The hippocampal dentate gyrus (DG) has emerged as a cornerstone of engram research. While DG fear-based engrams have been extensively studied, revealing principles of allocation, consolidation, retrieval, and valence switching, engrams encoding context-reward associations, particularly those involving drugs of abuse, remain comparatively underexplored. This knowledge gap has critical implications for understanding addiction, depression, and other disorders involving dysfunctional reward processing. In this review, we first establish the DG's unique anatomical and functional properties that pos

The elucidation of the functional neuroanatomy of human fear, or threat, extinction has started in the 2000s by a series of enthusiastically greeted functional magnetic resonance imaging (fMRI) studies that were able to translate findings from rodent research about an involvement of the ventromedial prefrontal cortex (vmPFC) and the hippocampus in fear extinction into human models. Enthusiasm has been painfully dampened by a meta-analysis of human fMRI studies by Fullana and colleagues in 2018 who showed that activation in these areas is inconsistent, sending shock waves through the extinction

The case to be reported presented with multiple motor tics. During the course of therapy, directed along behavioural lines, a model of the nature of the movements emerged which was later found to be strikingly similar to that described by Bliss (1980). The patient (J) complained of a "horrible" sensation, similar to an "itch", which always preceded the tic movements and which could be removed by movement. Occasionally, by supreme effort, the "itch" would be resisted and movement avoided. These sensations could thus be viewed as a trigger for the movements. The implications of such an analysis

BACKGROUND: This systematic review and meta-analysis evaluated whether people with obsessive-compulsive disorder (OCD) exhibit differences in associative learning.
METHODS: CINAHL, Cochrane Library, EMBASE, PsycINFO, PubMed, Scopus and Web of Science were searched for published peer-reviewed studies in English quantitatively examining the relationship between OCD and associative phenomena in humans. 5508 titles, 124 abstracts and 55 full texts were reviewed; citation searching identified 15 records. Sixty-six studies were included. Risk of bias was assessed and random-effects meta-analysis sy

BACKGROUND: Computational models suggest that both acute and chronic alcohol exposure impair fear extinction learning by destabilising amygdala circuitry, yet empirical validation has been limited.
OBJECTIVE: To synthesise recent circuit manipulation studies with advanced neural network modelling and highlight their joint implications for alcohol induced dysregulation of amygdala circuits, fear extinction, and related comorbidities.
METHODS: We review chemogenetic and optogenetic investigations targeting basolateral and central amygdala sub circuits, neuromodulatory systems, and stress hormo

Memories are multifaceted and can simultaneously contain positive and negative attributes. Here, we report that negative attributes of a mixed-valence memory dominate long-term recall. To induce a mixed-valence memory, running responses were randomly reinforced with either food (∼83% of trials) or footshock (∼17% of trials), or a noise conditioned stimulus (CS) was followed randomly with either food (∼80% of trials) or footshock (∼20% of trials). Control animals were consistently reinforced with only food. Mixed-valence training promoted unstable behavior (e.g., erratic approach and withdrawal

Protocadherin 9 (Pcdh9) is a member of the cadherin superfamily and is uniquely expressed in the vestibular and limbic systems; however, its physiological role remains unclear. Here, we studied the expression of Pcdh9 in the limbic system and phenotypes of Pcdh9-knock-out mice (Pcdh9 KO mice). Pcdh9 mRNA was expressed in the fear extinction neurons that express protein phosphatase 1 regulatory subunit 1 B (Ppp1r1b) in the posterior part of the basolateral amygdala (pBLA), as well as in the Cornu Ammonis (CA) and Dentate Gyrus (DG) neurons of the hippocampus. We show that the Pcdh9 protein was

BACKGROUND: Responding emotionally to danger is critical for survival. Normal functioning also requires flexible alteration of emotional responses when a threat becomes safe. Aberrant threat and safety learning occur in many psychiatric disorders, including posttraumatic stress disorder, obsessive-compulsive disorder, and schizophrenia, in which emotional responses can persist pathologically. While there is evidence that threat and safety learning can be modulated by the serotonin systems, there have been few studies in humans. We addressed a critical clinically relevant question: How does low

Four experiments with rat subjects asked whether a partial reinforcement extinction effect (PREE) occurs in taste aversion learning. The question has received little attention in the literature, and to our knowledge no taste aversion experiment has previously demonstrated a PREE. In each of the present experiments, experimental groups received a taste mixed in drinking water for 20 min; such taste exposures were sometimes paired with a lithium chloride (LiCl) injection and sometimes not. Control groups received only taste-LiCl pairings. There was evidence that each reinforced and non-reinforce

This study investigated the potential for recreational gamblers to respond as if certain types of losing slot machine outcomes were actually closer to a win than others (termed the near-miss effect). Exposure to conditional discrimination training and testing disrupted this effect for 10 of the 16 participants. These 10 participants demonstrated high percentages of conditional discrimination testing performance, and the remaining 6 participants failed the discrimination tests. The implications for a verbally based behavioral explanation of gambling are presented.

