A lipid nanoparticle encapsulated CPA-CTD mRNA vaccine provides protection against Clostridium perfringens-driven diseases.

INTRODUCTION: Clostridium perfringens (C. perfringens), a ubiquitous Gram-positive bacterium in the environment and mammalian gut flora, is a leading cause of enterotoxemia in animals, necrotizing enteritis in humans and animals, and gas gangrene in both, attributed to its diverse exotoxin profile. Alpha-toxin, a pivotal virulence factor produced by all C. perfringens serotypes, plays a central role in the pathogenicity of these diseases. METHODS: Here, we engineered a lipid nanoparticle encapsulated CPA-CTD mRNA vaccine targeting the conserved C-terminal domain of C. perfringens alpha-toxin

Etx-Y71A as a non-toxic mutant of Clostridium perfringens epsilon toxin induces protective immunity in mice and sheep.

Epsilon toxin (Etx) is an extremely potent toxin produced by Clostridium perfringens toxinotypes B and D, which cause fatal enterotoxemia in many livestock species, mainly sheep and goats. Our previous study demonstrated that the aromatic amino acid (AA) residue at position 71 in domain III of Etx is needed for its cytotoxic activity toward MDCK cells. Here, we first determined that Etx mutants with non-aromatic AA substitutions at Tyr71 lost lethality in mice, indicating that the aromatic AA residue at position 71 is a toxicity determinant of Etx in vivo. After intravenous injection with a hi

Bacillary hemoglobinuria in a free-ranging elk calf.

A dead elk (Cervus elaphus roosevelti) calf was diagnosed with bacillary hemoglobinuria, a toxemia caused by the bacterium Clostridium haemolyticum. The mortality occurred in southwest Washington, USA (46 degrees 13'N, 123 degrees 22'W), in an area in which several previous mortalities, suspected but not conclusively diagnosed to be either bacillary hemoglobinuria, enterotoxemia, or leptospirosis, occurred. This is the first reported incidence of mortality attributable to bacillary hemoglobinuria in free-ranging elk. Similar deaths of young elk in the area suggest that mortality from this dise

Dairy calf and replacement heifer mortality on a single intensively managed dairy farm in Jordan: A 3-year-long study (2016-2018).

BACKGROUND: Pre-weaning dairy calf and replacement heifer mortality represents significant economic loss, limits genetic improvement and growth of the herd, and indicates poor management and animal welfare status on the farm. AIM: Currently, the rates and causes of the dairy calf and replacement heifer mortality in Jordan are not known. Therefore, the objective of this study was to determine the incidence rates and causes of mortality of pre-weaning calves and replacement heifers in Jordan. In addition, the age and seasonal distribution of mortality are determined in the study. METHODS: Data

[Progress in research of Clostridium perfringens toxin].

Clostridium perfringens can produce many kinds of toxins and hydrolase, causing gas gangrene, enteritis and enterotoxemia in both human and animals. It is known that C. perfringens can produce more than 20 toxins and hydrolases. The different toxin types are associated with specific disease types. At present, molecular toxin-typing method by PCR has replaced the traditional serological typing method. In this study, we systematically summarize the types, basic characteristics, pathogenic mechanism and the relationship with disease of C. perfringens toxins to provide evidence for the establishme

Clostridium perfringens in animal disease: a review of current knowledge.

The diseases caused by various types of Clostridium perfringens are critically reviewed in the light of current knowledge. Particular emphasis is placed on information concerning these diseases in Canadian livestock. There are two etiologically clearly-defined acute C. perfringens diseases recognized in Canada: hemorrhagic enteritis of the new born calf, caused by C. perfringens type C, and enterotoxemia of sheep, caused by type D. Clostridium perfringens type A may play a role as a secondary pathological agent in various disease conditions, such as necrotic enteritis of chickens. It may also

[Toxins of Clostridium perfringens].

Clostridium perfringens is an anaerobic gram-positive spore-forming bacillus. It is one of the pathogens with larger distribution in the environment; it can be isolated from soil and water samples, which also belongs to the intestinal flora of animals and humans. However, on some occasions it can act as an opportunistic pathogen, causing diseases such as gas gangrene, enterotoxemia in sheep and goats and lamb dysentery, among others. In human beings, it is associated to diseases such as food poisoning, necrotic enterocolitis of the infant and necrotic enteritis or pigbel in Papua-New Guinea tr

Enterocolitis in goats associated with enterotoxaemia in the perspective of two toxins: Epsilon toxin and beta-2 toxin - An immunohistochemical and molecular study.

Caprine intestinal diseases associated with clostridia are generally caused by Cpa and Etx encoded alpha (α) and epsilon (ε) toxinotypes of Clostridium perfringens type D respectively. A recent study on goat enterocolitis, demonstrated that the incidence of Clostridium perfringens type-D toxinotype and beta 2 toxins is high, suggesting its role in enterocolitis and many other diseases of goats affecting intestinal tract. Considering this scenario, the present prevalence study was planned to screen the goat intestinal tissues for the presence of the epsilon toxin and beta 2 toxin. Tissue sectio

Vaccine Production to Protect Animals Against Pathogenic Clostridia.

Clostridium is a broad genus of anaerobic, spore-forming, rod-shaped, Gram-positive bacteria that can be found in different environments all around the world. The genus includes human and animal pathogens that produce potent exotoxins that cause rapid and potentially fatal diseases responsible for countless human casualties and billion-dollar annual loss to the agricultural sector. Diseases include botulism, tetanus, enterotoxemia, gas gangrene, necrotic enteritis, pseudomembranous colitis, blackleg, and black disease, which are caused by pathogenic Clostridium. Due to their ability to sporula

The pharmacological profile of mouse hind paw inflammation induced by staphylococcal enterotoxin type A.

OBJECTIVE AND DESIGN: The present paper examines the possible pharmacological mediators involved in mouse hind paw inflammation induced by staphylococcal enterotoxin type A (SEA). MATERIALS AND METHODS: The edema and the Evans blue exudation were measured in male Swiss mice (20-25 g) using methods described by Levy and Griswold, respectively. RESULTS: SEA (32 micrograms/paw) produced a biphasic, long-lasting, dose- and time-dependent edematogenic response. The acute phase edema was pronounced while the chronic edema was of a low intensity. Exudate was the principal component of the edema. Th

An outbreak of enterotoxaemia in suckling camels.

An outbreak of enterotoxaemia was observed for the first time in suckling camels in Saudi Arabia. The animals were weak, diarrhoeic and succumbed quickly to exertion. The main pathological findings were those of acute catarrhal enteritis and acute myocardial degeneration. Clostridium perfringens was isolated from the enteric lesions; Aeromonas hydrophila was also identified. The properties of both isolates were studied.
