Influence of protein formulation and carrier solution on asymmetrical flow field-flow fractionation: a case study of the plant-produced recombinant anthrax protective antigen pp-PA83.

Asymmetrical flow field-flow fractionation (afFFF) was used to investigate the properties of a plant-produced anthrax toxin protective antigen, pp-PA83. The afFFF fractogram consisted of two main peaks with molar masses similar to the molecular mass of pp-PA83 monomer. afFFF carrier solutions strongly influenced the ratio and the intensity of the two main peaks. These differences indicate that conformation changes in the pp-PA83 molecule occurred during the afFFF analysis. Similar fractograms were obtained for different pp-PA83 formulations when the afFFF carrier solution and the protein formu

Colloidal mercury (Hg) distribution in soil samples by sedimentation field-flow fractionation coupled to mercury cold vapour generation atomic absorption spectroscopy.

Diverse analytical techniques are available to determine the particle size distribution of potentially toxic elements in matrices of environmental interest such as soil, sediments, freshwater and groundwater. However, a single technique is often not exhaustive enough to determine both particle size distribution and element concentration. In the present work, the investigation of mercury in soil samples collected from a polluted industrial site was performed by using a new analytical approach which makes use of sedimentation field-flow fractionation (SdFFF) coupled to cold vapour generation ele

A comprehensive study on the relationship between structure and antioxidant activity of natural deep eutectic solvent extracted polysaccharides from Chinese yam peel using asymmetrical flow field-flow fractionation.

In this study, an ultrasound-assisted natural deep eutectic solvents (NADES) combined with enzymatic deproteinization was developed for extracting Chinese Yam peel polysaccharide (CYPP) from two varieties. The result demonstrated that the viscosity of NADES was the primary factor affecting the extraction rate of CYPP. Under optimized extraction conditions, the CYPP extraction rate obtained by NADES was 2.42-fold higher than that obtained by the water extraction method. Moreover, the relationship between the structure and antioxidant activity of CYPP was investigated using asymmetrical flow fie

Lipidomic alterations in lipoproteins of patients with mild cognitive impairment and Alzheimer's disease by asymmetrical flow field-flow fractionation and nanoflow ultrahigh performance liquid chromatography-tandem mass spectrometry.

Alzheimer's disease (AD) is an irreversible neurodegenerative disorder with the clinical symptom of the progressive loss of cognitive function and mild cognitive impairment (MCI) is a translational state between cognitive changes of normal aging and AD. Lipid metabolism and pathogenesis of Alzheimer's disease (AD) are closely linked. Despite obviously discrete lipidome constitutions across lipoproteins, lipidomic approaches of AD has been mostly conducted without considering lipoprotein-dependent alterations. This study introduces a combination of asymmetrical flow field-flow fractionation (AF

Elemental ratios for characterization of quantum-dots populations in complex mixtures by asymmetrical flow field-flow fractionation on-line coupled to fluorescence and inductively coupled plasma mass spectrometry.

Separation and identification of nanoparticles of different composition, with similar particle diameter, coexisting in heterogeneous suspensions of polymer-coated CdSe/ZnS quantum dots (QDs) have been thoroughly assessed by asymmetric flow field-flow fractionation (AF4) coupled on-line to fluorescence and inductively coupled plasma mass spectrometry (ICPMS) detectors. Chemical characterization of any previously on-line separated nanosized species was achieved by the measurement of the elemental molar ratios of every element involved in the synthesis of the QDs, using inorganic standards and ex

Cascade Filtration With PCR Detection and Field-Flow-Fractionation Online With ICP-MS for the Characterization of DNA Interaction With Suspended Particulate Matter.

The variety of applied antibiotics in animal and human medicine results in the release, development, and spread of relevant numbers of antibiotic resistance genes (ARGs) in the environment. The majority of ARGs are present in intracellular forms (in bacteria). Neglected aspects are extracellular variants of ARGs (eARGs) and their fragments, which have been detected in surface-water samples and sediments. The stability of eARGs is expected to be low; however, binding to particulate matter is likely to improve their stability and also affect their transport and dissemination behavior. Few studie

Asymmetrical flow field-flow fractionation and multi-angle laser light scattering: A new analytical approach for the characterisation of insect protein aggregation/polymerisation after heat treatment of Tenebrio molitor larvae.

