Synergistic Enhancement of Paramylon Production in Edible Microalga Euglena gracilis via Ethanol-Guaiacol Co-Regulation.

Biomass-derived growth stimulants are widely recognized as green and economical solutions that can significantly enhance microalgae culture efficiency and optimize the biomanufacturing process of target products. In this paper, we investigated the effect of ethanol synergized with guaiacol (GA) on biomass and β-1,3 glucan accumulation in edible microalgae, namely Euglena gracilis. The ethanol-induced mixotrophic mode significantly increased biomass and paramylon production by 12.68 and 6.43 times, respectively, compared to the autotrophic control group. GA further exerted toxic excitatory effe

Euglena gracilis Suppresses Cold Symptoms in Healthy Individuals: A Double-Blind, Randomized, Placebo-Controlled Trial.

The immune system functions to eliminate pathogens, such as viruses, and regulate physiological responses and pathways. General immunity is affected by various factors, including fatigue, stress, and aging. A weakened immune system reduces the ability of the body to eliminate pathogens, thereby increasing the risk of infections, including the common cold. Euglena gracilis is a microalga rich in various nutrients, including β-1,3-glucan paramylon, which aids in maintaining and regulating immune function. In the present study, we aimed to investigate the effects of E. gracilis administration on

Euglena gracilis Z and its carbohydrate storage substance relieve arthritis symptoms by modulating Th17 immunity.

Euglena gracilis Z is a microorganism classified as a microalga and is used as a food or nutritional supplement. Paramylon, the carbohydrate storage substance of E. gracilis Z, is reported to affect the immunological system. This study evaluated the symptom-relieving effects of E. gracilis Z and paramylon in rheumatoid arthritis in a collagen-induced arthritis mouse model. The efficacy of both substances was assessed based on clinical arthritis signs, as well as cytokine (interleukin [IL]-17, IL-6, and interferon [IFN]-γ) levels in lymphoid tissues. Additionally, the knee joints were harvested

Mapping the proteome landscape in Euglena gracilis: global, phospho, and ubiquitin proteomes under anaerobic response.

The photosynthetic flagellate Euglena gracilis (Euglena) exhibits a remarkable metabolic flexibility, enabling the dynamic coordination of energy production and resource partitioning during anaerobic adaptation. While the wax ester synthesis under hypoxia is well-documented, the regulatory interplay between protein abundance and post-translational modifications (PTMs) remains unexplored. A definitive understanding of this metabolic remodeling is hindered by the lack of high-depth proteomic resources and the fragmented genome annotations, which fail to capture Euglena's proteomic diversity and

Oral administration of Euglena gracilis paramylon ameliorates chemotherapy-induced leukocytopenia and gut dysbiosis in mice.

Euglena gracilis (EUG) is a food supplement rich in beta-glucans, which are stored in the form of granules called paramylon. We determined whether EUG improved chemotherapy-induced leukocytopenia and dysbiosis. Mice were orally administered EUG prior to gemcitabine treatment. Analyses of the blood cell count, leukocyte population in the spleen, granulocyte/macrophage-colony-stimulating factor (GM-CSF) production by splenocytes, and fecal microbiome were conducted. The recovery of total leukocytes, neutrophils, and monocytes was accelerated after a single gemcitabine treatment. A more rapid lym

β-glucans from Euglena gracilis or Saccharomyces cerevisiae effects on immunity and inflammatory parameters in dogs.

Considering the differences in molecular structure and function, the effects of β-1,3-glucans from Euglena gracilis and β-1,3/1,6-glucans from Saccharomyces cerevisiae on immune and inflammatory activities in dogs were compared. Four diets were compared: control without β-glucans (CON), 0.15 mg/kg BW/day of β-1,3/1,6-glucans (Β-Y15), 0.15 mg/kg BW/day of β-1,3-glucans (Β-S15), and 0.30 mg/kg BW/day of β-1,3-glucans (Β-S30). Thirty-two healthy dogs (eight per diet) were organized in a block design. All animals were fed CON for a 42-day washout period and then sorted into one of four diets for 4

Elucidation of Enzymatic Routes Underlying Odd-Chain Fatty Acid Synthesis and Propionate Assimilation in Euglena gracilis.

Wax esters (WEs) produced by Euglena gracilis under anaerobic conditions contain significant proportions of odd-chain fatty acids and fatty alcohols (30%-40%). Propionyl-CoA, the primer for odd-chain fatty acid synthesis, is generally thought to be generated via the methylmalonyl-CoA pathway; however, the enzymatic basis of this pathway in E. gracilis remains unclear. Here, we generated knockout (KO) mutants of candidate enzymes in the methylmalonyl-CoA pathway-succinyl-CoA synthetase (SCS), methylmalonyl-CoA mutase (MCM), methylmalonyl-CoA epimerase (MCE), and propionyl-CoA carboxylase (PCC)-

Structural basis of diadinoxanthin-Chl a/b-binding proteins in the photosystem I supercomplex of Euglena gracilis.

