Fate of Cd during microbial Fe(III) mineral reduction by a novel and Cd-tolerant Geobacter species.

Fe(III) (oxyhydr)oxides affect the mobility of contaminants in the environment by providing reactive surfaces for sorption. This includes the toxic metal cadmium (Cd), which prevails in agricultural soils and is taken up by crops. Fe(III)-reducing bacteria can mobilize such contaminants by Fe(III) mineral dissolution or immobilize them by sorption to or coprecipitation with secondary Fe minerals. To date, not much is known about the fate of Fe(III) mineral-associated Cd during microbial Fe(III) reduction. Here, we describe the isolation of a new Geobacter sp. strain Cd1 from a Cd-contaminated

One-megadalton metalloenzyme complex in Geobacter metallireducens involved in benzene ring reduction beyond the biological redox window.

Reversible biological electron transfer usually occurs between redox couples at standard redox potentials ranging from +0.8 to -0.5 V. Dearomatizing benzoyl-CoA reductases (BCRs), key enzymes of the globally relevant microbial degradation of aromatic compounds at anoxic sites, catalyze a biological Birch reduction beyond the negative limit of this redox window. The structurally characterized BamBC subunits of class II BCRs accomplish benzene ring reduction at an active-site tungsten cofactor; however, the mechanism and components involved in the energetic coupling of endergonic benzene ring re

Bridging inner membrane and periplasm in Geobacter sulfurreducens: structural and biochemical insights into CbcA and its redox partners.

Biotechnological applications such as bioremediation, bioenergy production, and microbial electrosynthesis are emerging as sustainable alternatives to conventional and environmentally harmful industrial practices. Advancing these technologies requires a deeper understanding of extracellular electron transfer, a process mediated by a network of redox partners bridging the inner membrane and the extracellular environment in electroactive bacteria. The CbcBA complex, a quinol:cytochrome c oxidoreductase from Geobacter sulfurreducens, is essential for the reduction of extracellular metal oxides an

Unveiling the synergistic regulation of Co-metabolic substrates and Fe(III) on tetrabromobisphenol A degradation by Geobacter sulfurreducens PCA.

Tetrabromobisphenol A (TBBPA) poses a significant environmental threat due to its persistence and toxicity. This study investigated the anaerobic degradation of TBBPA by Geobacter sulfurreducens PCA, focusing on the synergistic effects of co-metabolic substrates (glucose and humic acid) and Fe(III) sources (ferric citrate and ferrihydrite). Results demonstrated that both substrates significantly enhanced TBBPA degradation, with optimal doses of 4.0 g/L for glucose (83.31% degradation) and 0.4 g/L for humic acid (80.70% degradation). Kinetic analysis revealed that glucose primarily increased th

The Combined Effect of Hg(II) Speciation, Thiol Metabolism, and Cell Physiology on Methylmercury Formation by Geobacter sulfurreducens.

The chemical and biological factors controlling microbial formation of methylmercury (MeHg) are widely studied separately, but the combined effects of these factors are largely unknown. We examined how the chemical speciation of divalent, inorganic mercury (Hg(II)), as controlled by low-molecular-mass thiols, and cell physiology govern MeHg formation by Geobacter sulfurreducens. We compared MeHg formation with and without addition of exogenous cysteine (Cys) to experimental assays with varying nutrient and bacterial metabolite concentrations. Cysteine additions initially (0-2 h) enhanced MeHg

Regulation of two highly similar genes, omcB and omcC, in a 10 kb chromosomal duplication in Geobacter sulfurreducens.

The Fe(III)-reducing micro-organism Geobacter sulfurreducens requires an outer-membrane c-type cytochrome, OmcB, for Fe(III) reduction, but a related cytochrome, OmcC, which is 73 % identical to OmcB, is not required. The omcB and omcC genes are part of a tandem chromosomal duplication consisting of two repeated clusters of four genes. The 2.7 kb sequences preceding omcB and omcC are identical with the exception of a single base pair change. Studies that combined genetic, Northern blotting and primer extension analyses demonstrated that both omcB and omcC are transcribed as monocistronic and p

Fast and eco-friendly sulfidation of zero-valent iron using elemental sulfur and Geobacter sulfurreducens for groundwater remediation.

