A clostridial endo-beta-galactosidase that cleaves both blood group A and B glycotopes: the first member of a new glycoside hydrolase family, GH98.

We have isolated an endo-beta-galactosidase designated E-ABase from Clostridium perfringens ATCC 10543 capable of liberating both the A trisaccharide (A-Tri; GalNAcalpha1-->3(Fucalpha1-->2)Gal) and B trisaccharide (B-Tri; Galalpha1-->3(Fucalpha1-->2)Gal) from glycoconjugates containing blood group A and B glycotopes, respectively. We have subsequently cloned the gene (eabC) that encodes E-ABase from this organism. This gene was found to be identical to the CPE0329 gene of C. perfringens strain 13, whose product was labeled as a hypothetical protein (Shimizu, T., Ohtani, K., Hirakawa, H., Ohshi

α-N-acetylgalactosaminidase from infant-associated bifidobacteria belonging to novel glycoside hydrolase family 129 is implicated in alternative mucin degradation pathway.

Bifidobacteria inhabit the lower intestine of mammals including humans where the mucin gel layer forms a space for commensal bacteria. We previously identified that infant-associated bifidobacteria possess an extracellular membrane-bound endo-α-N-acetylgalactosaminidase (EngBF) that may be involved in degradation and assimilation of mucin-type oligosaccharides. However, EngBF is highly specific for core-1-type O-glycan (Galβ1-3GalNAcα1-Ser/Thr), also called T antigen, which is mainly attached onto gastroduodenal mucins. By contrast, core-3-type O-glycans (GlcNAcβ1-3GalNAcα1-Ser/Thr) are predom

Comparative transcriptomics of CAZy enzymes in white- and brown-rot agaricomycetes: Evolutionary insights into lignocellulose degradation and the relevance of GH16 glycoside hydrolase functional divergence.

White-rot and brown-rot fungi (WRF and BRF, respectively) decompose lignocellulose, the main structural component of plant biomass, through distinct mechanisms. This study examines the transcriptomic responses of three WRF species (Pleurotus ostreatus, Phanerochaete chrysosporium, and Heterobasidion irregulare) and two BRF species (Fomitopsis schrenkii and Rhodonia placenta) grown on poplar wood (W) and glucose (G) as sole carbon sources. RNA-seq analysis revealed upregulation of carbohydrate-active enzymes (CAZymes) linked to lignocellulose degradation. WRF displayed a broader enzymatic reper

Effect of glycoside hydrolase 18 chitinase on alternately β-1,4-linked glucose and N-acetylglucosamine derived from the sulfur-oxidizing bacterium Thiothrix nivea.

The filamentous sulfur-oxidizing bacterium Thiothrix nivea produces glucosaminoglucan (GG), which is β-1,4-linked copolymer of glucose and glucosamine. GG is a major component of the sheath which covers the bacterial filament, and the bacterium uses CO2 and organic acids as carbon sources in the presence of H2S. Similar to chitin, N-acetylated GG (NGG) can be degraded by lysozyme. However, it has not yet been confirmed whether NGG can be degraded by chitinase. Glycoside hydrolase 18 (GH18) chitinase is present in mammals and is produced by various microorganisms. In this study, the degradation

Glycoside hydrolase-mediated utilization of Poria cocos polysaccharide enriches Lactobacillus gasseri and activates the AhR-IL-22 axis to attenuate DSS-induced colitis.

Polysaccharides derived from edible fungi can alleviate ulcerative colitis (UC) by enriching beneficial probiotics. However, the mechanisms underlying their interaction with probiotics remain unclear. Here, The current study investigated how Poria cocos polysaccharide (PCP) enriches L. gasseri and the effects of L. gasseri on dextran sulfate sodium (DSS)-induced UC. Genomic analyses predicted multiple polysaccharide utilization loci in L. gasseri, and growth kinetics together with gene expression assays confirmed that PCP promoted L. gasseri growth and upregulated glycoside hydrolases (GH1 and

Biochemical and structural characterization of AmyY from Alkalimonas sp. NCh-2 reveals a class of metal-free α-amylases in glycoside hydrolase family 13.

α-Amylases (EC 3.2.1.1) are among the most important industrial enzymes in starch saccharification, detergent, paper, and textile industries. Canonical α-amylases typically contain Ca2+ in their structure, which is not directly involved in catalysis but essential in maintaining the structural integrity of α-amylases. Ca2+ ions are easily removed by chelating reagents, which may affect the activity and stability of α-amylases and thereby hinder their applications in some industries. Here, we identified a metal-free α-amylase, AmyY, from the bacterium Alkalimonas sp. NCh-2 isolated from an alkal

Azithromycin, an Antimicrobial Agent, Targets Glycoside Hydrolase Family 35 and Exhibits Potent Curative Activity Against Tea Plant Diseases Caused by Lasiodiplodia theobromae.

