A Polygenic Lipodystrophy Genetic Risk Score Characterizes Risk Independent of BMI in the Diabetes Prevention Program.

CONTEXT: There is substantial heterogeneity in insulin sensitivity, and genetics may suggest possible mechanisms by which common variants influence this trait. OBJECTIVES: We aimed to evaluate an 11-variant polygenic lipodystrophy genetic risk score (GRS) for association with anthropometric, glycemic and metabolic traits in the Diabetes Prevention Program (DPP). In secondary analyses, we tested the association of the GRS with cardiovascular risk factors in the DPP. DESIGN: In 2713 DPP participants, we evaluated a validated GRS of 11 common variants associated with fasting insulin-based measu

Association of a Pathway-Specific Genetic Risk Score With Risk of Radiation-Associated Contralateral Breast Cancer.

IMPORTANCE: Radiation therapy for breast cancer is associated with increased risk of a second primary contralateral breast cancer, but the genetic factors modifying this association are not well understood. OBJECTIVE: To determine whether a genetic risk score comprising single nucleotide polymorphisms in the nonhomologous end-joining DNA repair pathway is associated with radiation-associated contralateral breast cancer. DESIGN, SETTING, AND PARTICIPANTS: This case-control study included a case group of women with contralateral breast cancer that was diagnosed at least 1 year after a first pr

Family history and genetic risk score combined to guide cancer risk stratification: A prospective cohort study.

Family history (FH) of cancer and polygenic risk scores (PRS) are pivotal for cancer risk assessment, yet their combined impact remains unclear. Participants in the UK Biobank (UKB) were recruited between 2006 and 2010, with complete follow-up data updated until February 2020 for Scotland and January 2021 for England and Wales. Using UKB data (N = 442,399), we constructed PRS and incidence-weighted overall cancer PRS (CPRS). FH was assessed through self-reported standardized questions. Among 202,801 men (34.6% with FH) and 239,598 women (42.0% with FH), Cox regression was used to examine the a

The inherited genetic contribution and polygenic risk score for risk of CLL and MBL: a narrative review.

Chronic lymphocytic leukemia (CLL) is a neoplasm of B-cells in the blood and monoclonal B-cell lymphocytosis (MBL) is a precursor state to CLL. This narrative review provides an overview of the genetic studies that identified 43 common variants associated with risk of CLL among individuals of European ancestry. Emerging studies found that ∼50% of these variants are associated with MBL risk. Moreover, the polygenic risk score (PRS) calculated from these CLL variants has been shown to be a robust predictor for both CLL and MBL risk among European ancestry individuals but a weak predictor among A

Genetic Susceptibility to Nephrolithiasis: A Genome-Phenome Approach with Polygenic Risk Score Analysis.

INTRODUCTION: Kidney stone disease (KSD) is multifactorial, and the genetic basis is poorly understood among the East Asian population. This study aims to elucidate comprehensive genetic factors associated with KSD. METHODS: We conducted a genome-wide association study (GWAS) on Taiwanese Han Chinese individuals from the Taiwan Biobank to investigate genetic predispositions to KSD. A total of 108,483 participants were included, of whom 7,635 reported having KSD. A phenome-wide association study (PheWAS) leveraging the National Health Insurance Research Database was used to verify GWAS finding

Composite microRNA-genetic risk score model links to migraine and implicates its pathogenesis.

The neurobiological mechanisms driving the ictal-interictal fluctuations and the chronification of migraine remain elusive. We aimed to construct a composite genetic-microRNA (miRNA) model that could reflect the dynamic perturbations of the disease course and inform the pathogenesis of migraine. We prospectively recruited four groups of participants, including interictal episodic migraine (i.e. headache free for >72 h, apart from prior and subsequent attacks), ictal episodic migraine (i.e. during moderate to severe migraine attacks), chronic migraine and controls in the discovery cohort. Contribution of a genetic risk score to ethnic differences in fatty liver disease.

BACKGROUND AND AIMS: Susceptibility to fatty liver disease (FLD) varies among individuals and between racial/ethnic groups. Several genetic variants influence FLD risk, but whether these variants explain racial/ethnic differences in FLD prevalence is unclear. We examined the contribution of genetic risk factors to racial/ethnic-specific differences in FLD. METHODS: A case-control study comparing FLD patients (n = 1194) and population-based controls (n = 3120) was performed. Patient characteristics, FLD risk variants (PNPLA3-rs738409 + rs6006460, TM6SF2-rs58542926, HSD17B13-rs80182459 + rs7261

Genetic risk score as a predictor of gestational diabetes in Central European Caucasians.

Gestational diabetes mellitus (GDM) is a common complication of pregnancy associated with perinatal complications and increased future risk of T2DM. GDM has a polygenic background partly overlapping with T2DM, and using a genetic risk score (GRS) that captures complex genetics may improve early detection of individuals at risk. SNPs previously associated with T2DM in the Czech population (21 loci) have been examined in a study comprising Caucasian (i) pregnant women with GDM (N = 416), (ii) non-diabetic population controls (N = 1170) and (iii) T2DM patients (N = 359) to analyse (di)similaritie

A Polygenic Risk Score for Predicting Racial and Genetic Susceptibility to Prurigo Nodularis.

