Set1 is a conserved histone H3 lysine 4 (H3K4) methyltransferase that exists as a multisubunit complex. Although H3K4 methylation is located on many actively transcribed genes, few studies have established a direct connection showing that loss of Set1 and H3K4 methylation results in a phenotype caused by disruption of gene expression. In this study, we determined that cells lacking Set1 or Set1 complex members that disrupt H3K4 methylation have a growth defect when grown in the presence of the antifungal drug Brefeldin A (BFA), indicating that H3K4 methylation is needed for BFA resistance. Coccidioidomycosis is a systemic mycosis caused by the dimorphic fungi Coccidioides spp. The treatment for chronic and/or disseminated coccidioidomycosis can be prolonged and complicated. Therefore, the search for new drugs is necessary. Farnesol is a precursor in the sterol biosynthesis pathway that has been shown to present antifungal activity. Thus, the objective of this study was to evaluate the in vitro antifungal activity of farnesol alone and in combination with antifungal agents against clinical and environmental strains of Coccidioides posadasii as well as to determine their effect on

Giant vesicles formed of 1,2-dipalmitoylphosphatidylcholine (DPPC) and sterols (cholesterol or ergosterol) in water and water/ethanol solutions have been used to examine the effect of sterol composition and ethanol concentration on the area compressibility modulus (K(a)), overall mechanical behavior, vesicle morphology, and induction of lipid alkyl chain interdigitation. Our results from micropipette aspiration suggest that cholesterol and ergosterol impact the order and microstructure of the gel (L(beta)') phase DPPC membrane. At low concentration (10-15 mol%) these sterols disrupt the long-r

Rearranged steroids featuring contracted or modified B-ring architectures, such as the unprecedented 6/5/6/5-fused tetrahydrofuran framework in calvatianone, represent challenging targets within the rigid steroidal scaffold. Herein, we describe a unified synthetic strategy enabling efficient access to calvatianone (1) and herbarulide (2), as well as the first synthesis of (22E,24R)-3β,5α-dihydroxy-ergosta-7,22-dien-6,5-olide (3) and its C5-epimer from readily available ergosterol. The key feature of this approach is a nonradical C5-C6 oxidative cleavage enabled by protecting-group modulation,

Leishmania protozoan parasites (Trypanosomatidae family) are the causative agents of cutaneous, mucocutaneous and visceral leishmaniasis worldwide. While these diseases are associated with significant morbidity and mortality, there are few adequate treatments available. Sterol 14alpha-demethylase (CYP51) in the parasite sterol biosynthesis pathway has been the focus of considerable interest as a novel drug target in Leishmania. However, its essentiality in Leishmania donovani has yet to be determined. Here, we use a dual biological and pharmacological approach to demonstrate that CYP51 is indi

This study was undertaken to study the antifungal activity of Pleurotus cystidiosus against Colletotrichum gloeosporioides. This was achieved by fractionating the mushroom, P. cystidiosus initially to acetone (A), dichloromethane (D), and hexane (H) and studying the antifungal activity using the standard poisoned food technique. All the test solutions used were in the concentration of 20,000 ppm. The percentage inhibition of extracts A, D, and H was 12, 7, and 0.4%, respectively. Antifungal assay guided fractionation of the most active extract A resulted in four fractions; A1, A2, A3, and A4 h

A series of novel ergosterol pyridine derivatives were synthesized via a three-component reaction and their antiproliferative activity was preliminarily evaluated against two human cancer cell lines. Structure-activity relationship (SAR) studies revealed that the pyridine ring is more favorable than the phenyl ring for enhancing biological activity. Among them, Compound 5 g displayed the most potent antiproliferative activity against MDA-MB-231 cells (IC50 = 9.28 μM), surpassing that of ergosterol (IC50 > 32 μM). Further mechanistic studies revealed that compound 5 g inhibited colony formation

The increasing antifungal resistance in Candida species highlights the critical need for new medications with unique mechanisms and antibiofilm properties. In this study, indole-thiosemicarbazide compounds were designed, synthesized, and evaluated against both sensitive and resistant strains of Candida. Compounds 3b, 3d, and 3i were most potent and among them, 3d and 3i showed strong activity against C. albicans, with MICs of 2.44 μg/mL, similar to fluconazole's 2.0 μg/mL. Compound 3b had broad-spectrum activity with MICs of 4.88 μg/mL against C. albicans, C. tropicalis, and C. auris. These co

BACKGROUND: Trichophyton rubrum, among other dermatophytes, is a major causative agent for superficial dermatomycoses like onychomycosis and tinea pedis, especially among pediatric and geriatric populations. Ellagic acid (EA) and shikonin (SK) have been reported to have many bioactivities, including antifungal activity. However, the mechanism of EA and SK on Trichophyton rubrum has not yet been reported.
OBJECTIVES: The purposes of this study were to evaluate the antifungal activities of EA and SK against Trichophyton rubrum and to illuminate the underlying action mechanisms.
METHODS: The ef

