Urinary GAGs from patients affected with Osteogenesis Imperfecta (O.I.) type I, type II and type III - according to Sillence et al. (1979) - have been investigated. Galactosamine to glucosamine ratio resulted significantly decreased in O.I. type II and III, whereas smaller differences in the mildest type of the disease were observed. Cellulose polyacetate electrophoresis of urinary GAGs purified from some patients showed the presence of a slowly moving polysaccharide substance, which did not appear in normal subjects. Moreover some pathological fractions, mainly constituted of ChS on the basis

The basic defects of a number of genetic disorders affecting the connective tissue have been elucidiated during the last few years. A similar progress in understanding the pathobiochemistry of other dermatological diseases is expected. The present review summarizes our knowledge about structure, synthesis, degradation, macromolecular organization and possible function of glycosaminoglycans, which are an important constitutent of the ground substance in the skin. The difficulties of relating the clinical symptoms of diseases with the basic defects are examplified for the mucopolysaccharidoses.

Glycosaminoglycans (GAGs) are linear, negatively charged biopolymers that modulate complex biological processes, such as blood coagulation, immune regulation, or viral entry. Their sulfation pattern and chain length govern how strongly they bind to other physiologically relevant species. Most of these interactions rely on electrostatic forces facilitated by the strong polyanionic properties of GAGs; therefore, considering these from a polyelectrolyte vantage point can help understand how such charge-based, often transient interactions contribute to physiological and pathological processes. Whi…

Glycosaminoglycans (GAGs) are long blocks of negatively charged polysaccharides. They are one of the major components of the extracellular matrix and play multiple roles in different tissues and organs. The accumulation of undegraded GAGs causes mucopolysaccharidoses (MPS). GAGs are associated with other pathological conditions such as osteoarthritis, inflammation, diabetes mellitus, spinal cord injury, and cancer. The need for further understanding of GAG functions and mechanisms of action boosted the development of qualitative and quantitative (alcian blue, toluidine blue, paper and thin lay…

Osteoarthritis (OA) influences the levels of free intraarticular glucosaminoglycans (GAG). Little is known about the direction--decrease/increase--of these changes, and information on the correlation between GAG levels and the degree of OA is sparse. Objectives of this study were to investigate the correlation between intraarticular levels of sulphated and unsulphated GAG and the degree of experimental OA, the time course of these changes and whether GAG might be useful as a marker for OA. Twenty-one sheep were randomly assigned to three groups: (1) transsection of the posterolateral bundle of

BACKGROUND: Since Brockhurst reported the connection between uveal effusion in nanophthalmic eyes and their scleral alterations and treated them with vortex vein decompression or sclerectomy, many observers have found abnormal accumulation of glycosaminoglycans (GAGs) in nanophthalmic sclera. These GAG abnormalities were thought to effect the collagen changes, though it was not clear which GAGs were changed.
METHODS: GAGs were isolated and their contents were determined in scleral specimens from three nanophthalmic patients and five age-matched controls, using electrophoresis and the cetylpyr…

Histochemical examination of odontogenic cyst connective tissue has demonstrated that hyaluronic acid is the predominant glycosaminoglycan present with lesser amounts of the sulphated glycosaminoglycans. Mast cells were frequently observed in the connective tissue, particularly just beneath the epithelium and may be responsible for the enhanced staining for heparin observed in this area. Local tissue metabolism and inflammation will contribute to the release of glycosaminoglycans from connective tissue into the lumenal fluid. Staining of glycosaminoglycans was generally minimal in the epitheli…

Zoonotic diseases are infectious diseases that pass from animals to humans. These include diseases caused by viruses, bacteria, fungi, and p … e in frequency. Because of this trend, new treatments and prevention strategies are being explored. Glycosaminoglycans (GAGs) are complex linear polysaccharides that are ubiquitously present on the surfaces of most human and animal cells. In many infectious diseases, the interactions between GAGs and zoonotic pathogens correspond to the first contact that results in …

Glycosaminoglycans (GAGs), known to be present in airway mucus, are macromolecules with a variety of structural and biological functions. In the present work, we used fluorophore-assisted carbohydrate electrophoresis (FACE) to identify and relatively quantify GAGs in human tracheal aspirates (HTA) obtained from healthy volunteers. Primary cultures of normal human bronchial epithelial (NHBE) and submucosal gland (SMG) cells were used to assess their differential contribution to GAGs in mucus. Distribution was further assessed by immunofluorescence in human trachea tissue sections and in cell cu…

1. Certain compounds (e.g., the immunomodulator tilorone and congeners) are able to induce lysosomal storage of sulphated glycosaminoglycans (GAG), thus, producing cytological and biochemical alterations reminiscent of the inherited mucopolysaccharidoses. The drug-induced GAG storage has been studied in cultured fibroblasts of several species and in rats, and it is likely to occur also in humans. 2. The cytological hallmarks of GAG storage are enlarged lysosomes congested with material that is intensely stained by cationic dyes. With respect to fixation techniques, one has to keep in mind that…

BACKGROUND: It has been observed that there may be some connection between scleral collagen changes, glycosaminoglycan (GAG) changes and nanophthalmic uveal effusion. In a previous paper we reported increased dermatan sulfate (DS) and chondroitin sulfate (CS) contents in nanophthalmos sclera using biochemical analysis. In the present work we used immunohistochemical methods to assess the GAG changes in nanophthalmos sclera.
METHODS: Scleral specimens from three nanophthalmic patients and five controls were processed to immunostaining with peroxidase-antiperoxidase (PAP) methods. Monoclonal an

Interaction of glycosaminoglycans (GAG) with cationic fluorochromes, which are used for binding with DNA, was studied in biological material (urine, extract from hyalin cartilage) obtained from patients with hereditary mucopolysaccharidoses and from healthy persons. Among the four fluorochromes studied in the reactions with standard GAG's, DAPI proved to be the most suitable fluorochrome. Quantitative fluorimetric technique enabled to estimate the GAG concentration and to evaluate the individual content of heparan-SO4 and keratan-SO4 in biological sources. Interaction of DAPI with GAG was elec…

Glycosaminoglycans (GAG) in pterygium tissues were analyzed and the results were compared with those in the normal human conjunctiva. A marked difference in the GAG composition was found: 78% of the total hexosamines of GAG from pterygium was glucosamine, whereas 98% of those of the conjunctiva was galactosamine. Pterygium contained mainly hyaluronic acid (48.8%), followed by chondroitin sulfate A, chondroitin sulfate C, chondroitin and dermatan sulfate in this order. The normal conjunctiva contained mainly chondroitin sulfate C (73.5%), followed by chondroitin sulfate A and chondroitin sulfat…
