Gallamine exerts biphasic allosteric effects at muscarinic receptors.

Although gallamine and a number of other compounds have been reported to slow the rate of dissociation of labeled ligands, especially [3H]N-methylscopolamine (NMS), from muscarinic receptors of heart and brain, there has been some dispute as to whether the dissociation of [3H]quinuclidinyl benzilate (QNB) is subject to such allosteric regulation. The present studies were intended to determine whether past discrepancies might be due to differences between tissues. We have found that gallamine modulates the dissociation of [3H]QNB from muscarinic receptors of the heart in a biphasic manner. Low

The interaction of nicotinic receptor antagonists at muscarinic receptors.

1. The interaction of some neuromuscular blocking drugs such as gallamine, pancuronium and stercuronium with muscarinic receptors has several features which distinguish these compounds from competitive antagonists in functional and binding studies. 2. They also differentiate between muscarinic receptors and may produce effects on ion channels.

Biochemical changes following suxamethonium. Serum myoglobin, potassium and creatinine kinase changes before commencement of surgery.

Forty patients were investigated for serum myoglobin changes following induction of anaesthesia but before the commencement of surgery. Blood was drawn for potassium, creatinine kinase and serum myoglobin immediately prior to and 5, 10 and 20 minutes after administration of thiopentone 4 mg/kg and suxamethonium 1.2 mg/kg. Twenty patients were given either 2 mg alcuronium or 20 mg gallamine as pretreatment 2 to 3 minutes before the suxamethonium to reduce the fasciculations. Anaesthesia was maintained with artificial ventilation and alcuronium, or spontaneous ventilation with halothane. Serum m

Effects of combined carotid chemoreceptor and atrial receptor stimulation on renal blood flow in anaesthetized dogs.

In dogs anaesthetized with chloralose and artificially ventilated, carotid chemoreceptors were stimulated by changing the perfusate of the vascularly isolated carotid bifurcations from arterial to venous blood. Left atrial receptors were stimulated by distending balloons in two pulmonary vein-left atrial junctions and in this left atrial appendage. The left renal blood flow was measured by an electromagnetic flow meter at a constant systemic (renal) arterial pressure in preparations in which heart rate changes were prevented by administration of propranolol hydrochloride (0.5 mg kg-1) and atro

Monosynaptic rubrospinal projections to distal forelimb motoneurons in the cat.

In the present electrophysiological experiments, we examined whether or not there are short-latency excitatory postsynaptic potentials (EPSPs) suggesting monosynaptic rubro- and corticomotoneuronal connections in the cervical spinal segments in cats. Under pentobarbital anesthesia and immobilization with gallamine triethiodide, intracellular recordings were made from forelimb motoneurons (Mns) following stimulation of the red nucleus (RN) and the cerebral peduncle (CP). Mns comprising the musculocutaneous nerve (Musc. Mns), the radial nerve (Rad. Mns), the ulnar nerve (Uln. Mns), and the media

Functional evidence of excitatory M1 receptors in the rabbit airway.

The effect of vagally and acetylcholine (ACh)-induced bronchoconstrictions was assessed by an increase in the slowly adapting pulmonary stretch receptor (SAR) activity during both inflation and deflation and the rise in total lung resistance (R(L)). Those responses were compared before and after pirenzepine (PZ, M1 selective) with or without propranolol (a beta adrenoreceptor blocker), gallamine (M2 selective), 4-DAMP (M3 selective), hexamethonium (C6, a ganglion blocker) and atropine (a nonselective muscarinic receptor antagonist). The SAR activity was recorded from the cut left vagus nerve,

Spontaneous synchronized neural activity in decerebrate gallamine-paralysed cats.

In decerebrate cats paralysed with gallamine, over a period of several days there develops a remarkable synchronization of discharge in widely different motor nerves throughout the body, including intercostal nerves and limb nerves. These discharges are also in synchrony with slow waves approximately 100 ms in duration in the inferior olive. The slow waves and discharges are at first irregular and only weakly synchronized, but become increasingly strongly synchronized and by about the fourth day exhibit a strong 6-8 Hz rhythm. The degree of synchronization is greater the lower the end-tidal CO

Acetylcholine is released from in vitro cat carotid bodies during hypoxic stimulation.

Previous pharmacological, immunocytochemical, electrophysiological, and microfluorometric studies have suggested that acetylcholine (ACh) is a critically important excitatory transmitter in the chemotransduction of hypoxia by the cat carotid body (CB). With the use of HPLC this study shows that the in vitro cat CB releases ACh under normoxic conditions; this release is increased when the CB is challenged with hypoxia. The preliminary observation that greater amounts of ACh are liberated in the presence of gallamine and AFDX116 suggests the presence of functioning M2 muscarinic receptors on the

Allosteric regulation of the binding of [3H]acetylcholine to m2 muscarinic receptors.