Tea disease caused by Lasiodiplodia theobromae is an emerging fungal disease that significantly reduces the yield and quality of tea in tea-producing regions owing to the lack of effective control methods. In this study, we evaluated the antifungal activity of azithromycin, a macrolide antibiotic, against L. theobromae. In vitro assays demonstrated strong inhibitory activity, with a half-maximal effective concentration (EC50) of 140.61 μg/ml, and in vivo application resulted in a 54.30% lesion inhibition rate at 800 μg/ml against tea leaf spot. A novel β-1,2-mannosidase GH130-BG31 from glycoside hydrolase family 130 targets healthcare-associated bacterial biofilms and potentiates antibiotic activity.

Hospital-acquired infections (HAIs) pose a major global health challenge, largely due to persistent biofilms formed by multidrug-resistant pathogens. In this study, we investigate GH130-BG31, a β-1,2-mannosidase from the GH130 glycoside hydrolase family, and demonstrate for the first time that it exhibits strong activity against mannose-rich biofilms associated with HAIs. By targeting β-1,2-mannosidic linkages in exopolysaccharides, GH130-BG31 inhibits 80% of biofilm formation in Escherichia coli and Staphylococcus epidermidis at 10 μg/mL, and in Klebsiella pneumoniae at 100 μg/mL. Expression, purification, crystallization and preliminary X-ray diffraction analysis of Aspergillus terreus endo-β-1,4-glucanase from glycoside hydrolase family 12.

Endoglucanases are important enzymes that are involved in the modification and degradation of cellulose. Filamentous fungi such as Aspergillus terreus are effective biomass degraders in nature owing to their capacity to produce an enzymatic arsenal of glycoside hydrolases, including endoglucanase from glycoside hydrolase family 12 (GH12). The A. terreus GH12 endoglucanase was cloned and overexpressed in A. nidulans, purified and crystallized. A single crystal was obtained from a solution consisting of 2 M ammonium sulfate, 5%(v/v) 2-propanol. X-ray diffraction data were collected to a resoluti

Structural analysis of a glycoside hydrolase family 43 arabinoxylan arabinofuranohydrolase in complex with xylotetraose reveals a different binding mechanism compared with other members of the same family.

AXHs (arabinoxylan arabinofuranohydrolases) are alpha-L-arabinofuranosidases that specifically hydrolyse the glycosidic bond between arabinofuranosyl substituents and xylopyranosyl backbone residues of arabinoxylan. Bacillus subtilis was recently shown to produce an AXH that cleaves arabinose units from O-2- or O-3-mono-substituted xylose residues: BsAXH-m2,3 (B. subtilis AXH-m2,3). Crystallographic analysis reveals a two-domain structure for this enzyme: a catalytic domain displaying a five-bladed beta-propeller fold characteristic of GH (glycoside hydrolase) family 43 and a CBM (carbohydrate

Crystallographic analysis shows substrate binding at the -3 to +1 active-site subsites and at the surface of glycoside hydrolase family 11 endo-1,4-beta-xylanases.

GH 11 (glycoside hydrolase family 11) xylanases are predominant enzymes in the hydrolysis of heteroxylan, an abundant structural polysaccharide in the plant cell wall. To gain more insight into the protein-ligand interactions of the glycone as well as the aglycone subsites of these enzymes, catalytically incompetent mutants of the Bacillus subtilis and Aspergillus niger xylanases were crystallized, soaked with xylo-oligosaccharides and subjected to X-ray analysis. For both xylanases, there was clear density for xylose residues in the -1 and -2 subsites. In addition, for the B. subtilis xylanas

Novel metabolic adaptation driven by glycoside hydrolase family 25 protein contributes to increasing trimethoprim-sulfamethoxazole resistance in clinical human Brucella melitensis isolates in China.

Brucellosis, caused by Brucella spp., is a globally zoonotic disease that results in substantial economic losses and public health concerns. Although antibiotic-resistant Brucella strains have been reported worldwide, the current status and underlying mechanisms of resistance among Chinese isolates remain poorly characterized. In this study, we analyzed 636 clinical human isolates of B. melitensis from China using genomic sequencing, transcriptomic sequencing, and neural network prediction to identify key determinants and mechanisms of antibiotic resistance. Cellulophaga algicola alginate lyase and Pseudomonas aeruginosa Psl glycoside hydrolase inhibit biofilm formation by Pseudomonas aeruginosa CF2843 on three-dimensional aggregates of lung epithelial cells.

