Glycosylation is an important post-translational modification catalyzed by glycosyltransferases (GTs), which comprise a large group of enzymes with diversified specificity. Most GTs show high specificity toward donor and acceptor molecules, but some can recognize several donor or acceptor molecules and/or create novel glycosidic linkage types. This promiscuity has profound implications for cellular processes, influencing signaling pathways, protein stability, and disease progression. GT promiscuity may result from the structural peculiarities of the enzyme and acceptor, modulation by a ligand,

Streptococcus agalactiae [group B Streptococcus (GBS)] is a leading cause of neonatal meningitis, with late-onset disease (LOD) occurring after gastrointestinal tract colonization in infants. Bacterial membrane lipids are essential for host-pathogen interactions, and the functions of glycolipids are yet to be fully elucidated. GBS synthesizes three major glycolipids: glucosyl-diacylglycerol (Glc-DAG), diglucosyl-DAG (Glc2-DAG), and lysyl-Glc-DAG (Lys-Glc-DAG). Here, we identify the enzyme, IagB, as responsible for biosynthesis of Glc-DAG, the precursor for the two other glycolipids in GBS. To

The abilities of lectins to combine specifically with particular carbohydrate residues have made them valuable probes in the analysis of the surface components of intestinal epithelial cells. Fluorescence microscopy studies using FITC-labeled lectins have shown that the surfaces of these cells change as the cells move up the villi and that these changes are dependent upon the region of intestine in which the villi are located. The lectins have been used in radioassays developed to quantitate these cell surface components and have proven useful in monitoring the extraction and purification of t

A simple and efficient method to synthesize the immunogenic glycolipid BbGL1 is introduced. Two simple steps were required to obtain the desired product in good yield. First, a highly efficient glycosylation of cholesterol using galactosyl trichloroacetimidate as a donor was performed to produce cholesteryl-β-d-galactoside. Finally, an efficient palmitoylation on the C6-OH of the galactose of the synthesized saponin using sym-collidine and acyl chloride under microwave heating that produced BbGL1 in good yield. The procedure is a convenient and cheaper alternative to the reported procedures al

Distinct glycolipid profiles are described in microorganisms, which have been shown to modulate the innate immune system. We tested the hypothesis that glycosphingolipids from Paracoccidioides brasiliensis have immunomodulatory properties on monocytes and dendritic cells of two groups of healthy individuals, one cured of paracoccidioidomycosis in the past (CUR-I) and the other nonexposed to P. brasiliensis (HNE-I). Two classes of glycosphingolipids purified from yeast cells were evaluated: a neutral glycosphingolipid, monohexosylceramide (CMH), and acidic glycosylinositolphosphorylceramides (G

Autoantibodies to glycans present on glycolipids mediate the postinfectious paralytic disease, Guillain-Barré syndrome (GBS). These glycans are also found on lipo-oligosaccharides (LOSs) of GBS-inducing microbes, suggesting molecular mimicry as a mechanism for disease induction. How B lymphocyte tolerance to self-glycans is regulated during the initiation phase of the disease is currently under investigation. The discovery of antiglycolipid antibodies that bind to heteromeric glycolipid complexes has generated new insights in this field. Heteromeric complexes are structurally distinct glycolip

Medical interest in glycolipids has been mainly directed to the rare and complex glycosphingolipid storage disorders that are principally caused by unitary deficiencies of lysosomal acid hydrolases. However, glycolipids are critical components of cell membranes and occur within newly described membrane domains known as lipid rafts. Glycolipids are components of important antigen systems and membrane receptors; they participate in intracellular signalling mechanisms and may be presented to the immune system in the context of the novel CD1 molecules present on T lymphocytes. A knowledge of their

Diabetes and obesity have become the most popular metabolic diseases in the world. A large number of previous studies have shown that glucose and lipid metabolism disorder is an important risk factor and a main cause of diabetes and obesity. Schistosoma is a parasite transmitted by freshwater snails. It can induce a series of inflammatory and immune reactions after infecting the human body, causing schistosomiasis. However, in recent years, studies have found that Schistosoma infection or Schistosoma related products can improve or prevent some immune and inflammatory diseases, such as severe

BACKGROUND: Human trichomoniasis is the most common non-viral sexually transmitted disease, yet immune responses are not well studied.
METHODS: Since the Trichomonas vaginalis lipophosphoglycan (TvLPG) is an important virulence factor, a bank of eight monoclonal antibodies was generated to define the antigen in clinical isolates. The TvLPG-specific antibody response of women who were culture positive (n=33) or negative (n=33) for T vaginalis infection was determined by isotype-specific ELISA.
RESULTS: The bank of monoclonal antibodies reacted with conserved surface TvLPG epitopes in 27 isola

