Drug absorption in gastrointestinal disease with particular reference to malabsorption syndromes.

There is a considerable range in the dose of many drugs that is required to produce a given pharmacological effect in an individual patient. This individual variation in dose requirement is sometimes reflected in the wide scatter in the steady state plasma concentration that follows the same oral dose of a drug given to any group of subjects. Such individual differences are largely due to variation in the rate of elimination of drugs. Gastrointestinal disease may also alter oral dose requirements by producing variation in both the amount and rate of drug absorption. These changes may be reflec

Gastrointestinal function, divalent metal transporter-1 expression and intestinal iron absorption.

Iron absorption involves two carriers, one involved in the uptake of iron across the microvillus membrane of the enterocyte and the other in its transfer to the plasma at the basolateral surface. The uptake phase is thought to involve divalent metal transporter-1 (DMT1) which may move from the cytoplasm to the microvillus membrane under conditions of iron deficiency. To examine this possibility we used fasted animals previously fed an iron-deficient diet and then gavaged with iron. We measured the processes of iron absorption using in vivo gut sacs and correlated the changes observed with the

Folic acid-PEO-labeled liposomes to improve gastrointestinal absorption of encapsulated agents.

The design of targeted oral liposomes is anticipated to improve the systemic delivery of poorly absorbed agents, such as proteins and peptides. A poly(ethylene oxide) (PEO)-folic acid (FA) derivative was prepared and evaluated for improving liposome transport across a model gastrointestinal cell line (Caco-2). FA-PEO-cholesterol (Chol) derivatives were synthesized and adsorbed at liposome surfaces encapsulating Texas Red((R))-Dextran 3000 (TR-dex), a poorly-absorbed, neutral, hydrophilic, large molecular weight (M(w)) marker. Apparent permeabilities (P(app)) of Caco-2 cells to FA-PEO conjugate

Bioactive peptide release and the absorption tracking of casein in the gastrointestinal digestion of rats.

Bovine casein is considered as an important source of many bioactive peptides (BAPs), which can also be produced via in vitro simulated gastrointestinal hydrolysis. To perform their physiological functions, some active peptides need to pass through the intestinal epithelial barrier and keep their structural integrity after oral administration. Owing to the complexity of in vivo digestion and absorption, there have been few studies in this area. In this study, casein was labeled with FITC to trace its digestion and absorption in Sprague Dawley (SD) rats. Gastric juice, intestinal fluid, blood,

Impact of gastrointestinal physiology on drug absorption in special populations--An UNGAP review.

The release and absorption profile of an oral medication is influenced by the physicochemical properties of the drug and its formulation, as well as by the anatomy and physiology of the gastrointestinal (GI) tract. During drug development the bioavailability of a new drug is typically assessed in early clinical studies in a healthy adult population. However, many disease conditions are associated with an alteration of the anatomy and/or physiology of the GI tract. The same holds true for some subpopulations, such as paediatric or elderly patients, or populations with different ethnicity. The v

Study on the absorption of corosolic acid in the gastrointestinal tract and its metabolites in rats.

Corosolic acid (CRA) has been widely used as a food supplement. However, its pharmacokinetic behavior still needs to be explored. In this study, the absorption of CRA in stomach and intestine were investigated by in situ gastric absorption and in situ single-pass perfusion, respectively. Furthermore, the metabolites of CRA in rat plasma, bile, and urine were identified by UPLC-QTOF-MS. The enzymes responsible for its metabolism were explored by rat liver microsome (RLMs). The effects of plasma containing metabolites on cancer cell growth and glucose consumption were evaluated by HT29 and HepG2

Changes in the Expression of Aquaporin-3 in the Gastrointestinal Tract Affect Drug Absorption.

Aquaporin-3 (AQP3) plays an important role in water transport in the gastrointestinal (GI) tract. In this study, we conducted a Caco-2 cell permeability assay to examine how changes in the expression and function of AQP3 affect the rate at which a drug is absorbed via passive transport in the GI tract. When the function of AQP3 was inhibited by mercuric chloride or phloretin, there was no change in warfarin permeability. In contrast, when the expression of AQP3 protein was decreased by prostaglandin E₂ (PGE₂) treatment, warfarin permeability increased to approximately twice the control level,

[Characterization of drug absorption from the gastrointestinal tract with the use of numeric deconvolution].

The present investigation is concerned with the absorption of drugs from the gastrointestinal tract. The duration of absorption of drugs was calculated using numerical deconvolution. Clinical data were obtained from the literature. The absorption of 36 different drugs is characterized. The results show that the duration of absorption is not uniform. Penicillins and tetracyclines are absorbed for about 3 hours after application. Most drugs are absorbed during 4 to 6 hours. A few drugs, e.g. theophylline or metoprolol, are absorbed for more than 12 hours from the gastrointestinal tract.

Enhanced gastrointestinal absorption of drugs in rats pretreated with the synthetic immunomodulator, levamisole.

The effect of levamisole on drug absorption from the rat small intestine has been investigated by means of an in-situ recirculation technique. The absorption of salicylic acid, sulphanilamide and aminopyrine was significantly increased by the intraperitoneal administration of levamisole (2 mg) 1 day before the absorption studies, but there was no significant effect on absorption from the small intestine of indomethacin, bromthymol blue, sulphafurazole (sulfisoxazole), quinine, sulphanilic acid, phenol red (phenolsulfonphthalein), L-tryptophan and fluorescein isothiocyanate-dextrans. Drug absorption from the gastrointestinal tract and immunity: the mechanism of the decreased absorption of salicylic acid during systemic anaphylaxis. II.

