Measles pneumonia in a newborn.

We report a 21-day-old preterm infant who had severe respiratory distress of 6 days' duration and whose lungs revealed a giant cell pneumonia at necropsy. Measles antigen was demonstrated in mononuclear and multinucleated epithelial cells of the lung by immunoperoxidase staining. We recommend the immunostaining procedure to differentiate measles from other viruses, such as parainfluenza 2 or 3, and respiratory syncytial virus, all of which may produce giant cell pneumonias.

Usurpativirus massiliensis, a new giant virus related to clandestinovirus.

Giant virus discovery in 2003 revolutionized the virus paradigm. In 2015, we introduced a new host, Vermamoeba vermiformis, that led to the discovery of Faustovirus, Orpheovirus and two giant viruses (Clandestinovirus and a faustovirus) having distinct cytopathic effects despite being co-isolated from the same environmental sample and culture. This raised concerns about the possibility that the most "discreet" virus be overlooked. Here we report on Usurpativirus, a new giant virus closely related to Clandestinovirus and co-isolated with Faustovirus LCD7 in V. vermiformis. Its non-lytic replica

Differentiation of smallpox and camelpox viruses in cultures of human and monkey cells.

The cytopathic effect of smallpox and camelpox viruses has been compared in cell cultures derived from human and monkey tissues. The two viruses could easily be distinguished in HeLa, GMK-AH1, BSC-1 and WISH cells, camelpox producing multinucleate giant cells and smallpox producing rounding up of individual cells. In other cells (LLC-MK2, HEK and 2RhK) the differences were not so marked, and in some (VERO, HEL, K and HuTh) no differences were seen.

Combination of steroid with azathioprine in treatment of giant cell autoimmune hepatitis.

Giant cell hepatitis is a rare disorder after the newborn period. Drugs, autoimmunity, and viruses (lately, paramyxovirus infection) have been implicated in its etiology. Without treatment, liver dysfunction is progressive and fatal. Immunosuppression with steroids and azathioprine has been demonstrated to sustain improvement in the disease. In this report, a one-year-old boy who has giant cell hepatitis with Coombs' positive hemolytic anemia and anti-smooth muscle antibodies is presented, and the course of the disease and the patient's response to treatment with steroid and azathioprine is re

Refining a giant virus lineage: a novel order unifying Mamonoviridae and "Manesviridae," unveiled by the discovery of furtivovirus.

UNLABELLED: The evolutionary origins and taxonomic framework of giant viruses related to the family Mamonoviridae and its relative group, including clandestinovirus, remain unclassified due to gaps in genome size and host range between these two groups. This study aimed to address this gap by integrating our newly isolated virus with publicly available metagenome-assembled genomes (MAGs) to construct a more robust phylogenetic framework. Here, we report the isolation and characterization of a new giant virus, furtivovirus, using the unicellular amoeba Vermamoeba vermiformis as a host. Furtivov

Evidence for the acquisition of a proteorhodopsin-like rhodopsin by a chrysophyte-infecting giant virus.

Chrysophytes are nanoflagellate protists widespread in aquatic ecosystems with diverse trophic roles as primary producers and bacterivores. Molecular evidence suggests that chrysophytes are commonly infected by giant viruses, but isolates of such virus-host systems have not been reported. Here, we describe the first cultivated chrysophyte-infecting virus, Chrysophyceae Clade H virus SA1 (ChrysoHV), isolated along with its phago-mixotrophic host alga from surface waters in the tropical North Pacific Ocean. The ChrysoHV capsid (290 ± 40 nm diameter) is associated with a loose, sac-like membrane

Molecular surveillance of adenovirus and coronavirus in Procellariiformes, Brazil: Detection of new divergent adenoviruses.

