A new series of quinoxaline derivatives, 2a-4b, were synthesized and their anxiolytic potential was evaluated in vivo using elevated plus maze (EPM), open field (OF) and light-dark box (LDB) techniques. According to the results of the EPM, four active compounds were found in 2a, 2b, 2c, 4b. Their anxiolytic properties were confirmed in terms of LDB and the most active was compound 2b. In the OF, only 2c had an influence on the locomotor activity of the rodents. Thus, the most promising substance was determined; this was 2b, which has the structure of 2-(2-{[3-(4-tert-butylphenyl)quinoxaline-2-

INTRODUCTION: Although the transition of a lead candidate into a drug is currently structured by well-defined milestone, it is still most challenging and offers no guarantee in success to the end. In fact, ligand-based pharmacophore modeling has become a key motive force for retrieving potential leads across several therapeutic areas.
METHODS: An urgent need towards the development of novel antidepressant agents led us to generate a pharmacophore model from an existing 44 compounds dataset. The best model with one hydrophobic, two ring aromatic, and one positive ionization features was chosen

The present study tested the effects of cannabidiol (CBD) alone, Δ-9-tetrahydrocannabinol (THC) alone, and CBD and THC in combination (15:1 ratio) in the elevated plus maze (EPM), a test useful for the study of anxiety. In dose-response studies, adult, male CD1 mice were injected intraperitoneally with (1) CBD alone (0-96 mg/kg), (2) THC alone (0-6.4 mg/kg) or (3) CBD+THC in a 15:1 combination (0.0 + 0.0 mg/kg to 96.0 + 6.4 mg/kg). Diazepam (2.5 mg/kg) was also tested as a positive control. It was found that diazepam significantly increased open arm time in the EPM. CBD alone had no significan

Ultrasonic vocalizations (USV) are emitted by both young pups and adult rats to convey positive or negative emotional states. These USV manifestations are contingent on factors including developmental stage, situational requirements, and individual dispositions. Pups emit 40-kHz USV when separated from their mother and litter, which function to elicit maternal care. Conversely, adult rats can produce 50-kHz USV in response to stimuli that elicit reward-related states, including natural rewards, stimulant drugs, and reward-predictive stimuli. The present study aims to investigate whether pup 40

Caffeine is a commonly used stimulant of the central nervous system that reduces fatigue, increases alertness, and exerts positive effects on emotion through actions on various brain structures. High doses of caffeine can cause headaches, heart palpitations, hyperactivity, and anxiety symptoms. Consequently, reducing the consumption of stimulant substances, such as sugar and caffeine, is proposed to ameliorate symptoms of premenstrual syndrome in women. The administration of steroid hormones has been suggested to modulate the effects of caffeine, but unknown is whether endogenous hormone varia

Diazepam has been broadly accepted as an anxiolytic drug and is often used as a positive control in behavioral experiments with mice. However, as opposed to this general assumption, the effect of diazepam on mouse behavior can be considered rather controversial from an evidence point of view. Here we revisit this issue by studying the effect of diazepam on a benchmark task in the preclinical anxiety literature: the elevated plus maze. We evaluated the minute-by-minute time-course of the diazepam effect along the 10 min of the task at three different doses (0.5, 1 and 2 mg/kg i.p. 30 min before

We examined the activities of serotonin (5-HT) neurons in the dorsal raphe nucleus (DRN) and corticotropin-releasing factor (CRF) neurons in the hypothalamic paraventricular nucleus (PVN) during acute treadmill running at different speeds (control, low, high) and durations (15, 30, 60 min) in male Wistar rats using c-Fos/5-HT or CRF immunohistochemistry. We also performed elevated plus maze test (EPM) and forced swim test (FST) after acute treadmill running in rats. Acute treadmill running at low speed, regardless of exercise duration, significantly increased c-Fos expression in 5-HT neurons i

This report evaluates the effects of chlordiazepoxide, a benzodiazepine commonly prescribed to manage anxiety-related disorders in adolescent/pediatric populations, on elevated plus maze (EPM) performance in juvenile mice. This approach was taken because chlordiazepoxide produces anxiolytic-like effects in multiple models in adult rodents, however, less is known about the behavioral effects of this benzodiazepine in juveniles. Thus, we administered a single intraperitoneal injection of chlordiazepoxide (0, 5, or 10 mg/kg) to postnatal day 35 male C57BL/6 mice. Thirty minutes later, mice were a

RATIONALE: Cannabidiol (CBD), the major non-psychoactive constituent of cannabis, has therapeutic potential for the treatment of anxiety. Most preclinical studies investigate only acute effects of CBD and only in males, yet the drug is most likely to be used over a sustained period in clinical practice.
OBJECTIVES: The objectives of this study were to investigate the anxiolytic-like effect of CBD in female rats compared to males and to determine whether the responsiveness of females was influenced by the stage of the estrous cycle.
METHODS: We carried out experiments to compare the effect of

While several pieces of evidence link obesity and mood disorders in menopause, the mechanisms involved are not yet fully understood. We have previously demonstrated that Ginkgo biloba extract (GbE) both attenuated diet-induced obesity of male rats and restored serotonin-induced hypophagia in ovariectomized female rats. The present study aimed at exploring whether GbE treatment ameliorates ovariectomy-related obesity and anxious/depressive-like behaviours. Wistar female rats were either ovariectomized (OVX) or sham-operated (Sham). After 2 months, either 500 mg/kg of GbE or vehicle were adminis

