Peritoneal Inflammation in PD-Related Peritonitis Induces Systemic Eryptosis: In Vitro and In Vivo Assessments.

Erythrocytes (RBCs) have a highly specialized and organized membrane structure and undergo programmed cell death, known as eryptosis. Our preliminary data show a significant increase in the eryptosis during peritoneal dialysis (PD)-associated peritonitis. The objectives of the present study were assessment of the incrementation of eryptosis in PD patients with peritonitis, evaluation of the relationship between systemic eryptosis in peritonitis and specific peritonitis biomarkers in PD effluent (PDE), and confirmation of the induction of eryptosis by peritonitis in a vitro setting. We enrolled

Metabolic Exhaustion, p38 MAPK/Rac1 GTPase/MLKL Signaling, and Ca2+ Influx Mediate Solasodine-Triggered Eryptosis.

Solasodine (SOL) is a steroidal glycoalkaloid that shows a wide range of biological effects, most notably anticancer activities. Eryptosis is the programmed cell death of erythrocytes that leads to anemia. Specifically, chemotherapy-induced anemia can be precipitated in part by eryptosis triggered by anticancer agents. We hypothesize that the cytotoxic effects of SOL observed in cancer cells are nonspecific and extend to human erythrocytes. Eryptosis following SOL exposure was detected by fluorescence-assorted cell sorting analysis in erythrocytes from healthy volunteers. Phosphatidylserine (P

Ferutinin induces in vitro eryptosis/erythroptosis in human erythrocytes through membrane permeabilization and calcium influx.

Ferutinin, isolated from the root of Ferula hermonis and proposed to be used as an antiosteoporosis phytoestrogen, has death promoting activities in a number of cancer cells. However, the effect of ferutinin on the induction of apoptosis in human red blood cells (RBCs), also known as eryptosis or erythroptosis, remains unclear. Given that ferutinin is a small molecule that can induce apoptosis in the cancer cells by opening the mitochondrial permeability transition pores, we therefore hypothesized that the effect of ferutinin to elicit apoptosis in human RBCs devoid of mitochondria would be mi

The CILLO-E Hypothesis: Erythrocyte-Driven Acidosis and Early Eryptosis as Drivers of Cancer-Associated Anemia.

BACKGROUND: The tumor microenvironment (TME) is characterized by high lactate and proton accumulation resulting from glycolytic metabolism. While acidosis is known to influence immune and stromal cells, its direct effects on erythrocytes-the most abundant circulating cells-remain underexplored. METHODS: An integrative review of cancer metabolism, erythrocyte physiology, and lactate transport systems was conducted using PubMed and Web of Science. From this synthesis, the CILLO-E hypothesis (Cancer-Induced Lactate Load on Erythrocytes) was formulated. RESULTS: The hypothesis proposes that lact

The Prognostic Value of Eryptosis Parameters in the Cerebrospinal Fluid for Cerebral Vasospasm and Delayed Cerebral Ischemia Formation.

BACKGROUND: Delayed cerebral ischemia (DCI) and cerebral vasospasm (VS.) contribute to poor outcomes in patients with aneurysmal subarachnoid hemorrhage (aSAH). The pathophysiology of DCI is not fully understood, and this has hindered the adoption of a uniform definition. Reliable diagnostic tests and effective evidence-based treatment are lacking. This study explored the possibility of using eryptosis parameters in the cerebrospinal fluid (CSF) as a marker for early detection of VS and DCI. METHODS: Twenty-one SAH patients were recruited and treated at Kharkiv Regional Hospital. The occurren

Stimulation of Eryptosis and Hemolysis by Adrenic Acid Involves Oxidative Stress, Calcium Elevation, and Metabolic Collapse.

Omega-6 polyunsaturated fats (ω-6 PUFAs) are vital for many physiological functions, but their impact on cardiovascular disease (CVD) risk is controversial. Eryptosis alters blood viscosity by providing a procoagulant surface and leads to anemia, which is a recognized risk factor for CVD. This study examines the toxic mechanisms of adrenic acid (ADR), an ω-6 PUFA enriched in inflammatory and oxidative conditions, in red blood cells (RBCs). Purified RBCs were prepared from healthy volunteers and treated with 10-100 μM of ADR for 24 h at 37 °C under various physiological conditions. Eryptotic ma

Anacardic acid, a histone acetyltransferase inhibitor, stimulates hemolysis and eryptosis through energy depletion and calcium accumulation.

