Emerging technologies to increase gastrointestinal transit times of drug delivery systems.

Apart from already established technologies to increase gastrointestinal transit times, including devices rapidly increasing in size once they have reached the stomach in order to retard the passage through the pylorus, formulations that float on gastric fluids and mucoadhesive drug delivery systems adhering to the gastrointestinal mucosa, there are new technologies emerging that might be game changing. They include mucus permeating nanocarriers that are able to diffuse deeply into the mucus gel layer of the gastric and intestinal mucosa remaining there for a prolonged time period (i), charge-

Effect of Laparoscopic-assisted Gastropexy on Gastrointestinal Transit Time in Dogs.

BACKGROUND: Prophylactic gastropexy has been promoted as a means of preventing gastric volvulus during gastric dilatation and volvulus (GDV) syndrome. Little is known about the impact of gastropexy on gastrointestinal transit time. HYPOTHESIS: Laparoscopic-assisted gastropexy (LAG) will not alter gastrointestinal transit times when comparing gastric (GET), small and large bowel (SLBTT), and whole gut transit times (TTT) before and after surgery. ANIMALS: 10 healthy client-owned large-breed dogs. METHODS: Prospective clinical trial. Before surgery, all dogs underwent physical examination and

Effect of Schistosoma mansoni infection on physiological gastrointestinal transit and contractility.

The work aimed to study the effect of Schistosoma mansoni (S. mansoni) on gastrointestinal transit and contractility of the colonic muscles of two subgroups of experimental mice, infected by 50 & 200 cercaria/mouse respevtively, at 8th & 12th week postinfection (PI). In addition, the histopathologic changes in the colon, and the immunological changes of the host were studied at different durations. At 8th weeks PI, in both subgroups, gastrointestinal transit was statistically significant decreased, in concurrent with statistically significant increase in the colonic muscle contractility compar

Gastrointestinal transit monitoring and absorption of controlled-release pellets of diltiazem.

The gastrointestinal (GI) transit and absorption of a multiparticulate controlled-release diltiazem formulation were investigated with reference to an innovator preparation. Transit of controlled-release pellets in GI tract was monitored using two marker drugs, namely paracetamol and sulfasalazine. Both formulations had little intersubject variation in gastric emptying and small intestine transit time. In both formulations, about 51% to 64% of the drug was absorbed while pellets were in the small intestine and the remaining amount while in the colon. The results found in this study were compar

Prolonged Gastrointestinal Transit Times and Dysmotility in m.3243A>G Mitochondrial Disease.

BACKGROUND: Gastrointestinal (GI) symptoms are frequently reported from carriers of the pathogenic mitochondrial DNA variant m.3243A>G, causing decreased mitochondrial adenosine triphosphate (ATP) production. ATP deficiency may adversely affect both autonomic neurogenic and myogenic regulation of GI motility, contributing to the symptoms. This study examined if carriers of m.3243A>G exhibit GI dysmotility, characterized as prolonged segmental transit times and decreased motility indices. METHODS: Twenty-two Danish carriers of m.3243A>G and 22 healthy, sex- and age-matched controls with body m

Effect of alvimopan and codeine on gastrointestinal transit: a randomized controlled study.

UNLABELLED: background & aims: Opiate bowel dysfunction is a significant clinical problem. Our aim was to evaluate the ability of a peripheral mu-opioid antagonist, alvimopan, to reverse the effect of codeine on gastric, small-bowel, and colonic transit time in healthy volunteers. METHODS: Seventy-four healthy participants (43 women) were randomized in a double-blind, placebo-controlled manner to 1 of 4 groups: alvimopan 12 mg twice daily in the presence and absence of codeine sulfate 30 mg 4 times/day, or codeine or placebo alone. Gastric emptying, small-bowel, and colonic transit were measu

Gastrointestinal transit in cirrhotic patients: effect of hepatic encephalopathy and its treatment.

Chronic hepatic encephalopathy is highly responsive to changes in diet, to antibiotic therapy and to ingestion of nondigestible disaccharides. The precise pathophysiology of chronic hepatic encephalopathy in individual cases is highly variable, although ammonia toxicity and production of neurotransmitterlike substances in the gut have been proposed to contribute to the overall syndrome of chronic hepatic encephalopathy. The support for this hypothesis is based on the empiric observation that reduction in protein intake, a catharsis or both are effective treatments for chronic hepatic encephalo

Proximal colonic response and gastrointestinal transit after high and low fat meals.

The fat component of meals has been thought to make a major contribution to the colonic response to feeding. We have combined gamma scintigraphy and radiotelemetry to noninvasively study the response of the normally inaccessible proximal colon after ingestion of either a high or low fat meal. Separate studies were performed to measure the rate of passage of the same meals through the whole gut. Gastric emptying and small bowel transit of the two meals to the colon was similar, 50% of meal marker reaching the ascending colon 4.8 +/- 0.2 and 4.5 +/- 0.3 hr after the high and low fat meals respec

Vagotomy inhibits the effect of neurotensin on gastrointestinal transit in the rat.