Resurgence is an increase in a previously reduced target behavior with worsening conditions for a more recently reinforced alternative behavior. Alternating sessions in which alternative reinforcement is available (i.e., "on" sessions) versus extinction (i.e., "off" sessions) during treatment (i.e., contingency discrimination training [CDT]) reduces resurgence. This experiment examined whether CDT could mitigate resurgence without complete extinction during "off" sessions. Target lever pressing of rats was reinforced in baseline and then extinguished during treatment. In treatment, all groups

Activation of N-methyl-D-aspartate (NMDA) receptor can facilitate the extinction of various maladaptive memories. Sodium benzoate (NaB) has been known to enhance a naturally occurring full agonist on the glycine binding site of the NMDA receptor. This study aimed to test whether systemic NaB treatment can affect the extinction of a cocaine-supported memory, the cocaine-induced conditioned place preference (CPP). Following the establishment of the cocaine (10 mg/kg/conditioning × 3)-induced CPP, an extinction protocol, consisting of two consecutive extinction training bouts at an 8-h interval,

We test the hypothesis that the cerebellar cortex is required for the extinction of conditioned eyelid responses in rabbits trained using standard Pavlovian delay procedures. Following 10 daily training sessions during which rabbits achieved asymptotic performance, lesions of the ipsilateral hemisphere of the cerebellar cortex were made by aspiration. The target of these lesions was the anterior lobe, as suggested by previous observations that this region is necessary for the learning-dependent timing of conditioned eyelid responses (Perrett et al., 1993). We report that anterior lobe damage,

Squirrel monkeys pressed a lever to produce food on several fixed-ratio schedules; they could also bite a rubber hose. Biting attack occurred during the postreinforcement pause and in early portions of the ratio response run. Also, biting attacks increased after transitions to higher values of the fixed-ratio requirement and in extinction. The results show that extinction-induced aggression effects occur in primates.

Fear extinction is a phenomenon that involves a gradual reduction in conditioned fear responses through repeated exposure to fear-inducing cues. Functional brain connectivity assessments, such as functional magnetic resonance imaging (fMRI), provide valuable insights into how brain regions communicate during these processes. Stress, a ubiquitous aspect of life, influences fear learning and extinction by changing the activity of the amygdala, prefrontal cortex, and hippocampus, leading to enhanced fear responses and/or impaired extinction. Glucocorticoid receptors (GRs) are key to the stress re

Although exposure-based therapy has been found to be effective at alleviating symptoms of social anxiety disorder, it often does not lead to full remission, and relapse after treatment is common. Exposure therapy is based on theoretical principles of extinction of conditioned fear responses. However, there are inconsistencies in findings across experiments that have investigated the effect of social anxiety on threat conditioning and extinction processes. This systematic review and meta-analysis aimed to examine whether elevated levels of social anxiety are associated with abnormalities in thr

Hyperactive orbitofrontal cortical activation is commonly seen in patients of obsessive-compulsive disorder (OCD). Previous studies from our laboratory showed that for rats with aberrant activation of the orbitofrontal cortex (OFC) during the extinction phase, they were unable to use contexts as the reference for proper retrieval of fear memory during renewal test. This result supported the phenomenon that many OCD patients show poor regulation of fear-related behavior. Since there are robust anatomical connections of the OFC with the fear-circuit, we aim to further examine whether the OFC is

A critical developmental task is learning what constitutes reliable threat and safety signals in the environment. In humans, atypical fear learning processes are implicated in many mental health conditions, particularly fear and anxiety disorders, pointing to the potential for laboratory measures of fear learning to facilitate early identification of at-risk individuals. This chapter reviews studies of fear learning and extinction learning that incorporate peripheral measures of psychophysiological response and include a developmental sample. Broadly, these studies indicate substantial consist

The intercalated cells of the amygdala (ITCs) are a fundamental processing structure in the amygdala that remain relatively understudied. They are phylogenetically conserved from insectivores through primates, inhibitory, and project to several of the main processing and output stations of the amygdala and basal forebrain. Through these connections, the ITCs are best known for their role in conditioned fear, where they are required for fear extinction learning and recall. Prior work on ITC connectivity is limited, and thus holistic characterization of their afferent and efferent connectivity i

Fear-related disorders, mainly phobias and post-traumatic stress disorder, are highly prevalent, debilitating disorders that pose a significant public health problem. They are characterized by aberrant processing of aversive experiences and dysregulated fear extinction, leading to excessive expression of fear and diminished quality of life. The gold standard for treating fear-related disorders is extinction-based exposure therapy (ET), shown to be ineffective for up to 35% of subjects. Moreover, ET combined with traditional pharmacological treatments for fear-related disorders, such as selecti
