Environmental and Genetic Determinants of Ankylosing Spondylitis.

Exposure to heavy metals and lifestyle factors like smoking contribute to the production of free oxygen radicals. This fact, combined with a lowered total antioxidant status, can induce even more damage in the development of ankylosing spondylitis (AS). Despite the fact that some researchers are looking for more genetic factors underlying AS, most studies focus on polymorphisms within the genes encoding the human leukocyte antigen (HLA) system. The biggest challenge is finding the effective treatment of the disease. Genetic factors and the influence of oxidative stress, mineral metabolism diso

Genetic determinants of thyroid function in children.

OBJECTIVE: Genome-wide association studies in adults have identified 42 loci associated with thyroid stimulating hormone (TSH) and 21 loci associated with free thyroxine (FT4) concentrations. While biologically plausible, age-dependent effects have not been assessed. We aimed to study the association of previously identified genetic determinants of TSH and FT4 with TSH and FT4 concentrations in newborns and (pre)school children. METHODS: We selected participants from three population-based prospective cohorts with data on genetic variants and thyroid function: Generation R (N = 2169 children,

Genetic Determinants of Antibiotic Resistance in Francisella.

Tularemia, caused by Francisella tularensis, is endemic to the northern hemisphere. This zoonotic organism has historically been developed into a biological weapon. For this Tier 1, Category A select agent, it is important to expand our understanding of its mechanisms of antibiotic resistance (AMR). Francisella is unlike many Gram-negative organisms in that it does not have significant plasmid mobility, and does not express AMR mechanisms on plasmids; thus plasmid-mediated resistance does not occur naturally. It is possible to artificially introduce plasmids with AMR markers for cloning and ge

Dilated Cardiomyopathy: Genetic Determinants and Mechanisms.

Nonischemic dilated cardiomyopathy (DCM) often has a genetic pathogenesis. Because of the large number of genes and alleles attributed to DCM, comprehensive genetic testing encompasses ever-increasing gene panels. Genetic diagnosis can help predict prognosis, especially with regard to arrhythmia risk for certain subtypes. Moreover, cascade genetic testing in family members can identify those who are at risk or with early stage disease, offering the opportunity for early intervention. This review will address diagnosis and management of DCM, including the role of genetic evaluation. We will als

[GENETIC AND EPIGENETIC DETERMINANTS OF ALZHEIMER'S DISEASE].

Alzheimer's disease (AD) is one of the most important economic, medical and social problems in modern society. Numerous studies have highlighted the role of genetic and epigenetic factors in this disease. Increased risk of AD is associated with certain polymorphic variants of the APOE gene, as well as some other, less affecting genes. In the review, recent studies demonstrating the role of genetic and epigenetic factors in the etiology and pathogenesis of AD are discussed.

Overview of the genetic determinants of primary aldosteronism.

Primary aldosteronism is the most common cause of secondary hypertension. The syndrome accounts for 10% of all cases of hypertension and is primarily caused by bilateral adrenal hyperplasia or aldosterone-producing adenoma. Over the last few years, the use of exome sequencing has significantly improved our understanding of this syndrome. Somatic mutations in the KCNJ5, ATP1A1, ATP2B3 or CACNA1D genes are present in more than half of all cases of aldosterone-producing adenoma (~40%, ~6%, ~1% and ~8%, respectively). Germline gain-of-function mutations in KCNJ5 are now known to cause familial hyp

Genetic determinants of "cognitive impairment, no dementia".

Dementia is a heritable condition with devastating effects on both patients and their caregivers. Studies have identified genetic variants associated with increased susceptibility to late-onset Alzheimer disease (AD)-related dementia; however, no studies have assessed whether genetic variation is associated with the early stages of cognitive decline. Given that cerebrovascular disease is an established mechanism in which chronic ischemia increases susceptibility to dementia, we assessed whether genetic variation associated with either cardio-metabolic or AD-related traits is associated with an

Genetically determined encephalopathy: Rett syndrome.

Rett syndrome (RTT) is a severe neurodevelopmental disorder primarily affecting females that has an incidence of 1:10000 female births, one of the most common genetic causes of severe mental retardation in females. Development is apparently normal for the first 6-18 months until fine and gross motor skills and social interaction are lost, and stereotypic hand movements develop. Progression and severity of the classical form of RTT are most variable, and there are a number of atypical variants, including congenital, early onset seizure, preserved speech variant, and "forme fruste." Mutations in

2006 update on genetic determinants of osteoporosis.

