The DJ-1 superfamily member Hsp31 repairs proteins from glycation by methylglyoxal and glyoxal.

Hsp31 belongs to the PfpI/Hsp31/DJ-1 superfamily, and has been reported to display chaperone, peptidase and glutathione-independent glyoxalase activities. Here, we show that Hsp31 repairs glyoxal- and methylglyoxal-glycated amino acids and proteins and releases repaired proteins and lactate or glycolate, respectively. Hsp31 deglycates cysteine, arginine and lysine by acting on early glycation intermediates (hemithioacetals and aminocarbinols) and prevents the formation of Schiff bases and advanced glycation endproducts. Hsp31 repairs glycated serum albumin, glyceraldehyde-3-phosphate dehydroge

A glyoxal-specific aldehyde signaling axis in Pseudomonas aeruginosa that influences quorum sensing and infection.

The universally conserved α-oxoaldehydes glyoxal (GO) and methylglyoxal (MGO) are toxic metabolic byproducts whose accumulation can lead to cell death. In the absence of a known, natural inducer of the GO-specific response in prokaryotes, we exploited RNA-seq to define a GO response in the bacterial pathogen Pseudomonas aeruginosa. The highest upregulated operon consisted of the known glyoxalase (gloA2) and an antibiotic monooxygenase (ABM) domain of unknown function - renamed here Aldehyde responsive quorum-sensing Inhibitor (ArqI). The arqI-gloA2 operon is highly specific to GO induction and

The Pyruvate-Glyoxalate Pathway as a Toxicity Assessment Tool of Xenobiotics: Lessons from Prebiotic Chemistry.

There is an urgent need to evaluate the toxicity of xenobiotics and environmental mixtures for preventing loss in water quality for the sustainability of aquatic ecosystems. A simple prebiotic chemical pathway based on malate formation from pyruvate (pyr) and glyoxalate (glyox) is proposed as a quick and cheap screening tool for toxicity assessment. The assay is based on the pyr and glyox (aldol) condensation reactions, leading to biologically relevant precursors such as oxaloacetate and malate. Incubation of pyr and glyox at 40-70 °C in the presence of reduced iron Fe(II) led to malate format

An anthracenecarboximide-guanidine fluorescent probe for selective detection of glyoxals under weak acidic conditions.

An anthracenecarboximide-guanidine based turn-on fluorescent probe ANC-DCP-1 for selective detection of glyoxals (methylglyoxal and glyoxal, GOS) over formaldehyde under weak acidic conditions around pH 6.0 was reported. The probe showed great potential in studying relative GOS levels in weak acidic biological fluids such as in urine for diabetic diagnosis and prognosis, and also found application in the food industry such as for fast unique manuka factor (UMF) scale determination of Manuka honey.

Glyoxal is a superior fixative to formaldehyde in promoting antigenicity and structural integrity in murine cardiac tissues.

BACKGROUND: Immunofluorescence (IF) is an essential technique for evaluating histological and biochemical changes in tissue specimens. A critical step in IF is sample fixation, typically achieved using formaldehyde-based fixatives, such as 4 % paraformaldehyde (PFA) or 10 % formalin. However, these fixatives are prone to over-fixation, which can alter antigenicity and promote artifacts. This study investigated glyoxal, a two‑carbon dialdehyde, as a potential alternative fixative for murine cardiac tissues for IF and crosslinking immunoprecipitation-mass spectrometry (xIP-MS) applications. Copper(I)-catalyzed tandem synthesis of 2-acylquinolines from 2-ethynylanilines and glyoxals.

An efficient one-step synthesis of 2-acylquinolines using a copper-catalyzed tandem reaction of 2-ethynylanilines with glyoxals in the presence of piperidine has been developed. This new protocol successfully avoids multi-step operation and the use of highly toxic cyanides required in traditional methods, and provides a practical tool for synthetic and pharmaceutical chemists. Various 2-acylquinolines are obtained with perfect regioselectivity in moderate to good yields (up to 86%). The potential synthetic utility of this method is exemplified by a large-scale experiment and synthetic transfor

The effects of acetaldehyde, glyoxal and acetic acid on the heterogeneous reaction of nitrogen dioxide on gamma-alumina.

