PURPOSE: To present a case of glioblastoma multiforme which initially presented with only ophthalmic manifestations.
OBSERVATIONS: A 48-year-old man presented with decreased vision and pain with eye movements of the right eye. MRI of the brain showed increased T2/FLAIR signal involving the right optic nerve with no other identified abnormalities. He was treated with intravenous steroids for presumed optic neuritis. His visual acuity then rapidly worsened to no light perception, with new orbital apex symptoms including central retinal artery and vein occlusions and inferior division third and

BACKGROUND: Dp71 is the most abundant dystrophin (DMD) gene product in the nervous system. Mutation in the Dp71 coding region is associated with cognitive disturbances in Duchenne muscular dystrophy (DMD) patients, but the function of dystrophin Dp71 in tumor progression remains to be established. This study investigated Dp71 expression in glioblastoma, the most common and aggressive primary tumor of the central nervous system (CNS).
METHODS: Dp71 expression was analyzed by immunofluorescence, immunohistochemistry, RT-PCR, and immunoblotting in glioblastoma cell lines and cells isolated from

BACKGROUND: Glioblastoma multiforme (GBM) is a highly malignant glial tumour classified by the World Health Organization (WHO) as a stage IV astrocytoma. It varies in shape and size and can be cystic, vascular and necrotic. It often appears as a ring-enhancing lesion on magnetic resonance imaging (MRI). The most common symptoms of GBM, such as headache, vomiting and seizures, are due to increased intracranial pressure. The objective of this case report is to describe an atypical presentation of GBM.
CASE REPORT: A 53-year-old woman of Italian origin presented with a 2-week history of lack of

Glioblastoma is the most common and most aggressive primary brain tumour. Standard of care consists of surgical resection followed by radiotherapy and concomitant and maintenance temozolomide (temozolomide/radiotherapy→temozolomide). Corticosteroids are commonly used perioperatively to control cerebral oedema and are frequently continued throughout subsequent treatment, notably radiotherapy, for amelioration of side effects. The effects of corticosteroids such as dexamethasone on cell growth in glioma models and on patient survival have remained controversial. We performed a retrospective anal

BACKGROUND: Patients with glioblastoma (GBM) are at increased risk of initial and recurrent venous thromboembolism (VTE) but rates of recurrence and real-world treatment choices are incompletely understood.
OBJECTIVES: We aim to describe the treatment of incident VTE, report incidence and risk factors for recurrence.
PATIENTS/METHODS: We conducted a retrospective cohort study of consecutive Cleveland Clinic patients with GBM presenting with objectively diagnosed deep vein thrombosis (DVT) or pulmonary embolism (PE) from 2007 to 2013 with at least 6-month follow-up. We collected information o

To inform clinical management of glioblastoma patients, we estimated the relative prevalence (present at glioblastoma diagnosis) and incidence (newly diagnosed) of comorbid conditions among these patients and their matched controls. We identified 2,424 glioblastoma patients registered in the Swedish National Cancer Registry between 1993 and 2006. Next, 12,120 randomly sampled population-based controls were individually matched to cases on age, sex and calendar year of diagnosis. We then evaluated patient discharge data for selected potential comorbid conditions. Seizures (odds ratio (OR) 31.6,

Guillain-Barré syndrome (GBS) is defined as an acute demyelinating peripheral neuropathy. We describe a case of GBS in a patient with glioblastoma undergoing chemotherapy treatment. A 57 year old woman diagnosed with glioblastoma developed a subacute progressive history of bilateral symmetric numbness of her fingers and toes, belt-type neuropathic pain, a left facial droop and upper and lower extremity muscle weakness. There was no evidence of a tumor mass or leptomeningeal disease in the spine. Electrophysiological studies confirmed the diagnosis. Although rare, GBS should be considered in pr

We report a 33-year-old Chinese gentleman who presented with visual epilepsy and symptoms of raised intracranial pressure in which clinical examination revealed normal visual fields and acuity despite Magnetic Resonance Imaging (MRI) brain showing large contrast enhancing mass at the right occipital lobe. Craniotomy and excision of tumour was done and the histology confirmed glioblastoma multiforme (GBM). He completed radiotherapy and recovered well except developing left inferior homonymous quadrantropia post operatively which improved with time.

Glioblastoma multiforme is a rapidly growing anaplastic primary tumor of the central nervous system. It is frequently associated with neurofibromatosis or other hamartomatous syndromes, and in such kindreds may be found in several generations. We describe a highly inbred family in which glioblastoma multiforme presents in males only as an isolated central nervous system neoplasm on the right side, without evidence of other underlying genetic disease. It is speculated that in this family the tumor develops as the consequence of an autosomal recessive or an X-linked recessive mutation. In additi

PURPOSE: Glioblastoma (GBM) is the most common and aggressive primary brain tumor. Human GBM tumorspheres (TS) are essential for preclinical drug screening and establishing patient-derived xenograft (PDX) models, but their derivation is often limited to fresh tissue. Whether TS from cryopreserved tissues retain comparable molecular and biological properties to those from fresh tissues remains underexplored. We hypothesized that TS from cryopreserved tissues could provide a reliable alternative for TS derivation, thereby expanding accessibility for GBM research.
METHODS: TS isolation rates wer

