Mutation of the imprinted gene OsEMF2a induces autonomous endosperm development and delayed cellularization in rice.

In angiosperms, endosperm development comprises a series of developmental transitions controlled by genetic and epigenetic mechanisms that are initiated after double fertilization. Polycomb repressive complex 2 (PRC2) is a key component of these mechanisms that mediate histone H3 lysine 27 trimethylation (H3K27me3); the action of PRC2 is well described in Arabidopsis thaliana but remains uncertain in cereals. In this study, we demonstrate that mutation of the rice (Oryza sativa) gene EMBRYONIC FLOWER2a (OsEMF2a), encoding a zinc-finger containing component of PRC2, causes an autonomous endospe

Atypical E2F transcription factors couple Polycomb Repressive Complex 2 to endosperm cell division and differentiation.

Flowering plant endosperm and mammalian placenta represent convergent evolutionary products sharing conserved developmental programmes, embryonic nutrient transport and genomic imprinting. Polycomb Repressive Complex 2 (PRC2)-mediated H3K27me3 silencing restricts precocious tissue overproliferation in both lineages, yet the underlying molecular mechanism remains undefined. Here we show that mutations in ancestral maize ZmFIE2, not its derived paralogue ZmFIE1, abrogates global H3K27me3 levels, resulting in aberrant cell proliferation, impaired differentiation in early endosperm and seed lethal

Endosperm-specific expression of a yeast thiamin transporter enhances vitamin B1 content in white rice.

Vitamin B1 is a vital co-factor in cellular metabolism, but it must be obtained through the diet in humans. Polished (white) rice, a dietary staple for much of the global population, contains very low levels of vitamin B1, and this contributes to widespread thiamin deficiency in regions that rely heavily on rice. To address this issue, we engineered rice to express the Saccharomyces cerevisiae thiamin transporter gene THI7 under the control of the rice endosperm-specific GLUTELIN1 (GT1) promoter. In transgenic lines with moderate-to-high THI7 expression, we found significantly increased thiami

Sorbitol dehydrogenase aids grain-fill and integrates carbohydrate metabolism with redox in the hypoxic maize endosperm.

Extreme low oxygen is typical of microenvironments inside developing grains, thus requiring metabolic adaptations to hypoxia. Potential contributions from sorbitol dehydrogenase (SDH) are tested here for maize (Zea mays) since the reversible reaction (fructose + NADH ↔ sorbitol + NAD+) can balance both redox state and metabolism during assimilate import into kernels. To do so, we developed and analyzed maize sdh1 mutants and over-expression lines. Dysfunction of Sdh1 decreased seed weight by 17%, impaired starch accumulation, and enhanced levels of glucose, fructose, and sucrose. Redox balance

NMR spectroscopy and chemical studies of an arabinan-rich system from the endosperm of the seed of Gleditsia triacanthos.

Exhaustive extraction of the endosperm from the seed of Gleditsia triacanthos using water at room temperature and 50 degrees C left a residue, which was further extracted at 95 degrees C. Precipitation of this extract with 2-propanol yielded major amounts of galactomannan components, while the supernatant was mainly composed of arabinose-rich constituents. Two fractions were obtained by anion-exchange chromatography. The fraction that eluted with water is an arabinan with (1-->5) alpha-L linkages and branching mainly on C-2, accompanied with equal amounts of a low-galactose galactomannan oligo

HvPHO1 Controls a Glucose 1-Phosphate-SnRK1 Signalling Module That Coordinates Starch Accumulation in Barley Endosperm.

Developing barley grains must balance the use of imported sucrose between soluble sugars and storage starch, but how this balance is controlled remains unclear. Here we show that loss of the plastidial α-glucan phosphorylase HvPHO1 causes starch and soluble sugars to accumulate concurrently in barley grains. Developing pho1 grains showed sustained increases in glucose 1-phosphate (G1P) and adenosine diphosphate glucose (ADP-glucose), and exogenous G1P also promoted starch accumulation, supporting a stimulatory effect of elevated G1P on starch synthesis. G1P directly bound sucrose non-fermentin

KIL transcription factors facilitate embryo growth in maize by promoting endosperm elimination via lytic cell death.

The endosperm is a transient nutritive tissue in plant seeds. During maize (Zea mays) grain development, 2 distinct endosperm cell death processes occur: in 1 process, the endosperm adjacent to the embryo scutellum (EAS) is completely dismantled; in the other, the starchy endosperm (SE) retains nutrient-packed cell corpses after grain filling. Here, we show that SE cell death degrades some organelles including the mitochondria and the endoplasmic reticulum, while preserving protein bodies, starch granules, and chromatin. In contrast, EAS cells undergo lytic cell death to remobilize stored nutr

A wheat-specific proline-rich protein regulates grain weight through a ubiquitin-mediated delay of endosperm cellularization.

Grain weight is a key determinant of wheat yield, yet the developmental mechanisms that define this trait remain poorly understood. Here, we identify TaPGW1-A1, a wheat-specific proline-rich gene that regulates grain weight. TaPGW1-A1 is a recently evolved gene found only in wheat. It is highly expressed in early developing grains, and its overexpression delays endosperm cellularization and prolongs the syncytial phase, resulting in an 8% increase in field-based grain weight. TaPGW1-A1 protein directly interacts with the ubiquitin receptor TaDSK2a (Dominant Suppressor of KAR2 homolog) via its

Maize ATR safeguards genome stability during kernel development to prevent early endosperm endocycle onset and cell death.

