Iminosugar-based galactoside mimics as inhibitors of galactocerebrosidase: SAR studies and comparison with other lysosomal galactosidases.

Several families of iminosugar-based galactoside mimics were designed, synthesized, and evaluated as galactocerebrosidase (GALC) inhibitors. They were also tested as inhibitors of lysosomal β- and α-galactosidases in order to find new potent and selective pharmacological chaperones for treatment of the lysosomal storage disorder, Krabbe disease. Whereas 1-C-alkyl imino-L-arabinitols are totally inactive toward the three enzymes, 1-C-alkyl imino-D-galactitols were found to be active only toward α-galactosidase A. Finally, 1-N-iminosugars provided the best results, as 4-epi-isofagomine was found

Galactosemia: how does long-term treatment change the outcome?

In galactosemic subjects, treatment prevents liver and kidney failure, brain damage and cataracts, but total exclusion of galactose from the diet does not ensure the absence of all pathology. Early and well-treated children show satisfactory general health and growth, make reasonable though suboptimal intellectual progress, are prone to speech defects, and commonly experience visual perceptual difficulties and some social maladjustment. Two different metabolites are potentially toxic: galactitol is responsible for the cataracts while galactose-1-phosphate causes the rest of the pathology. As b

Hypergonadotropic hypogonadism in female patients with galactosemia.

We evaluated gonadal function in 18 female and eight male patients with galactosemia due to transferase deficiency; it was normal in the males, but 12 females had signs of hypergonadotropic hypogonadism. All female patients had a 46,XX karyotype, normal levels of thyroid hormone and prolactin, and no anti-ovarian antibodies. The biologic activity of urinary gonadotropins was normal. Ultrasonography of the pelvis revealed that ovarian tissue was diminished or absent. Total estrogens increased in one of two patients after administration of human menopausal gonadotropin. The frequency of hypergon

Characterisation of a Listeria monocytogenes mutant deficient in D-arabitol fermentation.

We selected and analysed a Tn917-lac Listeria monocytogenes mutant deficient in D-arabitol fermentation. Comparison of the 310-aa-long translated partial sequence of the disrupted gene with known proteins showed similarity with the phosphotransferase system galactitol-specific enzyme IIC component of the alkaliphilic Bacillus halodurans (50% identity) and of Escherichia coli (36% identity). Fermentation of 18 other carbohydrates was unimpaired, suggesting the specificity of this transmembrane permease IIC for the pentitol D-arabitol. The deficiency in D-arabitol fermentation did not alter L. m

Onychomycosis caused by Trichosporon mucoides.

A case of onychomycosis caused by Trichosporon mucoides in a man with diabetes is presented. The infection was characterized by a brown-black pigmentation of the nail plates and subungual hyperkeratosis of the first three toes of both feet. Onychogryphosis was also visible on the third left toe. Direct microscopic examinations revealed wide and septate hyphae and spores. Three cultures on Sabouraud-gentamicin-chloramphenicol 2 agar and chromID Candida agar produced white, creamy, and smooth colonies that were judged to be morphologically typical of T. mucoides. Microscopic examinations of the

Diagnosis of inherited disorders of galactose metabolism.

Galactose metabolism occurs through an evolutionarily conserved pathway in which galactose and uridine diphosphoglucose are converted to glucose-1-phosphate and uridine diphosphogalactose through the action of three sequential enzymes: galactokinase (GALK, EC 2.7.1.6), galactose-1-phosphate uridyltransferase (GALT, EC 2.7.7.12), and uridine phosphogalactose 4'-epimerase (GALE, EC 5.1.3.2). Inborn errors of galactose metabolism occur with impaired activity for each of the enzymes. Classical galactosemia is the most common and the most severe of these diseases and is caused by deficiency of the

A geographic approach to senile cataracts: possible links with milk consumption, lactase activity, and galactose metabolism.

Examined in this article is presently available evidence for the hypothesis that some types of senile cataracts may be brought on by decades-long consumption of milk and milk products. The author approaches the question from a background of research in the geography and history of dairying as these relate to present-day differences among the world's peoples in prevalence of primary adult lactose malabsorption, which is based on a deficiency of the enzyme lactase in adulthood. Among peoples who have consumed milk in lactose-rich forms over a long historical period, there seems to have been a mu

Combined phytohaemagglutinin and dianhydrodulcitol treatment of lymphocytic choriomeningitis virus infection in mice.

The course of intracerebral lymphocytic choriomeningitis (LCM) virus infection was studied in mice treated simultaneously with dianhydrodulcitol (DAD) and phytohaemagglutinin (PHA). Earlier experiments revealed that DAD decreased and PHA enhanced the cellular immune response of mice to LCM virus infection. On applying the treatments simultaneously they inhibited each other and neither the decreasing effect of DAD nor the enhancing effect of PHA on the cellular immune response could be observed.

Two class II D-tagatose-bisphosphate aldolases from enteric bacteria.

