The projection of different retinal ganglion cell classes to the dorsal lateral geniculate nucleus in the hooded rat.

The morphology of retinal ganglion cells which project to different parts of the dorsal lateral geniculate nucleus (DLG) in the hooded rat has been investigated. Small amounts of a retrograde tracer (horseradish peroxidase) were injected into the DLG, then labelled retinal ganglion cells were examined in retinal wholemounts. After injections into different parts of the DLG, differences were noted in the size, morphology and retinal distribution of labelled retinal ganglion cells. Specifically, after injections into the antero-ventral part of the DLG labelled retinal ganglion cells were spread

The transcription factor Phox2b distinguishes between oral and non-oral sensory neurons in the geniculate ganglion.

Many basic characteristics of gustatory neurons remain unknown, partly due to the absence of specific markers. Some neurons in the geniculate ganglion project to taste regions in the oral cavity, whereas others innervate the outer ear. We hypothesized that the transcription factor Phox2b would identify oral cavity-projecting neurons in the geniculate ganglion. To test this possibility, we characterized mice in which Phox2b-Cre mediated gene recombination labeled neurons with tdTomato. Nerve labeling revealed that all taste neurons projecting through the chorda tympani (27%) and greater superfi

Changes in transient receptor potential channels in the rat geniculate ganglion after chorda tympani nerve injury.

We reported differential expression of the transient receptor potential vanilloid 1 (TRPV1), the transient receptor potential ankyrin 1 (TRPA1), and the (TRPM8) in the geniculate ganglions (GGs) of naive rats. In medical practice, the chorda tympani nerve (CTN) is injured in some patients during middle-ear surgery, and results in tongue numbness and taste disorder. We investigated changes in the expression of these receptors in GGs after CTN injury. In naive-rat GGs, 11.4, 11.8, and 0.5% of neurons were found to express the TRPV1, the TRPA1, the TRPM8, respectively. At 3 days after CTN injury,

Biophysical properties and responses to glutamate receptor agonists of identified subpopulations of rat geniculate ganglion neurons.

The goal of the current study was to evaluate the electrophysiological properties and responses to glutamate receptor agonists of rat geniculate ganglion (GG) neurons innervating the tongue. Subpopulations of GG neurons were labeled by injecting Fluoro-Gold (FG) or True Blue chloride into the anterior tongue and soft palate (AT and SP neurons) and applying FG crystals to the posterior auricular branch of the facial nerve (PA neurons). Three to 12 days later, the GG neurons were acutely isolated and patch clamped. Although many biophysical properties of the AT, SP and PA neurons were similar, s

Ribosomal Profiling of the Geniculate Ganglion Identifies the Receptor ALK as Critical for the Development and Maintenance of Oral Sensory Neurons.

The geniculate ganglion (GG) consists of two populations of neurons, sensory neurons that innervate the oral cavity and express the transcription factor Phox2b and somatosensory neurons that innervate the pinna and express Brn3a To identify signaling pathways necessary for oral sensory neuron development and physiology, the translatome of Phox2b+ GG neurons was selectively profiled using the RiboTag method. From this analysis, anaplastic lymphoma kinase (ALK) was identified as one of the most highly enriched tyrosine kinase receptors. In situ hybridization revealed that Alk was expressed in ne

A new entity in the differential diagnosis of geniculate ganglion tumours: fibrous connective tissue lesion of the facial nerve.

Differential diagnosis of geniculate ganglion tumours includes chiefly schwannomas, haemangiomas and meningiomas. We report the case of a patient whose clinical and imaging findings mimicked the presentation of a facial nerve schwannoma.Pathological studies revealed a lesion with nerve bundles unstructured by intense collagenisation. Consequently, it was called fibrous connective tissue lesion of the facial nerve.

Polymerase chain reaction amplification of herpes simplex viral DNA from the geniculate ganglion of a patient with Bell's palsy.