Since the identification of vitamin D gene, the first one characterised by being related to osteoporosis, a high number of genetic studies, especially association studies, emerged to elucidate the genetic complex pattern of this disease. Osteoporosis is a skeletal chronic multifactorial disease, characterised by abnormal low bone mass and microarchitectural deterioration of bone tissue. It is getting a more and more important health problem, affecting a larger number of people, especially in developed countries. Studies demonstrated that variations in several genes, involved in bone metabolism

[Which malignant melanomas are genetically determined?].

There are familial forms in 1%-11% of all malignant melanomas. The familial type of malignant melanoma, however, is heterogeneous and not clearly defined. Three groups of familial malignant melanoma may be distinguished: malignant melanoma without additional skin disorders, melanoma associated with disturbances in the pigmentation system (including albinism, congenital nevi, and the dysplastic nevus syndrome), and malignant melanoma in other forms of genodermatosis. The most frequent variant of familial malignant melanoma is dysplastic nevus syndrome (a skin disease whose biologic and clinic i

[Genetic determinants of antibiotic resistance in enterobacteria].

Antibiotic resistance of enterobacterial strains from population isolated in Krasnodar region is rather often controlled by the "plasmid" genes. The conclusion is based on using the colony hybridization with [32P]-DNA fragments of plasmids, carrying the genetic determinants of antibiotic resistance, as a method for antibiotic resistance, genes screening. Kanamycin resistance in the majority of strains is coded by APH (3') II gene, streptomycin resistance by APH (3") gene, chloramphenicol resistance by CATI, sulphonilamide resistance by DHPS type II gene. Tetracycline resistance of the studied

Genetically determined chromosome instability syndromes.

Spontaneously increased chromosomal instability is well documented in the three autosomal recessive diseases, Fanconi's anemia (FA), Bloom's syndrome (BS), and ataxia telangiectasia (AT). Other conditions have been reported to be associated with chromosomal breakage. Some are still single observations: in Werner's syndrome only fibroblasts are affected, and systemic sclerosis may not be an inherited disease. Various aspects of FA, BS, and AT are discussed which have emerged since recent reviews have been published. The differential diagnosis in FA has become more important than it was in the p

Primary sex determination: genetics and biochemistry.

On the basis of widespread phylogenetic conservatism, it has been proposed that serologically-defined H-Y antigen is the inducer of primary sex differentiation in mammals, causing the initially indifferent gonad to become a testis rather than an ovary. The proposal has withstood extensive testing in a variety of biological circumstances: XX males have testes and are H-Y+ and fertile XY females lack testicular tissue and are H-Y-; soluble H-Y antigen induces testicular organogenesis in XX indifferent gonads of the fetal calf in culture; H-Y antibody blocks tubular reaggregation of dispersed XY

Genetic Determination of Maternal Behaviour in Pigs.

In sows, maternal behaviour is a commercially important trait which affects sow and piglet welfare and the quality and safety of human-animal interactions, especially if free farrowing of sows without crates is in place. The aim of this study was to evaluate maternal behaviour and to estimate its quantitative genetic parameters in Swiss Landrace sows (SLR). The lactating sows, with an average lactation period of 29-30 days and different parities (one to eleven) at four nucleus farms of the Swiss pig breeding company Suisag underwent behaviour tests and observations during two 12-week periods.

Genetic determinants of pancreatitis risk in hypertriglyceridemia.

PURPOSE OF REVIEW: In recent years, studies have shed light on the concept of risk heterogeneity among patients with severe hypertriglyceridemia (HTG). Several clinical risk factors for acute pancreatitis have been identified in this population, but the importance of different genetic factors above and beyond triglyceride concentration remains unclear. This review endeavours to summarize recent developments in this field. RECENT FINDINGS: Recent studies suggest that the molecular basis of severe HTG (polygenic susceptibility vs. rare pathogenic variants) can modulate the risk of acute pancrea

Genetic Determinants of Peripheral Artery Disease.

Peripheral artery disease-atherosclerosis of the abdominal aorta and lower extremity vascular bed-is a complex disease with both environmental and genetic determinants. Unmitigated disease is associated with major functional decline and can lead to chronic limb-threatening ischemia, amputation, and increased mortality. Over the last 10 years, major advances have been made in identifying the genetic basis of this common, complex disease. In this review, we provide an overview of the primary types of genetic analyses performed for peripheral artery disease, including heritability and linkage stu
