Efcab7 deletion sensitizes mice to the teratogenic effects of gastrulation-stage alcohol exposure.

Alcohol exposure during the gastrulation stage of development can disrupt Sonic hedgehog (Shh) signaling and cause eye, craniofacial, and brain defects. One of the genes that regulates Shh signaling is Efcab7, which encodes a protein that facilitates the actions of Smoothened (Smo), a critical component of the Shh pathway. Previous work from our lab has demonstrated that Efcab7 is differentially expressed between two sub-strains of C57BL/6 mice that differ in their sensitivity to gastrulation-stage alcohol exposure. The more alcohol-sensitive C57BL/6 J mice express lower levels of Efcab7 durin

Convergence and extension at gastrulation require a myosin IIB-dependent cortical actin network.

Force-producing convergence (narrowing) and extension (lengthening) of tissues by active intercalation of cells along the axis of convergence play a major role in axial morphogenesis during embryo development in both vertebrates and invertebrates, and failure of these processes in human embryos leads to defects including spina bifida and anencephaly. Here we use Xenopus laevis, a system in which the polarized cell motility that drives this active cell intercalation has been related to the development of forces that close the blastopore and elongate the body axis, to examine the role of myosin

Time-specific effects of ethanol exposure on cranial nerve nuclei: gastrulation and neuronogenesis.

During the development of the central nervous system, neurons pass through critical periods or periods of vulnerability. We explored periods of vulnerability for cranial nerve nuclei by determining the effects of acute exposure to ethanol during development on the number of neurons in mature brainstem. Long-Evans rats were injected with 2.9 g ethanol/kg body weight on one day between gestational day (G) 7 and G13, inclusive. Two hours later, animals received a second injection of 1.45 g/kg. Controls were injected with equivalent volumes of saline. Brainstems of 31-day-old offspring were cryose

Collinear activation of Hoxb genes during gastrulation is linked to mesoderm cell ingression.

The vertebral column exhibits segmentation and regionalization along the antero-posterior axis. During embryogenesis, the rhythmic production of the precursors of the vertebrae, the somites, imposes a segmented aspect to the spine, whereas the spine's regional differentiation is controlled by Hox genes. Here we show that in the paraxial mesoderm, Hoxb genes are first activated in a temporal collinear fashion in precursors located in the epiblast lateral to the primitive streak. Our data suggest that collinear activation of Hoxb genes regulates the flux of cells from the epiblast to the streak

Migrating mesoderm cells self-organize into a dynamic meshwork structure during chick gastrulation.

Migration of cell populations is a fundamental process in morphogenesis and disease. The mechanisms of collective cell migration of epithelial cell populations have been well studied. It remains unclear, however, how the highly motile mesenchymal cells, which migrate extensively throughout the embryo, are connected with each other and coordinated as a collective. During chick gastrulation, cells emerging from the primitive streak and migrating in the 3D space between ectoderm and endoderm (mesoderm region) exhibit a novel form of collective migration. Using live imaging and quantitative analys

Alteration of the anterior-posterior embryonic axis: the pattern of gastrulation in macrocephalic frog embryos.

The production of an enlarged head or macrocephaly in frog embryos can be achieved by interspecific hybridization or by injection of the contents of the germinal vesicle (GV), the large nucleus of immature oocytes, into the blastocoels of embryos before they gastrulate. The macrocephalic embryos have large suckers and their neural tubes are larger anteriorly but smaller posteriorly as compared to controls. This abnormal syndrome has previously been thought to arise as a result of an axial structure determinant present in the germinal vesicle. When examined during gastrulation, however, Xenopus

Critical periods for alcohol teratogenesis in mice, with special reference to the gastrulation stage of embryogenesis.

Studies directed towards determining a 'critical' developmental period for the induction of neural, ocular and craniofacial malformations typical of those in severe forms of the fetal alcohol syndrome (FAS) are described here. Sequential developmental analyses of C57Bl/6J mice whose mothers had been given alcohol either intraperitoneally or by gastric intubation on the seventh day of pregnancy (a time corresponding to three weeks after fertilization in humans) illustrate that early deficiencies in the anterior aspect of the embryonic disc of embryos at the gastrulation stage lead to defects ch

From Gastrulation to Malformation: Understanding the Embryology of Split Cord Malformations for Obstetric Practice.