Adenoviruses are widespread DNA viruses that infect a broad range of vertebrate hosts and are recognized for their long-term host coevolution, remarkable genetic diversity, and broad pathogenic potential. Coronaviruses circulate among wild birds due to their evolutionary potential and are relevant to animal and public health. However, the diversity and ecological role of these viruses in pelagic seabirds remain poorly understood. This study provides the first molecular survey of adenoviruses and coronaviruses in Procellariiformes from the South Atlantic. Fifty stranded individuals representing

Detection and infection dynamics of Marseillevirus-like viruses in Aedes aegypti: Evidence for vertical and human exposure pathways.

Mosquito-specific viruses (MSVs) have garnered increasing interest, particularly those phylogenetically related to arboviruses. Among them, giant viruses of the phylum Nucleocytoviricota, such as Marseillevirus, have been detected in mosquito populations. However, the infection dynamics of these viruses, especially between mosquitoes and vertebrate hosts, remain poorly understood. This study aimed to identify the presence of giant viruses in Aedes aegypti (Ae. aegypti) mosquitoes and to investigate their potential to persist across mosquito generations and infect humans, using Marseilles-like

Giant cell arteritis, polymyalgia rheumatica, and viral hypotheses: a multicenter, prospective case-control study. Groupe de Recherche sur l'Artérite à Cellules Géantes.

OBJECTIVE: Although suspected, a viral etiology has never been proven in giant cell arteritis (GCA). We tested for viruses known to induce multinucleated giant cells in human pathology, which include the parainfluenza viruses (HPIV), respiratory syncytial virus, measles virus, herpesviruses type 1 and 2, and the Epstein-Barr virus. METHODS: A multicenter case-control study on incident cases of temporal arteritis (TA) and polymyalgia rheumatica (PMR). Population based age and sex matched controls were randomly selected. Serological tests for IgG and IgM directed against the viruses listed abov

Investigation of coronaviruses, paramyxoviruses, and poxviruses in free-ranging anteaters and armadillos: a search for the unknown in Brazil.

Xenarthrans face increasing viral threats from anthropogenic pressures and domestic animals. In this study, we investigated the presence of coronaviruses, paramyxoviruses, and poxviruses in anteaters and armadillos from Brazilian biomes. We screened 236 individuals for these viruses by molecular methods. Canine distemper virus (CDV) was identified in four giant anteaters and one six-banded armadillo. A poxvirus, showing 88% sequence identity to molluscum contagiosum virus, was detected in one giant anteater. No coronaviruses were detected. These findings expand our knowledge on the host range

Variations of the Virome in Raw and Treated Water: A One-Year Follow-Up at Six Different Drinking Water Treatment Plants.

Little is known about virome changes in raw and drinking water over time, and differences between raw water sources and treatment technologies. This study used metagenomics to assess viruses prevalent in raw and drinking water samples over 1 year from six Swedish drinking water treatment plants (DWTPs) with varying treatment barriers and with different raw water sources. Sequences homologous to known viruses in the raw water samples were detected by amplification and next-generation sequencing and classified into 152 different virus species belonging to 76 virus families/orders. The majority w

[Pathologic anatomy of cytomegalovirus encephalomyelitis and varicella-zona virus encephalomyelitis].

Cytomegalovirus (CMV) infection of the nervous system is frequent in acquired immunodeficiency syndrome (AIDS) and can be responsible for encephalitis, encephalomyelitis, meningoradiculitis or polyradiculo-neuropathy. Encephalitis is characterized at microscopy by its periventricular and cerebellar location, and by the presence of cytomegalic cells, containing intranuclear and/or intracytoplasmic inclusions, microglial nodules and necrotic foci. The virus can infect almost all types of cells. Coexistence of CMV and HIV has been observed in giant cells of macrophagic origin. It has been suggest

Viral cysteine proteinases.

Dozens of novel cysteine proteinases have been identified in positive single-stranded RNA viruses and, for the first time, in large double-stranded DNA viruses. The majority of these proteins are distantly related to papain or chymotrypsin and may be direct descendants of primordial proteolytic enzymes. Virus genome synthesis and expression, virion formation, virion entry into the host cell, as well as cellular architecture and functioning can be under the control of viral cysteine proteinases during infection. RNA virus proteinases mediate their liberation from giant multidomain precursors in

A comparative review of viral entry and attachment during large and giant dsDNA virus infections.

