Exotoxin activity associated with the Legionnaires disease bacterium.

Hemolysis occurred around growth of the Legionnaires disease bacterium on supplemented Mueller-Hinton agar containing sterile defibrinated blood from each of five mammalian species. Hemolysis was most pronounced with guinea pig or rabbit blood, was less intense with horse or sheep blood, and was slight with blood from a human donor. Sterile filtrates of allantoic fluid from embryonated hen's eggs that had been infected with this organism displayed hemolytic activity in a radial hemolysis assay with guinea pig cells in agar. Growth of the Legionnaires disease bacterium on F-G agar with 5% hen's

Prevalence and production of superantigen exotoxins by cystic fibrosis Staphylococcus aureus isolates.

Staphylococcus aureus (SA) is the most common cystic fibrosis (CF) lung pathogen that is uniquely capable of producing superantigen (SAg) exotoxins, which are recognized as the most potent activators of the immune system and inducers of inflammation. Herein, we analyzed the distribution of SA-SAgs among pediatric and adult CF clinical isolates. We then investigated the production of biologically active SA-SAgs by some of these clinical isolates and tested the stability of SA-SAg in CF sputum. We demonstrate that 60-80 % of pediatric and adult CF SA isolates carried at least one SA-SAg gene, wi

Staphylococcus aureus Exotoxins and Their Detection in the Dairy Industry and Mastitis.

Staphylococcus aureus constitutes a major food-borne pathogen, as well as one of the main causative agents of mastitis in dairy ruminants. This pathogen can produce a variety of extracellular toxins; these include the shock syndrome toxin 1 (TSST-1), exfoliative toxins, staphylococcal enterotoxins (SE), hemolysins, and leukocidins. S. aureus expresses many virulence proteins, involved in evading the host defenses, hence facilitating microbial colonization of the mammary glands of the animals. In addition, S. aureus exotoxins play a role in the development of both skin infections and mastitis.

Evaluation of the staphylococcal exotoxins and their specific IgE in childhood atopic dermatitis.

BACKGROUND: Superantigenic exotoxins produced by Staphylococcus aureus and their specific IgE antibodies are thought to be important precipitating factors of atopic dermatitis (AD), but there are few reports evaluating these 2 factors at the same time. OBJECTIVE: We examined whether the presence of the exotoxins sampled from the skin of patients with AD and the levels of anti-exotoxin IgE antibodies in their sera correlated with their severity of AD. METHODS: Patients with mild-to-severe AD, 1 to 22 years of age, were evaluated by using Leicester's scoring system. Specific IgE antibodies aga

[Detection of exotoxins of the pathogens of anaerobic gas infection using enzyme immunoassay].

To detect the exotoxins of the causative agents of the main anaerobic gas infection (Clostridium perfringens, C.oedematiens, C.histolyticum, C.septicum) a rapid and easily reproducible variant of the enzyme-linked immunosorbent assay (ELISA), based on the use of the sandwich modification activated due to avidin-biotin interaction, was proposed. The possibility of using the avidin-biotin variant of ELISA for the detection of C.perfringens, C.oedematiens, C.histolyticum and C.septicum toxins in experimental gas gangrene in guinea pigs was shown. The method made it possible to reproducibly detect

Nanoporous Hydrogels for the Observation of Anthrax Exotoxin Translocation Dynamics.

The ability to observe lethal anthrax exotoxins translocating through size-constricting nanopores in vitro, combined with detailed sequence and structural data, has aided in elucidated mechanisms of exotoxin cell entry and toxicity. However, due to limited observations of anthrax exotoxins translocating through protective antigen nanopores in vitro and the instability of protective antigen-functionalized suspended lipid bilayers, questions remain regarding the native mechanisms of cell entry. Nanoporous hydrogel membranes offer a robust tool for studying protein translocation with ensemble mea

Pfiesteria shumwayae kills fish by micropredation not exotoxin secretion.

Pfiesteria piscicida and P. shumwayae reportedly secrete potent exotoxins thought to cause fish lesion events, acute fish kills and human disease in mid-Atlantic USA estuaries. However, Pfiesteria toxins have never been isolated or characterized. We investigated mechanisms by which P. shumwayae kills fish using three different approaches. Here we show that larval fish bioassays conducted in tissue culture plates fitted with polycarbonate membrane inserts exhibited mortality (100%) only in treatments where fish and dinospores were in physical contact. No mortalities occurred in treatments where

Effect of Pseudomonas aeruginosa exotoxin A on inner ear function.

Electrophysiological changes were studied in the albino rat following instillation of Pseudomonas aeruginosa exotoxin A into the middle ear cavity through the tympanic membrane. Hearing threshold was measured by a burst-elicited, frequency-specific auditory brainstem response (ABR) technique prior to exposure, then 24 and 48 h, 5 days, 2 and 4 weeks after the toxin instillation. A single dose (1 microgram/20 microliters) of Pseudomonas aeruginosa exotoxin A raised the ABR threshold over the whole frequency range, by 5-25 dB, particularly in the high tones. All threshold shifts were of combined

Nucleotide sequence of streptococcal pyrogenic exotoxin type C.

The nucleotide sequence of the gene speC, encoding streptococcal pyrogenic exotoxin type C (SPE C), was determined. The gene encoded a mature protein of 208 amino acids, with a calculated molecular weight of 24,354. The mature amino acid sequence of SPE C was analyzed for homology with the amino acid sequences of streptococcal pyrogenic exotoxin type A, the staphylococcal enterotoxins, and toxic shock syndrome toxin-1. Of these, SPE C shared the greatest amount of homology with streptococcal exotoxin type A.