Escherichia coli, Salmonella enterica, Klebsiella pneumoniaeand Klebsiella oxytocawere found to contain two D-tagatose 1,6-bisphosphate (TagBP)-specific aldolases involved in catabolism of galactitol (genes gatY gatZ) and of N-acetyl-galactosamine and D-galactosamine (genes kbaY kbaZ,also called agaY agaZ). The two aldolases were closely related (> or = 53.8% identical amino acids) and could substitute for each other in vivo. The catalytic subunits GatY or KbaY alone were sufficient to show aldolase activity. Although substantially shorter than other aldolases (285 amino acids, instead of 358

Mutagenic activity of 6-azido deoxyhexoses and azido alcohols in Salmonella typhimurium and its inhibition by a structure-similar carbon source in the medium.

6-Azido-6-deoxy (AZd) derivatives of D-glucose, D-mannose, D-altrose, D-allose, L-idose, D-galactose, D-galactonic acid and D-galactitol, 3-azido-1,2-propanediol (azidoglycerol), 3,1-diazido-2-propanol (diazidoglycerol) and (at much higher doses) 2-azidoethanol were mutagenic in Salmonella typhimurium strains TA100 and TA1535. The mutagenic response was similar to that induced by sodium azide, i.e., the azido compounds failed to induce mutations in strain TA98, and mutagenesis was independent of plasmid pKM101, and independent of external activation. The specific mutagenicity (his+ rev/mmole)

The evaluation of therapeutic efficacy of hachimi-jio-gan (traditional Chinese medicine) to rat galactosemic cataract.

Hachimi-jio-gan (Rehmannia Eight Formula, pa-wei-ti-huang-wan or Bawei dihuang wan) is one of traditional Chinese medicines which has been used for treating various senile disease for a few hundred years. This drug was evaluated for its therapeutic efficacy to rat galactosemic cataract from the suppressive rate of variance of some biochemical parameters, whose variation with cataractogenesis or the advance of cataract have been reported already. The dose of 500mg of Hachimi-jio-gan/day/200g of rat body weight suppressed significantly the variations of hydration rate, Na/K ratios, and calcium i

Yunzhangomyces sedi sp. nov., a basidiomycetous yeast species isolated from phylloplane.

Two yeast strains, CICC 33533 and NYNU 26815, belonging to the basidiomycetous yeast genus Yunzhangomyces, were isolated from the leaf surfaces of Sedum aizoon and Sorbus alnifolia, respectively, collected in the Baotianman Nature Reserve, Henan Province, China. Phylogenetic analyses of the internal transcribed spacer (ITS) region and the D1/D2 domain of the large subunit rRNA gene, together with detailed phenotypic characterization, indicated that these two strains, along with an additional strain NRP040 isolated from a plant in Japan, represent a novel asexual species of the genus Yunzhangom

Glucose dependence of glycolysis, hexose monophosphate shunt activity, energy status, and the polyol pathway in retinas isolated from normal (nondiabetic) rats.

PURPOSE: To measure glucose-dependent metabolic activities and selected parameters of the polyol pathway in retinas isolated from normal rats to test the hypothesis recently proposed by Van den Enden et al that incubation of whole retinas for 2 hours with elevated concentrations of glucose results in activation of the polyol pathway, which is the cause of a redox imbalance, as measured by an increase in the retinal cytosolic lactate-pyruvate ratio and a diabetic-like state. METHODS: Retinas obtained from nondiabetic rats and separated from other ocular tissues were incubated for several hours

Rethinking Newborn Screening: A Case of GALM Deficiency.

Galactosemia is a group of hereditary disorders of galactose metabolism. A new type of galactosemia was discovered, caused by a deficiency in galactose mutarotase (GALM), which catalyzes the epimerization between beta- and alpha-D-galactose. All GALM-deficient patients reported in the literature (n = 44) had abnormal newborn screening (NBS) results or did not receive NBS (n = 2). We present the first patient with GALM deficiency who had negative NBS in the Netherlands and was identified at age 1.5 years during broad metabolic screening because of her global developmental delay, nystagmus, and

Novel Mutation in GALT Gene in Galactosemia Patient with Group B Streptococcus Meningitis and Acute Liver Failure.

Classic galactosemia is an autosomal recessive disorder caused by the deficiency of the enzyme galactose-1-phosphate uridyltransferase (GALT) involved in galactose metabolism. Bacterial infections are a known cause of early morbidity and mortality in children with classic galactosemia. The most common agent is Escherichia coli, but in rare situations, other bacteria are incriminated. We report a case of a three-week-old female patient with galactosemia, who presented with Group B Streptococcus (GBS) meningitis/sepsis. She received treatment with antibiotics, supportive therapy, and erythrocyte

Pathophysiology of impaired ovarian function in galactosaemia.

Classical galactosaemia is an inherited inborn error of the major galactose assimilation pathway, caused by galactose-1-phosphate uridyltransferase (GALT) deficiency. Many GALT mutations have been described, with different clinical consequences. In severe forms, newborns present with a life-threatening, acute toxic syndrome that rapidly regresses under a galactose-restricted diet. However, long-term complications, particularly cognitive and motor abnormalities, as well as hypergonadotrophic hypogonadism in female patients are still unavoidable.