Rats were intraperitoneally immunized with ovalbumin (egg albumin) with incomplete Freund's adjuvant, and the effect of intravenous challenge with ovalbumin on the intestinal blood flow was measured by means of hydrogen clearance method. The intestinal blood flow was significantly reduced by the antigen challenge in ovalbumin-immunized rats compared to the non-immunized rats. However, no significant change was observed on the intestinal blood flow in rats without challenge of the antigen. Moreover, the blood flow reduction was not found in ovalbumin-immunized rats challenged with bovine gamma-

Drug absorption from the gastrointestinal tract and immunity: the mechanism of the decreased absorption of salicylic acid during systemic anaphylaxis. I.

Previous studies have demonstrated the decrease of intestinal salicylic acid absorption in ovalbumin-immunized rats during systemic anaphylaxis. In the present study, the mechanism whereby systemic anaphylaxis interferes with the intestinal absorption of salicylic acid was studied. The pH of the luminal solution was not affected by the intravenous challenge with ovalbumin. A significant increase of the intraluminal protein was observed in rats under systemic anaphylaxis. However, there was no significant difference between ovalbumin-treated rats and saline-treated ones on the binding of salicy

Characterization of Gastrointestinal Absorption of Salmon Milt-Derived Oligodeoxyribonucleic Acids in Mice.

Dietary nucleic acids exert physiological effects beyond serving as substrates for nucleic acid synthesis; however, their exact molecular forms that are absorbed in the small intestine remain unclear. In this study, we investigated the absorption forms of food-derived DNA by comprehensively quantifying 31 nucleic acid monomers that permeated the intestinal membrane after oral administration, using LC-MS/MS. Hydrolyzed Oncorhynchus milt DNA (HD-omDNA; mainly 1-4-mer nucleotides), deoxyribonucleotides (dNTs), and deoxyribonucleosides (dNSs) were orally administered to mice, and blood samples wer

The gastrointestinal absorption characteristics and metabolic mechanisms of polysaccharides from Hohenbuehelia serotina.

In order to elucidate the digestion, absorption and metabolism mechanism, Hohenbuehelia serotina polysaccharides were successfully labelled by fluorescein isothiocyanate (FITC-Tyr-HSP), with the fluorescent substitution degree of 0.37 %. FITC-Tyr-HSP with excellent physiological stability possessed the significant fluorescent characteristics, which could be dynamically monitored by in vivo fluorescence imaging and high performance gel permeation chromatography-fluorescence detector (HPGPC-FLD). Through investigation of gastrointestinal digestion in vitro, the degradation of HSP was only occurr

Potential mechanisms by which Peceol increases the gastrointestinal absorption of amphotericin B.

PURPOSE: The purpose of this study was to ascertain how the incorporation of AmpB into a glyceride-rich excipient Peceol significantly increased Amphotericin B's (AmpB) gastrointestinal absorption in white male Sprague-Dawley rats. Based on preliminary studies, our working hypothesis was that incorporation of AmpB into mixed micelles composed of Peceol would significantly enhance gastro-intestinal (GI) tract absorption by increasing lymphatic drug transport and decreasing P-glycoprotein (PGP)-mediated drug efflux. METHODS: I. Lymphatic Transport STUDIES: Following an overnight fast (12-16 hr

Drug absorption in gastrointestinal disease and surgery. Clinical pharmacokinetic and therapeutic implications.

Drug absorption from the gastrointestinal (GI) tract and the impact of GI surgery and disease on drug absorption are discussed. Recommendations are made to manage problems of drug malabsorption. Absorption from the GI tract is a first-order process described by its rate and extent. GI surgery changes the anatomy of the GI tract and alters important variables in the absorption process. In the wake of procedures which diminish small bowel surface area, the extent of absorption of phenytoin, digoxin, cyclosporin, aciclovir, hydrochlorothiazide and certain oral contraceptives is reported to be red

The influence of individual gastrointestinal physiology parameters on prediction of regional drug absorption.

Physiologically based biopharmaceutics modeling (PBBM) is crucial for drug absorption prediction due to its ability to integrate drug-specific, physiological, and biopharmaceutic properties. Recent reports highlight the need to improve prediction accuracy for regional, colonic absorption, and extended-release (ER) formulations, often using average physiological data due to rare concurrent individual measurements (e.g., luminal pH and transit time). This investigation evaluated if in-vivo measured individual pH and transit time data could enhance absorption predictions. Metoprolol (BCS I), with

Insights into the Function of Aquaporins in Gastrointestinal Fluid Absorption and Secretion in Health and Disease.

Aquaporins (AQPs), transmembrane proteins permeable to water, are involved in gastrointestinal secretion. The secretory products of the glands are delivered either to some organ cavities for exocrine glands or to the bloodstream for endocrine glands. The main secretory glands being part of the gastrointestinal system are salivary glands, gastric glands, duodenal Brunner's gland, liver, bile ducts, gallbladder, intestinal goblet cells, exocrine and endocrine pancreas. Due to their expression in gastrointestinal exocrine and endocrine glands, AQPs fulfill important roles in the secretion of vari
