Acute early-life stress results in premature emergence of adult-like fear retention and extinction relapse in infant rats.

Recent studies have shown that chronic early life stress results in precocious expression of the adult-like phenotype of fear retention and inhibition. However, it is unknown whether the experience of acute early trauma has the same effects as exposure to chronic early stress. In the present study, a 24-hr period of maternal deprivation on postnatal day (P) 9 was used as an acute early life stressor. In infancy (P16-17), maternally deprived and standard-reared rats were conditioned to fear a noise paired with shock. In Experiments 1 and 2, fear to the noise was then extinguished before rats we

Freezing reaction in panic disorder patients associated with anticipatory anxiety.

BACKGROUND: Anticipatory anxiety can be described as a conditioned response with a defensive posture of freezing and autonomic activation. The purpose of this study was to assess the postural control analysis and autonomic activation in panic disorder (PD) patients presented with visual stimuli. METHODS: PD patients (n=29) and healthy controls (n=27) stood on a force platform while viewing a series of anxiogenic, mutilation, and neutral pictures. Skin conductance responses and the displacements of the center of pressure were measured. RESULTS: Overall, the PD patients demonstrated significan

Posterior parietal cortex mediates fear renewal in a novel context.

The return of fear following extinction therapy is an important issue associated with the treatment of many fear-related disorders. Fear renewal is a suitable model, with which context-dependent modulation of the fear response can be examined. In this model, any context outside of an extinction context (e.g., novel or familiar contexts) could evoke relapse of the fear response. However, brain regions associated with context-dependent modulation are not fully understood. The posterior parietal cortex (PPC) is considered a center for integrating multisensory information and making decisions. To

The association between serotonin transporter availability and the neural correlates of fear bradycardia.

Susceptibility to stress-related psychopathology is associated with reduced expression of the serotonin transporter (5-HTT), particularly in combination with stress exposure. Aberrant physiological and neuronal responses to threat may underlie this increased vulnerability. Here, implementing a cross-species approach, we investigated the association between 5-HTT expression and the neural correlates of fear bradycardia, a defensive response linked to vigilance and action preparation. We tested this during threat anticipation induced by a well-established fear conditioning paradigm applied in bo

Post-weaning housing conditions influence freezing during contextual fear conditioning in adult rats.

The present study aimed to investigate the influence of housing conditions on contextual fear memory malleability. Male Wistar rats were housed in enriched, standard, or impoverished conditions after weaning and remained in these conditions throughout the entire experiment. After six weeks into those housing conditions, all animals underwent a 3-day protocol including contextual fear conditioning (day 1), memory reactivation followed by systemic administration of midazolam or vehicle (day 2), and a retention test (day 3). Percentage freezing was used as a behavioral measure of contextual fear.

Remote fear memory is sensitive to reconditioning.

Exposure of some individuals to recurring traumatic events from the same perpetrator or situation, such as during child abuse or domestic violence, is quite prevalent. Studies have shown that the number of traumatic events experienced is positively related to the severity of post-traumatic stress disorder and other mental disorders. Using a contextual fear conditioning (Cond1) and reconditioning (Cond2) paradigm, which were separated by either 1 or 35 days, we examined fear responses to immediate extinction and retrieval-extinction procedures after repeated fear conditioning stress. Proven cardiac changes during death-feigning (tonic immobility) in rabbits (Oryctolagus cuniculus).

Tonic immobility (TI) is a response to fear or threat by remaining motionless, principally when attacked by predators from which there is no possibility of escape. Thus, here we demonstrate a way of easily reproducing this phenomenon in a laboratory setting and characterize the cardiac electromechanical alterations during TI. We observed a significant decrease in heart rate (HR) and changes of rhythm in electrocardiogram during TI in rabbits. Echocardiogram showed a significant increase in the left ventricle chamber diameter during systole and a consequent decrease in fractional shortening and

Passive Behavioral Patterns in Symptomatic WAG/Rij Rats: Dissociation From Absence Epilepsy and Sex-Dependent Link Between Early Fear Coping and Post-Stress Freezing.

