Identification of Ellagic Acid as a Natural GPR35 Agonist for Ulcerative Colitis Therapy.

The escalating global burden of Ulcerative Colitis (UC) underscores the urgent need for novel therapeutic strategies. Although dietary modulation is known to influence UC progression, the specific molecular mediators remain largely undefined. Recently, the G protein coupled receptor 35 (GPR35) has emerged as a promising target for maintaining gut homeostasis and promoting intestinal epithelium repair. Yet, whether the therapeutic benefits of dietary polyphenols are mediated through the direct activation of GPR35 remains unexplored. Here, the NanoLuc Binary Technology (NanoBiT) assay was first

In Vitro Combination of Ascorbic and Ellagic Acids in Sperm Oxidative Damage Inhibition.

It is known that an altered redox balance interferes with normal spermatic functions. Exposure to genotoxic substances capable of producing oxidative stress (OS) can cause infertility in humans. The use of antioxidants to reduce oxidative stress contributes to the improvement in reproductive function. This study focused on an antigenotoxic evaluation of ellagic acid (EA) and ascorbic acid (AA) in combination against benzene genotoxic action on human spermatozoa in vitro. In addition to the evaluation of sperm parameters, damage in sperm genetic material and intracellular ROS quantification wer

[Studies on procedure for sample preparation of ellagic acid in several kinds of foodstuffs].

A sample preparation for ellagic acid in several kinds of foodstuffs, which is included in the List of Existing Food Additives as natural antioxidants in Japan, functioning as an antioxidant, was studied. The solid samples were refluxed with methanol, and then the extract was refined using a solid-phase cartridge. The liquid samples were directly pretreated by solid-phase extraction. On the other hand, test solutions for cooking oils and fats were inapplicable to solid-phase extraction in this work, because the recovery tests from samples spiked with ellagic acid gave low recoveries. Consequen

The antioxidant and anticonvulsant effects of ellagic acid in kainic acid-induced temporal lobe epilepsy in mice.

Oxidative stress (OS) resulting from high levels of free radicals contributes to the initiation and progression of epilepsy. Temporal lobe epilepsy (TLE) is associated with alteration in the structure and function of the hippocampus and is modeled in mice using kainic acid (KA). In this study, the neuroprotective effect of ellagic acid (EA) on KA-induced epilepsy in mice was evaluated. Sixty male Swiss albino mice were assigned to six groups: I received normal saline (NS; 10 ml/kg, intraperitoneally (i.p.)); II, received KA (15 mg/kg, i.p.); III, received diazepam (20 mg/kg, i.p.) and KA (15 m

Ellagic acid mitigates rotavirus-induced intestinal injury via bidirectional "immune-microbiota" regulatory effect.

INTRODUCTION: Rotavirus (RV) is a major cause of childhood gastroenteritis, leading to intestinal damage, inflammation, and gut microbiota dysbiosis. This study investigated whether ellagic acid (EA), a natural polyphenol, can alleviate RV-induced intestinal injury by modulating both host immunity and the gut microbiota. METHODS: In this study, RV was used to infect BALB/c suckling mouse models to explore whether ellagic acid could alleviate intestinal damage following rotavirus infection through bidirectional regulation of "immunity and microbiota". The viral load of RV, the expression level

Ellagic Acid prevents vascular dysfunction in small mesenteric arteries of ovariectomized hypertensive rats.

Cardiovascular diseases rank the top causes of death worldwide, with a substantial increase in women compared to men. Such increase can beexplained by the drastic decrease in 17-β-estradiol hormone during menopause and associated with endothelium-dependent vascular dysfunction. The current treatments for cardiovascular diseases (e.g., hypertension), are only palliative and therefore, feasible, non-invasive options for preventing further vascular damage are needed. The polyphenol ellagic acid (EA) has risen as a candidate with possible vascular protection properties. This study evaluated the ef

Presynaptic Release-Regulating Alpha2 Autoreceptors: Potential Molecular Target for Ellagic Acid Nutraceutical Properties.

