Mitochondrial Omics in Macrophage Foam Cell Formation and Atherosclerosis.

Atherosclerotic cardiovascular disease (ASCVD) is a leading cause of death worldwide, with atherosclerosis serving as the core pathological mechanism driving ischemic heart disease, ischemic stroke, and peripheral artery disease. Macrophages play a central role in atherosclerosis by internalizing modified lipids and transforming them into foam cells, thereby driving plaque formation and progression. Mitochondrial metabolism is critically involved in regulating macrophage function, and targeting mitochondrial dysfunction may provide strategies for limiting plaque inflammation and improving plaq

Pica for foam rubber in patients with sickle cell disease.

We report three cases of pica for foam rubber among sickle cell disease patients. All three were African-American males, and at the times of initial presentation for the pica, two of them were 11 years of age and one was 15 years of age. In all cases, the pica reportedly had been occurring for at least several years. The foam rubber was most often obtained from furniture and mattresses, as well as from ironing-board pads, stereo speakers, and padded hair rollers. Reports from other researchers also suggest that this is not an uncommon type of pica. We discuss this problem from biologic, psycho

Direct Scaffold-Coupled Electrical Stimulation of Chondrogenic Progenitor Cells through Graphene Foam Bioscaffolds to Control the Mechanical Properties of Graphene Foam-Cell Composites.

Osteoarthritis, a major global cause of pain and disability, is driven by the irreversible degradation of hyaline cartilage in the joints. Cartilage tissue engineering presents a promising therapeutic avenue, but success hinges on replicating the native physiological environment to guide cellular behavior and generate tissue constructs that mimic natural cartilage. Although electrical stimulation has been shown to enhance chondrogenesis and extracellular matrix production in two-dimensional (2D) cultures, the mechanisms underlying these effects remain poorly understood, particularly in three-d

Restoring cholesterol efflux in vascular smooth muscle cells transitioning into foam cells through Liver X receptor activation.

Macrophage foam cells derived from vascular smooth muscle cells (VSMCs) account for 30-70 % of foam cells in atherosclerotic lesions. Liver X receptor (LXR) agonists promote high-density lipoprotein (HDL)-mediated cholesterol efflux from macrophages. This study aimed to investigate the effects of LXR activation on the reverse cholesterol transport (RCT) rate from VSMCs to feces in vivo. Both human and mouse VSMCs exhibited similar levels of cholesterol efflux when exposed to serum and HDL. However, cholesterol efflux was significantly reduced following methyl-β-cyclodextrin (MBD)-cholesterol l

Disturbed flow induces reprogramming of endothelial cells to immune-like and foam cells under hypercholesterolaemia during atherogenesis.

AIMS: Atherosclerosis occurs preferentially in the arteries exposed to disturbed flow (d-flow), while the stable flow (s-flow) regions are protected even under hypercholesterolaemic conditions. We recently showed that d-flow alone initiates flow-induced reprogramming of endothelial cells (FIRE), including the novel concept of partial endothelial-to-immune-cell-like transition (partial EndIT), but it was not validated using a genetic lineage-tracing model. In addition, the combined effect of d-flow and hypercholesterolaemia has not been tested. Here, we tested and validated the two-hit hypothes

Foam Cells: The Origin of Formation and Target for Plant-derived Bioactive Compounds.

Foam cells play a crucial role in the initiation and progression of atherosclerosis, a condition marked by the development and growth of plaques that narrow blood vessel lumens. This narrowing can prevent normal blood flow and, in severe cases, lead to plaque rupture and blood clot formation, which can cause stroke or myocardial infarction. The origin of foam cells is diverse, arising from monocytes, vascular smooth muscle cells, stem/progenitor cells, and dendritic and endothelial cells. In their attempt to eliminate excess lipoproteins and cholesterol, foam cells inadvertently contribute to

High density lipoprotein mediates selective reduction in cholesteryl esters from macrophage foam cells.