BACKGROUND: Acute stress exposure can lead to delayed behavioral changes, which are widely regarded as core endophenotypes of post-traumatic stress disorder (PTSD). Identifying pre-existing behavioral traits that predict later maladaptive responses is crucial. We investigated whether early fear-related passive coping behavior during an active avoidance task predicts subsequent freezing reactions in a safe, stimulus-free environment. This context-inappropriate fear pattern may model individual vulnerability to PTSD-like symptoms. We used Wistar Albino rats from Rijswijk (WAG/Rij rats), which ar

Improvement in γ-hydroxybutyrate-induced contextual fear memory deficit by systemic administration of NCS-382.

Low, nonsedative doses of γ-hydroxybutyric acid (GHB) produce short-term anterograde amnesia in humans and memory impairments in experimental animals. We have previously shown that acute systemic treatment of GHB in adolescent female rats impairs the acquisition, but not the expression, of contextual fear memory while sparing both the acquisition and the expression of auditory cued fear memory. In the brain, GHB binds to specific GHB-binding sites as well as to γ-aminobutyric acid type B (GABAB) receptors. Although many of the behavioral effects of GHB at high doses have been attributed to its

Lateralizing value of unilateral relative ictal immobility in patients with refractory focal seizures--Looking beyond unilateral automatisms.

PURPOSE: Ictal motor phenomena play a crucial role in the localization of seizure focus in the management of refractory focal epilepsy. While the importance of unilateral automatisms is well established, little attention is paid to the contralateral relatively immobile limb. In cases where automatisms mimic clonic or dystonic movements and in the absence of previously well-established signs, unilateral relative ictal immobility (RII) is potentially useful as a lateralizing sign. This study was carried out to examine the lateralizing value of this sign and to define its characteristics among pa

Expression of renewal is dependent on the extinction-test interval rather than the acquisition-extinction interval.

A recent finding suggested that when extinction occurs shortly after acquisition, renewal of an extinguished fear response (fear-potentiated startle) to a light conditioned stimulus (CS) is diminished (Myers, Ressler, & Davis, 2006). The present study attempted to extend this finding using a white-noise CS and freezing as the behavioral measure of fear. In Experiments 1A and 1B, we observed renewal whether extinction occurred 10 min or 24 hr after acquisition. In contrast, renewal was not observed if test occurred 10 min after extinction (Experiment 2). Experiment 3 demonstrated that expressio

Heart rate changes during freezing of gait in patients with Parkinson's disease.

Freezing of gait (FOG) is one of the most disabling symptoms that affect patients with Parkinson's disease (PD). Although the pathophysiology underlying FOG largely remains an enigma, several lines of evidence suggest that the autonomic nervous system might be involved. To this end, we tested the hypothesis that heart rate (HR) increases during FOG and, further, that HR increases just before FOG. To evaluate these hypotheses, 15 healthy older adults, 10 patients with PD who experienced FOG, and 10 patients who did not were studied. Patients with PD were tested during their "off" medication sta

Effects of exercise and environmental complexity on deficits in trace and contextual fear conditioning produced by neonatal alcohol exposure in rats.

In rodents, voluntary exercise and environmental complexity increases hippocampal neurogenesis and reverses spatial learning and long-term potentiation deficits in animals prenatally exposed to alcohol. The present experiment extended these findings to neonatal alcohol exposure and to delay, trace, and contextual fear conditioning. Rats were administered either 5.25 g/kg/day alcohol via gastric intubation or received sham-intubations (SI) between Postnatal Day (PD) 4 and 9 followed by either free access to a running wheel on PD 30-41 and housing in a complex environment on PD 42-72 (wheel-runn

A panic attack-like unusual stress reaction.

