Best practice for the size analysis of nanomedicines - An iron sucrose case study.

Iron sucrose is used in the parenteral treatment of iron deficiency anaemia. A number of iron sucrose similars have been developed alongside the original product. These products consist of colloidal iron with particles in the nanometre range stabilised by sucrose molecules. Dynamic light scattering (DLS) is the method of choice for the qualification of iron sucrose products in terms of particle size. However, the broad range of instrumentation and accessories available today for the execution of these measurements requires that best laboratory practice is established to ensure measurement comp

Sulfide silver autometallography to differentiate the ultrastructural localization of iron-carbohydrate complexes inside macrophages.

Iron is an essential element for numerous physiological processes in the human body, and maintaining proper iron homeostasis is critical for health. Iron imbalance can lead to conditions like iron deficiency anemia (IDA). Intravenous (IV) iron drugs, including iron-carbohydrate complexes such as iron sucrose (IS) and ferric carboxymaltose (FCM), are commonly used in the treatment of IDA. However, the cellular mechanisms underlying the uptake and intracellular fate of these complexes remain poorly understood despite over seven decades of clinical use. This study introduces a novel application o

[Treatments of the martial deficiencies: preparations available in France].

The financial advantage of the oral treatments of iron is minimised by their low bioavailibility (10 to 20%) and the fact that instructions for oral treatments are often badly followed by the patient (eg: tablets taken too far away from meals inducing frequent digestive problems). In France, since Maltofer (hydroxic ferric polymaltose) has been discontinued, Venofer (complex hydroxide ferric saccharose) is the only alternative intraveinous treatment with a cost which is now ten times higher than it was before. In foreign countries, other specialised products for intraveinous treatments (notabl

Real-World Comparative Effectiveness of Ferric Derisomaltose Versus Conventional Iron Treatment in Postpartum Hemorrhage-Related Anemia.

BACKGROUND: Postpartum hemorrhage (PPH) can lead to acute iron deficiency anemia, negatively affecting maternal health and infant care by contributing to fatigue, depression, and difficulties with breastfeeding. There is a clinical need for rapid and effective iron supplementation. OBJECTIVE: This study aimed to evaluate the real-world effectiveness of ferric derisomaltose (FDI), which allows for single high-dose intravenous administration, in comparison with conventional treatment combining intravenous and oral iron administration. METHODS: This retrospective observational study included po

Analytical quality by design in the development of a paper-based microfluidic device for iron (II) determination as an impurity in iron (III) pharmaceuticals.

A microfluidic paper-based analytical device (μPAD) was developed for the quantitation of ferrous ions (Fe2+) and total iron in iron sucrose administered in iron deficiency anemia. The elemental iron content is 20 mg/mL and USP established a limit of Fe2+ as 0.4 % (w/v). The μPAD allowed the quantitation of Fe2+ by complexation with 1,10-phenanthroline, forming an orange-red complex analysed by imageJ®. For total iron determination, ferric ion (Fe3+) was reduced to Fe2+ using ascorbic acid prior to complexation. The device was prepared on chromatographic paper by using the wax printing method.

Analysis of transition from intravenous iron sucrose to ferric carboxymaltose infusions in pediatric patients with intestinal failure.

BACKGROUND: Iron supplementation is required for pediatric patients with intestinal failure (IF). There is a paucity of literature on optimal iron formulation and outcomes in this patient population that requires ongoing supplementation. The aim of this study was to assess outcomes in pediatric patients with IF receiving iron sucrose (IS) vs ferric carboxymaltose (FCM) iron infusions. METHODS: This was a single-center observational cohort study of pediatric patients with IF requiring ongoing intravenous iron supplementation. Patients were transitioned from IS to FCM as iron therapy. Longitudi

Use of iron sucrose injection in anemia patients with reduced serum iron concentration during hospitalizations of digestive and liver diseases.

BACKGROUND: Anemia is one of the most common disorders in the world. Serum iron is an essential element for the synthesis of hemoglobin and contribution of the oxygen-carrying ability of red blood cells (RBCs). Iron sucrose injection may effectively correct iron deficiency, increase iron storage, and then improve anemia. The aim of the present study was to evaluate the therapeutic effect of iron sucrose injection in anemia patients with reduced serum iron concentration. METHODS: Overall, 95 anemia patients with digestive and/or liver diseases were included. They were divided according to the

The comparative safety of intravenous iron dextran, iron saccharate, and sodium ferric gluconate.

Intravenous iron treatment is an important component of anemia therapy for patients on dialysis. Until recently iron dextran was the only parenteral form of iron available in the United States. This drug has been associated with occasional serious adverse reactions, including full-blown anaphylaxis. In 1999 the Food and Drug Administration approved a second form of iron for intravenous administration, sodium ferric gluconate in sucrose. It is expected that by the time of this publication, a third agent, iron saccharate will also be approved. In this review the comparative safety of these three

A randomized, controlled, open label non-inferiority trial of intravenous ferric carboxymaltose versus iron sucrose in patients with iron deficiency anemia in China.

Iron deficiency (ID) and ID anemia (IDA) pose significant public health concerns in China. Although iron sucrose (IS) treatment is well-established in the country, ferric carboxymaltose (FCM) offers the advantage of higher doses and fewer infusions. This open label, randomized, controlled, non-inferiority trial was conducted at multiple sites in China to compare the outcomes of FCM (maximum of 2 doses, 500 or 1000 mg iron) and IS (up to 11 infusions, 200 mg iron) treatments in subjects with IDA. The primary endpoint was the achievement of hemoglobin (Hb) response (an increase of ⩾2 g/dL from b

Intravenous iron sucrose therapy for moderate to severe anaemia in pregnancy.

BACKGROUND & OBJECTIVES: Iron deficiency anaemia (IDA) is the most common nutritional deficiency in pregnancy. Prophylactic oral iron is recommended during pregnancy to meet the increased requirement. In India, women become pregnant with low baseline haemoglobin level resulting in high incidence of moderate to severe anaemia in pregnancy where oral iron therapy cannot meet the requirement. Pregnant women with moderate anaemia are to be treated with parentral iron therapy. This study was undertaken to evaluate the response and effect of intravenous iron sucrose complex (ISC) given to pregnant w

Ferumoxytol: a new era of iron deficiency anemia treatment for patients with chronic kidney disease.

Ferumoxytol is a new product approved for intravenous use by the US Food and Drug Administration (FDA) in the treatment of iron deficiency anemia in adults with chronic kidney disease. This approval was based on data from 3 open-label, randomized, controlled clinical trials. In all of these trials, ferumoxytol was well tolerated, and hemoglobin levels were significantly increased compared with those achieved by orally administered iron. Ferumoxytol, a superparamagnetic iron oxide coated with a carbohydrate shell, is also used as a magnetic resonance imaging (MRI) agent due to its magnetic prop