Heterogeneous reactions of nitrogen oxides on the surface of aluminium oxide result in the formation of adsorbed nitrite and nitrate. However, little is known about the effects of other species on these heterogeneous reactions and their products. In this study, diffuse reflectance infrared spectroscopy (DRIFTS) was used to analyze the process of the heterogeneous reaction of NO2 on the surface of aluminium oxide particles in the presence of pre-adsorbed organic species (acetaldehyde, glyoxal and acetic acid) at 298 K and reveal the influence of these organic species on the formation of adsorbe

Methylglyoxal and Glyoxal in High-Puff Disposable Electronic Cigarette Liquids: Unexpected Accumulation and Enhanced Cytotoxicity.

Despite concerns regarding the potential health risks associated with EC use, their design, puff count, and e-liquid formulations have continued to evolve. Aldehydes are a key class of thermal degradation byproducts generated during the vaporization of EC fluids. This study tested the hypothesis that aldehyde concentrations would be elevated in used ECs with high puff numbers. Devices from multiple brands were analyzed to assess the impact of extended use on aldehyde formation. E-fluids were extracted from field-collected and user-donated devices and analyzed using gas chromatography/mass spec

HDGF Protects Retinal Pigment Epithelium from Glyoxal-Induced Ferroptosis via SIRT1/PGC-1α/Nrf2 Pathway.

Age-related macular degeneration (AMD) is driven in part by the accumulation of reactive metabolites like glyoxal (GO), which induces retinal pigment epithelium (RPE) degeneration. Here, we demonstrate that GO triggers ferroptosis in human ARPE-19 cells, as characterized by iron-dependent lipid peroxidation, glutathione depletion, and reactive oxygen species (ROS) accumulation. This ferroptotic cell death is coupled with profound mitochondrial dysfunction, featuring network fragmentation and the downregulation of the key regulators MFN2, PGC-1α, and SIRT1. We identify hepatoma-derived growth f

Development of environmentally friendly glyoxal-based adhesives with outstanding water repellency utilizing wheat gluten protein.

To reduce the release of volatile organic compounds (VOCs) from formaldehyde-based adhesives at the source, the use of low-toxicity and biodegradable glyoxal instead of formaldehyde for the preparation of novel urea-glyoxal resins is a simple and promising strategy. The limited water resistance and adhesive strength of the new urea-glyoxal resins (UG) restrict their extensive application. This study prepared a high-performance, water-resistant WP-UG wood adhesive by combining UG prepolymer with wheat gluten protein (WP). FTIR, XRD, and XPS confirmed the existence of a chemical reaction between

Sensitive detection of glyoxal by cluster-mediated CH2Br2+ chemical ionization time-of-flight mass spectrometry.

Direct and rapid analysis of glyoxal by soft ionization mass spectrometry remains a great challenge due to its low ionization efficiency in existing soft ionization techniques, such as proton transfer reaction (PTR) and photoionization (PI). In this work, we developed a new VUV lamp-based cluster-mediated CH2Br2+ chemical ionization (CMCI) source for time-of-flight mass spectrometry (TOFMS), which was accomplished by employing photoionization-generated CH2Br2+ as the reactant ion and co-sampling of glyoxal with high-concentration ethanol (EtOH). The signal intensity of glyoxal could be enhance

Chemical ionization of glyoxal and formaldehyde with H3O+ ions using SIFT-MS under variable system humidity.

Glyoxal (C2H2O2) is a highly reactive molecule present at trace levels in specific gaseous environments. For analyses by chemical ionization mass spectrometry, it is important to understand the gas-phase chemistry initiated by reactions of H3O+ ions with C2H2O2 molecules in the presence of water vapour. This chemistry was studied at variable humidity using a selected ion flow tube, SIFT. The initial step is a proton transfer reaction forming protonated glyoxal C2H3O2+. The second step, in the presence of water vapour, is the association forming C2H3O2+(H2O) and interestingly also protonated fo

Non-toxic acid-free glyoxal fixative for veterinary gross specimen preservation, histopathology, immunohistochemistry, and molecular analysis.

