Identification of lipids in the cuticle of the parasitic nematode Anisakis simplex and the somatic tissues of the Atlantic cod Gadus morhua.

The main aim of this work was to assign the cuticular lipids identified in a parasitic nematode and to distinguish those originating from its host. The hypothesis that long-chained fatty acids and sterols are imported by the parasite in the absence of certain enzymes was also tested. The organisms (Anisakis simplex and Gadus morhua) were extracted in petroleum ether and dichloromethane. Matrix-assisted laser desorption time-of-flight mass spectrometry (MALDI-TOF) was used to identify unknown components, and electrospray ionization mass spectrometry (ESI/MS) to verify recognized groups of lipid

Exophthalmia in wild-caught cod (Gadus morhua L.): development of a secondary barotrauma effect in captivity.

Capture-based aquaculture (CBA) of Atlantic cod (Gadus morhua) has become increasingly important in recent years, and increased attention is being paid to animal welfare issues linked to these activities. Earlier studies showed that some cod develop secondary exophthalmia in captivity. This study investigated the development of secondary exophthalmia in two groups of wild-caught cod, one of which was exposed to rapid decompression causing acute barotrauma (treatment group) while the other was not (control group). Photographs and radiographs before and up to 33 days after barotrauma revealed a

Early Nutrition Impacts on Growth, Skeletal Anomalies and Organ Ontogeny in Larval Atlantic Cod (Gadus morhua).

Early life nutrition is a critical factor influencing subsequent performance and quality, including skeletal development, in farmed Atlantic cod (Gadus morhua). This study investigated the effects of a novel start-feed protocol utilizing barnacle nauplii and plankton eggs and two experimental microdiets on larval survival, growth, skeletal anomalies, and organ ontogeny. Atlantic cod larvae were reared using three feeding protocols (COM, D1, and D2): COM used enriched rotifers and a commercial microdiet, while D1 and D2 protocols incorporated blue mussel eggs (Cryo-µ) and barnacle nauplii (Cryo

Identification of umami peptides based on virtual screening and molecular docking from Atlantic cod (Gadus morhua).

Umami peptides have currently become the research focus in the food umami science field and the key direction for umami agent development. This is because umami peptides have good processing characteristics, umami and nutritional values. We here used virtual screening (including online enzymolysis through ExPASy PeptideCutter, bioactivity screening using the PeptideRanker, toxicity and physicochemical property prediction using Innovagen and ToxinPred software), molecular docking, and electronic tongue analysis to identify umami peptides generated from Atlantic cod myosin. Twenty-three putative

Comparison of metazoan parasites of Atlantic cod, Gadus morhua, from three geographical areas of coastal Newfoundland.

This study compared metazoan parasites in the digestive tract of Atlantic cod, Gadus morhua , captured from 3 geographical areas off the northeastern (NE), southeastern (SE), and southwestern (SW) coasts of Newfoundland. Samples were obtained by line trawl and frozen after capture, and the digestive tract was subsequently examined for metazoan parasites following conventional parasitological methods. Mean abundance of nematodes, but not the trematodes, and an acanthocephalan was significantly greater in cod taken from the NE than from the SE or SW areas, which were generally similar. Four spec

Detection of chemical warfare agent related phenylarsenic compounds and multibiomarker responses in cod (Gadus morhua) from munition dumpsites.

Recently, sea-dumped chemical weapons (CWs) containing toxic chemical warfare agents (CWAs) have raised international attention. It is well known that CWAs are leaking from corroded munitions causing a risk to the surrounding marine environment, while the impact on marine biota is still unknown. In this study, cod (Gadus morhua) was used as a model species to study the possible bioaccumulation of phenylarsenic CWAs and their negative effects at multiple levels of biological organization on fish living in the vicinity of a major CWs dumpsite in the Bornholm Basin in the Baltic Sea. In total, 14

Experimental Challenge of Atlantic Cod (Gadus morhua) with a Brucella pinnipedialis Strain from Hooded Seal (Cystophora cristata).

