Guided Electrokinetic Assembly of Functionalized Latex Beads for Fluorescence Signal Enhancement Using AC Electro-Osmosis.

Fluorescence-based immunoassays are widely used for sensitive and specific biomarker detection; however, further improvements in sensitivity remain desirable for detecting low-abundance analytes without increasing assay complexity. In this work, we present a proof-of-concept demonstration of guided electrokinetic assembly of functionalized latex beads as a post-assay signal enhancement strategy using alternating-current electro-osmosis (ACEO). Using multiple short-end injections to develop fast electrophoretic separations--applications in iodide analysis.

The aim of this study was to develop a fast CE separation method by using multiple short-end injections in a capillary coated with quaternary ammonium chitosan (HACC), in order to determine the iodide content of pharmaceutical formulations. The BGE was composed of 20 mM tris(hydroxymethyl)aminomethane and 11 mM hydrochloric acid, at pH 8. The internal standard used was thiocyanate. Separations were performed in a fused silica capillary (32 cm total length, 8.5 cm effective length and 50 μm i.d.) coated with HACC and direct UV detection at 220 nm. EOF was modified by flushing the capillary with