Bell's palsy is the most common cause of facial paralysis. In this study, we demonstrate the presence of herpes simplex viral type 1 (HSV-1) genomic DNA in the geniculate ganglion of a patient who had Bell's palsy. This association suggests that in this patient, HSV-1 may have caused Bell's palsy. If HSV-1 is a cause of Bell's palsy, treatment with acyclovir may be beneficial. Additional studies should be done to establish the prevalence of HSV-1 as an etiologic agent of Bell's palsy.

Temporal Lobe Retraction Provides Better Surgical Exposure of the Peri-Geniculate Ganglion for Facial Nerve Decompression via Transmastoid Approach.

PURPOSE: For the exposure of the labyrinthine segment of the facial nerve, transmastoid approach is not usually considered due to being situated behind the superior semicircular canal. To obtain a better view and bigger field for manipulation in the peri-geniculate area during facial nerve decompression, retraction of temporal lobe after bony removal of tegmen mastoideum was designed via transmastoid approach. MATERIALS AND METHODS: Fifteen patients with traumatic facial paralysis [House-Brackmann (HB) grade IV-VI], 3 patients with Bell's palsy (HB grade V-VI), and 2 patients with herpes zost

Transdural Stimulation for the Identification of a Dehiscent Geniculate Ganglion in Intradural Middle Cranial Fossa Surgery: A Proof-of-Principle Study.

PURPOSE: Facial nerve injury is a potential complication in intradural middle cranial fossa surgery, particularly because of dehiscence of the geniculate ganglion, exposing the nerve to thermal injury. Identifying the geniculate ganglion during surgical approaches is crucial for preserving facial nerve function. This study aimed to evaluate the feasibility and efficacy of transdural stimulation in identifying the geniculate ganglion during intradural approaches to improve the preservation of facial nerve function. METHODS: We conducted a retrospective analysis on patients who underwent surger

EGR4 is critical for cell-fate determination and phenotypic maintenance of geniculate ganglion neurons underlying sweet and umami taste.

The sense of taste starts with activation of receptor cells in taste buds by chemical stimuli which then communicate this signal via innervating oral sensory neurons to the CNS. The cell bodies of oral sensory neurons reside in the geniculate ganglion (GG) and nodose/petrosal/jugular ganglion. The geniculate ganglion contains two main neuronal populations: BRN3A+ somatosensory neurons that innervate the pinna and PHOX2B+ sensory neurons that innervate the oral cavity. While much is known about the different taste bud cell subtypes, considerably less is known about the molecular identities of P

Islet-2 Is Required for the Survival of Islet-Expressing Retinal Ganglion Cells but Not for Their Pathfinding to the Contralateral Dorsal Lateral Geniculate Nucleus.

Retinal ganglion cells (RGCs) exhibit remarkable diversity owing to their expression of developmentally expressed transcription factors. Many transcription factors are expressed in unique RGC populations, but their roles within these populations remain undiscovered. The transcription factor Islet-2 (Isl2) is expressed in approximately 30%-40% of contralateral projecting RGCs. Previous work by others found increased ipsilateral innervation of the dorsal lateral geniculate nucleus (dLGN) in Isl2 mutant mice, implicating Isl2 in promoting a contralateral RGC axon trajectory. Since Isl2 mutant mic

Morphology and distribution of retinal ganglion cells projecting to different layers of the dorsal lateral geniculate nucleus in normal and Siamese cats.

Electrophoretic injections of horseradish peroxidase were made into physiologically characterized sites within the different layers of the dorsal lateral geniculate nuclei (LGNd) of normal and Siamese cats. The histochemical procedures used stained the cell bodies, dendrites, and axons of retrogradely labeled ganglion cells. In both normal and Siamese cats, only alpha and beta ganglion cells are labeled by injections restricted to the A laminae. In normal cats, the alpha/beta ratios (number of labeled alpha cells/number of labeled alpha + beta cells) resulting from injections into lamina A inc