Pseudomonas aeruginosa is an opportunistic pathogen that produces a biofilm containing the polysaccharides, alginate, Psl, and Pel, and causes chronic lung infection in cystic fibrosis patients. Others and we have previously explored the use of alginate lyases in inhibiting P. aeruginosa biofilm formation on plastic and lung epithelial cell monolayers. We now employ a more physiologically representative model system, i.e., three-dimensional aggregates of A549 lung epithelial cells cultured under conditions of microgravity in a rotary cell culture system to mimic the natural lung environment, a

Biofilm Dispersal and Wound Infection Clearance With Preclinical Debridement Agents.

Biofilms complicate wound care by causing recurrent infections that are often resistant to debridement and are highly antibiotic-tolerant. We investigated whether the addition of a biofilm dispersal agent could improve the efficacy of debridement. The previous studies have indicated that a glycoside hydrolase cocktail of alpha-amylase and cellulase can act as a potent biofilm dispersal agent. With in vitro and ex vivo Pseudomonas aeruginosa biofilm models, we compared glycoside hydrolases against other, clinically relevant, enzymatic debridement agents (papain, bromelain, and collagenase). Rapid acting α-transglucosidase from Aspergillus carbonarius: Cloning, characterization, in-silico analyses and synthesis of stable vitamin-C analogue using the recombinant enzyme.

The transglycosylation activity of many glycoside hydrolases (GH) can potentially synthesise di, oligo, polysaccharides, or glycosylate various non-carbohydrate moieties, increasing their bioavailability and solubility. While the transglycosylation potential of glycoside hydrolases makes them attractive candidates in glyco-synthesis due to their broad substrate specificity and ability to utilise simple sugar donors, the major challenge is the identification of potent transglucosidases for specific reactions. In this study, a GH 31 α-transglucosidase/glucosidase produced by Aspergillus carbonar

Fluorescence-quenched substrates for live cell imaging of human glucocerebrosidase activity.

Deficiency of the lysosomal glycoside hydrolase glucocerebrosidase (GCase) leads to abnormal accumulation of glucosyl ceramide in lysosomes and the development of the lysosomal storage disease known as Gaucher's disease. More recently, mutations in the GBA1 gene that encodes GCase have been uncovered as a major genetic risk factor for Parkinson's disease (PD). Current therapeutic strategies to increase GCase activity in lysosomes involve enzyme replacement therapy (ERT) and molecular chaperone therapy. One challenge associated with developing and optimizing these therapies is the difficulty in

1H, 15N and 13C backbone and side-chain resonance assignments of a family 32 carbohydrate-binding module from the Clostridium perfringens NagH.

The Gram-positive anaerobe Clostridium perfringens is an opportunistic bacterial pathogen that secretes a battery of enzymes involved in glycan degradation. These glycoside hydrolases are thought to be involved in turnover of mucosal layer glycans, and in the spread of major toxins commonly associated with the development of gastrointestinal diseases and gas gangrene in humans. These enzymes employ multi-modularity and carbohydrate-binding function to degrade extracellular eukaryotic host sugars. Here, we report the full (1)H, (15)N and (13)C chemical shift resonance assignments of the first f

Imaging Staphylococcus aureus biofilms in vivo reveals neutrophil exclusion by PNAG exopolysaccharide.

Staphylococcus aureus forms biofilms on biotic and abiotic surfaces, enabling persistent infections that evade immune clearance. Although many S. aureus strains can produce the biofilm-associated exopolysaccharide, poly-β-1,6-N-acetyl-D-glucosamine (PNAG), they often form protein-dominated biofilm matrices in vitro, leaving a role for PNAG unclear. Using intravital imaging in a foreign-body infection model, we found that PNAG in biofilms hinders neutrophil access and delays bacterial clearance. Neutrophil elastase was crucial for eventual biofilm clearance. In vivo PNAG labeling revealed that

Mechanistic Insights into β‑1,6-Branch Recognition and Orientation-Selective Hydrolysis by Endo-β-1,3-Glucanase Using Synthetic β‑Glucan Probes.

Synthetic Strategies and Immunological Evaluation of C-Linked α-Galactosylceramide Analogs.

Invariant natural killer T (iNKT) cells recognize lipid antigens, such as α-galactosylceramide (α-GalCer), presented on CD1d, leading to the production of cytokines. C-Glycoside analogs of α-GalCer have garnered attention due to their stability in vivo and their superior therapeutic effects in disease models, including anti-malarial and anti-tumor activity, compared to native α-GalCer. The carbon linkage of C-glycosides is fundamentally inert to glycosyl hydrolases, but the loss of hydrogen-bonding ability and changes in conformational distribution may influence the biological activity.