OBJECTIVES: Although berberine (BBR) is reported to exhibit weak activity against Candida albicans, combined use of BBR and fluconazole (FLC) showed synergism against FLC-resistant C. albicans in vitro. The aim of this study was to explore the synergistic antifungal mechanism of FLC and BBR in Saccharomyces cerevisiae, a typical fungal cell model.
METHODS: Biochemical and genetic analyses were performed to investigate the probable antifungal role of the combined use of BBR and FLC in S. cerevisiae.
RESULTS: FLC led to elevated cell membrane permeability in the wild-type S. cerevisiae BY4741,

INTRODUCTION: Naegleria fowleri is a pathogenic free-living amoeba that causes primary amoebic meningoencephalitis, a rare and difficult-to-diagnose form of meningitis. By testing patient cerebrospinal fluid (CSF) for ergosterol, a membrane component unique to fungi and amoeba, cases of primary amoebic meningoencephalitis may be distinguished from cases of bacterial and viral meningitis.
METHODS: Pooled CSF was fortified with ergosterol, extracted, filtered, and measured by liquid chromatography-tandem mass spectrometry. Measurement method precision, sensitivity, recovery, and stability were

Ergosterol has traditionally been used as a proxy to estimate fungal biomass as it is almost exclusively found in fungal lipid membranes. Ergosterol determination has been mostly used for fungal samples from terrestrial, freshwater, salt marsh- and mangrove-dominated environments or to describe fungal degradation of plant matter. In the open ocean, however, the expected concentrations of ergosterol are orders of magnitude lower than in terrestrial or macrophyte-dominated coastal systems. Consequently, the fungal biomass in the open ocean remains largely unknown. Recent evidence based on micros

Superficial mycoses caused by Trichophyton rubrum are among the most common infections worldwide. T. rubrum infections are difficult to treat and are often associated with recurrences after interruption of the antifungal therapy. Nevertheless, reports on T. rubrum resistance to commonly used antifungal drugs are rare. In this study, we compared the in vitro resistance frequencies and development of resistance to terbinafine, itraconazole, amorolfine, and ciclopirox in T. rubrum. Results demonstrated that naturally occurring mutants were isolated at a frequency of 10(-7) for itraconazole and 10

Aspergillus fumigatus is the most common airborne fungal pathogen for humans. In this mold, iron starvation induces production of the siderophore triacetylfusarinine C (TAFC). Here we demonstrate a link between TAFC and ergosterol biosynthetic pathways, which are both critical for virulence and treatment of fungal infections. Consistent with mevalonate being a limiting prerequisite for TAFC biosynthesis, we observed increased expression of 3-hydroxy-3-methyl-glutaryl (HMG)-CoA reductase (Hmg1) under iron starvation, reduced TAFC biosynthesis following lovastatin-mediated Hmg1 inhibition, and i

Concerns with fungal plant pathogen resistance to fungicides require sustainable alternatives. Model yeast lipid membranes were used to assess the role of ergosterol in the activity of antimicrobials known to affect biomembranes. Results showed that nystatin decreased yeast liposome stability as ergosterol content increased, whereas iturin effects were consistently high, but did not appear to be affected by ergosterol content. Nisin tended to decrease yeast liposome stability as ergosterol content increased, albeit much less than nystatin, and caused increased liposome size at lower ergosterol

Mushrooms are a rich source of steroidal compounds and have attracted attention from scientists for long. Recently, we found that several naturally occurring steroidal compounds including chaxine B (1) could be generated non-enzymatically from ergosterol, a major component of the fungal cell membrane. Here, we conducted further analyses to determine that the reactions require radical intermediates by using the radical trap TEMPO but proceed without light or heat. Steric hindrance led to the formation of only a limited number of adducts. We then conducted computational analyses of the reaction

The increasing fungicide resistance in pathogenic fungi presents a serious challenge to global agricultural production and food security. Phytopathogens like Penicillium italicum and Colletotrichum musae are great threats to economically important crop production. Citrus, a globally important fruit crop, is highly susceptible to post-harvest infections by P. italicum, while the huge post-harvest losses of bananas (Musa spp.) suffer from anthracnose caused by C. musae. In this study, aspergopyranoid A (1), a new γ-pyrone-ergosterol hybrid with a 6/6/6/6/5/6 hexacyclic skeleton, was isolated and

Candida albicans, the most common opportunistic pathogenic fungus, is also the main pathogenic organism for oral candidiasis. This condition is particularly prevalent among the elderly, children, and individuals undergoing radiotherapy or suffering from HIV. The lack of new antifungal drugs, and drug resistance coupled with the side effects of current antifungal agents have increased the challenges of clinical antifungal therapies. Polyenes, including amphotericin B and nystatin, are clinical fungicidal drugs, however, their side effects and low solubility have limited their clinical applicati

Increasing the targeting ability of antifungal proteins towards specific components of fungal cells has the potential to improve their antifungal activity and reduce harmful effects to nontarget cells. To obtain effective disease resistance genes against cotton Verticillium wilt, we constructed several fusion genes, in which binding domains targeting chitin, sphingolipid or ergosterol in the fungal cell wall or cell membrane were individually fused to the antifungal peptide BbAFP1 from entomopathogenic fungus Beauveria bassiana. Transient expression of fusion genes in cotton cotyledons indicat