Ever since the seminal studies of Hans Selye, activation of hypothalamus-pituitary-adrenal (HPA) axis is emblematic of stress. Consequently, the lack of HPA axis responses following the undisputable psychological stress of a panic attack stands out as one of the most intriguing findings of contemporary psychiatry. On the other hand, the defensive behaviors and aversive emotions produced by stimulation of the dorsal periaqueductal gray matter (DPAG) have been proposed as a model of panic attacks. Therefore, we examined whether the plasma levels of 'stress hormones' corticotropin and prolactin s

Gastrin-releasing peptide is essential for generalization of auditory conditioned fear under stress.

Fear generalization, which allows animals to respond adaptively to cues similar to original threatening ones, is generally beneficial for survival. However, an inability to distinguish between threat and safety, leading to the overgeneralization of fear to non-threatening stimuli, is maladaptive and is implicated in anxiety disorders such as post-traumatic stress disorder (PTSD). The neuropeptide gastrin-releasing peptide (GRP) is known to modulate fear memory under stress, yet its role in response to intense aversive stimuli remains less understood. In this study, we used GRP knockout (Grp-/-

Conditioned Contextual Freezing is A Neurobehavioral Biomarker of Axonal Injury Indicated by Reduced Fractional Anisotropy in A Mouse Model of Blast-Induced Mild Traumatic Brain Injury.

Mild traumatic brain injury (TBI) is an important public health problem generated by closed head injury. This study is focused on the impact of blast-induced mild TBI on auditory trace and delay fear conditioning, models of declarative and non-declarative memory, respectively, and the correlation of conditioned freezing and fractional anisotropy, a measure of axonal state. A supersonic helium pressure wave was generated by a shock tube to blast 8-week-old male mice on Day 1 for 1.4 msec with an incident pressure of 16 psi, corresponding to a reflected pressure of 56.9 psi at the mouse head. Intermittent hypoxia improves behavioral and adrenal gland dysfunction induced by posttraumatic stress disorder in rats.

Nonpharmacological treatments of stress-induced disorders are promising, since they enhance endogenous stress defense systems, are free of side effects, and have few contraindications. The present study tested the hypothesis that intermittent hypoxia conditioning (IHC) ameliorates behavioral, biochemical, and morphological signs of experimental posttraumatic stress disorder (PTSD) induced in rats with a model of predator stress (10-day exposure to cat urine scent, 15 min daily followed by 14 days of stress-free rest). After the last day of stress exposure, rats were conditioned in an altitude

Behavioral tagging in infant rats.

Recent studies have shown that exposure to a novel environment may stabilize the persistence of weak memories, a phenomenon often attributed to a process referred to as "behavioral tagging." While this phenomenon has been repeatedly demonstrated in adult animals, no studies to date have examined whether it occurs in infant animals, which is surprising given that infants exhibit an impaired ability to form long-term memories (LTMs). In the present study, infant (i.e., postnatal day (P) 17) rats were placed in a context and repeatedly shocked. Infant rats given brief open field exposure 1 h, but

Social buffering-mediated fear response modulation: Investigating synaptic activity changes in the prelimbic cortex of the medial prefrontal cortex.

Social buffering is a phenomenon in which stress is ameliorated when the subject is accompanied by a conspecific animal while being exposed to distressing stimuli. Although evidence indicates that the prelimbic cortex of the medial prefrontal cortex (mPFC PL) plays a prominent role in social buffering, its underlying synaptic mechanisms remain unclear. In this study, we observed synaptic activity changes in the mPFC PL using whole-cell patch-clamp recording and investigated the relationship between synaptic activity and social buffering under fear-exposed conditions. In the passive avoidance t

Targeting fear memories: Examining pharmacological disruption in a generalized fear framework.

Labilization-reconsolidation, which relies on retrieval, has been considered an opportunity to attenuate the negative aspects of traumatic memories. A therapeutic strategy based on reconsolidation blockade is deemed more effective than current therapies relying on memory extinction. Nevertheless, extremely stressful memories frequently prove resistant to this process. Here, after inducing robust fear memory in mice through strong fear conditioning, we examined the possibility of rendering it susceptible to pharmacological modulation based on the degree of generalized fear (GF). To achieve this
