Mechanisms and inhibitors of ferroptosis in psoriasis.

Psoriasis is a chronic inflammatory skin disease that features localized or widespread erythema, papules, and scaling. It is common worldwide and may be distributed throughout the whole body. The pathogenesis of psoriasis is quite complex and the result of the interplay of genetic, environmental and immune factors. Ferroptosis is an iron-dependent programmed death that is different from cell senescence, apoptosis, pyroptosis and other forms of cell death. Ferroptosis involves three core metabolites, iron, lipids, and reactive oxygen species (ROS), and it is primarily driven by lipid peroxidati

Cancer: The role of iron and ferroptosis.

Iron is an essential element for virtually all living things. Body iron levels are tightly controlled as both increased iron levels and iron deficiency are associated with many clinical conditions. Increased iron levels are associated with a worse prognosis in some cancers, so understanding the role of iron in cancer development has thus been an active area of research. Regulated forms of cell death are important in development and disease pathogenesis. In this Medicine in Focus review article, we discuss the role of iron in cancer, and ferroptosis, a new form of iron-regulated cell death trig

Ferroptosis in Liver Diseases: An Overview.

Ferroptosis is an iron-dependent form of cell death characterized by intracellular lipid peroxide accumulation and redox imbalance. Ferroptosis shows specific biological and morphological features when compared to the other cell death patterns. The loss of lipid peroxide repair activity by glutathione peroxidase 4 (GPX4), the presence of redox-active iron and the oxidation of polyunsaturated fatty acid (PUFA)-containing phospholipids are considered as distinct fingerprints of ferroptosis. Several pathways, including amino acid and iron metabolism, ferritinophagy, cell adhesion, p53, Keap1/Nrf2

Study on the Synergistic Mechanism of Photodynamic Therapy Combined With Ferroptosis Inducer to Induce Ferroptosis in Cholangiocarcinoma.

BACKGROUND: Photodynamic therapy (PDT) induced lipid peroxidation reaction can lead to necrosis and apoptosis of extrahepatic cholangiocarcinoma (ECC) cells, reducing the tumor load. However, the depth of action of PDT is shallow, and its therapy efficacy is weak, making it difficult to achieve eradication even with multiple treatments. OBJECTIVES: This study aims to investigate the mechanism and main pathways of ferroptosis in cholangiocarcinoma under Hematoporphyrin-mediated photodynamic therapy, and to compare the effects of different ferroptosis inducers on photodynamic therapy-induced fe

Decoding ferroptosis for cancer therapy.

Resisting cell death is a pivotal hallmark of cancer and one of several increasingly actionable functional capabilities acquired by cancer cells to sustain their malignant state. Since the early 2000s, the discovery of multiple regulated cell death programmes has intensified interest in targeting these maladaptive traits that cancer cells employ to resist cellular demise. Among these, ferroptosis - the lethal outcome of iron-dependent (phospho)lipid peroxidation - stands apart from other regulated cell death mechanisms, as it is persistently suppressed while lacking an activating signal. In ca

Ferroptosis: mechanisms and therapeutic targets.

Ferroptosis is a nonapoptotic form of cell death characterized by iron-dependent lipid peroxidation in membrane phospholipids. Since its identification in 2012, extensive research has unveiled its involvement in the pathophysiology of numerous diseases, including cancers, neurodegenerative disorders, organ injuries, infectious diseases, autoimmune conditions, metabolic disorders, and skin diseases. Oxidizable lipids, overload iron, and compromised antioxidant systems are known as critical prerequisites for driving overwhelming lipid peroxidation, ultimately leading to plasma membrane rupture a

COX15 deficiency causes oocyte ferroptosis.

Mitochondria play diverse roles in mammalian physiology. The architecture, activity, and physiological functions of mitochondria in oocytes are largely different from those in somatic cells, but the mitochondrial proteins related to oocyte quality and reproductive longevity remain largely unknown. Here, using whole-exome sequencing data from 1,024 women (characterized by oocyte maturation arrest and degenerated or morphologically abnormal oocytes) and 2,868 healthy controls, we performed a population and gene-based burden test for mitochondrial genes and identified a candidate gene, cytochrome

Iron, ferroptosis, and ischemic stroke.