Glioblastoma (GBM) represents one of the prototypical immune-cold tumors, characterized by profound immune suppression, T-cell exclusion, low neoantigen burden, and a highly immunosuppressive myeloid-dominant tumor microenvironment (TME). Despite advances in immunotherapy, including immune checkpoint blockade (ICB), CAR-T cells, and cancer vaccines, clinical benefits remain limited. This review synthesizes emerging evidence on multi-modal strategies aimed at reprogramming the cold TME into an immunologically active state. We highlight innate immune agonists, oncolytic virotherapy, precision na

Glioblastoma (GBM) remains one of the most aggressive and lethal brain tumors in adults, characterized by extensive heterogeneity, robust therapeutic resistance, and a dismal prognosis despite maximal surgical resection, radiotherapy, and chemotherapy. A defining hallmark of GBM is its complex tumor microenvironment (TME), a dynamic ecosystem comprising immune cells, vascular networks, extracellular matrix, and various stromal components that collectively drive tumor progression and therapeutic evasion. Within this intricate niche, astrocytes, traditionally regarded as passive support cells in

BACKGROUND: Dexamethasone-induced regression of an intracranial space-occupying lesion is commonly characteristic of primary central nervous system lymphoma (PCNSL). However, dexamethasone does not have an established chemotherapeutic role in glioblastoma multiforme (GBM). This is a report on dexamethasone-induced regression in GBM with the aim of exploring the mechanisms behind the phenomenon.
CASE DESCRIPTION: We present the case of a 63-year-old male presenting with status epilepticus. Initial imaging showed a high-grade glioma with significant vasogenic edema. Following 23 days of dexamet

Glioblastoma (GBM) remains a therapeutic challenge due to its heterogeneity and plasticity, which drive treatment resistance, especially when compounded by interactions with the brain microenvironment. Recent preclinical evidence indicates that trifluoperazine (TFP) inhibits treatment-induced malignant reprogramming of tumour cells, potentially helping to reduce tumour plasticity. TFP targets calmodulin, dopamine receptors, and stress-responsive proteins (nuclear protein 1, NUPR1). Through these mechanisms, TFP has been shown to reduce tumour growth, sensitise tumours to chemoradiotherapy, and

Sudden-onset anomic aphasia is a unique symptom that is suggestive of an acute etiology. This case presents a sudden-onset focal neurological deficit with an underlying brain tumor. A 68-year-old female awoke with sudden-onset anomic aphasia, with mild hypertension as her only medical history. After an initial stroke workup was unremarkable, magnetic resonance imaging found a focal lesion on the left temporal lobe. An electroencephalogram showed lateralized periodic discharge and a focal area of increased epileptic potential in the left temporal lobe. Brain biopsy revealed World Health Organiz

Glioblastoma (GB) is the most common and deadly type of primary malignant brain tumor with an average patient survival of only 15-17 months. GBs typically have hypoxic regions associated with aggressiveness and chemoresistance. Using patient derived GB cells, we characterized how GB responds to hypoxia. We noted a hypoxia-dependent glycolytic switch characterized by the up-regulation of HK2, PFKFB3, PFKFB4, LDHA, PDK1, SLC2A1/GLUT-1, CA9/CAIX, and SLC16A3/MCT-4. Moreover, many proangiogenic genes and proteins, including VEGFA, VEGFC, VEGFD, PGF/PlGF, ADM, ANGPTL4, and SERPINE1/PAI-1 were up-re

Glioblastoma multiforme (GBM) is commonly known as the most aggressive primary CNS tumor in adults. The mean survival of it is 14 to 15 months, following the standard therapy from surgery, chemotherapy, to radiotherapy. Efforts in recent decades have brought many novel therapies to light, however, with limitations. In this paper, authors reviewed current treatments for GBM besides surgery. In the past decades, only radiotherapy, temozolomide (TMZ), and tumor treating field (TTF) were approved by FDA. Though promising in preclinical experiments, therapeutic effects of other novel treatments inc

OBJECTIVES: Recently, the total area of the inflammatory infiltrate and the percentage of inflammatory infiltrate have been proposed as novel histopathological parameters to improve the stratification of patients with Sjögren's syndrome (SS) in clinical trials. Both these parameters provide a more accurate assessment of the extent of the infiltrate in minor salivary gland biopsies (MSGBs) and may overcome the bias related to the Focus score (FS). To date, however, only few studies have investigated their clinical value and feasibility. In this study we revised consecutive MSGBs obtained routin

Temozolomide (TMZ) is an oral alkylating agent used to treat glioblastoma multiforme (GBM) and astrocytomas. However, at least 50% of TMZ treated patients do not respond to TMZ. This is due primarily to the over-expression of O6-methylguanine methyltransferase (MGMT) and/or lack of a DNA repair pathway in GBM cells. Multiple GBM cell lines are known to contain TMZ resistant cells and several acquired TMZ resistant GBM cell lines have been developed for use in experiments designed to define the mechanism of TMZ resistance and the testing of potential therapeutics. However, the characteristics o
