[Simultaneous determination of eight furocoumarines in cosmetics by high performance liquid chromatography and verification by liquid chromatography-tandem mass spectrometry].

A method using high performance liquid chromatography (HPLC) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) was developed for the simultaneous determination of eight furocoumarines (8-hydroxypsoralen, psoralen, isopsoralen, 8-methoxypsoralen, 5-methoxypsoralen, trioxsalen, imperatorin and isoimperatorin) in cosmetics. The cosmetic samples, including cream, lotion, shampoo, powder and lipstick, were supersonically extracted with appropriate solvents. The extract was centrifuged, and the supernatant was filtered through a membrane, and then separated on an Agilent Zorbax SB-Phenyl

Imperatorin a furocoumarin inhibits periplasmic Cu-Zn SOD of Shigella dysenteriae their by modulates its resistance towards phagocytosis during host pathogen interaction.

Shigella dysenteriae continues to be a major health problem, which leads to death, due to diarrhoea and dysentery, predominantly in children below the age of 5. Bacterial invasion of the colonic epithelium leads to severe inflammation together with bacterial dissemination generates abscesses and ulcerations. Periplasmic copper, zinc super oxide dismutase of Shigella protects it from exogenous superoxide produced by host, during its invasion. Hence, in present study an attempt was made to study the effect of aqueous extract of Aegle marmelos on host and pathogen defence. Histology analysis of r

DNA damage induced by 4,6,8,9-tetramethyl-2H-furo[2,3-h]quinolin-2-one, a new furocoumarin analog: biological consequences.

4,6,8,9-Tetramethyl-2H-furo[2,3-h]quinolin-2-one (HFQ) and its isomer FQ (1,4,6,8-tetramethyl-2H-furo[2,3-h]quinolin-2-one) showed very strong antiproliferative activity in mammalian cells, about two times greater than 8-methoxypsoralen (8-MOP). Both compounds induced DNA-protein cross-links (DPC) but not interstrand cross-links. The FQ generated DPC in a biphotonic process, yielding a new kind of diadduct, whereas HFQ induced DPC by a monophotonic one, probably without its physical participation in the covalent bridge. These lesions gave different toxic responses. Sensitization of FQ led to e

Dietary Intake of Coumarins and Furocoumarins through Citrus Beverages: A Detailed Estimation by a HPLC-MS/MS Method Combined with the Linear Retention Index System.

Official regulations concerning the maximum number of substances in food are introduced as a consequence of possible adverse effects, after oral administration. In this regard, analytical methods are necessary in order to determine specific targets. Among oxygen heterocyclic compounds (OHCs, that are furocoumarins, coumarins and polymethoxyflavones), only coumarin is subject to restriction by the Regulation (EC) No 1334/2008 of the European Parliament. Furocoumarins are known for their phototoxicity and other side effects due to their dietary intake; however, an official limit about the maximu

Photoallergic contact dermatitis to 8-methoxypsoralen in Ficus carica.

BACKGROUND: Photocontact dermatitis to Ficus carica is induced by furocoumarins present in sap. These substances are generally considered to cause phototoxic reactions. OBJECTIVES: We conducted a patch test and histopathological study of patients with phytophoto contact dermatitis from the fig tree to evaluate the mechanism underlying the photoreaction. PATIENTS AND METHODS: Patch and photopatch testing with serial dilutions of two natural furocoumarins [5-methoxypsoralen and 8-methoxypsoralen (8-MOP)] contained in plant sap were performed in 47 patients. A synthetic furocoumarin, 4,5',8-tri

Psoralens sensitize glutathione photooxidation in vitro.

In vitro experiments are reported showing that psoralens and other furocoumarins of current pharmacological interest, e.g., angelicin and 4,6,4'-trimethylangelicin, all have, to a variable extent, the ability to sensitize the photooxidation of glutathione in ethanol/phosphate buffer with pyrex-filtered ultraviolet light. Besides substrate concentration and the nature of the furocoumarin used, the rate of the sensitized reaction is markedly dependent on the partial pressure of oxygen and the pH of the medium, being progressively faster on passing from pH 5 to pH 8.5. Scavengers of superoxide io

Absence of phototoxicity/photoirritation potential of bergamottin determined In Vitro using OECD TG 432.