Cholera toxin (CT) is the etiological agent of cholera. Here we report that multiple classes of fucosylated glycoconjugates function in CT binding and intoxication of intestinal epithelial cells. In Colo205 cells, knockout of B3GNT5, the enzyme required for synthesis of lacto- and neolacto-series glycosphingolipids (GSLs), reduces CT binding but sensitizes cells to intoxication. Overexpressing B3GNT5 to generate more fucosylated GSLs confers protection against intoxication, indicating that fucosylated GSLs act as decoy receptors for CT. Knockout (KO) of B3GALT5 causes increased production of f

Novel mycoloyl glycolipids with short carbon chains were isolated and purified from Rhodococcus sp. 4306, a soil origin of Actinomycetales. Their chemical structures were identified as trehalose 6,6'-dimycolate (TDM), trehalose 6-monomycolate, glucose 6-monomycolate, mannose 6-monomycolate and fructose 6-monomycolate. The length of carbon chains and number of double bonds of mycolic acids were C(34), C(36)and C(38)saturated, monoenoic and dienoic molecular species, which were much shorter than those of Mycobacterium tuberculosis (C(78-88)monoenoic and dienoic). Among them, only TDM could induc

Immunoglobulins from patients with paraproteinemic polyneuropathy were screened for reactivity with nerve and brain glycolipids by ELISA and/or a thin-layer-chromatogram-overlay technique. The myelin-associated glycoprotein (MAG) has been shown to be an antigen in many neuropathy patients with IgM gammopathy, but this study focused on seven neuropathy patients in which the IgM paraproteins had been shown not to react with this glycoprotein. Five of these seven had IgM that reacted with components in the acidic glycolipid fraction of human sciatic nerve, and three of these IgMs also reacted wit

Glycolipids are sugar-based amphiphiles that play crucial roles in many biological processes. Under thermotropic and lyotropic conditions, glycolipids self-assemble into a variety of mesophases, including cocontinuous network phases, such as the double gyroid. In this work, a two-stage molecular dynamics simulation workflow is developed to probe network formation for solvent-free amphiphiles at different temperatures. In the first stage, the structural evolution of systems initiated in lamellar and hexagonally packed cylinder arrangements is examined as an indicator of the likelihood of networ

Immunobiological activity of selected decyl and (thio)dodecyl hexopyranosides based on d-glucose, d-galactose, N-acetyl d-glucosamine and d-mannose and their effect on leukemia cell lines L1210 and HL-60 and Candida's biofilm were studied. Alkyl d-glucosides and d-galactosides showed mainly similar antiproliferative properties on leukemia cell lines, while N-acetyl d-glucosaminides revealed diverse properties with lower efficacy. Also, the cytokine response of RAW 264.7 macrophages was significantly influenced by the type of sugar moiety and (thio)alkyl chain length. Contrary to the proliferat

The neutral and acidic fraction glycolipids of Echinococcus granulosus metacestode tissue compartments were isolated, defined by their chromatographic and antigenic properties, and assessed as to their efficacy as antigens in the serodiagnosis of human hepatic cystic and alveolar echinococcosis, and other helminthiases. Analyses were accomplished by thin-layer chromatography immunostaining and ELISA. The neutral glycolipid fraction's major carbohydrate epitope was the same as or very similar to that of Taenia crassiceps neutral glyco(sphingo)lipids, as represented by the 'neogala'-series core

A Mycobacterium leprae-specific phenolic glycolipid antigen was purified from Formalin-fixed liver preserved from an advanced lepromatous leprosy patient. Its chemical and immunological properties were compared with those of phenolic glycolipid-I obtained from M. leprae-infected armadillo liver. Based on the findings that the glycolipids from the two sources have the same thin-layer chromatographic properties, infrared absorption spectrum, sugar composition, and seroreactivity, we conclude that large quantities of the phenolic glycolipid-I antigen are produced in human lepromatous leprosy lesi

Antibodies to glycolipids likely play a pathogenic role in the development of immune-mediated neuropathy. Guillain-Barre syndrome (GBS), an acute immune-mediated polyradiculoneuropathy, often has IgG antibodies to gangliosides, sialic acid-containing glycolipids. The heterogeneity of ganglioside expression in the peripheral nervous system (PNS) may underlie the differential clinical manifestation of GBS variants. Recent studies show that anti-ganglioside antibody-mediated complement activation plays a key role in the development of GBS and related disorders. Futher, complement-independent nerv

Disorders of lysosomal metabolism often involve the accumulation of specific types of glycolipid, particularly gangliosides, because of either degradative failure or other currently unknown mechanisms. Although the precise role of gangliosides in cells remains enigmatic, the presence of specific abnormalities secondary to ganglioside accumulation in lysosomal diseases has suggested important biological functions. Chief among these is the growth of new dendrites on particular classes of mature neurons secondary to an increase in GM2 ganglioside. That GM2 has also been shown to be elevated in no
