The leaf-monkeys, Presbytis cristata and Presbytis melalophos, experimentally infected with subperiodic Brugia malayi, have been used for studies on the pathoimmunology of the infection and the screening of potential filaricides during the last 6-8 years, and considerable information on the pattern of microfilaraemia and adult worm recoveries have been obtained. The prepatent periods in 97 P. cristata and 45 P. melalophos, each infected with about 200 infective larvae, were similar, these being approximately 70 and 68 days respectively. Although all infected animals became microfilaraemic, the

Onchocerciasis is a disease that engages the attention of most researchers. Presently, there is not an ideal onchocerciatic agent, hence the search must continues. Consequently alpha, alpha-trehalose-6-phosphate was synthesised and assessed for onchocerciatic activity against O. volvulus; using diethylcarbamizine citrate as the control drug. Results from this study showed that alpha, alpha-trehalose-6-phosphate is a glucose analogue with effective micro and macro-filaricidal agent, better than that of the control drug. The inhibitory action of this compound on enzyme trehalase is a postulate f

Several drugs were investigated for action against Onchocerca volvulus in chimpanzees. Pentamidine at 4 mg/kg daily for 16 days had no filaricidal action; nor did stibocaptate at 10 mg/kg monthly for 8 months. Hoechst 33258, a substituted bis-benzamidazole, showed micro- and macrofilaricidal activity; F151 (an arsenical compound) showed a purely macrofilaricidal action; and Compound E (a reaction product of Hoechst 33258 and F151) was micro- and macrofilaricidal. The toxicity of the three last-mentioned compounds was high, and further trials were not made. Nifurtimox, at doses of up to 40 mg/k

The present investigation deals with the evaluation of antifilarial efficacy of liposome entrapped antiwolbachial antibiotics doxycycline and rifampicin (5 doses at 10 mg/kg, subcutaneously for 15 days) alone and/or in combination with standard filaricide diethylcarbamazine (DEC) against human lymphatic filariid Brugia malayi in rodent host Mastomys coucha. The delivery system maintained the sustained release of antibiotics up to 48 h and significantly (P < 0.05) augmented the antifilarial potential of these antibiotics over their free administration. A combination of DEC with each entrapped a

5-fluorouracil and 5-fluorodeoxyuridine at 30 mg/kg body weight daily for four days inhibit microfilarial production in Brugia pahangi in the jird. Disruption of intrauterine embryogenesis was observed in treated female worms but the compounds were not macrofilaricidal or microfilaricidal under the conditions employed. 5-fluorocytosine possessed no filaricidal or embryostatic activity. The inhibition of microfilaria production by 5-fluorouracil was temporary and larval production was resumed within nine weeks. The compound also inhibited the development of B. pahangi and Dirofilaria immitis la

Previous results have revealed the antifilarial activities of crude extracts and pure compounds from some Cameroonian medicinal plants against Onchocerca volvulus and Onchocerca gutturosa. In our efforts to find new filaricidal agents against adult male O. gutturosa worms, we have isolated and screened three compounds: polycarpol and polyveoline from Polyalthia suaveolens (Annonaceae) and 3-O-acetyl aleuritolic acid from Discoglypremna caloneura (Euphorbiaceae). Only polycarpol and 3-O-acetyl aleuritolic acid exhibited significant inhibitory activities on the vitality of adult male worms of O.

Glucose-6-phosphate dehydrogenase (E.C. 1.1.1.49) was partially purified from the extracts of adult Onchocerca volvulus by affinity chromatography on 2'5'ADP-Sepharose-4B. Kinetic studies revealed a typical bell-shaped pH profile with an optimum lying between pH 7.3 and 7.8. The apparent Km for glucose-6-phosphate was 5.66 x 10(-5) M, whereas that for NADP was 2.17 x 10(-6) M. Suramin, a filaricidal drug, inhibited the enzyme competitively with respect to NADP as a substrate: the apparent Ki values were 2.23 x 10(-6) M and 4.21 x 10(-7) M, respectively, for the crude and purified enzyme prepar

Control and treatment of onchocerciasis, a devastating tropical filarial disease caused by Onchocerca volvulus, rely solely on the community directed treatment with ivermectin. However, ivermectin is only microfilaricidal with evidence of resistance of the parasite among other limitations, which necessitate the search for new efficacious and safe filaricides. Ten synthetic thienylazoryl dyes were screened in vitro against adult and microfilariae worm stages of Onchocerca ochengi based on worm motility and MTT formazan assay. Cytotoxicity of active compounds was assessed on monkey kidney epithe

Presbytis cristata monkeys infected through the inoculation of between 200 and 400 subperiodic Brugia malayi infective larvae (L3) in the right thigh, in both thighs or in the dorsum of the right foot were followed up for varying periods of up to about 8 months after infection. All 148 inoculated animals became patent, with mean prepatent periods being between 66 and 76 days. In animals injected in the thigh, the patterns of microfilaraemia were similar, there being a rapid rise in the geometric mean counts (GMCs) of microfilariae during the first 10-12 weeks of patency, which then plateaued a

