Neurodevelopmental disorder associated with gene ARF3: A case report.

We present a case study of a patient exhibiting acquired microcephaly along with global developmental delay and drug-resistant epilepsy. Brain magnetic resonance imaging revealed distinctive features, including a Z-shaped morphology of the brainstem, volumetric reduction of white matter, diffuse thinning of the corpus callosum, and partial fusion of the cerebellar hemispheres at their most cranial portion. Whole-exome sequencing uncovered a pathogenic variant in the ARF3 gene c.200A>T, p.(Asp67Val). The neurodevelopmental disorder associated with the ARF3 gene is exceptionally rare, with only

First report of a Mexican family with mutation in the CDH1 gene.

BACKGROUND: Germline mutations in E-cadherin (CDH1) gene are associated with autosomal-dominantly inherited cancer syndrome characterized by diffuse gastric cancer, lobular breast cancer, and in some families, cleft lip/palate. However, there may be generations in which these neoplasms do not occur at all in a family and later on, one or another carcinoma arises, which makes it difficult for physicians to think about hereditary origin. METHODS: We report the first Mexican family with CDH1 mutation (variant c.377del). RESULTS: An asymptomatic young woman underwent a search for mutations in su

EXPANDED PHENOTYPE OF TMEM67 GENE MUTATION (CASE REPORT).

Human ciliopathies are a class of multi-organ genetic disorders caused by defects of proteins expressed at the primary cilium, an organelle present on the cell surface of almost all cell types. Thus far, dozens of causative genes for ciliopathies have been identified and many of them are known to cause allelic disease. Of particular interest is the TMEM67 gene, encoding the transmembrane protein meckelin. The involvement of the mutant TMEM67 gene is known to be associated with a broad range of clinical presentations, namely Joubert syndrome 6 (JBTS6), nephronophthisis 11 (NPHP11), Bardet-Biede

Quantitative PET Reporter Gene Imaging with [11C]Trimethoprim.

There is a need for improved methods to image genetically engineered cells, including immune cells used for cell-based therapy. Given the genetic manipulation inherent to gene therapy, the use of a reporter protein is a logical solution and positron emission tomography (PET) can provide the desired sensitivity and spatial localization. We developed a broadly applicable PET imaging strategy based on the small bacterial protein E. coli dihydrofolate reductase (Ec dhfr) and its highly specific small molecule inhibitor, trimethoprim (TMP). The difference in TMP affinity for bacterial compared to m

LHX4 Gene Alterations: Patient Report and Review of the Literature.

LHX4 mutations are rare in combined pituitary hormone deficiency, and even rarer in isolated GHD. We describe a 14 years old boy who was referred for investigation of short stature. Convergent strabismus, nystagmus was present. At the age of 5 years his gait was unstable. A progressive myopathy ensued. Tests of pituitary reserve showed partial IGHD (8.2 ng/ml). Other pituitary hormones were within normal range. Muscle biopsy showed congenital myopathy of undefined etiology. MRI of the brain revealed the empty sella syndrome. Targeted resequencing with a panel containing probe sets for enrichme

Case Report: a family report of hereditary pancreatitis caused by SPINK1 and PRSS1 gene mutations.

Hereditary pancreatitis (HP) is a rare genetic disorder of the pancreas. We report a case of a 20-year-old woman presenting with classic features of lean diabetes mellitus, chronic diarrhea, and diabetic ketoacidosis but notably without abdominal pain. Imaging revealed pancreatic calcification. Genetic testing identified pathogenic compound mutations in SPINK1 (c.194+2T>C) and PRSS1 (c.623G>C), confirming a diagnosis of HP. Family screening showed her mother carried a homozygous SPINK1 mutation, while her siblings variably carried heterozygous SPINK1 and/or PRSS1 mutations. All family members

Predicted Effector Gene Aggregation, Standards and Unified Schema (PEGASUS): A Community Framework for Effector Gene Reporting.

Genome-wide association studies (GWAS) increasingly report predicted effector genes (PEGs) - genes hypothesised to mediate the biological effects of associated variants. These function as key outputs for advancing variant-to-function research, mechanistic understanding, and therapeutic discovery. However, the rapid growth of PEG lists has not been matched by standards for organising, annotating, and reporting these predictions. As shown by recent landscape analyses, PEG lists vary widely in methodology, evidence definition, nomenclature, provenance tracking, and data structure, limiting intero

Reappraisal of Reported Genes for Sudden Arrhythmic Death: Evidence-Based Evaluation of Gene Validity for Brugada Syndrome.