Neurotensin has previously been shown to delay gastric emptying, gastrointestinal transit and ileo-caecal emptying in the rat. To investigate the vagal influence on these effects of neurotensin, separate groups of rats were operated with combined vagotomy and pyloroplasty or with pyloroplasty alone and compared to a group of normal rats. All animals were supplied with a permanent gastrointestinal catheter and a venous catheter. After operation the rats were allowed to recover for 7 days, and were fasted for 24 h prior to the experiments. A radioactive marker of 1.0-0.5 ml Na2(51)CrO4 in isoton

Transit time changes with age in the gastrointestinal tract of the rat.

Using 85Sr-labelled microspheres as nonabsorbable markers, the effect of age on gastrointestinal propulsion motility was determined in conscious rats. There was no significant correlation between the gastric emptying rate or the propulsion motility of the small intestine and the age of the rats. However, the transit time of the intestinal content in the caecum in adult (17- to 20-month-old) rats was about fivefold the value found in weanling (19-day-old) rats. Also the transit time of the large intestine was found to be prolonged by about 70% during the life span investigated.

Hydrophilic Polymers Enable Gastrointestinal Transit Monitoring by Fluorine-19 Magnetic Resonance Imaging.

Gastrointestinal (GI) transit shapes nutrient absorption and oral drug delivery. However, GI anatomy, function, and motility disorders are primarily examined by radiographic imaging, which exposes patients to ionizing radiation and yields only coarse proxies of how a meal moves through the gut. By contrast, fluorine-19 (19F) MRI offers background-free, quantitative tracking, but most oral 19F agents are hydrophobic and phase-separate from chyme, generating layering artifacts, which spoil motility and anatomy readouts. Here, we developed nonresorbable, hydrophilic fluorinated polymer tracers mi

Disturbances of gastrointestinal transit and autonomic functions in postural orthostatic tachycardia syndrome.

BACKGROUND: Gastrointestinal symptoms are common in the postural orthostatic tachycardia syndrome (POTS). However, few studies have evaluated gastrointestinal transit in POTS. Our primary objectives were to evaluate gastrointestinal emptying and the relationship with autonomic dysfunctions in POTS. METHODS: We reviewed the complete medical records of all patients aged 18 years and older with POTS diagnosed by a standardized autonomic reflex screen who also had a scintigraphic assessment of gastrointestinal transit at Mayo Clinic Rochester between 1998 and 2012. Effect of daikenchuto (TU-100) on gastrointestinal and colonic transit in humans.

Daikenchuto (TU-100) is a traditional Japanese (Kampo) medicine used to treat postoperative ileus. TU-100 dose dependently increases gastrointestinal (GI) motility by modulating cholinergic and serotonergic mechanisms in animal studies. The aim of this study was to evaluate the effects of orally administered TU-100 on GI and colonic transit and bowel function in healthy humans. In a randomized, parallel-group, double-blind, placebo-controlled, dose-response study, 60 healthy subjects were randomly assigned to placebo or TU-100 2.5 g or 5 g tid ingested immediately before meals for 5 consecutiv

Effect of tolterodine on gastrointestinal transit and bowel habits in healthy subjects.

Clinical trials and observations suggest that constipation is an uncommon side effect of treating overactive bladder with the muscarinic receptor antagonist tolterodine. Because muscarinic antagonism inhibits gastrointestinal motor activity, we evaluated the effects of tolterodine on bowel habits, gastrointestinal and colonic transit in healthy subjects. In this double-blind study, 36 healthy subjects were randomized to tolterodine extended release (ER, 4 mg daily) or placebo for 6 days. Gastric emptying (GE), small bowel and colonic transit were assessed on days 4-6 by scintigraphy. Bowel hab

Effects of oleanolic acid glycosides on gastrointestinal transit and ileus in mice.

The effects of various oleanolic acid glycosides obtained from medicinal herbs on gastrointestinal transit (GIT) and ileus were investigated in fasted mice. Ileus was induced by the peritoneal-irritation or by the laparotomy with manipulation. One hour after the oral administration, three oleanolic acid 3-O-monodesmosides (oleanolic acid 3-O-glucuronide (3, 50 mg/kg), momordin Ic (4, 25 and 50 mg/kg), and momordin I (6, 25 mg/kg)) significantly accelerated GIT, but two oleanolic acid 3-O-monodesmosides (28-deglucosyl-chikusetsusaponins IV (8) and V (10)), oleanolic acid 3,28-O-bisdesmosides (m

Factors determining gastrointestinal transit time of several markers in the domestic fowl.

The aim of this study was to find out how marker characteristics could affect digestive transit time in Gallus gallus. One soluble marker, Cr-EDTA, and two insoluble markers, Cr2O3 and chromium-mordanted plant cells of two sizes, were used. Three- to six-week-old chickens were killed in series after the oral administration of the markers at intervals of 0, 0.5, 1, 2, 3, 5, 7, and 9 h. The amount of chromium in each digestive segment was determined by atomic absorption. There were some differences in the initial distribution of markers; whereas almost the total amount of the chromium-mordanted