A competitive immunosorbent assay for detection of Pseudomonas pseudomallei exotoxin.

The development of monoclonal antibody and enzyme-linked immunosorbent assay (ELISA) techniques has made possible the detection of specific antigens at extremely low concentrations. Diagnosis of recalcitrant diseases such as melioidosis depends upon either early isolation and identification of the causative organism or the identification of a specific marker antigen, Pseudomonas pseudomallei exotoxin, in serum; the latter is better because it allows more rapid and simple diagnosis. A method of detecting exotoxin concentrations of greater than 16 ng/ml by an ELISA based on a monoclonal antitoxi

Staphylococcal pyrogenic exotoxin type C: further characterization.

Staphylococcal pyrogenic exotoxin type C was purified by differential precipitation with ethanol and resolubilization in water followed by thin-layer isoelectric focusing. The purified toxin migrated as a homogeneous protein when re-electrofocused in polyacrylamide (isoelectric point, 7.2), when subjected to sodium dodecyl sulfate polyacrylamide gel electrophoresis (molecular weight, 22,000 daltons), and when reacted against hyperimmune antisera in an Ouchterlony immunodiffusion plate. A 2200-fold purification of the toxin could be obtained. Pyrogenic exotoxin C was elaborated by all of 44 Sta

Purification and physicochemical and biological characterization of a staphylococcal pyrogenic exotoxin.

A staphylococcal pyrogenic exotoxin was purified and characterized biochemically and biologically. The organism producing the toxin was a group I Staphylococcus aureus strain which was isolated from a vaginal infection of a patient with mucocutaneous lymph node syndrome (Kawasaki's disease). The possible association of the toxin with the disease syndrome is discussed. The toxin was purified from cell-free culture supernatant fluids by means of differential precipitation with ethanol and resolubilization in pyrogen-free distilled water followed by preparative thin-layer isoelectric focusing. Production of pyrogenic exotoxin by groups of streptococci: association with group A.

Several groups of streptococci were tested for production of pyrogenic exotoxins (SPE) with Ouchterlony immunodiffusion, a newly developed passive hemagglutination inhibition assay, and an assay for pyrogenicity and capacity to enhance lethal endotoxin shock. With use of these assays, 68 (91%) of 75 group A streptococcal strains were positive for one or more of SPE types A, B, and C; seven were negative for both the known SPE types and antigenically unrelated pyrogenic exotoxins. Group A strains producing both SPE B and C were the most common, and strains producing A alone or AB and AC togethe

Interaction of Bacterial Exotoxins with Neutrophil Extracellular Traps: Impact for the Infected Host.

Since their discovery in 2004, neutrophil extracellular traps (NETs) have been characterized as a fundamental host innate immune defense against various pathogens. Released in response to infectious and pro-inflammatory stimuli, NETs can immobilize invading pathogens within a fibrous matrix consisting of DNA, histones, and antimicrobial peptides. Conversely, excessive or dysregulated NET release may hold a variety of detrimental consequences for the host. A fine balance between NET formation and elimination is necessary to sustain a protective effect during infectious challenge. In recent year

Engineered liposomes sequester bacterial exotoxins and protect from severe invasive infections in mice.

Gram-positive bacterial pathogens that secrete cytotoxic pore-forming toxins, such as Staphylococcus aureus and Streptococcus pneumoniae, cause a substantial burden of disease. Inspired by the principles that govern natural toxin-host interactions, we have engineered artificial liposomes that are tailored to effectively compete with host cells for toxin binding. Liposome-bound toxins are unable to lyse mammalian cells in vitro. We use these artificial liposomes as decoy targets to sequester bacterial toxins that are produced during active infection in vivo. Administration of artificial liposom

Detection of staphylococcal exotoxins in antrochoanal polyps and chronic rhinosinusitis with nasal polyps.

OBJECTIVE: Staphylococcal exotoxins have been reported in the pathogenesis of many chronic inflammatory diseases. Recent reports have hypothesized that staphylococcal exotoxins might be related to inflammatory mucosal changes seen in chronic sinusitis with nasal polyps (CRS-NPs). Staphylococcal exotoxins have the capacity to act as superantigens (SAgs), bypassing normal antigen processing and directly stimulating a massive inflammatory response. The objective of this study was to analyze polyp tissue samples from patients with antrochoanal polyps (ACPs) and CRS-NPs for the presence of staphylo

Congenital streptococcal toxic shock syndrome with absence of antibodies against streptococcal pyrogenic exotoxins.

Congenital infection with group A beta-hemolytic streptococcus was complicated by toxic shock syndrome in a neonate. We hypothesize that the severity of the clinical syndrome was related to the streptococcal pyrogenic exotoxin in the absence of corresponding antibodies. The outcome may have been favorably influenced by the antibodies to streptococcal pyrogenic exotoxin present in the immunoglobulins given as treatment.

From endotoxin to exotoxin: De's rich legacy to cholera.

Between 1951 and 1959, Sambhu Nath De made crucial discoveries on the pathogenesis of cholera that changed the course of our understanding of the disease. The discovery that cholera is caused by a potent exotoxin (cholera enterotoxin) affecting intestinal permeability, the demonstration that bacteria-free culture filtrates of Vibrio cholerae were enterotoxic, and the development of a reproducible animal model for the disease are considered milestones in the history of the fight against cholera. In this commentary, a classic article by De & Chatterje published in 1953 and its public health and