Over 30 million people suffer from the consequences of ischemic stroke. The precise molecular mechanism of neuronal damage during ischemic stroke remains unclear; therefore, the effective treatment of post-ischemic stroke remains a critical challenge. Recently, iron has emerged as a crucial factor in post-reperfusion injuries, participating in cell peroxidation, excitotoxicity, and a distinctive cell death pathway, namely, ferroptosis. Since iron is tightly regulated in the brain and important for brain functions, the imbalance of its metabolism, including its overload and deficiency, has been

Ferroptosis: past, present and future.

Ferroptosis is a new type of cell death that was discovered in recent years and is usually accompanied by a large amount of iron accumulation and lipid peroxidation during the cell death process; the occurrence of ferroptosis is iron-dependent. Ferroptosis-inducing factors can directly or indirectly affect glutathione peroxidase through different pathways, resulting in a decrease in antioxidant capacity and accumulation of lipid reactive oxygen species (ROS) in cells, ultimately leading to oxidative cell death. Recent studies have shown that ferroptosis is closely related to the pathophysiolog

Ferroptosis at the crossroads of epilepsy and neurodegeneration.

Ferroptosis, an iron-dependent form of regulated cell death characterized by lipid peroxidation, has emerged as a potential contributor to neuronal injury across neurological disorders. Increasing evidence suggests that ferroptotic mechanisms may participate in the pathophysiology of epilepsy, linking sustained oxidative stress, iron dysregulation, and progressive neuronal loss. Mechanistically, ferroptosis is driven by disruption of the canonical system Xc--glutathione-glutathione peroxidase 4 axis, resulting in impaired detoxification of lipid hydroperoxides and membrane damage accumulation.

Studying Ferroptosis in the Tumor Microenvironment.

Despite strong preclinical evidence, ferroptosis-based strategies have not yet translated into the clinic, in part due to the disconnect between conventional in vitro models and the complex tumor microenvironment. In this commentary, we consider ferroptosis as a microenvironmentally regulated process and call for multidisciplinary approaches that bridge foundational ferroptosis biology with clinically actionable interventions grounded in the context of the tumor microenvironment.

Ferroptosis in liver biology and diseases.

Ferroptosis is a form of non-apoptotic cell death driven by iron-dependent lipid peroxidation (LPO). Ferroptosis is closely associated with hepatic metabolic pathways, particularly those involving iron, fatty acids, amino acids, and redox changes. This review aims to provide a comprehensive overview of the current molecular pathways and regulatory mechanisms underlying iron homeostasis and intracellular compartmentation in liver biology, emphasizing the roles of ferroptosis in several liver diseases, such as ischemia-reperfusion injury, drug-induced liver injury, viral hepatitis, metabolic dys

Role of ferroptosis in gastric cancer.

Gastric cancer remains a globally prevalent malignancy with significant clinical burdens. The iron-mediated cell death mechanism known as ferroptosis has gained substantial attention in cancer studies owing to its distinctive biological characteristics and disease-related implications. This review delineates the molecular machinery driving ferroptosis and systematically analyzes its dual roles in gastric carcinogenesis-both as a tumor-suppressive mechanism and a vulnerability axis during metastasis. Furthermore, we summarize the multi-layered regulatory networks governing ferroptosis, encompas

Ferroptosis at the intersection of radiotherapy and immunotherapy.

Immunotherapy and radiotherapy are widely used in the treatments of various cancers. However, their application remains limited in certain cancer types because only a small number of patients respond to monotherapy. Notably, these two modalities can exert synergistic anti-cancer effects when combined. A growing body of evidence suggests that ferroptosis plays a crucial role in such combination therapies. Specifically, immunotherapy-activated CD8+ T cells and radiotherapy-induced ataxia-telangiectasia mutated gene (ATM) activation (associated with DNA damage) can synergistically enhance tumor l