Formaldehyde-based fixation is the most used chemical system for histopathological examination worldwide. However, its toxicity is well known, and preservation of gross features, proteins, and nucleic acids is not optimal. Alternative fixatives resulting in similar morphological tissue quality and costs, but with reduced toxicity and with better preservation of gross features, proteins, and nucleic acids would increase operator safety and application possibilities in pathology. This multi-institutional study aimed to compare the morphological, histochemical, immunohistochemical (IHC), and mole

Rapid formation of N ε-(carboxymethyl)lysine (CML) from ribose depends on glyoxal production by oxidation.

N ε-(Carboxymethyl)lysine (CML) is a major advanced glycation end-product (AGE) involved in protein dysfunction and inflammation in vivo. Its accumulation increases with age and is enhanced with the pathogenesis of diabetic complications. Therefore, the pathways involved in CML formation should be elucidated to understand the pathological conditions involved in CML. Ribose is widely used in glycation research because it shows a high reactivity with proteins to form AGEs. We previously demonstrated that ribose generates CML more rapidly than other reducing sugars, such as glucose; however, the

Mechanism of Carbon Skeleton Formation of 2,3,5-Trimethylpyrazine via a Conversion Reaction between Methylglyoxal and Glyoxal.

Maillard flavor compounds, such as 2,3,5-trimethylpyrazine, have been frequently identified in thermally processed food products, such as popcorn and peanuts. However, the origin of the carbon atoms in 2,3,5-trimethylpyrazine has not been clearly elucidated. Herein, a model reaction showed that precursor methylglyoxal and intermediates glyoxal and formaldehyde contributed to the formation of 2,3,5-trimethylpyrazine via a conversion reaction between methylglyoxal and glyoxal. In addition, carbon module labeling technology and model response validation experiments indicated that this transformat

Analysis of the binding of warfarin to glyoxal- and methylglyoxal-modified human serum albumin by ultrafast affinity extraction.

Ultrafast affinity extraction (UAE) and affinity microcolumns containing immobilized human serum albumin (HSA) were employed to evaluate the effect of advanced stage glycation on HSA and its binding to warfarin, a common site-specific probe for Sudlow site I of this protein. The modification of HSA by glyoxal (GO) and methylglyoxal (MGO) was considered, where GO and MGO are known to be important in the formation of many types of advanced glycation end products. Free drug fractions were measured by UAE for warfarin in solutions containing normal HSA or HSA that had been modified by GO or MGO at

Flow injection chemiluminescence analysis of some penicillins by their sensitizing effect on the potassium permanganate-glyoxal reaction.

A new chemiluminescence method using flow injection is described for the determination of four penicillins, namely: phenoxymethylpenicillin potassium, amoxicillin, ampicillin, and ampicillin sodium. The method is based on sensitizing effect of these drugs on the chemiluminescence reaction of potassium permanganate in sulfuric acid with glyoxal. The different experimental parameters affecting the chemiluminescence intensity were carefully studied and incorporated into the procedure. The method allows the determination of 0.1-1.0 microg/ml phenoxymethylpenicillin potassium, 0.1-1.0 microg/ml amo

A Glyoxal Based Co-Condensation Adhesive with Excellent Water Resistance Using Chitosan and Starch as Enhanced Agents.

To address the hazards posed by formaldehyde emissions from wood-based products to human health and the indoor environment, research on wood adhesives has focused on developing green and eco-friendly alternatives. However, the limited water resistance and bonding strength of bio-based or glyoxal-based adhesives have hindered their practical application. In this work, a co-condensation method was employed to prepare glyoxal-based co-condensation adhesive incorporating starch and a small amount of chitosan as synergistic reinforcing agents to enhance their cross-linking extent. Considering cost