Sulfidated zero-valent iron (S-ZVI) with high electron selectivity and reactivity toward contaminants is a promising reagent for groundwater remediation. However, the widely-used chemical and mechanical sulfidation methods to prepare S-ZVI are often chemical- or energy-intensive. Biogenic sulfidation method relying on sulfate-reducing bacteria (SRB) are considered as green methods, yet are time-consuming and may release toxic and odorous hydrogen sulfide gas. Instead, elemental sulfur (S0) as sulfur precursor may avoid prolonged preparation time and hydrogen sulfide emission. However, elementa

Environmental considerations and bioremediation applications of the metal-, nitrate-, and organohalide-respiring bacterium Trichlorobacter lovleyi (formerly Geobacter lovleyi).

Trichlorobacter lovleyi (formerly Geobacter lovleyi) is a gram-negative bacterium that can couple the oxidation of acetate or other small organic acids with the reduction of soluble and insoluble electron acceptors, such as chlorinated solvents, heavy metals, and nitrate. Evidence has shown that T. lovleyi plays a direct role in nitrogen, iron, and carbon cycling, and its versatile metabolism can be leveraged for environmental biotechnology applications. However, the contributions of T. lovleyi to treatment of contaminants at groundwater sites or electron transfer in bioelectrochemical systems

Reduction of iron oxide nanoparticles by Geobacter sulfurreducens PCA involves outer membrane proteins and secreted redox-active substances.

Fe reducing bacteria (FRB), through extracellular electron transfer (EET) pathway, can reduce Fe(III) nanoparticles, thereby affecting the migration, transformation, and degradation of pollutants. However, the interaction of Fe(III) nanoparticles with the most commonly identified FRB, Geobacter sulfurreducens PCA, remains poorly understood. Herein, we demonstrated that the synergistic role of outer membrane proteins and periplasmic proteins in the EET process for α-Fe2O3, Fe3O4, and β-FeOOH nanoparticles by construction of multiple gene knockout strain. oxpG (involved in the type II secretion

Biochemical characterization and mercury methylation capacity of Geobacter sulfurreducens biofilms grown in media containing iron hydroxide or fumarate.

Geobacter species are common in iron-rich environments and can contribute to formation of methylmercury (MeHg), a neurotoxic compound with high bioaccumulation potential formed as a result of bacterial and archaeal physiological activity. Geobacter sulfurreducens can utilize various electron acceptors for growth including iron hydroxides or fumarate. However, it remains poorly understood how the growth on these compounds affects physiological properties of bacterial cells in biofilms, including the capacity to produce MeHg. The purpose of this study was to determine changes in the biochemical

Enhancement of direct interspecies electron transfer and methane production by co-culture of dual Methanosarcina species and Geobacter metallireducens.

Anaerobic digestion is a key technology for converting organic waste into methane, offering significant potential for renewable energy production and waste management. While the addition of conductive materials has been shown to improve direct interspecies electron transfer (DIET), their application faces challenges like biofouling, environmental risks, and increased operational costs. This study investigated the effects of co-culturing dual Methanosarcina (Methanosarcina barkeri and Methanosarcina acetivorans) and Geobacter metallireducens (DM-G) to enhance DIET and methane production without

The exacerbation of mercury methylation by Geobacter sulfurreducens PCA in a freshwater algae-bacteria symbiotic system throughout the lifetime of algae.

Mine-polluted wastewater with mercury (Hg) poses severe environmental pollution since Hg(II) can be converted to highly neurotoxic methylmercury (MeHg) under anaerobic conditions. Previous studies on Hg methylation have focused on aquatic sediments, but few have investigated the MeHg formation in water layers containing algae. In this study, we investigated the dynamic effect of algae on Hg methylation throughout the lifetime of algae. We found that Chlorella pyrenoidosa was a non-methylating alga and exhibited good tolerance to Hg stress (1-20 μg/L); thus Hg(II) could not inhibit the process

The shuttling effects and associated mechanisms of different types of iron oxide nanoparticles for Cu(II) reduction by Geobacter sulfurreducens.