Three monoclonal antibodies directed towards antigens of Onchocerca gibsoni were used in antigen detection ELISAs to detect parasite antigens in sera from 100 cattle infected with O. gibsoni, in trials with the filaricidal compounds CGP 6140, CGP 20309, CGP 20376, CGP 21833, CGP 24589 and CGP 26702. Measurable levels of parasite antigens were highly variable, both within and between treatment and control groups of animals, with no consistent trends which related to time after treatment, micro or macrofilaricidal effects against O. gibsoni, or dose rate for any of the compounds used. It was con

Increasing concentrations of levamisole and of mebendazole were applied to 1 eye in groups of 4 patients with ocular onchocerciasis in northern Cameroon. No effect resulted from up to 3.0% mebendazole suspensions, but 3.0% levamisole solutions rapidly caused entry of microfilariae, straightening out and subsequent opacification of previously curled-up living microfilariae, the rapid formation of typical limbal globular infiltrates, and the subsequent formation of fluffy opacities around the microfilariae. These changes are typical of all other drugs so far studied that have a microfilaricidal

The presence of 5'-nucleotidase was demonstrated in Onchocerca volvulus and Dirofilaria immitis; the bulk of activity was found in the particulate fraction. The enzyme of filarial worms exhibited a broad pH-optimum between 6.4 and 8.0 and substrate specificity for nucleotides compared to glucose-6-phosphate and p-nitrophenyl phosphate. The apparent Km-values for AMP were found to be 0.15 mM and 0.22 mM for the enzyme from O. volvulus and D. immitis, respectively. The activity of 5'-nucleotidase from both filarial worms was effectively inhibited by the filaricidal compound CGP 8065, a dithiocar

It is possible to infect jirds with Brugia pahangi by three methods. Infective larvae (L3) can be injected either intraperitoneally (ip), when adults develop in the peritoneal cavity, or sub-cutaneously (sc), when they develop in the lymphatics or the heart and blood vessels associated with the lungs. Alternatively adult worms which have been grown in the peritoneal cavities of jirds can be implanted into the peritoneal cavities of other jirds. This latter system has been widely used for screening for new filaricides. We have compared the activity of 9 macrofilaricidal compounds against these

The present work seeks to explore the antifilarial activity of biopolymer functionalized gold nanoparticles (AuNPs) against human filarial parasite (Wuchereria bancrofti) through Nrf2 signaling for the first time. A natural polymer, chitosan is used along with Terminalia chebula extract to synthesize AuNPs following the principles of green chemistry. The probable mode of action of AuNPs as filaricidal agent has been investigated in detail using model filarial parasite, Setaria cervi (bovine parasite). Biopolymers inspired AuNPs exhibit superior antifilarial activity against both human and bovi

Cases of onchocerciasis were treated with three drugs, Antrypol, Pentostam and Hetrazan. Antrypol proved undoubtedly effective, producing disappearance of the microfilariae and onchocercal nodules, and improvement in cases of defective vision with anterior ocular manifestations, but little or no improvement in cases with well-defined posterior manifestations. The severe toxic effects of the drug, however, are sometimes fatal, and therefore preclude its use. Pentostam, although non-toxic, did not prove effective in the doses tried; it may be of some use in very large doses. Hetrazan was given t

To assess the filaricidal activity and clinical safety of albendazole in human loiasis, a double-blind, placebo-controlled study was conducted in an endemic area in Benin, Africa. Twenty-three men with microfilaremia (100-30,000/mL) were randomly assigned to receive albendazole (200 mg; n = 11) or placebo (n = 12) twice daily for 21 days; 1 patient from each group withdrew from the study. There were no clinical adverse effects and no observed hepatotoxicity, renal toxicity, or hematologic abnormalities attributable to the drug. In the albendazole group, microfilarial levels began to decrease a

The effect of some known antifilarial drugs on naturally-occurring filariae in squirrel monkeys, Saimiri sciureus, was evaluated. Four drugs, dithiazanine iodide, naphuride sodium, thiacetarsamide sodium, and diethylcarbamazine were tested against Dipetalonema gracile and Tetrapetalonema marmosetae. Dipetalonema caudispina was included in some of the studies using diethylcarbamazine. These are the three most commonly encountered filarial species in squirrel monkeys. Dithiazanine iodide proved to be unsuitable for use because of lack of acceptance by the monkeys. Naphuride sodium (10 mg/kg/week

BACKGROUND: New drugs effective against adult filariae (macrofilaricides) would accelerate the elimination of lymphatic filariasis and onchocerciasis. Anti-Onchocerca drug development is hampered by the lack of a facile model. We postulated that SCID mice could be developed as a fmacrofilaricide screening model.
METHODS: The filaricides: albendazole (ABZ), diethylcarbamazine (DEC), flubendazole (FBZ), ivermectin (IVM) and the anti-Wolbachia macrofilaricide, minocycline (MIN) were tested in Brugia malayi (Bm)-parasitized BALB/c SCID mice vs vehicle control (VC). Responses were compared to BALB

BACKGROUND: Lymphatic filariasis is a pervasive and life-threatening disease for human beings. Currently, 893 million people in 49 countries worldwide affected by lymphatic filariasis as per WHO statistics. The concealed aspects of lymphatic diseases such as delayed disease detection, inappropriate disease imaging, the geographical outbreak of infection, and lack of preventive chemotherapy have brought this epidemic to the edge of Neglected Tropical Diseases. Many medications and natural bioactive substances have seen to promote filaricidal activity against the target parasitic species. Howeve