Understanding the structural modifications of insect proteins during the transformation processes used for extract preparation is essential for optimising their functionalities and obtaining high added-value proteins. From this perspective and in addition to classical analytical approaches, we developed an original methodology based on the implementation of Asymmetrical Flow Field-Flow Fractionation and Multi-Angle Laser Light Scattering (A4F-MALLS) coupling to quantify and characterise the aggregation/polymerisation phenomena of Tenebrio molitor larvae proteins after heat treatment (from 65 t

Insight into the formation and biological effects of natural organic matter corona on silver nanoparticles in water environment using biased cyclical electrical field-flow fractionation.

Natural organic matter (NOM) readily interacts with nanoparticles, leading to the formation of NOM corona structures on their surface. NOM corona formation is closely related to the surface coatings and bioavailability of nanoparticles. However, the mechanism underlying NOM corona formation on silver nanoparticles (AgNPs) remains largely unknown due to the lack of effective analytical methods for identifying the changes in the AgNP surface. Herein, the separation ability of biased cyclical electrical field-flow fractionation (BCyElFFF) for same-sized polyvinyl pyrrolidone-coated and poly(ethyl

Asymmetric-flow field-flow fractionation for measuring particle size, drug loading and (in)stability of nanopharmaceuticals. The joint view of European Union Nanomedicine Characterization Laboratory and National Cancer Institute - Nanotechnology Characterization Laboratory.

Asymmetric-flow field-flow fractionation (AF4) has been recognized as an invaluable tool for the characterisation of particle size, polydispersity, drug loading and stability of nanopharmaceuticals. However, the application of robust and high quality standard operating procedures (SOPs) is critical for accurate measurements, especially as these complex drug nanoformulations are most often inherently polydisperse. In this review we describe a unique international collaboration that lead to the development of a robust SOP for the measurement of physical-chemical properties of nanopharmaceuticals

Resolving measurement artifacts of field-flow fractionation coupled to conventional dynamic light scattering detection by using phase and spatially-resolved dynamic light scattering to improve size accuracy.

The use of dynamic light scattering (DLS) as an online detector in asymmetrical flow field-flow fractionation (AF4 ‒ DLS) for size characterization is known to introduce measurement bias for particles with hydrodynamic size exceeding ∼100 nm. This study demonstrates that the accuracy of hydrodynamic size measurements in AF4 ‒ DLS is significantly improved by applying advanced phase and spatially-resolved DLS (PhaSR-DLS) detection. PhaSR-DLS overcomes two limitations of conventional DLS: (i) overestimation of particle size due to 'number fluctuations' at low particle concentrations, causing 'U-

Separation and preparation of nanoparticles of urban dust for biological studies.

Nanoparticles (NPs) of urban dust pose a potential threat to public health. Nevertheless, this issue remains largely unexplored due to a lack of biological research related to these NPs. This may be attributed to the complexity of the separation, characterization, analysis, and subsequent preparation of NPs of urban dust for biological studies. In the present work, the methodology for the separation and preparation of NPs of urban dust for biological assays has been developed. The isolation of NPs from bulk samples of urban dust has been carried out using coiled tube field-flow fractionation,

Flash flow pyrolysis: mimicking flash vacuum pyrolysis in a high-temperature/high-pressure liquid-phase microreactor environment.

Flash vacuum pyrolysis (FVP) is a gas-phase continuous-flow technique where a substrate is sublimed through a hot quartz tube under high vacuum at temperatures of 400-1100 °C. Thermal activation occurs mainly by molecule-wall collisions with contact times in the region of milliseconds. As a preparative method, FVP is used mainly to induce intramolecular high-temperature transformations leading to products that cannot easily be obtained by other methods. It is demonstrated herein that liquid-phase high-temperature/high-pressure (high-T/p) microreactor conditions (160-350 °C, 90-180 bar) employi

AF4-UV-MALS-ICP-MS/MS, spICP-MS, and STEM-EDX for the Characterization of Metal-Containing Nanoparticles in Gas Condensates from Petroleum Hydrocarbon Samples.

