Loa loa: development to the infective stage in an American deerfly, Chrysops atlanticus.

Chrysops atlanticus, an American species of deerfly collected along the Mississippi Gulf Coast, will support the development of the microfilaria of human Loa loa to the infective stage. Development takes place in the fat body of the fly and requires 9 to 10 days of development and pattern of morphogenesis of the parasite in C. atlanticus is virtually identical to that described in natural African vectors. Chrysops atlanticus will support the development of large numbers of L. loa to the infective stage without apparent ill effects. Two uninfected patas nonkeys each were given 75 third-stage la

Effects of atropine and pyridostigmine in heat-stressed patas monkeys.

The effects of a single intramuscular atropine injection (0.03 mg.kg-1) and a chronic oral pyridostigmine treatment (0.4 mg.kg-1, 3 times/day over a period of 7 d) on the thermoregulatory effector responses of unanesthetized patas monkeys were investigated using indirect calorimetry. The effects of atropine treatment on the thermoregulatory effector responses of patas monkeys exposed to 25 degrees and 35 degrees C were qualitatively similar but quantitatively greater at 35 degrees C. At 35 degrees C atropine decreased sweating (Esw) 52%, increased rectal temperature (Tre), mean skin temperatur

Pathology of gestational choriocarcinoma induced in patas monkeys by ethylnitrosourea given during pregnancy.

A rapidly fatal neoplastic disease with histological and clinical features resembling gestational choriocarcinoma in humans has been observed in patas monkeys. Timed pregnant females were given ethylnitrosourea (ENU) intravenously at doses of 0.1 to 0.4 mmol/kg body weight, beginning on day 30 of gestation and continuing weekly for a total of 12 injections. Of 59 monkeys given ENU during pregnancy, four of 12 subjected to the highest dose and three of the remaining 47 given lower doses died of choriocarcinoma within six months of cessation of ENU exposure. Death was usually caused by exsanguin

Intestinal parasite infection in non-human primates from The Gambia, West Africa, and their relationship to human activity.

In many areas of The Gambia, West Africa, population crowding in a degraded environment has forced close interactions of diurnal primate species with humans. We assessed intestinal parasitic infection prevalence and diversity in 4 diurnal non-human primate (NHP) species, Chlorocebus sabaeus, Erythrocebus patas, Papio papio and Piliocolobus badius across 13 sampling sites. The effect of human activity, determined by the human activity index, and NHP group size on parasite richness was assessed using a generalized linear mixed model (GLMM). The most common protozoa identified were Entamoeba coli

Effects of CSF ANG II and AVP on sweating in the heat-stressed patas monkey.

Increasing cerebrospinal fluid [Na+] reduces sweat rate (msw) in the heat-stressed patas monkey (Erythrocebus patas). This study determined the potential role of two neuropeptides, angiotensin II (ANG II) and arginine vasopressin (AVP), in mediating this response. Artificial cerebrospinal fluid, containing either ANG II or AVP, was infused into the third cerebral ventricle of lenperone-tranquilized monkeys (n = 4) exposed to 41 +/- 2 degrees C. Solutions were infused at 16.5 microliters/min for 25 min (total vol approximately 413 microliters). ANG II (1.25, 2.5, 5, and 10 ng/microliters) tende

Edema resulting from experimental filariasis.

Domestic cats and patas monkeys were infected with Brugia malayi so that the worms localized in the regional lymphatics of the hind legs. Reaction to the filarial parasites resulted in visible local edema in cats and disruption of normal lymph flow in the monkeys. Edematous tissue was examined grossly and by light and electron microscopy. Lymph flow patterns were examined by direct observation following injection of lymph staining dye and reflection of the skin, by X-ray following injection of radio-opaque contrast media, and by lymphscintigraphy after subcutaneous injection of radioisotopes.

Use of 99mTc-sulfur colloid to assess lymphatic dysfunction in filarial infection.

Investigations of lymphatic dysfunction in animals infected with filarial parasites has been hampered by a paucity of techniques to measure efficiency of lymphatic drainage. In this study a 99mTc-sulfur colloid technique was used to assess the efficiency of lymphatic drainage in Patas monkeys infected with filarial nematodes. In all 15 uninfected hind limbs there was rapid and consistent appearance of labeled colloid in the primary lymph node (popliteal) and subsequently in the secondary nodes (abdomino-pelvic) in 11 of 15 limbs. In contrast, in all eight limbs tested 1-9 months after infectio

Retinal detachment after choroidal ischemia.

Injection of a 15-micron microsphere suspension through one or two vortex veins of nine monkey eyes caused various degrees of sectorial choroidal ischemia, which were documented by fluorescein angiography and electron microscopy. The severity of the lesions to the fundus depended on the volume of microspheres injected (0.4 to 1.6 ml of a suspension of 600,000 microspheres/ml). Three hours after embolization white patches appeared in the retinal pigment epithelium as well as a posterior pole serous retinal detachment in five eyes. Delayed choroidal filling was noted in the quadrant involved, bu

Development and larval morphology of Loa loa in experimental primate hosts.