The elevated plus maze is a prominent and well-documented test for studying anxiety in rodents. Fish are becoming more prevalent in studies of anxiety, yet the elevated plus maze has not been adapted and validated for fish. In the present study, we created an aquatic version of the elevated plus maze called the 'submerged plus maze,' which is shaped like a plus symbol with four arms alternating between black and transparent walls. We used convict cichlid fish (Amatitlania nigrofasciata) and administered diazepam to validate the apparatus for studying anxiety-like behaviour. After diazepam expo

Special features of anxious behavior in the elevated plus maze test and acoustic startle response were analyzed in 11 inbred mouse strains. A significant influence of the genotype both on the startle amplitude and behavior in the elevated plus maze was found. However, analysis of covariance did not reveal a genotype-related association between anxiety and startle amplitude. The data indicates that the fear-induced acoustic startle response and anxious behavior in the elevated plus maze (agoraphobia) are not genetically related.

Anxiety- and stress-related disorders are commonly treated using combinations of centrally acting drugs. However, the behavioral outcomes of interactions between GABAergic and glutamatergic systems remain incompletely characterized. The present study investigated the effects of flumazenil, ketamine and midazolam, and their combinations, on anxiety-like behavior in adult male rats using aversive 22-kHz ultrasonic vocalization (AUV) and the elevated plus maze (EPM). Systemic administration of flumazenil (0.1-10 mg/kg, i.p.) produced a significant, dose-dependent reduction of AUV in Sprague Dawle

To investigate whether an elevated plus-maze consisting of two open and two closed arms could be used as a model of anxiety in the mouse, NIH Swiss mice were tested in the apparatus immediately after a holeboard test. Factor analysis of data from undrugged animals tested in the holeboard and plus-maze yielded three orthogonal factors interpreted as assessing anxiety, directed exploration and locomotion. Anxiolytic drugs (chlordiazepoxide, sodium pentobarbital and ethanol) increased the proportion of time spent on the open arms, and anxiogenic drugs (FG 7142, caffeine and picrotoxin) reduced th

OBJECTIVES: To evaluate hypnosedative action of Zopiclone by using animal models for hypnosis and sedation (anxiolysis); and to further evaluate whether this hypnosedation impairs memory-learning in albino mice like conventional hypnosedatives.
METHODS: For evaluation of hypnosedation, following experiments were performed in albino mice: (1) righting reflex test, (2) pentobarbitone sleeping time potentiation, (3) open field apparatus behavior, and (4) elevated plus maze performance. For evaluation of effects on impairment of memory-learning, elevated plus maze retention test was performed in

Prenatal stress and maternal separation are used in a large number of studies on early adversity consequences and present some similarities in their effects. The present work investigates the behavioral effects of these two procedures on two models of anxiety: the elevated plus-maze and the elevated T-maze. During pregnancy, female rats were submitted to uncontrollable electric foot shock sessions every other day or kept undisturbed. After delivery, litters from undisturbed dams were submitted to either 180-min daily periods of maternal separations from the 3-14th postnatal days or maintained

1. Different animal tests model different anxiety disorders. Thus, the social interaction test is a model of generalised anxiety disorder, plus-maze Trial 1 models elements of panic disorder and Trial 2 in the elevated plus-maze is a model of specific phobia. 2. Studies of the neuroanatomical and neurochemical pathways controlling behaviour in these different tests provides information on the neurobiological mechanisms modulating anxiety disorders. 3. In the social interaction test, nicotine and 8-OH-DPAT had anxiogenic effects when injected into the dorsal hippocampus or the lateral septum. 4

Anxiety is highly common, and stress is a major trigger for anxiety. Anxiety includes heightened threat assessment and avoidance, but we do not fully understand which components are sensitive to stress. Rodents show a balance of exploration and avoidance that incorporates threat assessment prior to making the relatively risky decision to explore an open area. The purpose of this study was to determine if stress impacts risk assessment and if this is tied to the effects of stress on exploration. The present study used elevated plus maze (EPM) to test the effects of repeated social defeat stress

Analyses of the behavioral reactions of rodents to predators have greatly contributed to the understanding of defense-related human psychopathologies such as anxiety and panic. We here investigated the behavioral consequences of exposing male Wistar rats to a live cat using the elevated T-maze test of anxiety. This test allows the measurement of two defensive responses: inhibitory avoidance and escape, which in terms of pathology have been associated with generalized anxiety and panic disorders, respectively. For comparative reasons, the effects of exposure to the cat were also assessed in the

BACKGROUND: Avoidance behavior is a hallmark of panic disorder (PD), yet experimental evidence remains limited due to challenges in assessing unconditioned avoidance without specific threats. The human Elevated Plus Maze (EPM), implemented in mixed reality, provides an ecologically valid paradigm to measure approach-avoidance behavior under controlled conditions.
METHODS: Twenty PD patients and 20 healthy controls (HC) were tested on the human EPM. Approach-avoidance behavior was assessed alongside subjective anxiety, tension, and cardiovascular responses across multiple time points. Hormonal