Iron oxide nanoparticles (IONPs), commonly occurring in soils, aquifers and subsurface sediments, may serve as important electron shuttles for the biotransformation of coexisting toxic metals. Here, we explored the impact of different IONPs (low-crystallinity goethite and ferrihydrite, high-crystallinity magnetite and hematite) on the reduction of Cu(II) by Geobacter sulfurreducens and the associated electron shuttle mechanisms. All four IONPs tested can function as electron shuttles to enhance long distance electron transfer from bacteria to Cu(II). Upon IONPs addition, the rate of Cu(II) red

Inhibition of Geobacter dechlorinators at elevated trichloroethene concentrations is explained by a reduced activity rather than by an enhanced cell decay.

Microbial dechlorination of trichloroethene (TCE) is inhibited at elevated TCE concentrations. A batch experiment and modeling analysis were performed to examine whether this self-inhibition is related to an enhanced cell decay or a reduced dechlorination activity at increasing TCE concentrations. The batch experiment combined four different initial TCE concentrations (1.4-3.0 mM) and three different inoculation densities (4.0 × 10(5) to 4.0 × 10(7)Geobacter cells·mL(-1)). Chlorinated ethene concentrations and Geobacter 16S rRNA gene copy numbers were measured. The time required for complete c

Exposure of Geobacter-enriched electro-active biofilm to [BMIM][NTf2] ionic liquid inhibits current generation in a bioelectrochemical system.

In this work, the effect of 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide ([BMIM][NTf2]) ionic liquid (IL) on the behavior of bioelectrochemical systems (BES) was studied in operando. Geobacter-enriched (>60%) biofilms were formed on the anodes (+0.2 V vs. Ag/AgCl potential) and their response to adding [BMIM][NTf2] in 0, 0.22, 0.44 and 0.65 mM concentrations to the BES anolytes was evaluated with acetate substrate. The average peak current densities (7.32; 5.52; 4.24; 2.04 A m-2, respectively) showed decreasing values with increasing IL concentration. By replacing the anolytes

Characterization of the inner membrane cytochrome ImcH from Geobacter reveals its importance for extracellular electron transfer and energy conservation.

Electroactive bacteria combine the oxidation of carbon substrates with an extracellular electron transfer (EET) process that discharges electrons to an electron acceptor outside the cell. This process involves electron transfer through consecutive redox proteins that efficiently connect the inner membrane to the cell exterior. In this study, we isolated and characterized the quinone-interacting membrane cytochrome c ImcH from Geobacter sulfurreducens, which is involved in the EET process to high redox potential acceptors. Spectroscopic and electrochemical studies show that ImcH hemes have low

Motile Geobacter dechlorinators migrate into a model source zone of trichloroethene dense non-aqueous phase liquid: experimental evaluation and modeling.

Microbial migration towards a trichloroethene (TCE) dense non-aqueous phase liquid (DNAPL) could facilitate the bioaugmentation of TCE DNAPL source zones. This study characterized the motility of the Geobacter dechlorinators in a TCE to cis-dichloroethene dechlorinating KB-1(™) subculture. No chemotaxis towards or away from TCE was found using an agarose in-plug bridge method. A second experiment placed an inoculated aqueous layer on top of a sterile sand layer and showed that Geobacter migrated several centimeters in the sand layer in just 7days. Enhancement effects of dissolved organic matter leached from sewage sludge on microbial reduction and immobilization of Cr(VI) by Geobacter sulfurreducens.

Sewage sludge has a high concentration of dissolved organic matter (DOM) which contains compounds that can serve as electron donors or shuttles for metal reduction by dissimilatory metal reducing bacteria (DMRB). In this study, Cr(VI) removal by G. sulfurreducens, a common DMRB present in anaerobic soils, was examined in the presence or absence of sludge DOM. Two different types of sludge DOM were tested; composted sludge DOM (C-DOM) and anaerobically digested sludge DOM (A-DOM). Both sludge DOMs enhanced Cr(VI) reduction by G. sulfurreducens, but C-DOM was more effective likely because it had