Histone acetyltransferase (HAT) inhibitors are promising epigenetic anticancer therapeutics. However, challenges remain regarding tissue specificity and off-target toxicity. This study presents a preclinical safety assessment of the HAT inhibitor anacardic acid (AA) in erythrocytes, given that anaemia is a clinically significant complication of chemotherapy. Freshly isolated erythrocytes were cultured in vitro with and without 1-10 µmol L-1 of AA, then markers and mechanisms of cell death were examined by fluorescence-activated cell sorting, spectrophotometry, and potentiometry. Antiproliferative Wnt inhibitor wogonin prevents eryptosis following ionophoric challenge, hyperosmotic shock, oxidative stress, and metabolic deprivation.

Anemia is a common complication of chemotherapy and may arise due to premature or suicidal death of red blood cells (RBCs). Prevention of RBC death thus lends itself as a promising strategy to counteract anemia. Wogonin (WGN; 5,7-dihydroxy-8-methoxyflavone) is a Wnt inhibitor derived from Scutellaria baicalensis plant with potent cytotoxic and antitumor potential. However, the nature of interaction of WGN with human RBCs is unknown. RBCs from healthy participants were exposed to different hemolytic and eryptotic stimuli for 24 or 48 hr at 37°C in the presence and absence of 100 μM WGN. Calcium

Mitochondrial toxin betulinic acid induces in vitro eryptosis in human red blood cells through membrane permeabilization.

Betulinic acid (BA), a compound isolated from the bark of white birch (Betula pubescens), was reported to induce apoptosis in many types of cancer through mitochondrial dysfunction with low side effects in normal cells. Because of these features, BA is regarded as a potential anti-cancer agent. However, the effect of BA on the induction of cell death in human erythrocytes remains unknown. Given that BA is a mitochondrial toxin and mitochondria are the central cell death regulator, we hypothesized that BA is unable to elicit apoptosis (also known as eryptosis or erythroptosis) in human erythroc

Modulation of Host Cell Death Signaling Platforms by Plasmodium falciparum: Implications for Eryptosis Regulation and Parasite Survival.

Programmed cell death pathways in Plasmodium falciparum remain conceptually fragmented. Over decades, researchers have applied metazoan apoptotic, autophagic, and necrotic markers to this deep-branching protozoan, frequently clashing with the reality that the parasite lacks the canonical genetic machinery (such as true caspases or death receptors) found in multicellular eukaryotes. In this work, we shift the focus from the parasite's disputed intrinsic death machinery to the host-parasite interaction arena: the active manipulation of the host erythrocyte's autonomous suicide program, eryptosis

Para-tertiary butyl catechol induces eryptosis in vitro via oxidative stress and hemoglobin leakage in human erythrocytes.

Exposure of human population to industrial chemicals is believed as a significant contributing factor to the outgrowth of occupational diseases especially in developing countries due to improper safety measures and sanitary conditions. Para-tertiary butylcatechol (PTBC) widely employed in petrochemical, thermofax and phototypesetting industries, induces melanocytotoxicity and contact dermatitis leading to occupational leukoderma/vitiligo. Few vitiligo patients were reported for oxidative stress-induced hemolytic anemia and thrombocytopenia, however its impact on blood components is still not c

Red blood cell in preeclampsia: attenuated nitric oxide generation and enhanced reactive oxygen species formation and eryptosis.

Preeclampsia (PE) pathogenesis is strongly related to diminished nitric oxide (NO) bioavailability and enhanced oxidative stress. Emerging evidence suggests that red blood cells (RBCs) eNOS enzyme contributes to systemic NO bioavailability by its ability of both NO and ROS generation. We aimed to investigate RBC eNOS enzyme activity, NO and ROS generation capacity, eryptosis index and aggregation levels in preeclamptic and uncomplicated pregnant women. Fifty-eight PE patients and 36 healthy pregnant women were included to the investigation. RBC eNOS enzyme activity, intracellular NO, calcium a

Dual protection by naringin and dexrazoxane against iron-induced erythrocyte damage: Exploring their synergistic regulation of ferroptosis and eryptosis.

