Effect of IgY on Periodontitis and Halitosis Induced by Fusobacterium nucleatum.

Fusobacterium nucleatum is a morbific agent in periodontitis and halitosis. Egg yolk antibody (IgY) was obtained from egg yolks from chickens stimulated with F. nucleatum. This study was to assess the effectiveness of IgY on periodontitis and halitosis caused by F. nucleatum in vitro and in vivo. The growth of F. nucleatum was inhibited (p <0. 05) by different concentrations of IgY in vitro and the results of a Halimeter show volatile sulfur compounds (VSCs) were reduced to 904 ± 57 ppb at a concentration 40 mg/ml of IgY. The changes of fatty acids of F. nucleatum were determined using GC-MS.

Tracking Surface Protein Assembly and Peptidoglycan Biosynthesis in Fusobacterium nucleatum.

Fusobacterium nucleatum is a Gram-negative, obligate anaerobe and oncogenic oral bacterium that employs a diverse array of surface-exposed adhesins to colonize extra-oral tissues. Among these, Fusobacterial adhesin A (FadA) is a well-characterized virulence factor known for its ability to target epithelial and endothelial tight junctions, disrupt barrier integrity, and activate pro-tumorigenic signaling pathways implicated in colorectal cancer. FadA also facilitates placental and fetal colonization, contributing to adverse pregnancy outcomes such as preterm birth and stillbirth. While its func

Modeling Fusobacterium nucleatum-Driven Colorectal Cancer in Apc-Mutant Mice.

Colorectal cancer (CRC) typically initiates with mutations in the adenomatous polyposis coli (Apc) gene, followed by the influence of environmental factors. Among these, Fusobacterium nucleatum has emerged as an important CRC-enriched microbial species with demonstrated tumor-promoting potential. To investigate the contribution of F. nucleatum in the context of oncogenic mutations, physiologically relevant mouse models are essential. While Apcmin/+ mice have been employed to study F. nucleatum-associated CRC, prior studies have not systematically evaluated the effects of different bacterial in

Fusobacterium nucleatum-Induced Pyopneumothorax: A Rare but Serious Clinical Entity.

Fusobacterium nucleatum (F. nucleatum) is a Gram-negative, anaerobic bacterium predominantly found in the oral cavity, gastrointestinal tract, and urogenital system. It accounts for less than 1% of anaerobic bacterial infections in clinical settings and can lead to a spectrum of severe infections. This report details the case of a 55-year-old male who presented with a one-month history of productive cough, acute right-sided chest pain, dyspnea, and fever. Initial examinations showed elevated inflammatory markers and a right-sided pleural effusion on chest CT. Despite empirical therapy, the pat

Fusobacterium nucleatum-triggered neutrophil extracellular traps facilitate colorectal carcinoma progression.

BACKGROUND: Fusobacterium nucleatum (Fn) acts as a procarcinogenic bacterium in colorectal carcinoma (CRC) by regulating the inflammatory tumor microenvironment (TME). Neutrophil extracellular traps (NETs), which can be generated by persistent inflammation, have been recently considered to be significant contributors in promoting cancer progression. However, whether NETs are implicated in Fn-related carcinogenesis is still poorly characterized. Here, we explored the role of NETs in Fn-related CRC as well as their potential clinical significance. METHODS: Fn was measured in tissue specimens an

Aspirin Modulation of the Colorectal Cancer-Associated Microbe Fusobacterium nucleatum.

Aspirin is a chemopreventive agent for colorectal adenoma and cancer (CRC) that, like many drugs inclusive of chemotherapeutics, has been investigated for its effects on bacterial growth and virulence gene expression. Given the evolving recognition of the roles for bacteria in CRC, in this work, we investigate the effects of aspirin with a focus on one oncomicrobe-Fusobacterium nucleatum We show that aspirin and its primary metabolite salicylic acid alter F. nucleatum strain Fn7-1 growth in culture and that aspirin can effectively kill both actively growing and stationary Fn7-1. We also demons

Fusobacterium nucleatum Secretes Outer Membrane Vesicles and Promotes Intestinal Inflammation.

Multiple studies have implicated microbes in the development of inflammation, but the mechanisms remain unknown. Bacteria in the genus Fusobacterium have been identified in the intestinal mucosa of patients with digestive diseases; thus, we hypothesized that Fusobacterium nucleatum promotes intestinal inflammation. The addition of >50 kDa F. nucleatum conditioned media, which contain outer membrane vesicles (OMVs), to colonic epithelial cells stimulated secretion of the proinflammatory cytokines interleukin-8 (IL-8) and tumor necrosis factor (TNF). In addition, purified F. nucleatum OMVs, but

Fusobacterium nucleatum in Colorectal Carcinoma Tissue According to Tumor Location.