BACKGROUND: Split cord malformation (SCM) is an uncommon congenital anomaly in which the spinal cord is longitudinally divided into two hemicords separated by a fibrous, cartilaginous, or osseous septum. Historically described as diastematomyelia or diplomyelia, the unified classification proposed by Pang et al. [Neurosurgery. 1992;31(3):451-80] distinguishes two main subtypes: type I, characterized by two hemicords within separate dural sacs divided by a rigid spur, and type II, in which hemicords share a single dural sac separated by a fibrous band. Composite SCM, involving multiple nonconti

Effects of alcohol on the transcriptome, methylome and metabolome of in vitro gastrulating human embryonic cells.

Prenatal alcohol exposure (PAE) affects embryonic development, causing a variable fetal alcohol spectrum disorder (FASD) phenotype with neurodevelopmental disorders and birth defects. To explore the effects of PAE on gastrulation, we used an in vitro model with subchronic moderate (20 mM) and severe (70 mM) ethanol exposures during the differentiation of human embryonic stem cells into germ layer cells. We analyzed genome-wide gene expression (mRNA sequencing), DNA methylation (EPIC Illumina microarrays) and metabolome (non-targeted LC-MS) of the endodermal, mesodermal and ectodermal cells. Th

Zic2-associated holoprosencephaly is caused by a transient defect in the organizer region during gastrulation.

The putative transcription factor ZIC2 is associated with a defect of forebrain development, known as Holoprosencephaly (HPE), in humans and mouse, yet the mechanism by which aberrant ZIC2 function causes classical HPE is unexplained. The zinc finger domain of all mammalian Zic genes is highly homologous with that of the Gli genes, which are transcriptional mediators of Shh signalling. Mutations in Shh and many other Hh pathway members cause HPE and it has been proposed that Zic2 acts within the Shh pathway to cause HPE. We have investigated the embryological cause of Zic2-associated HPE and t

Wnt/planar cell polarity signaling controls morphogenetic movements of gastrulation and neural tube closure.

Gastrulation and neurulation are successive morphogenetic processes that play key roles in shaping the basic embryonic body plan. Importantly, they operate through common cellular and molecular mechanisms to set up the three spatially organized germ layers and to close the neural tube. During gastrulation and neurulation, convergent extension movements driven by cell intercalation and oriented cell division generate major forces to narrow the germ layers along the mediolateral axis and elongate the embryo in the anteroposterior direction. Apical constriction also makes an important contributio

Sequential developmental changes in holoprosencephalic mouse embryos exposed to ethanol during the gastrulation period.

BACKGROUND: Prenatal exposure to ethanol induces holoprosencephalic malformations in both humans and laboratory animals. However, its teratogenic window for inducing holoprosencephaly is narrow, and the teratogenic mechanism is not well understood. In the present study, we examined the morphological changes in the craniofacial structures of mouse embryos/fetuses at intervals following ethanol treatment and evaluated gene expression patterns in the embryos. METHODS: Pregnant C57BL/6J mice were given two doses of ethanol (30 mg/kg in total) on the morning (7:00 and 11:00 AM) of day 7. The fetus

Exogenous retinoic acid during gastrulation induces cartilaginous and other craniofacial defects in Fundulus heteroclitus.

Embryonic levels of retinoic acid (RA) and the response of cells to RA are critical to the normal development of vertebrates. To understand the effects of RA signaling in Fundulus heteroclitus, we exposed embryos to a range of RA concentrations of 2 h during gastrulation. Embryos exposed to low concentrations of RA (10(-10)-10(-7) M) develop normally, whereas those exposed to higher concentrations (5 x 10(-7)-10(-4) M) develop characteristic dose-dependent defects. We describe, in detail, four stages of development that represent morphological effects of RA on (1) cell death and defects in the

Tetrabromobisphenol S-induced developmental toxicity in zebrafish embryos: impacts on cytoskeletal proteins during gastrulation.