Prurigo nodularis (PN) is an understudied inflammatory skin disease characterized by pruritic, hyperkeratotic nodules. Identifying the genetic factors underlying PN could help to better understand its etiology and guide the development of therapies. In this study, we developed a polygenic risk score that predicts a diagnosis of PN (OR = 1.41, P = 1.6 × 10-5) in two independent and continentally distinct populations. We also performed GWASs, which uncovered genetic variants associated with PN, including one near PLCB4 (rs6039266: OR = 3.15, P = 4.8 × 10-8) and others near TXNRD1 (rs34217906: OR

Genetic Risk Score for Coronary Disease Identifies Predispositions to Cardiovascular and Noncardiovascular Diseases.

BACKGROUND: The taxonomy of cardiovascular (CV) diseases is divided into a broad spectrum of clinical entities. Many such diseases coincide in specific patient groups and suggest shared predisposition. OBJECTIVES: This study focused on coronary artery disease (CAD) and investigated the genetic relationship to CV and non-CV diseases with reported CAD comorbidity. METHODS: This study examined 425,196 UK Biobank participants to determine a genetic risk score (GRS) based on 300 CAD associated variants (CAD-GRS). This score was associated with 22 traits, including risk factors, diseases secondary

Body mass index and depressive symptoms: instrumental-variables regression with genetic risk score.

The causal role of obesity in the development of depression remains uncertain. We applied instrumental-variables regression (Mendelian randomization) to examine the association of adolescent and adult body mass index (BMI) with adult depressive symptoms. Participants were from the Young Finns prospective cohort study (n = 1731 persons, 2844 person-observations), with repeated measurements of BMI and depressive symptoms (modified Beck's Depression Inventory). Genetic risk score of 31 single nucleotide polymorphisms previously identified as robust genetic markers of body weight was used as a pro

Association between genetic risk score and periodontitis onset and progression: a pilot study.

AIM: Recent research has focused attention on the single nucleotide polymorphisms (SNPs) involved in the host response in periodontitis. However, so as to combine the relatively small effects of individual genes the use of multi locus genetic risk (GRS) has been proposed. This study aims to evaluate whether the genetic risk score may predict periodontitis onset and progression. MATERIALS AND METHODS: Fifty patients were divided into various groups according to periodontal status. Total DNA was isolated from epithelial oral cells by a masked operator and the selected SNPs were analysed. A GRS

Family score as an indicator of genetic risk of primary open-angle glaucoma.

OBJECTIVES: To assess the genetic risk of open-angle glaucoma (OAG) in individuals by calculating a family score (FS), which summarizes the information of all relatives including their disease status, age, sex, and degree of kinship and to examine the genetic contribution to OAG with and without an increased intraocular pressure. METHODS: Case and control probands, derived from the Rotterdam Study, underwent the same ophthalmologic examination as their relatives. The FS of each proband was the sum of the differences between observed and expected values of OAG for all relatives. The FSs were c

Exploratory analysis of the genetics of impulse control disorders in Parkinson's disease using genetic risk scores.

OBJECTIVE: To study the association between impulse control disorders (ICDs) in Parkinson's disease (PD) and genetic risk scores (GRS) for 40 known or putative risk factors (e.g. depression, personality traits). BACKGROUND: In absence of published genome-wide association studies (GWAS), little is known about the genetics of ICDs in PD. GRS of related phenotypes, for which large GWAS are available, may help shed light on the genetic contributors of ICDs in PD. METHODS: We searched for GWAS on European ancestry populations with summary statistics publicly available for a broad range of phenoty

Use of a Maternal Genetic Risk Score to Derive and Test a Genetically Customized Fetal Growth Curve.

OBJECTIVE: Our aim was to determine whether using a maternal genetic risk score for birth weight (GRSBW) to genetically customize a fetal growth curve would improve model fit and identification of perinatal morbidity. METHODS: We performed a secondary analysis of a prospective cohort of individuals with singleton deliveries at or after 24 weeks of gestation, with ultrasound data and maternal genotypes. In random training (∼60%) and testing (∼40%) subgroups, we computed maternal GRSBW using 73 birth weight-associated single-nucleotide polymorphisms. In the training cohort, we performed log-lin

Genetic risk score analysis indicates migraine with and without comorbid depression are genetically different disorders.

Migraine and major depressive disorder (MDD) are comorbid, moderately heritable and to some extent influenced by the same genes. In a previous paper, we suggested the possibility of causality (one trait causing the other) underlying this comorbidity. We present a new application of polygenic (genetic risk) score analysis to investigate the mechanisms underlying the genetic overlap of migraine and MDD. Genetic risk scores were constructed based on data from two discovery samples in which genome-wide association analyses (GWA) were performed for migraine and MDD, respectively. The Australian Twi

Genetic risk scores associated with temperament clusters in Finnish depression patients.

OBJECTIVE: Cloninger's temperament dimensions have been studied widely in relation to genetics. In this study, we examined Cloninger's temperament dimensions grouped with cluster analyses and their association with single nucleotide polymorphisms (SNPs). This study included 212 genotyped Finnish patients from the Ostrobothnia Depression Study. METHODS: The temperament clusters were analysed at baseline and at six weeks from the beginning of the depression intervention study. We selected depression-related catecholamine and serotonin genes based on a literature search, and 59 SNPs from ten dif

Assessing the clinical utility of genetic risk scores for targeted cancer screening.

BACKGROUND: Genome-wide association studies have identified thousands of disease-associated single nucleotide polymorphisms (SNPs). A subset of these SNPs may be additively combined to generate genetic risk scores (GRSs) that confer risk for a specific disease. Although the clinical validity of GRSs to predict risk of specific diseases has been well established, there is still a great need to determine their clinical utility by applying GRSs in primary care for cancer risk assessment and targeted intervention. METHODS: This clinical study involved 281 primary care patients without a personal