Experimental infections with Loa loa were established in laboratory primates by inoculation of third-stage larvae recovered from either Chrysops silacea infected after engorging on a human volunteer with loiasis, or Chrysops atlanticus infected after blood feeding on experimentally infected primates. The third molt occurred at 16 to 20 days postinoculation of infective larvae, and the fourth at about 50 days. Early larval development was characterized by rapid growth and marked differentiation of the reproductive systems. Differences in the body's growth rate between the sexes were seen as ear

The immune response to nematode parasites: modulation of mast cell numbers and function during Strongyloides stercoralis infections in nonhuman primates.

Mucosal mast cell numbers are modulated in the intestines of rodents during parasitic infections. These mast cells can degranulate in response to worm antigens, and this event has been suggested to play a protective role for the host. To examine whether mast cells in higher animals play a role in protecting from disseminated parasitic disease, mast cell numbers and responsiveness to parasite antigens were evaluated in 5 Erythrocebus patas infected with the human intestinal nematode Strongyloides stercoralis. Initial infection and subsequent challenge infections were associated with increase in

Epikeratophakia: the surgical correction of aphakia. II. Preliminary results in a non-human primate model.

We have developed a new form of refractive surgery, epikeratophakia, in which a disc of donor corneal tissue is lathed to a specific dioptric power and sutured on top of the recipient's cornea after the recipient epithelium has been removed. The optical correction obtained with epikeratophakia should be similar in quality to that of a contact lens, but would be permanent and would require no maintenance. A non-human primate model was used to test the predictability, stability, and clarity of epikeratophakia grafts. After surgery, the grafts did not always re-epithelialize readily; this problem

Molecular Detection and Genetic Diversity of Cytomegaloviruses and Lymphocryptoviruses in Free-Roaming and Captive African Green Monkeys (Chlorocebus sabaeus).

To date, limited information is available on cytomegalovirus (CMV) and lymphocryptovirus (LCV) from Chlorocebus monkeys. We report here high detection rates of herpesviruses in free-roaming African green monkeys (AGMs, Chlorocebus sabaeus) (26.4%, 23/87) and in captive AGMs (75%, 3/4) with respiratory disease on the Caribbean Island of St. Kitts. LCV (81.25%) was more prevalent than CMV (18.75%) in the AGMs. Applying a bigenic PCR approach (targeting DNA polymerase (DPOL) and glycoprotein B (gB) genes), long sequences were obtained from representative AGM CMV (KNA-SD6) and LCV (KNA-E4, -N6 and

Fetal patas monkeys sustain mitochondrial toxicity as a result of in utero zidovudine exposure.

Mitochondrial toxicity was examined in near-term fetuses of pregnant Erythrocebus patas monkeys given human equivalent doses of 3'-azido-3'deoxythymidine (AZT) during the second half of gestation. Pregnant monkeys were dosed daily with 10 or 40 mg AZT, equivalent to about 21% and 86% of the daily AZT dose (500 mg) given to HIV-1-positive pregnant women to prevent maternal-fetal virus transmission. The fetal tissues examined include heart and skeletal muscle, which have high energy requirements, and placenta, which is less dependent on mitochondrial integrity. Slot blot quantitation of mitochon

Imidazenil, a new anxiolytic and anticonvulsant drug, attenuates a benzodiazepine-induced cognition deficit in monkeys.

Imidazenil is a highly potent partial allosteric modulator of gamma-aminobutyric acid action at a great variety of gamma-aminobutyric acid(A) receptors, whereas alprazolam is a full allosteric modulator at these receptors. Data in rats indicate that imidazenil, like alprazolam, has pharmacological effects in animals indicative of anxiolytic, antipanic and anticonvulsant activity in humans, but unlike alprazolam, in animals imidazenil does not produce ataxia, sedation, tolerance or dependence nor does it potentiate the effects of ethanol. To extend the study of the imidazenil pharmacological pr

Investigation of a possible yellow fever epidemic and serosurvey for flavivirus infections in northern Cameroon, 1984.

A cluster of fatal hepatitis cases in northern Cameroon in 1984 stimulated a field investigation to rule out an epidemic of yellow fever. A serosurvey of villages in the extreme north of the country, in a Sudan savanna (SS) phytogeographical zone, disclosed no evidence of recent yellow fever infection. However, further south, in a Guinea savanna (GS) phytogeographical zone, serological evidence was found of endemic yellow fever virus transmission. The results indicate a potential for epidemic spread of yellow fever virus from the southern GS zone to the nothern SS zone of Cameroon, where immun

Two injections of the purified molecule with Muramyl-Di-Peptide (MDP) as adjuvant resulted in a high level antibody response in the five immunized animals and in a significant reduction in worm fecundity compared to the controls which received adjuvant alone. Mean levels of daily egg excretion in urine an faeces were reduced by respectively 55% and 74% although perfusion revealed tha
