How to combine the exigencies of histopathological and forensic-genetic investigation: a preliminary study on the use of medicolegal FFPE samples for DNA analysis.

In forensic autopsies, tissue samples are routinely collected, fixed in formalin, and embedded in paraffin (FFPE) to ensure long-term preservation for histological examination. In some cases, these specimens may represent the only remaining biological material available for postmortem genetic analysis. However, the use of FFPE samples in forensic genetics poses significant challenges, as formalin fixation compromises DNA integrity and reduces the likelihood of obtaining complete genetic profiles. Moreover, relatively few experimental studies in the scientific literature have addressed the inte

Genetic analysis of 22 pigmentation-associated SNPs in the Pashtun population of Khyber Pakhtunkhwa, Pakistan: Implications for forensic DNA phenotyping.

The prediction of externally visible characteristics (EVCs) from DNA, known as forensic DNA phenotyping (FDP), has emerged as a powerful tool in forensic investigations. The HIrisPlex system, comprising 24 SNPs for eye and hair color prediction, has been extensively validated in European populations but shows reduced accuracy in South Asian populations. This study genotyped 22 pigmentation-associated SNPs using the SNaPshot minisequencing platform in 146 unrelated Pashtun individuals from Khyber Pakhtunkhwa (KP), Pakistan, to evaluate their forensic utility and phenotypic association in this u

Free-ranging Eurasian otter (Lutra lutra) with rare osteoblastic osteosarcoma: a forensic and genetic investigation from central Italy.

BACKGROUND: The Eurasian otter Lutra lutra experienced a dramatic decline in Europe during the 20th century. The urbanization of human population following the abandonment of countryside as well as reintroduction plans led to a recent recovery in many areas. CASE PRESENTATION: We investigated both cause of death and genetic origins of a recently discovered dead otter specimen in Tuscany, a region of central Italy where the species disappeared three decades ago. The necropsy was performed using a forensic approach. We also sequenced three gene fragments [cytochrome-oxidase subunit I (COXI), cyt

Benefits and Limitations Associated with the Use of Alternative Biological Matrices in Forensic Toxicology, as Well as Genetic Markers of Poisoning and Psychoactive Substance Abuse.

In forensic toxicology, the analysis of blood and urine is regarded as the so-called "gold standard." However, when forensic experts are confronted with advanced postmortem changes, it becomes necessary to secure and examine alternative matrices. Suitable specimens in cases of advanced putrefactive decomposition may include: skin appendages (hair and nails), bones, bone marrow, cartilage tissue, teeth, antemortem fingerprints, cerebrospinal fluid, vitreous humor, breast milk, meconium, placenta and umbilical cord tissue, oral fluid, sweat, and other evidentiary materials. These tissues, owing

The contribution of seawater in DNA degradation: Comparison of genetic results from skeletal remains affected by different taphonomic conditions in forensic contexts.

In the field of forensic genetics, the examination of compromised biological samples is a common investigative challenge. It is crucial to thoroughly assess the thanatological and taphonomic events affecting human samples for genetic analysis, including the environment surrounding the corpses or the skeletal remains, which, in the present cases, is represented by seawater. This study aims to compare different conditions to determine the extent to which cadaveric submersion in seawater affects endogenous DNA, by evaluating the degree of degradation and nucleic acid quantity. The samples selecte

Genetic origin and forensic analysis of han populations from four districts in Shanghai, China, based on Y-chromosome STR and SNP markers.

Y-short tandem repeats (STRs) and Y-single nucleotide polymorphisms (SNPs) on the Y chromosome are genetic markers unique to men. They are widely used in identification, forensic medicine and paternal lineage tracing. Because of their strict paternal genetic characteristics, they have become a powerful means to trace the evolution of paternal lineages and populations. In this study, 38 Y-STR loci and 250 Y-SNP loci were genotyped, and a dataset was constructed for Qingpu Han (n = 396), Songjiang Han (n = 386), Jinshan Han (n = 384), and Fengxian Han (n = 372) populations in Shanghai. FIGG at 5: An update on U.S. public perspectives on forensic investigative genetic genealogy five years after its introduction to criminal investigations.