3,3',5,5'-Tetrabromobisphenol S (TBBPS) is an emerging brominated flame retardant that is widely detected in the environment, yet its toxic effects remain poorly understood. The objective of this study was to use zebrafish as a model and determine the effects of TBBPS exposure on gastrulation stages of embryogenesis. We initiated TBBPS exposures (0 or 40 µM) at 0.75 h post fertilization (hpf), phenotyped hourly through cleavage, blastula, and gastrula stages, and showed that TBBPS-treated embryos exhibited delay in development beginning at ∼4 hpf, a stage corresponding to early gastrulation, w

Dynamic WT1 expression during gastrulation specifies peritoneal smooth muscle fate independently of mesothelial fate.

The Wilms' tumor protein (WT1) was previously linked to the mesothelial and vascular smooth muscle cell (vSMC) lineage in the mouse intestine, with evidence suggesting that intestinal vSMCs arise from the mesothelium. Here, we report that WT1 is already expressed, unexpectedly, during mouse gastrulation, in cells that specify a population of SMC precursor cells in the lateral plate mesoderm. Tamoxifen-induced genetic lineage tracing of Wt1-expressing cells revealed that vSMC and visceral smooth muscle cells (visSMCs) of the foetal mid-gut, but not mesothelial cells, were labelled after tamoxif

Intracellular calcium signal at the leading edge regulates mesodermal sheet migration during Xenopus gastrulation.

During the gastrulation stage in animal embryogenesis, the cells leading the axial mesoderm migrate toward the anterior side of the embryo, vigorously extending cell protrusions such as lamellipodia. It is thought that the leading cells sense gradients of chemoattractants emanating from the ectodermal cells and translate them to initiate and maintain the cell movements necessary for gastrulation. However, it is unclear how the extracellular information is converted to the intracellular chemical reactions that lead to motion. Here we demonstrated that intracellular Ca2+ levels in the protrusion

A cell-based boundary model of gastrulation by unipolar ingression in the hydrozoan cnidarian Clytia hemisphaerica.

In Cnidaria, modes of gastrulation to produce the two body layers vary greatly between species. In the hydrozoan species Clytia hemisphaerica gastrulation involves unipolar ingression of presumptive endoderm cells from an oral domain of the blastula, followed by migration of these cells to fill the blastocoel with concomitant narrowing of the gastrula and elongation along the oral-aboral axis. We developed a 2D computational boundary model capable of simulating the morphogenetic changes during embryonic development from early blastula stage to the end of gastrulation. Cells are modeled as poly

The patterns of birthmarks suggest a novel population of melanocyte precursors arising around the time of gastrulation.

Systematic work in the mouse and chicken has mapped out two neural crest-derived pathways of melanocyte precursor migration. With these in mind, this study reappraises the patterns of congenital pigmentary disorders in humans and identifies three recurrent patterns consistent across genetically different diseases. Only two of these are seen in diseases known to be melanocyte cell-autonomous. The segmental pattern correlates well with the classical dorsolateral population from animal studies, demonstrating respect of the midline, cranio-caudal axial mixing, unilateral migration and involvement

Rapid induction of apoptosis in gastrulating mouse embryos by ethanol and its prevention by HB-EGF.

BACKGROUND: Ethanol exposure during gastrulation and early neurulation induces apoptosis within certain embryonic cell populations, leading to craniofacial and neurological defects. There is currently little information about the initial kinetics of ethanol-induced apoptosis, and interest in the ability of endogenous survival factors to moderate apoptosis is growing. Ethanol alters intracellular signaling, leading to cell death in chick embryos, suggesting that apoptosis could occur rapidly and that signaling pathways activated by survival factors might reduce apoptosis. METHODS: Pregnant mic

Inactivation of mouse Twisted gastrulation reveals its role in promoting Bmp4 activity during forebrain development.

Twisted gastrulation (Tsg) is a secreted protein that regulates Bmp signaling in the extracellular space through its direct interaction with Bmp/Dpp and Chordin (Chd)/Short gastrulation (Sog). The ternary complex of Tsg/Chd/Bmp is cleaved by the metalloprotease Tolloid (Tld)/Xolloid (Xld). Studies in Drosophila, Xenopus and zebrafish suggest that Tsg can act both as an anti-Bmp and as a pro-Bmp. We have analyzed Tsg loss-of-function in the mouse. Tsg homozygous mutants are viable but of smaller size and display mild vertebral abnormalities and osteoporosis. We provide evidence that Tsg interac