To elucidate an anti-atherogenic nature of high density lipoprotein (HDL) at cellular level, its in vitro effect on macrophage foam cells was examined. Rat peritoneal macrophages were converted to foam cells by incubation with [3H]cholesterol-labeled acetylated LDL. HDL addition to these foam cells resulted in a reduction in cellular radioactive cholesteryl esters (CE) as well as its CE mass. The radioactive free cholesterol (FC) was similarly reduced with time, whereas its FC mass level was unaltered. Other lipoproteins such as very low density lipoprotein and low density lipoprotein also red

The cell origins of foam cell and lipid metabolism regulated by mechanical stress in atherosclerosis.

Atherosclerosis is an inflammatory disease initiated by endothelial activation, in which lipoprotein, cholesterol, extracellular matrix, and various types of immune and non-immune cells are accumulated and formed into plaques on the arterial wall suffering from disturbed flow, characterized by low and oscillating shear stress. Foam cells are a major cellular component in atherosclerotic plaques, which play an indispensable role in the occurrence, development and rupture of atherosclerotic plaques. It was previously believed that foam cells were derived from macrophages or smooth muscle cells,

Hypertensive Pressure Mechanosensing Alone Triggers Lipid Droplet Accumulation and Transdifferentiation of Vascular Smooth Muscle Cells to Foam Cells.

Arterial Vascular smooth muscle cells (VSMCs) play a central role in the onset and progression of atherosclerosis. Upon exposure to pathological stimuli, they can take on alternative phenotypes that, among others, have been described as macrophage like, or foam cells. VSMC foam cells make up >50% of all arterial foam cells and have been suggested to retain an even higher proportion of the cell stored lipid droplets, further leading to apoptosis, secondary necrosis, and an inflammatory response. However, the mechanism of VSMC foam cell formation is still unclear. Here, it is identified that mec

Foam cell-derived exosomes: Messengers between atherosclerosis and microglia.

Atherosclerosis (AS) is an independent risk factor for vascular cognitive impairment (VCI). Zhang et al.1 revealed that foam cell-derived exosomes transmit redox imbalance and metabolic defects to microglia via the miR-101-3p-Nrf2-Slc2a1 axis, causing microglial dysfunction and exacerbating VCI, uncovering a peripheral-brain link and potential therapeutic targets for AS-induced VCI.

Signaling events in pathogen-induced macrophage foam cell formation.

Macrophage foam cell formation is a key event in atherosclerosis. Several triggers induce low-density lipoprotein (LDL) uptake by macrophages to create foam cells, including infections with Porphyromonas gingivalis and Chlamydia pneumoniae, two pathogens that have been linked to atherosclerosis. While gene regulation during foam cell formation has been examined, comparative investigations to identify shared and specific pathogen-elicited molecular events relevant to foam cell formation are not well documented. We infected mouse bone marrow-derived macrophages with P. gingivalis or C. pneumonia

Inflammation-induced foam cell formation in chronic inflammatory disease.

Atherosclerosis is the leading cause of cardiovascular disease and is both a metabolic and inflammatory disease. Two models describe early events initiating atherosclerotic plaque formation, whereby foam cells form in response to hyperlipidaemia or inflammation-associated stimuli. Although these models are inextricably linked and not mutually exclusive, identifying the unique contribution of each in different disease settings remains an important question. Circulating monocytes are key mediators of atherogenesis in both models as precursors to lipid-laden foam cells formed in response to eithe

The pro-fibrotic and anti-inflammatory foam cell macrophage paradox.

The formation of foamy macrophages by sequestering extracellular modified lipids is a key event in atherosclerosis. However, there is controversy about the effects of lipid loading on macrophage phenotype, with in vitro evidence suggesting either pro- or anti-inflammatory consequences. To investigate this in vivo we compared the transcriptomes of foamy and non-foamy macrophages that accumulate in experimental subcutaneous granulomas in fat-fed ApoE null mice or normal chow-fed wild-type mice, respectively. Consistent with previous studies in peritoneal macrophages from LDL receptor null mice (

Mechanical properties of open-cell foam synthetic thoracic vertebrae.