Polyphenol ellagic acid (EA) possesses antioxidant, anti-inflammatory, anti-carcinogenic, anti-diabetic and cardio protection activities, making it an interesting multi-targeting profile. EA also controls the central nervous system (CNS), since it was proven to reduce the immobility time of mice in both the forced swimming and the tail-suspension tests, with an efficiency comparable to that of classic antidepressants. Interestingly, the anti-depressant-like effect was almost nulled by the concomitant administration of selective antagonists of the noradrenergic receptors, suggesting the involve

Ellagic acid facilitates indomethacin-induced gastric ulcer healing via COX-2 up-regulation.

The mechanism of indomethacin-induced gastric ulcer healing by ellagic acid (EA) in experimental mice model is described in our study. Ulcer index (UI) and myeloperoxidase (MPO) activity of the stomach tissues showed maximum ulceration on the third day after indomethacin (18 mg/kg, single dose) administration. Preliminary observation of UI and MPO activity suggests that EA possesses ulcer-healing activity. Other anti-ulcer parameters such as the levels of prostaglandin E(2), cyclooxygenase (COX) 1 and 2 enzymes, anti-inflammatory cytokines [interleukin (IL)-4 and -5], pro-angiogenic factors, e

Multifunctional bioactivity of ellagic acid and pomegranate oil: selective anticancer in melanoma models.

Melasma and skin cancer require effective and safe topical treatments. The aim of this study was to investigate the potential cytotoxic and antioxidant effects attributed to exposure to ellagic acid (EA) and pomegranate oil (PO) using human keratinocytes (HaCaT) and melanoma cell lines (A375 and B16F10). Cell viability assay (MTT) demonstrated that EA and PO (5% or 10%) were not cytotoxic to HaCaT cells at 24 hr, but induced significant time-dependent cytotoxicity in melanoma cells after 72 hr. In DCFH-DA assay, PO exhibited greater potential in mitigating production of reactive oxygen species

Dietary ellagic acid inhibiting gastrointestinal pathogens by modulation of microbiome-metabolite-immune axis.

Antibiotic-induced depletion of the gut microbiota facilitated the colonization of vancomycin-resistant Enterococci (VRE) in the gastrointestinal tract, and then increased patients' susceptibility to secondary infections. Ellagic acid, a major constituent of fruits and nuts, showed various bioactivities except for antibacterial. Interestingly, it promoted the recovery of gut microbiota, enhanced microbial diversity and stimulated the proliferation of probiotic gut microbes, and then ameliorated the overgrowth of pathogens in vivo in our experiment. Moreover, ellagic acid activated Gpr41 and Gp

Ellagic Acid Combined with Tacrolimus Showed Synergistic Cell Growth Inhibition in Fission Yeast.

Calcineurin (CN) is a conserved Ca2+-calmodulin activated protein phosphatase, which plays important roles in immune regulation, cardiac hypertrophy, and apoptosis in humans. In pathogenic fungi, CN is essential for stress survival, sexual development, and virulence. The immunosuppressant tacrolimus (FK506) is a specific inhibitor of CN in humans and fungi including nonpathogenic fission yeast. Although calcineurin inhibition by FK506 or CN deletion in fission yeast does not induce growth defects, treatment with some anti-fungal drugs such as micafungin and valproic acid, induced synthetic let

Glutamate metabolism reprogramming is involved in the inhibitory effect of ellagic acid on rotavirus replication.

INTRODUCTION: Rotavirus (RV) is an important zoonotic pathogen that causes acute diarrhea in infants and young animals. There are several limitations in the prevention and control strategies currently, which result in an urgent need to develop innovative natural active substances against RV. This study aims to explore the RV infection preventive mechanism of ellagic acid (EA) and determine whether it exerts its effect by regulating the glutamine-glutamate metabolic axis. METHODS: RV-infected Balb/c mice and IEC-6 cell models were established to conduct mechanistic research by employing untarg

Ellagic Acid as a Promising Antifungal Agent: A Review of Mechanisms, Synergy, and Formulation Strategies.

Ellagic acid (EA), a naturally occurring phenolic compound, has garnered significant interest as a potential antifungal agent owing to increasing fungal resistance and a scarce therapeutic pipeline. This review consolidates the evidence of the broad-spectrum activity of EA against critical priority pathogens, including Candida auris and Cryptococcus neoformans. We highlight its multi-target mechanisms of action, such as the impairment of cell wall integrity and plasma membrane disruption resulting from the inhibition of ergosterol biosynthesis, and inhibition of key enzymes, such as laccase.