The phototoxic potential of a number of furocoumarins is well established. On the other hand, studies have shown that bergamottin, a furocoumarin containing a bulky, hydrophobic side chain, has significantly less or is even absent of phototoxicity potential. The OECD Test Guideline 432 3T3/Neutral Red Uptake (NRU) in vitro phototoxicity test has shown to be a highly predictive test for identifying compounds that exhibit no phototoxicological potential. In this study using OECD 432, the established phototoxic furocoumarin 5-methoxypsoralen (5-MOP), 8-methoxypsoralen (8-MOP) and psoralen were ph

Assessing phytophotodermatitis: boy with erythema and blisters on both hands.

Phytophotodermatitis (PPD) is a phototoxic reaction which occurs when the skin comes in contact with a photosensitizer and is subsequently exposed to radiation. PPD is often seen in people handling furocoumarin-containing products, such as agricultural workers, bartenders, florists, and gardeners. It may also be seen in beachgoers, athletes, and children. The pattern of the lesions usually resembles streaks; the hands and mouth are most commonly affected due to eating and handling of the offending furocoumarin-containing agents.

Not as Bland as You May Think: Celery (Apium graveolens) Commonly Induces Phytophotodermatitis.

Often consumed for its nutritious benefits, celery (Apium graveolens) contains furocoumarins and psoralens that have photosensitizing properties and can induce phytophotodermatitis (PPD) when exposed to UV light. Clinical manifestations of PPD include an eruption of vesicles in sun-exposed areas. When consumed as part of a typical diet, the amount of furocoumarins in celery generally is insufficient to induce PPD; however, individuals who regularly handle celery or consume produce that is infected with fungus as well as those who use tanning beds are at increased risk for this painful reaction

Exogenous Phytohormone Activators Enhance Rice Resistance to Brown Planthoppers by Boosting the Phenylpropanoid Pathway under Elevated CO2.

Studies indicate that elevated CO2 (eCO2) exacerbates brown planthopper (BPH, Nilaparvata lugens) damage to rice compared to ambient CO2 (aCO2), highlighting the need for enhanced pest resistance strategies under climate change. The phenylpropanoid pathway, activated by phytohormones, synthesizes defense compounds in plants. This study investigated exogenous abscisic acid (ABA) and strigolactone analogue (GR24) treatments on rice defense against BPH under aCO2 and eCO2. Results showed that ABA and GR24 suppressed BPH feeding and survival, particularly under eCO2, while activating the phenylpro

In several patients the effect of a phototoxic reaction to the juice of fresh figs (ficus carica) was observed as a striped pigmentation on the arms (after rubbing in the fruit juice followed by exposure to the sun), or as a patchy pigmentation of the face after eating fresh figs. Reference is made to the pathogenetic identity of this furocoumarin phototoxic reaction and the clinical transition of ficus dermatitis both to "Berloque dermatitis" (from the oil of types of citrus) and to bullous meadow dermatitis (from the juice of types of heracleum). Reference is also made to the similarity of t

Supercritical fluid chromatography-tandem mass spectrometry of oxygen heterocyclic compounds in Citrus essential oils.

Oxygen heterocyclic compounds are secondary metabolites mainly present in the non-volatile fraction of cold-pressed Citrus essential oils. Under this denomination are included coumarins, furocoumarins, and polymethoxyflavones. These compounds possess numerous beneficial properties for human health, but the ingestion of large amounts of coumarins is often related to toxic effects, whereas the phototoxicity caused by furocoumarins and UVA exposure has been well known for a long time. This research has been aimed at the validation of an analytical approach, based on supercritical fluid chromatogr

The Curious Cases of Burn by Fig Tree Leaves.

Phytophotodermatitis is a condition which is caused by contact with some plants containing furocoumarins. Furocoumarins in sap of fig tree are the main cause of its irritability when come in contact with the skin. The main symptoms are burning sensation and pain, itchy erythema, and edema, which usually begin 24 h after exposure. The leaf and root sap of the fig tree are the most potent parts causing irritant reaction. We report four children who were playing with fig tree leaves during a family picnic. They were referred to a burn center with second-degree burn in about 10% of total body surf

Phytophotodermatitis.

Phytophotodermatitis is a phototoxic cutaneous eruption due to skin exposure to furocourmarins combined with ultraviolet light. Bizzare linear patterns, ranging from erythema to bullae with residual hyperpigmentaion, is the clinical clue to this diagnosis. Avoidance of furocoumarins in direct sunlight can prevent recurrences.