Pathology has not been observed in true seals infected with Brucella pinnipedialis. A lack of intracellular survival and multiplication of B. pinnipedialis in hooded seal (Cystophora cristata) macrophages in vitro indicates a lack of chronic infection in hooded seals. Both epidemiology and bacteriological patterns in the hooded seal point to a transient infection of environmental origin, possibly through the food chain. To analyse the potential role of fish in the transmission of B. pinnipedialis, Atlantic cod (Gadus morhua) were injected intraperitoneally with 7.5 x 107 bacteria of a hooded s

Historical DNA reveals the demographic history of Atlantic cod (Gadus morhua) in medieval and early modern Iceland.

Atlantic cod (Gadus morhua) vertebrae from archaeological sites were used to study the history of the Icelandic Atlantic cod population in the time period of 1500-1990. Specifically, we used coalescence modelling to estimate population size and fluctuations from the sequence diversity at the cytochrome b (cytb) and Pantophysin I (PanI) loci. The models are consistent with an expanding population during the warm medieval period, large historical effective population size (NE), a marked bottleneck event at 1400-1500 and a decrease in NE in early modern times. The model results are corroborated b

Regional changes in vertebra morphology during ontogeny reflect the life history of Atlantic cod (Gadus morhua L.).

This study examined vertebra formation, morphology, regional characters, and bending properties of the vertebral column of Atlantic cod throughout its life cycle (0-6 years). The first structure to form was the foremost neural arch, 21 days post hatching (dph), and the first vertebra centrum to form - as a chordacentrum - was the 3rd centrum at 28 dph. Thereafter, the notochord centra developed in a regular sequence towards the head and caudal fin. All vertebrae were formed within 50 dph. The vertebral column consisted of 52 (± 2) vertebrae (V) and could be divided into four distinct regions:

Genome-wide identification of the Hsp70 gene family in Atlantic cod (Gadus morhua) and its response to pathogenic stress.

Heat shock protein 70 (Hsp70) is a critical molecular chaperone in most eukaryotes and prokaryotes, significantly contributing to several physiological and biochemical processes and regulating stress response to external stimuli. Gadus morhua is a fish species with exceptionally high nutritional value and significant economic importance, making it highly meaningful to study its response strategies under pathogenic stress. However, systematic analyses of Hsp70 in G. morhua have yet to be performed. In this study, we identified 24 family members of Hsp70 genes from G. morhua and analysed their p

Toxicokinetic of perfluorinated compounds - A study of liver sequestration in Baltic cod (Gadus morhua callarias) and human dietary exposure.

This study assessed the concentrations and temporal trends of perfluorinated compounds (PFCs) in the muscle and liver tissues of Baltic cod (Gadus morhua) from 2017 to 2023. The Environmental Quality Standard (EQS) for biota and the Estimated Weekly Intake (EWI) for human consumers were calculated to evaluate potential health risks associated with PFC exposure. Significant variations in PFC concentrations were observed across different compounds and sampling years. Perfluorooctanoic acid (PFOA) had a mean concentration ranging from 0.03 ng g⁻1 wet weight (ww) to 0.17 ng g⁻1 ww (mean: 0.093 ng

Composition and structure of the parasite faunas of cod, Gadus morhua L. (Teleostei: Gadidae), in the North East Atlantic.

BACKGROUND: Although numerous studies on parasites of the Atlantic cod, Gadus morhua L. have been conducted in the North Atlantic, comparative analyses on local cod parasite faunas are virtually lacking. The present study is based on examination of large samples of cod from six geographical areas of the North East Atlantic which yielded abundant baseline data on parasite distribution and abundance. MATERIALS AND METHODS: A total of 826 fish was sampled in the Baltic, Celtic, Irish and North seas, Icelandic waters and Trondheimsfjord (Norway) in 2002 (spring and autumn) and 2003 (spring). The

IgM-mediated protection drives early B-cell activation and mucosal containment of Vibrio anguillarum in Atlantic cod (Gadus morhua).