BACKGROUND: Implicit in the genetic evaluation of patients with suspected genetic diseases is the assumption that the genes evaluated are causative for the disease based on robust scientific and statistical evidence. However, in the past 20 years, considerable variability has existed in the study design and quality of evidence supporting reported gene-disease associations, raising concerns of the validity of many published disease-causing genes. Brugada syndrome (BrS) is an arrhythmia syndrome with a risk of sudden death. More than 20 genes have been reported to cause BrS and are assessed rout

A reporter gene assay to assess the molecular mechanisms of xenobiotic-dependent induction of the human CYP3A4 gene in vitro.

1. A plasmid containing 1 kb of the CYP3A4 regulatory (promoter) region coupled to a reporter gene for secretary placental alkaline phosphatase (SPAP) was transfected into HepG2 cells. Transfected cells were dosed with several known inducers of CYP3A4 and the levels of SPAP were measured. The effect of co-transfecting a plasmid encoding the human glucocorticoid receptor on reporter gene activity was also examined. 2. Dexamethasone induced CYP3A4-dependent reporter gene expression in a concentration-dependent manner and induction was approximately doubled in the presence of the glucocorticoid r

Nephrocalcinosis in a Child with Sotos Syndrome: A Case Report of Contiguous Gene Syndrome Encompassing NSD1 and SLC34A1 Genes.

Background: Nephrocalcinosis, characterized by the deposition of calcium salts within the renal parenchyma, is frequently identified incidentally in pediatric patients and may be associated with underlying genetic disorders. Sotos syndrome, a rare congenital overgrowth condition associated with neurodevelopment delay and congenital defects caused by mutations or deletions in the NSD1 gene, has been sporadically linked to renal abnormalities, including nephrocalcinosis. Clinical presentation: We report a case of a male patient with Sotos syndrome and concurrent nephrocalcinosis, in whom genetic

Bronchiectasis in a child with a homozygous DCDC2 gene mutation: A case report.

The DCDC2 (Doublecortin Domain-Containing Protein 2) gene encodes a ciliary protein involved in microtubule polymerization and ciliary assembly. Although DCDC2 has been associated primarily with neuronal migration and ciliopathies affecting the liver and kidneys, its role in pulmonary disease has not been clearly defined. Ciliary dysfunction, particularly in motile cilia of the respiratory tract, can impair mucociliary clearance and lead to bronchiectasis, as seen in primary ciliary dyskinesia. Here, we report a rare case of bronchiectasis associated with a pathogenic DCDC2 (NM_016356.5):c.558

A PET reporter ligand for quantitative imaging of gene expression in the brain.

The lack of brain penetrant and biologically stable positron emission tomography reporter systems hampers the development of neurological disease models and the monitoring of gene delivery because existing approaches depend on endogenous receptors that vary unpredictably in pathology. HaloTag, a fully exogenous protein label that forms rapid and irreversible bonds with synthetic ligands, provides a modular platform for engineering reporter probes with defined chemical properties. Here we developed a fluorine-18-labelled small-molecule HaloTag ligand optimized for brain entry and covalent reten

The Spermidine Synthase Gene as a Reporter of Transcription Inhibition in Escherichia coli.

Antimicrobial resistance is a major threat to modern society and healthcare, as it severely compromises the efficacy of standard antibiotic treatments. To meet the ever-increasing demand for novel antimicrobial drugs, it is crucial to develop new strategies for screening antimicrobial compounds and improve existing high-throughput techniques. Reporter systems that employ specific genetic markers are powerful tools not only for detecting antimicrobial activity of the substance being studied, but also for identifying the potential mechanism of its action. Among other metabolic pathways, RNA bios

Unilateral craniosynostosis associated with ZIC1 gene mutation: a case report.

Craniosynostosis is one of the most common craniofacial anomalies, resulting from premature fusion of one or more cranial sutures. While most cases are sporadic, a significant number have a genetic etiology, including monogenic mutations. Coronal synostosis, in particular, is frequently associated with genetic variants. Mutations in the zinc finger protein of cerebellum 1 (ZIC1) gene have recently been recognized as a rare cause of coronal craniosynostosis. We report an 11-month-old female infant with a ZIC1 mutation presenting with unilateral left coronal craniosynostosis, microcephaly, and m