The ataxia-telangiectasia mutated (ATM) and ATM and Rad3-related (ATR) kinases coordinate the DNA damage response. The roles described for Arabidopsis thaliana ATR and ATM are assumed to be conserved over other plant species, but molecular evidence is scarce. Here, we demonstrate that the functions of ATR and ATM are only partially conserved between Arabidopsis and maize (Zea mays). In both species, ATR and ATM play a key role in DNA repair and cell cycle checkpoint activation, but whereas Arabidopsis plants do not suffer from the absence of ATR under control growth conditions, maize mutant pl

Functional delineation of rice MADS29 reveals its role in embryo and endosperm development by affecting hormone homeostasis.

Rice MADS29 has recently been reported to cause programmed cell death of maternal tissues, the nucellus, and the nucellar projection during early stages of seed development. However, analyses involving OsMADS29 protein expression domains and characterization of OsMADS29 gain-of-function and knockdown phenotypes revealed novel aspects of its function in maintaining hormone homeostasis, which may have a role in the development of embryo and plastid differentiation and starch filling in endosperm cells. The MADS29 transcripts accumulated to high levels soon after fertilization; however, protein a

The ZmmiR169o/ZmNF-YA13 module orchestrates maize kernel texture by fine-tuning starch biosynthesis and endosperm architecture.

BACKGROUND: Kernel texture is a key agronomic trait determining grain quality and storage stability in maize. The ratio of vitreous endosperm is the principal metric for kernel texture, and elucidating the key genes governing its formation is vital for maize breeding. Although microRNAs (miRNAs) play important regulatory roles in seed development, their functions in controlling grain quality traits, especially endosperm texture, remain poorly understood. RESULTS: Here, we systematically dissect the regulatory role of the ZmmiR169o/ZmNF-YA13 module in maize kernel texture. Our results show tha

Molecular Characterization of capulet2 reveals the importance of ANAPHASE PROMOTING COMPLEX 6 maternal expression in Endosperm Development.

Flowering plants are characterized by a double fertilization event, and the fertilized female gametes develop into the endosperm and embryo. Genomic imprinting promotes parental allele-specific gene expression in the endosperm by epigenetic modifications such as DNA methylation. Similarly, gametophyte maternal effects influence gene function in the female gametophyte that affects development of the endosperm and embryo post-fertilization. While many imprinted genes do not display a seed phenotype upon mutation, gametophyte maternal effect mutants are characterized by distorted seed development

RNA-Binding Protein RBP-P Is Required for Glutelin and Prolamine mRNA Localization in Rice Endosperm Cells.

In developing rice (Oryza sativa) endosperm, mRNAs of the major storage proteins, glutelin and prolamine, are transported and anchored to distinct subdomains of the cortical endoplasmic reticulum. RNA binding protein RBP-P binds to both glutelin and prolamine mRNAs, suggesting a role in some aspect of their RNA metabolism. Here, we show that rice lines expressing mutant RBP-P mislocalize both glutelin and prolamine mRNAs. Different mutant RBP-P proteins exhibited varying degrees of reduced RNA binding and/or protein-protein interaction properties, which may account for the mislocalization of s

Bioengineered 'golden' indica rice cultivars with beta-carotene metabolism in the endosperm with hygromycin and mannose selection systems.

Vitamin-A deficiency (VAD) is a major malnutrition problem in South Asia, where indica rice is the staple food. Indica-type rice varieties feed more than 2 billion people. Hence, we introduced a combination of transgenes using the biolistic system of transformation enabling biosynthesis of provitamin A in the endosperm of several indica rice cultivars adapted to diverse ecosystems of different countries. The rice seed-specific glutelin promoter (Gt-1 P) was used to drive the expression of phytoene synthase (psy), while lycopene beta-cyclase (lcy) and phytoene desaturase (crtI), fused to the tr

Polycomb protein ZmEMF1a restricts endosperm proliferation and directs differentiation via stage-specific H2Aub1 and H3K27me3 landscapes in maize.

Polycomb group (PcG) proteins serve as pivotal epigenetic repressors that govern the transcriptional programs underlying cell growth and differentiation. However, their functional roles in maize endosperm remain largely unexplored. Here, we characterize the recessive maize small-kernel mutant sks1, which exhibits persistent endosperm cell hyperproliferation and compromised cell expansion during grain filling. Map-based cloning reveals that SKS1 encodes ZmEMF1a, a PcG protein that physically interacts with subunits of both PRC1 and PRC2. Integrated ChIP-seq and RNA-seq analyses were performed t

Characterization of rice endosperm-derived antidepressant-like peptide (REAP): An orally active novel tridecapeptide derived from rice protein.

It is ideal to ingest bioactive substances from daily foods to stay healthy. Rice is the staple food for almost half of the human population. We found that an orally administered enzymatic digest of rice endosperm protein exhibits antidepressant-like effects in the tail suspension test (TST) using mice. A comprehensive peptide analysis of the digest using liquid chromatography-tandem mass spectrometry was performed, and a tridecapeptide QQFLPEGQSQSQK, detected in the digest, was chemosynthesized. Oral administration of the tridecapeptide exhibited antidepressant-like effects at a low dose comp

Corn grain endosperm type and brown midrib 3 corn silage: feeding behavior and milk yield of lactating cows.

Interactions of endosperm type of corn grain and the brown midrib 3 mutation (bm3) in corn silage on feeding behavior, productivity, energy balance, and plasma metabolites of lactating dairy cows were evaluated. Eight ruminally and duodenally cannulated cows (72 +/- 8 d in milk; mean +/- SD) were used in a duplicated 4 x 4 Latin square design experiment with a 2 x 2 factorial arrangement of treatments. Treatments were corn grain endosperm type (floury or vitreous), and corn silage type (bm3 or isogenic control). Diets contained 26% neutral detergent fiber (NDF) and 30% starch. Floury endosperm