This study presents comprehensive morphological and mechanical properties (static, dynamic) of open-cell rigid foams (Pacific Research Laboratories Inc. Vashon, WA) and a synthetic vertebral body derived from each of the foams. Synthetic vertebrae were comprised of a cylindrical open-cell foam core enclosed by a fiberglass resin cortex. The open-cell rigid foam was shown to have similar morphology and porosity as human vertebral cancellous bone, and exhibited a crush or fracture consolidation band typical of open-celled materials and cancellous bone. However, the foam material density was 40%

Lactate promotes macrophage-derived foam cell formation and atherosclerosis.

BACKGROUND AND AIMS: Diabetes accelerates atherosclerosis progression by disrupting multiple metabolic pathways. However, the role and mechanisms of lactate-a glycolytic byproduct significantly elevated in diabetic patients-in atherosclerosis remain poorly understood. We hypothesized that lactate promotes atherosclerosis by regulating macrophage foam cell formation through histone lactylation. METHODS: We evaluated the association between lactate levels, lipid metabolism disorders, and atherosclerotic progression using clinical samples and mouse models. The thoracic aortas of atherosclerotic

High-Expansion Open-Cell Polylactide Foams Prepared by Microcellular Foaming Based on Stereocomplexation Mechanism with Outstanding Oil-Water Separation.

Biodegradable polylactic acid (PLA) foams with open-cell structures are good candidates for oil-water separation. However, the foaming of PLA with high-expansion and uniform cell morphology by the traditional supercritical carbon dioxide microcellular foaming method remains a big challenge due to its low melting strength. Herein, a green facile strategy for the fabrication of open-cell fully biodegradable PLA-based foams is proposed by introducing the unique stereocomplexation mechanism between PLLA and synthesized star-shaped PDLA for the first time. A series of star-shaped PDLA with eight ar

Platelet extracellular vesicles drive monocyte differentiation into inflammatory foam cells in peripheral arterial disease.

Inflammatory foam cells (IFC) derived from monocytes drive inflammation within the atherosclerotic plaque. During their differentiation they utilise scavenger receptors to assimilate oxidised low-density lipoproteins (OxLDL), storing the modified protein and lipid cargo in lysosomal storage depots and lipid droplets. The cues supporting their differentiation, and interventions that reverse their inflammatory phenotype are however, poorly understood. Here we show that in age and sex matched healthy donors, CD14+ /CD16- (classical), but not CD14+/CD16+ (non-classical and intermediate) monocytes

Increased atherosclerosis and expression of inflammarafts in macrophage foam cells in AIBP-deficient mice.

Atherosclerotic lesions comprise different populations of macrophages, including lipid-laden macrophage foam cells and non-foamy, inflammatory macrophages, which play distinct roles in disease progression. Non-foamy macrophages express higher levels of inflammarafts – enlarged, cholesterol-rich lipid rafts hosting assemblies of inflammatory receptors – compared to foam cells in atherosclerotic lesions of Ldlr−/− mice. Apolipoprotein A-I binding protein (AIBP) has been shown to control lipid raft dynamics. This study investigated the effect of systemic AIBP deficiency on inflammaraft expression

Contribution of monocyte-derived macrophages and smooth muscle cells to arterial foam cell formation.

Smooth muscle cells (SMCs) are the main cell type in intimal thickenings and some stages of human atherosclerosis. Like monocyte-derived macrophages, SMCs accumulate excess lipids and contribute to the total intimal foam cell population. In contrast, apolipoprotein (Apo)E-deficient and LDL receptor-deficient mice develop atherosclerotic lesions that are macrophage- as opposed to SMC-rich. The lesser contribution of SMCs to lesion development in these mouse models has distracted attention away from the importance of SMC cholesterol homeostasis in the artery wall. Intimal SMCs accumulate excess