BACKGROUND: Atlantic cod lack functional MHC class II and CD4, raising fundamental questions about how vaccination generates protection in this species. METHODS: We combined single-cell transcriptomic profiling of splenic cells with qRT-PCR across complementary active vaccination and passive serum-transfer experiments to define cellular and transcriptional correlates of immunity to Vibrio anguillarum. RESULTS: Bath-vaccinated fish and recipients of immune serum showed effective containment of infection, with bacterial signals largely restricted to gills and minimal detection in spleen or hea

Atlantic cod (Gadus morhua) larvae are attracted by low-frequency noise simulating that of operating offshore wind farms.

The number and size of offshore wind (OW) turbines is increasing rapidly. OW turbines produce continuous, low-frequency noise that could impact marine fish dispersing/migrating through the facilities. Any such impact would be relevant for larval stages, which have limited possibility to swim away from OW facilities. If directional movement of fish larvae at sea is impacted by low-frequency continuous sound is unknown. We observe the behavior of Atlantic cod larvae (N = 89) in response to low-frequency sound while they are drifting in a Norwegian fjord inside transparent drifting chambers. We t

Eye fluke infection status in Baltic cod, Gadus morhua, after three decades and their use as ecological indicators.

Eye flukes of the genus Diplostomum were recorded with a prevalence of 7.4% and a mean intensity of 11.9 (range 1-75) parasites per fish in eye lenses of a total of 188 Atlantic cod, Gadus morhua, sampled in the Southeastern Baltic Sea from March 2013 to February 2014. A slight decrease of infection level, among the fish with body length ranging from 30 to 89.5 cm, was found when data were compared to a corresponding survey in the 1980s. Due to imprecise species identification of eye flukes based on morphometric analyses we present, as a baseline for further studies, rDNA sequences from a subs

Prolonged SDA and reduced digestive efficiency under elevated CO2 may explain reduced growth in Atlantic cod (Gadus morhua).

Land-based aquaculture systems expose fish to elevated dissolved CO2 levels, a factor that is correlated with reduced growth, feed conversion efficiency and body condition index. The physiological basis underlying the pathological effects of environmental hypercapnia is poorly understood, in particular for marine fish species. We investigated whether changes in energy expenditure and the specific dynamic action (SDA) of digestion and assimilation could account for the lower growth of adult Atlantic cod (Gadus morhua) under environmental hypercapnia. Fish acclimated to a partial pressure of 800

Optimized conditions for primary culture of pituitary cells from the Atlantic cod (Gadus morhua). The importance of osmolality, pCO₂, and pH.

Protocols for primary cultures of teleost cells are commonly only moderately adjusted from similar protocols for mammalian cells, the main adjustment often being of temperature. Because aquatic habitats are in general colder than mammalian body temperatures and teleosts have gills in direct contact with water, pH and buffer capacity of blood and extracellular fluid are different in fish and mammals. Plasma osmolality is generally higher in marine teleosts than in mammals. Using Atlantic cod (Gadus morhua) as a model, we have optimized these physiological parameters to maintain primary pituitar

Unraveling the Influence of Behavioural Ecotypes on Fish Gut Microbiome: Focus on the Atlantic cod (Gadus morhua) in Icelandic Waters.

This study investigated the gut microbiome of Atlantic cod in northern Icelandic waters to assess how resident (coastal) and migratory (frontal) behavioural ecotypes influence gut microbial community composition, with a focus on the effects of ecological adaptation and the potential drivers of microbial shifts within this species. A total of 81 intestinal samples from coastal and frontal Atlantic cod collected in northern Iceland in December 2021 and January 2024 were analysed using 16S rRNA gene metabarcoding.