Viruses enter host cells via several mechanisms, including endocytosis, macropinocytosis, and phagocytosis. They can also fuse at the plasma membrane and can spread within the host via cell-to-cell fusion or syncytia. The mechanism used by a given viral strain depends on its external topology and proteome and the type of cell being entered. This comparative review discusses the cellular attachment receptors and entry pathways of dsDNA viruses belonging to the families Adenoviridae, Baculoviridae, Herpesviridae and nucleocytoplasmic large DNA viruses (NCLDVs) belonging to the families Ascovirid

A new cell line from the embryonic tissues of Culex tritaeniorhynchus and its susceptibility to certain flaviviruses.

A new cell line from the embryonic tissue of Culex tritaeniorhynchus mosquito was established. Morphological studies carried out at the 45th passage level (P-45) showed four different cell types viz. epithelial-like cells, fibroblast-like cells, giant cells and vacuolated cells. Karyological studies indicated diploid (2n = 6) chromosomes in majority of cells irrespective of passage level. A twelve-fold increase of cell number was observed in 10 days at P-49. The cells could be preserved in liquid nitrogen for more than 40 months. Isoenzyme profile analysis with four enzymes clearly indicated t

Epigenome analysis of an algae-infecting giant virus reveals a unique methylation motif catalogue.

DNA methylation can epigenetically alter gene expression and serve as a mechanism for genomic stabilization. Advancements in long-read sequencing technology have allowed for increased exploration into the methylation profiles of various organisms, including viruses. Studies into the Nucleocytoviricota phylum of giant dsDNA viruses have revealed unique strategies for genomic methylation. However, given the diversity across this phylum, further inquiries into specific lineages are necessary. Kratosvirus quantuckense (formerly known as Aureococcus anophagefferens Virus, AaV) is predicted to encod

Primary biliary cirrhosis with multinucleated hepatocellular giant cells: implications for pathogenesis of primary biliary cirrhosis.

Histological and clinical features of syncytial giant cell hepatitis (GCH) are rarely observed in adults, and the disease has been associated with several autoimmune disorders and drug reactions. We describe here the case of a 62-year-old woman who presented with evidence of severe acute hepatocellular injury and cholestasis. Serum work-up demonstrated antimitochondrial antibodies specific for primary biliary cirrhosis (PBC) autoantigens, whereas markers of viral infection including hepatitis viruses, paramyxovirus and measles virus were negative. Liver histology revealed the presence of multi

Syncytium-forming HSV-1 in cancer gene therapy: From molecular mechanisms to clinical translation.

Gene therapy has emerged as a promising strategy for cancer treatment, yet challenges in efficient gene delivery remain a major barrier. Herpes simplex virus type 1 (HSV-1), as an oncolytic virus, has garnered attention for its potential in cancer therapy due to its replicative capacity, large genomic payload, and relatively low toxicity. Notably, syncytium-forming HSV-1 (SF-HSV-1) not only exhibits enhanced and sustained antitumor efficacy but also triggers profound immune responses. However, the exact molecular mechanisms orchestrating HSV-1-induced syncytium formation, its resulting cytotox

No evidence of parvovirus B19, Chlamydia pneumoniae or human herpes virus infection in temporal artery biopsies in patients with giant cell arteritis.

OBJECTIVES: Recent studies have suggested that infective agents may be involved in the pathogenesis of giant cell arteritis (GCA), in particular Chlamydia pneumoniae and parvovirus B19. We investigated temporal arteries from patients with GCA for these infections as well as human herpes viruses using the polymerase chain reaction (PCR). METHODS: Thirty temporal artery biopsies from 30 patients suspected of having GCA within a period of 1 yr were examined. Thirteen patients had classical GCA, two had biopsy-negative GCA, 10 patients had polymyalgia rheumatica and five patients had other condit