Iron cellular accumulation is linked to ferroptosis, which induces cell injury. Conventional iron chelators often elicit side effects and encourage the exploration of naringin (NAR) as a potential protective agent against ferroptosis. Erythrocytes are a simple cellular model for exploring the beneficial effects of bioactive compounds. This study aims to investigate the protective effects of NAR against ferrous sulfate (FeSO4) induced ferroptosis. FeSO4 and hydrogen peroxide were applied to erythrocytes to induce ferroptosis either in the presence or absence of NAR, dexrazoxane (DEX), or their

Protective effects of selenium and vitamin E supplementation on eryptosis, oxidative stress, and inflammation in end-stage renal disease patients.

End-stage renal disease (ESRD) involves chronic microinflammation, oxidative stress, and increased red blood cell (RBC) destruction. This study examined their effects on erythrocyte integrity and oxidative stress in ESRD patients. Seventy ESRD patients were enrolled and received selenium (200 µg) and vitamin E (400 units) supplementation for 60 days. Phosphatidylserine exposure on RBCs was assessed to evaluate eryptosis. Serum levels of malondialdehyde (MDA), glutathione peroxidase (GPx) activity, ferritin, interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α), and iron (Fe) were measured.

Chloroquine induces eryptosis in P. falciparum-infected red blood cells and the release of extracellular vesicles with a unique protein profile.

INTRODUCTION: Malaria is a vector-borne parasitic disease that affects millions worldwide. To achieve the objective set by the World Health Organization of reducing malaria cases by 2030, antimalarial drugs with novel modes of action are required. Previously, a novel mechanism of action of chloroquine (CQ) was reported involving features of programmed cell death in the parasite, mainly characterized by calcium efflux from digestive vacuole permeabilization. Increased intracellular calcium induces suicidal death of erythrocytes, a process known as eryptosis. This study aimed to identify the hal

Quercetin protects red blood cells from aggregation, eryptosis, and delayed hemolysis caused by cell-free histones through sialic acid interaction.

Histones are nuclear proteins and play a vital role in regulating gene expression. Cell free histones in the circulation proved as damage-associated molecular patterns and were positively associated with various inflammatory disease conditions. Also, cell-free histones directly induce toxicity toward blood cells and are implicated in vascular dysfunction. However, cell-free histones are known to induce RBCs dysfunction in several histone-associated disorders, leading to hypoxia conditions. In this study, we aimed to understand the mechanism underlying the dysfunction of RBCs induced by histone

Linolenic acid stimulates eryptosis and hemolysis through oxidative stress and CK1α/MLKL: protective role of melatonin, urea, and polyethylene glycol.

Anticancer medications cause anemia in patients through ill-defined mechanisms, including hemolysis and eryptosis. Although α-linolenic acid (ALA) possesses anticancer properties against a variety of cancer cells, there is a dearth of evidence regarding how it modulates red blood cell (RBC) physiology. RBCs from healthy donors were subjected to anticancer concentrations of ALA (2.5, 5, 10, 20, 40, 80, and 100 μM) at 37 °C for 24 h, and colorimetric tests were used to determine hemolysis and acetylcholinesterase (AChE) activity. Meanwhile, flow cytometry was employed to identify eryptotic cells

Cell Death of P. vivax Blood Stages Occurs in Absence of Classical Apoptotic Events and Induces Eryptosis of Parasitized Host Cells.

Elucidation of pathways regulating parasite cell death is believed to contribute to identification of novel therapeutic targets for protozoan diseases, and in this context, apoptosis-like cell death has been reported in different groups of protozoa, in which metacaspases seem to play a role. In the genus Plasmodium, apoptotic markers have been detected in P. falciparum and P. berghei, and no study focusing on P. vivax cell death has been reported so far. In the present study, we investigated the susceptibility of P. vivax to undergo apoptotic cell death after incubating mature trophozoites wit

Complement inhibition by sutimlimab in cold agglutinin disease promotes red blood cell agglutination and may lead to circulatory complications, eryptosis and hemolysis.

BACKGROUND: Sutimlimab, an anti-C1s monoclonal antibody, is approved for the treatment of cold agglutinin disease (CAD). It prevents hemolysis by inhibiting the classical complement pathway. One of its off-target effects may be increasing circulatory complications that have not yet been characterized. METHODS: We report two patients with CAD who developed unusual widespread skin cyanosis and exacerbated acrocyanosis during sutimlimab treatment.