OBJECTIVES: Evidence suggests a possible role of Fusobacterium nucleatum in colorectal carcinogenesis, especially in right-sided proximal colorectum. Considering a change in bowel contents and microbiome from proximal to distal colorectal segments, we hypothesized that the proportion of colorectal carcinoma enriched with F. nucleatum might gradually increase along the bowel subsites from rectum to cecum. METHODS: A retrospective, cross-sectional analysis was conducted on 1,102 colon and rectal carcinomas in molecular pathological epidemiology databases of the Nurses' Health Study and the Heal

Fusobacterium nucleatum, inflammation, and immunity: the fire within human gut.

Fusobacterium nucleatum is an identified proinflammatory autochthonous bacterium implicated in human colorectal cancer. It is also abundantly found in patients suffering from chronic gut inflammation (inflammatory bowel disease), consequently contributing to the pathogenesis of colorectal cancer. Majority of the studies have reported that colorectal tumors/colorectal adenocarcinomas are highly enriched with F. nucleatum compared to noninvolved adjacent colonic tissue. During the course of multistep development of colorectal cancer, tumors have evolved many mechanisms to resist the antitumor im

Cellular Components Mediating Coadherence of Candida albicans and Fusobacterium nucleatum.

Candida albicans is an opportunistic fungal pathogen found as part of the normal oral flora. It can be coisolated with Fusobacterium nucleatum, an opportunistic bacterial pathogen, from oral disease sites, such as those involved in refractory periodontitis and pulp necrosis. The physical coadherence between these 2 clinically important microbes has been well documented and suggested to play a role in facilitating their oral colonization and colocalization and contributing to polymicrobial pathogenesis. Previous studies indicated that the physical interaction between C. albicans and F. nucleatu

Synergy in biofilm formation between Fusobacterium nucleatum and Prevotella species.

The formation of biofilm by anaerobic, Gram-negative bacteria in the subgingival crevice plays an important role in the development of chronic periodontitis. The aim of this study was to characterize the role of coaggregation between Fusobacterium nucleatum and Prevotella species in biofilm formation. Coaggregation between F. nucleatum and Prevotella species was determined by visual assay. Effect of co-culture of the species on biofilm formation was assessed by crystal violet staining. Effect of soluble factor on biofilm formation was also examined using culture supernatant and two-compartment

Nicotinamide attenuates the virulence of colorectal cancer-associated Fusobacterium nucleatum.

Fusobacterium nucleatum contributes to the progression of colorectal cancer (CRC). Nicotinamide (NAM), a safe, cost-effective, and water-soluble form of vitamin B3, has been shown to inhibit the virulence of various microorganisms. However, its effect on F. nucleatum pathogenicity remains unclear. In this study, we evaluated the effects of NAM on the pathogenic traits of F. nucleatum. A quarter of the minimum inhibitory concentration of NAM (~50 mM) was found to simultaneously inhibit bacterial growth, biofilm formation, and the adhesion and invasion of CRC cells. Transcriptomic analysis of F.

Vaccine Design against Fusobacterium nucleatum FadA Protein in Periodontal Infections.

Objectives: A common inflammatory condition affecting oral health is periodontitis, which can lead to tooth loss if not treated early, as it destroys the supporting structures of the teeth. Fusobacterium nucleatum is an orange-pigmented bacterium commonly referred to as "the bridging species" between early and late colonizers in the oral microbiota, and it is associated with the progression of periodontal disease. This study aimed to develop an effective and precise multi-epitope vaccine against F. nucleatum, as there is currently no approved vaccine for periodontal disease. Materials and Meth

PURPOSE OF REVIEW: Fusobacterium nucleatum (F. nucleatum), an anaerobic, gram-negative microbe, commonly found in human dental biofilm and the gut flora. It has long been known to have a higher concentration in periodontal disease and has recently been implicated in both oral and distant cancers such as colorectal, gastrointestinal, esophageal, breast, pancreatic hepatocellular, and genitourinary cancers. However, the mechanism of its involvement in the development of cancer has not been fully discussed. This review aims to cover biological molecular and clinical aspects of F. nucleatum and ca

Rectal Cancer Presenting with Absceding Infection Due to Fusobacterium nucleatum.

Intestinal microbiota such as Fusobacterium nucleatum play an important role in the pathogenesis of colorectal cancer. Here, we describe the case of a 47-year-old patient presenting with endophthalmitis and a liver abscess due to Fusobacterium nucleatum that prompted the diagnosis of colorectal cancer as the most likely source of infection. This case highlights that colorectal cancer needs to be considered in patients with systemic infection with Fusobacterium nucleatum and colonoscopy should be performed.

Fusobacterium nucleatum Metabolically Integrates Commensals and Pathogens in Oral Biofilms.

Fusobacterium nucleatum is a common constituent of the oral microbiota in both periodontal health and disease. Previously, we discovered ornithine cross-feeding between F. nucleatum and Streptococcus gordonii, where S. gordonii secretes ornithine via an arginine-ornithine antiporter (ArcD), which in turn supports the growth and biofilm development of F. nucleatum; however, broader metabolic aspects of F. nucleatum within polymicrobial communities and their impact on periodontal pathogenesis have not been addressed. Here, we show that when cocultured with S. gordonii, F. nucleatum increased ami
