Light is essential for life as it provides the energy for the growth of plants and photosynthetic microorganisms. Plants use the energy from light to fix carbon dioxide and produce oxygen, and their photosynthetic apparatus includes sensory systems that optimize light detection. For example, light is a signal that regulates plant growth and promotes the development and orientation of leaves so that they can optimize the capture of the light energy for photosynthesis. Light is also an environmental signal that is used by non-photosynthetic organisms for their orientation, to obtain food and cap

Fungal sexual reproduction requires complex cellular differentiation processes of hyphal cells. The plant pathogenic fungus Fusarium graminearum produces fruiting bodies called perithecia via sexual reproduction, and perithecia forcibly discharge ascospores into the air for disease initiation and propagation. Lipid metabolism and accumulation are closely related to perithecium formation, yet the molecular mechanisms that regulate these processes are largely unknown. Here, we report that a novel fungal specific bZIP transcription factor, F. graminearum perithecium overproducing 1 (Fpo1), plays

Ascochyta rabiei produces and accumulates one of the well-known fungal polyketides, 1,8-dihydroxynaphthalene-melanin pigment (DHN-melanin), in asexual and sexual fruiting bodies. Degenerate PCR primers were used to isolate an ArPKS1 of A. rabiei encoding a polypeptide with high similarity to polyketide synthase (PKS) involved in biosynthesis of DHN-melanin in other ascomycetous fungi. Site-directed mutagenesis of ArPKS1 in A. rabiei generated melanin-deficient pycnidial mutants but caused no significant reduction of pathogenicity to chickpea. Pycnidiospores in ArPKS1-mutant pycnidia showed hig

The growing demand for sustainable alternative proteins has intensified interest in fungal mycelia as a nutrient-dense biomass for food applications. This study compared Pleurotus ostreatus fruiting bodies with mycelia grown in liquid state on amaranth seed- and Bambara groundnut-based media, evaluating aroma-active volatiles and amino acid composition. Across 52 identified volatiles, C8 oxylipin-derived compounds dominated all matrices, with exceptionally high odour activity values (OAVs) for 1-octen-3-one (~4.1 × 103), 3-octanone (~1.5 × 103), 1-octen-3-ol (~8.3 × 102) and 3-octanol (~5.3 ×

The evaluation of germplasm resources is a critical foundation for the selection and breeding of superior cultivars of Ganoderma lucidum. In this study, 19 widely cultivated strains from Northeast China were systematically evaluated using an integrated approach combining somatic incompatibility tests, sequence-related amplified polymorphism (SRAP) markers, agronomic trait assessment, and comprehensive profiling of bioactive compounds and metal elements in both fruiting bodies and spore powder. Somatic incompatibility assays showed that most strain combinations produced distinct antagonistic re

We present a case of maxillary sinusitis caused by Schizophyllum commune, in a 50-year-old female. The patient presented with nasal obstruction, purulent nasal discharge from right side of the nose, cough, headache, and sneezing. Computed tomography revealed extensive opacity of the right maxillary sinus as well as erosion of the nasal wall and maxillary bone. Functional endoscopic sinus surgery was done, and fungal debris present on right side of the maxillary sinus was removed and sent to laboratory. Potassium hydroxide (KOH) examination of the nasal discharge showed hyaline, septate hyphae.

Both mercury (Hg) and selenium (Se) occur in many mushroom species, but the morphological distribution of these elements during different developmental stages of the fruiting bodies is not known. Although Amanita muscaria can be consumed after suitable processing, they are often ignored by mushroom foragers, leaving an abundance for investigative study. Multiple specimens in each of six developmental stages (button to fully mature) were collected in excellent condition during a single morning from the same forested location and composited. With an average of 30 specimens per composite, and low

Ergothioneine (EGT) is a thiol-histidine compound with antioxidant properties and an important dietary component of edible mushrooms. Rapid methods for preliminary screening of EGT-producing fungi remain limited. Here, we developed a thin-layer chromatography (TLC) method using iodine vapour visualization for screening and densitometric estimation of EGT-associated signals. Forty-three fungal isolates collected from edible mushrooms and wild fruiting bodies in Maynooth, Ireland, were screened, identifying eight putative EGT-positive isolates representing Pleurotus, Hypsizygus, Agaricus, Rhodoc

Sexual reproduction in an environmental pathogen helps maximize its lineage fitness to changing environment and the host. For the fungal pathogen Cryptococcus neoformans, sexual reproduction is proposed to have yielded hyper virulent and drug resistant variants. The life cycle of this pathogen commences with mating, followed by the yeast-hypha transition and hyphal growth, and it concludes with fruiting body differentiation and sporulation. How these sequential differentiation events are orchestrated to ensure developmental continuality is enigmatic. Here we revealed the genetic network of the

In this study, we analyzed bioactive compounds from six edible wild mushrooms: Amanita caesarea, Amanita vaginata, Amanita rubescens, Clitocybe gibba, Laccaria laccata, and Ramaria apiculata. The fruiting bodies were used to extract the compounds using methanol. Qualitative screening revealed that anthraquinones are predominantly found in R. apiculata. The highest amounts of flavonoids and total phenols, were found in A. caesarea, (17.4 mg GAE/g and 205.0 mg GAE/g, respectively). When quantifying antioxidant activity using DPPH, no differences were observed, except in C. gibba. However, with A

It is controversial how useful bioassays are for identifying the in vivo toxicity of hazardous environmental exposures. In this study, fruiting bodies of forest mushrooms (n = 46), indoor mold colonies (n = 412), fungal secondary metabolites (n = 18), xenobiotic chemicals such as biocides and detergents (n = 6), and methanol extracts of indoor dusts from urban buildings (n = 26) were screened with two different bioactivity assays: boar sperm motility inhibition (BSMI) and inhibition of cell proliferation (ICP) tests. For the forest mushrooms, the toxicity testing result was positive for 100% o

Digital phenotyping is increasingly recognized as an essential tool for the quantitative analysis of fungal morphology, particularly in controlled indoor cultivation systems where large numbers of fruiting bodies must be assessed consistently and non-destructively. While YOLOv8-based deep learning approaches have previously been applied in phenotypic analyses of edible mushrooms, the applicability of newer YOLO architectures to fungal phenotyping remains largely unexplored. In this study, we present a controlled-environment digital phenotyping framework for indoor mushroom cultivation and cond

Pleurotus ostreatus was cultivated on a commercially available wheat straw substrate enriched with Zn and Se. Various amounts of Zn (10, 50, and 100 mg) and Se (1, 5, and 10 mg) in suitable forms ((CH3COO)2Zn·2H2O, Zn(NO3)2·6H2O, and Na2SeO3·5H2O, respectively) were dissolved in 50 ml of deionized water and homogenously nebulized into the substrate block of 2.4 kg weight. The increase in the Zn content in fruiting bodies cultivated on the enriched substrate was relatively low compared with fruiting bodies cultivated on the substrate with no addition at the first flush. The application of diffe

Selenium-enriched Cordyceps militaris fruit bodies are industrially cultivated as functional food or medicinal food in China and southeast Asia. However, composition of selenium compounds and distribution of the main bioactive components are still unknown. In the selenium-enriched fruit bodies, the main soluble selenium compounds of low molecular weight were identified as SeMet (selenomethionine), and the main selenium compounds bound in proteins were identified as SeMet and SeCys (methylselenocysteine). Trace minerals as Se (selenium), Zn (zinc), Fe (iron) and the main active components as ad