The coupling of flow field flow fractionation (FlFFF) with ICP-MS/MS for the fractionation and analysis of natural nanoparticles (NPs) in environmental samples is becoming more popular. However, the applicability of this technique to nonaqueous samples, such as gas condensates from petroleum hydrocarbon samples, has not been reported yet. In this study, an asymmetric flow-field flow fractionation (AF4) system coupled with UV and MALS detectors has been optimized to perform the fractionation of natural NPs present in a gas condensate sample, using THF as the carrier liquid. Prior to this, STEM

Validation of an FFF-MALS Method to Characterize the Production and Functionalization of Outer-Membrane Vesicles for Conjugate Vaccines.

With the ongoing development of conjugate vaccines battling infectious diseases, there is a need for novel carriers. Although tetanus toxoid and CRM197 belong to the traditional carrier proteins, outer-membrane vesicles (OMVs) are an excellent alternative: in addition to their size, OMVs have self-adjuvanting properties due to the presence of genetically detoxified lipopolysaccharide (LPS) and are therefore ideal as a vaccine component or antigen carrier. An essential aspect of their development for vaccine products is characterization of OMVs with respect to size and purity. We report on the

Using inductively coupled plasma mass spectrometry for studying the association of gold nanoparticles with compounds of clinical interest.

The widespread use of gold nanoparticles (AuNPs) in biomedical and analytical applications arises from their versatile surface functionalization with clinically relevant compounds. Investigating their binding behavior using simple model compounds of clinical interest is essential for gaining fundamental insight into more complex interaction systems. Herein, single particle inductively coupled plasma mass spectrometry (SP-ICP-MS) and flow field-flow fractionation (FlFFF) coupled with an inductively coupled plasma mass spectrometry (ICP-MS) were employed to investigate the binding and aggregatio

In-Depth Characterization of PEGylated Liposomes: Using AF4 and HPLC-CAD in Tandem as a Strategy for Composition Assessment and Postinsertion Optimization.

The surface of nanoparticles for pharmaceutical use is crucial as it is the first interface that interacts with the biological environment in the body and therefore determines the fate of the nanoparticle itself. Aware of its key role, regulatory agencies urgently require rigorous quality control of the surface, including polyethylene glycol (PEG) coatings, a component widely used in nanomedicine, which is also present in recent vaccines against SARS-CoV-2, to ensure efficacy and safety. Building on a review of the literature and experimental testing, this study proposes a workflow to overcome

Evaluation of a combined fractionation and speciation approach for study of size-based distribution of organotin species on environmental colloids.

Results relating to the first original application of an analytical approach combining asymmetric flow field-flow fractionation (As-Fl-FFF) with multi-detection and chemical speciation for determination of organotins in a landfill leachate sample are presented. The speciation analysis involved off-line head-space solid-phase microextraction (HS-SPME)-gas chromatography with pulsed-flame photometric detection (GC-PFPD) performed after three consecutive collections of five different fractions of interest from the As-Fl-FFF system and cross-flow part (assumed to be representative of the <10 kDa p

A chromatographic approach for investigating the proliferation ability of native Saccharomyces cerevisiae yeast strains under varying temperatures and ethanol concentrations.

Native yeast strains have been proved to be of great importance for food industry. In the present work, two different Saccharomyces cerevisiae native yeast strains isolated from the must from Moschofilero and Agiorgitiko varieties, respectively, were studied in order to estimate the influence of temperature and ethanol concentration on their proliferation ability via asymmetric flow field-flow fractionation (AsFlFFF) technique. The growth rate of the yeast strains, was directly linked to the biomass production under these conditions and was finally investigated via the ability of AsFlFFF to se

Sedimentation studies on human amylin fail to detect low-molecular-weight oligomers.

Sedimentation velocity experiments show that only monomers coexist with amyloid fibrils of human islet amyloid-polypeptide. No oligomers containing <100 monomers could be detected, suggesting that the putative toxic oligomers are much larger than those found for the Alzheimer's peptide, Abeta(1-42).

Magnetically Sculpted Microfluidics for Continuous-Flow Fractionation of Cell Populations by EpCAM Expression Level.

Continuous-flow separation of magnetically labeled cells according to surface-marker expression levels is increasingly needed to study phenotypic heterogeneity and support downstream assays. Here, we present a microfluidic platform that uses spatially engineered soft magnetic strips (SMS) to sculpt lateral magnetic deflection fields for quantitative, label-guided cell fractionation. Under a uniform bias field, the SMS generates controllable magnetic gradients within the microchannel, producing distinct lateral velocities among EpCAM-labeled tumor cells that carry different Dynabead loads, whic