Euglena gracilis is a phototrophic flagellate that has evolved through secondary endosymbiotic events and belongs to green-lineage organisms. We report a unique structure of photosystem I-light harvesting complex I (PSI-LHCI) supercomplex from E. gracilis at a 2.23-angstrom resolution by cryo-electron microscopy. The supercomplex is composed of 8 core subunits and 16 LHCIs and exhibits distinctive structural features compared to its counterparts in green algae and plants. The LHCI subunits encircle the core complex, forming a two-layered arrangement that comprises six pairs of tightly packed h

Selective Quenching of Peracetic Acid by Sodium Dithionite Enables Rapid, Non-Thermal Sterilization for Euglena gracilis Cultivation.

Peracetic acid (PAA) has strong biocidal activity against bacteria, fungi, and spores, even with short contact times. PAA-mediated sterilization is therefore an attractive method for sterilization of growth media that have heat-labile components or when polymer-based equipment is used. However, residual PAA and co-existing hydrogen peroxide (H2O2) can inhibit the growth of cultivated species, necessitating a fast and reliable quenching strategy that does not require rinsing. In contrast to Fe-EDTA-based catalytic decomposition that is strongly influenced by pH, buffers, and organic nitrogen, w

Chemical cross-linking and its effect on fatty acid synthetase activity in intact chloroplasts from Euglena gracilis.

Intact chloroplasts were isolated from Euglena gracilis variety bacillaris, aliquots were exposed to several different chemical cross-linking reagents. The reagents penetrated the triple membrane of Euglena chloroplasts. This was shown by gradient acrylamide gel electrophoresis under denaturing conditions. The activity of the nonaggregated fatty acid synthetase of Euglena was located within the chloroplast stroma, and the effects of dimethylsuberimidate cross-linking on the activity of the enzyme system were examined. The acyl-carrier protein concentration in the chloroplast was measured at ab

Effects of orally administered Euglena gracilis and its reserve polysaccharide, paramylon, on gastric dysplasia in A4gnt knockout mice.

Euglena gracilis is widely utilized as food or supplement to promote human and animal health, as it contains rich nutrients. In this study, we administered spray-dried powder of E. gracilis and paramylon, β-glucan stored in E. gracilis cells, to A4gnt knockout (KO) mice. A4gnt KO mice are a mutant mouse model that spontaneously develops gastric cancer through hyperplasia-dysplasia-adenocarcinoma sequence in the antrum of the stomach, and we observed the effects of E. gracilis and paramylon on the early involvements of A4gnt KO mice. Male and female 10-week-old A4gnt KO mice and their age-match

Enhanced Biomass, Paramylon, and Lipids Production by Non-Axenic Cultivation of Euglena gracilis in Anaerobically Digested Livestock Wastewater.

Wastewater-based microalgal cultivation enables coupling environmental remediation with the production of sustainable, value-added biomass. In this study, Euglena gracilis was cultivated under non-axenic conditions in a 2% anaerobically digested livestock wastewater (LSWW)-based medium to enhance biomass accumulation, paramylon storage, and biodiesel precursor production, while simultaneously removing residual nitrogen and phosphorus. The LSWW medium was strongly phosphate-limited relative to ammoniacal nitrogen (N:P mass ratio ~39:1), which constrained growth. Adjustment of the N:P ratio to ~

Effect of ultrafine CO2 bubbles on Euglena gracilis Z growth with CO2 gas bubble size and chlorophyll content.

Microalgae have been explored as a viable alternative food source. Among them, Euglena gracilis stands out as a promising single-cell algae. However, the challenge lies in developing more efficient and cost-effective methods for industrial mass production of E. gracilis under controlled culture conditions. Our research aimed to address this by investigating the role of nanotechnology in using fine to ultra-fine bubble CO2 (FB-CO2)-ranging from micrometer to nanometer size-as feeding material to promote cell harvest of E. gracilis Z in autotrophic culture conditions. Our findings suggest that f

Cyanide insensitive iron superoxide dismutase in Euglena gracilis. Comparison of the reliabilities of different test systems for superoxide dismutases.

Two proteins (P1 and P2, with weights of 57,500 and 27,500 respectively) were isolated from Euglena gracilis. Both proteins show cyanide-insensitive superoxide dismutase activity in the "classical" superoxide dismutase assay, using xanthine-xanthine oxidase as O2.- generator. If O2.- is generated chemically (autoxidation of reduced anthraquinone), photochemically (illuminated riboflavine) or pulse radiolytically, only protein P1 but not P2 shows SOD activity. Protein P1 contains 1 g atom (determined: 0.82) iron (no Mn or Cu) per mole protein and may thus be defined as iron-superoxide dismutase