Muscarinic receptors of the m2 subtype expressed in Chinese hamster ovary cells were labeled with [methyl-3H]acetylcholine([3H]ACh), and the rate of dissociation in the presence and absence of several compounds known to exert allosteric effects on labeled antagonist binding was observed. At 25 degrees C, [3H]ACh bound to the receptors with a Kd of 1.2 nM and dissociated with a half-time of 1.6 min. This binding was sensitive to appropriate concentrations of guanine nucleotide and the muscarinic antagonist N-methylscopolamine (NMS). Gallamine, tetrahydroaminoacridine, physostigmine, obidoxime,

Vagal nerve contribution to the calcitonin-induced changes in intrapyloric pressure in anesthetized rats.

The effect of calcitonin on intrapyloric pressure was examined in urethane-anesthetized rats. The intrapyloric pressure was decreased dose dependently by intravenous (i.v.) administration of 0.002-20 U of calcitonin. The calcitonin-induced decreased response was abolished after the severance of bilateral vagal nerves at the cervical level. Ongoing efferent activity of vagal gastric nerve branches was decreased by i.v. administration of calcitonin. On the contrary i.v. administration of calcitonin increased intrapyloric pressure via vagal nerves in atropinized or gallaminized animals. The contr

Rats were rendered deficient in thiamine by feeding a synthetic diet free of the vitamin. Responses of the isolated heart to acetylcholine, adrenaline, noradrenaline and isoprenaline, and of the phrenic nerve-diaphragm preparation to tubocurarine, gallamine and eserine, were compared with responses of tissues obtained from littermate rats pair-fed an identical diet with the addition of 25 mug of thiamine hydrochloride per day. Changes in blood pressure induced by electrical stimulation of the femur in anesthetized rats.

The effects of electrical stimulation of the femur, on blood pressure, were examined in anesthetized rats. Two small holes, 3-4 mm apart, were manually drilled into the femur down to the bone marrow. Following this, two stainless-steel electrodes were inserted into the holes, and an electrical square wave current was passed between the electrodes. In central nervous system-intact rats, electrical stimulation of the femur at 5 and 10 mA at 20 Hz for 20 s produced an intensity-dependent decrease in mean arterial blood pressure. This response was abolished by severance of the femoral and sciatic

Effect of some cardiac and respiratory drugs on succinate-cytochrome c reductase.

Succinate-cytochrome c reductase was inhibited in vitro and in vivo by phenobarbitone, aminophylline and neostigmine using both 2,6-dichlorophenolindophenol (DCIP) and cytochrome c (cyt c) as substrates. The enzyme was also activated by gallamine towards both substrates. In vitro, phenobarbitone and aminophylline inhibited the enzyme with respect to the reduction of DCIP and cyt c in a non-competitive manner with Ki values of 1.5 x 10(-5) and 5.7 x 10(-5)M, respectively. Moreover, neostigmine competitively inhibited the enzyme towards both substrates with Ki values of 1.36 x 10(-5) and 1.50 x

Modulation by certain conserved aspartate residues of the allosteric interaction of gallamine at the m1 muscarinic receptor.

Muscarinic acetylcholine receptors belong to a superfamily of G-protein coupled receptors and contain within their structure several conserved aspartate residues. These residues have been implicated to play important roles in the interaction of agonists and their competitive antagonists with the receptor. In the present work, we investigated whether the same residues might also serve as important contact points for allosteric antagonists of muscarinic receptors, because the majority of these compounds are cationic in nature, or if such residues are involved in modification of receptor conforma

Interaction of cholinergic ligands with the purified acetylcholine receptor protein. I. Equilibrium binding studies.

We have studied the binding equilibria of two fluorescent ligands and several nonfluorescent cholinergic ligands with the purified acetylcholine receptor from Electrophorus electricus. The assay was based on the specifically cholinergic and reversible quenching of fluorescence observed upon complex formation between the receptor protein and N-7-(4-nitrobenzo-2-oxa-1,3-diazole)-5-aminopentanoic acid beta-(N-trimethylammonium) ethyl ester. This way, a large body of accurate, true equilibrium data was obtained. We find 1) all ligands studied compete for the same number of binding sites at the rec

Two allosteric modulators interact at a common site on cardiac muscarinic receptors.

The abilities of gallamine, obidoxime, tetrahydroaminoacridine (THA), and 8-(N,N-diethylamino)octyl-3,4,5-trimethoxybenzoate (TMB-8) to alter the rate of dissociation of N-[3H]methylscopolamine from rat cardiac muscarinic receptors were investigated. All four ligands monotonically slowed the dissociation, with the order of potency gallamine > TMB-8 > THA > obidoxime. There was a dramatic difference in the efficacy of these allosteric modulators.