In 2018, after law enforcement announced it had used a technique called forensic investigative genetic genealogy (FIGG) to identify the Golden State Killer, we conducted a U.S. general population survey and found most respondents supported using FIGG to solve violent crimes. Since then, FIGG has helped close hundreds of criminal cases, but it also has weathered controversies. On FIGG's fifth anniversary, we conducted an expanded, follow-up survey with U.S.-based participants to determine if public opinion had changed and found continued support for FIGG across most applications. The same propo

Forensic DNA typing in China.

In the field of forensic genetics, essential developmental impulses come from the advances of the molecular biology and human genome projects. This paper overviews existing technologies for forensic genetics in China and gives a perspective of forensic DNA analysis. In China, work has been done in the development of blood group serology of the conventional markers. Forensic scientists in China also contributed to the progress of DNA analysis by the validation of numerous test methods and by optimization of these methods. During these years, forensic DNA analysis in China has experienced tremen

Dangers resulting from DNA profiling of biological materials derived from patients after allogeneic hematopoietic stem cell transplantation (allo-HSCT) with regard to forensic genetic analysis.

The study documents the risk that comes with DNA analysis of materials derived from patients after allogeneic hematopoietic stem cell transplantation (allo-HSCT) in forensic genetics. DNA chimerism was studied in 30 patients after allo-HSCT, based on techniques applied in contemporary forensic genetics, i.e. real-time PCR and multiplex PCR-STR with the use of autosomal DNA as well as Y-DNA markers. The results revealed that the DNA profile of the recipient's blood was identical with the donor's in the majority of cases. Therefore, blood analysis can lead to false conclusions in personal identi

Role of forensic pathologists in mass disasters.

The forensic pathologist has always had a central role in the identification of the dead in every day practice, in accidents, and in disasters involving hundreds or thousands of victims. This role has changed in recent years, as advances in forensic odontology, genetics and anthropology have improved the chances of identifying victims beyond recognition. According to the Interpol DVI Guide, fingerprints, dental examination and DNA are the primary identifiers, and this has given new emphasis to the role of the forensic pathologist as the leader of a multidisciplinary team of experts in a disast

[Expert Consensus on Cause of Death Determinations in Antipsychotics-Related Deaths with Negative Autopsy Findings].

Antipsychotics are a category of psychotropic medications that often require long-term or even lifelong administration, and they are also routinely screened in forensic examinations. Clinically, the use of antipsychotics can usually cause observed signs, such as QT interval prolongation, supraventricular tachycardia and conduction block. A relatively high proportion of deaths among individuals taking these medications present as negative autopsies, making cause of death determination in cases involving antipsychotics particularly unique and challenging. At present, due to the lack of effective

pSTRminer: integrated bioinformatic software for genome-wide identification and population-scale evaluation of polymorphic short tandem repeats.

Animal forensic genetics plays a critical role in criminal investigations by providing crucial evidence through domestic animal individualization and wildlife species identification. While human forensic genetics benefits from standardized short tandem repeats (STR) genotyping systems, animal forensic applications encounter significant challenges, including the limited availability of validated STR markers, the prevalence of error-prone dinucleotide STRs (di-STRs), and insufficient integration of population data. To address these challenges, we developed pSTRminer, an integrated bioinformatic

Evaluation of the MiSeq FGx system for use in forensic casework.

Capillary electrophoresis (CE) is widely used in forensic genetics to study short tandem repeats (STRs). Recently, next-generation sequencing (NGS) platforms have facilitated the development of new strategies for forensic DNA typing. Several studies have shown that NGS successfully analyzes challenging samples. However, because NGS is complicated and time-consuming, it remains unclear whether NGS platforms offer significant advantages over CE for all forensic cases. Here, the MiSeq FGx system was used to test some cases that had previously been analyzed using CE. These cases included paternity

The "mystery" of the train station: The importance of a forensic approach to a rare case of survival after a train-pedestrian collision.

In the early morning a 28-year-old man was found lying on the tracks of a railway station with head injuries and fractures of the cervical spine resulting in permanent quadriplegia. He was in a club about 1 km away until about 2 h earlier and did not have any recollection of what could have happened. Was he the victim of an assault, did he fall down or was he hit by a passing train? The solution to this "mystery" came from a forensic evaluation that included the forensic branches of pathology, chemistry, merceology and genetics as well as the scene evaluation. Through these different steps, th
