Potent Antibiotics Active against Multidrug-Resistant Gram-Negative Bacteria.

The emergence of multidrug-resistant (MDR) Gram-negative bacteria has become a global problem. Among MDR Gram-negative bacteria, carbapenem-resistant Enterobacteriaceae (CRE), MDR Pseudomonas aeruginosa, and MDR Acinetobacter baumannii have limited treatment options and present serious threats. Therefore, strong countermeasures must be taken against these bacteria immediately. Accordingly, the focus of this review is on recent advances in the development of promising antibacterial agents against MDR Gram-negative bacteria. These agents include novel tetracyclines, polymyxins, β-lactams, β-lact

Chemogenomic Screen for Imipenem Resistance in Gram-Negative Bacteria.

Carbapenem-resistant Gram-negative bacteria are considered a major threat to global health. Imipenem (IMP) is used as a last line of treatment against these pathogens, but its efficacy is diminished by the emergence of resistance. We applied a whole-genome screen in Escherichia coli, Klebsiella pneumoniae, and Pseudomonas aeruginosa isolates that were submitted to chemical mutagenesis, selected for IMP resistance, and characterized by next-generation sequencing. A comparative analysis of IMP-resistant clones showed that most of the highly mutated genes shared by the three species encoded prote

Multidrug-resistant Gram-negative bacteria: a product of globalization.

Global trade and mobility of people has increased rapidly over the last 20 years. This has had profound consequences for the evolution and the movement of antibiotic resistance genes. There is increasing exposure of populations all around the world to resistant bacteria arising in the emerging economies. Arguably the most important development of the last two decades in the field of antibiotic resistance is the emergence and spread of extended-spectrum β-lactamases (ESBLs) of the CTX-M group. A consequence of the very high rates of ESBL production among Enterobacteriaceae in Asian countries is

Gram-negative bacteria in water distribution systems of hospitals.

A total of 67 samples of tap water were collected from faucets and showerheads in 6 hospitals located in the Lublin province (eastern Poland). The samples were examined for the presence and species composition of Legionella, Gram-negative bacteria belonging to family Enterobacteriaceae (GNB-E) and Gram-negative bacteria not belonging to family Enterobacteriaceae (GNB-NE), by filtering through cellulose filters and culture on respectively GVPC, EMB and tryptic soya agar media. On average, Legionella was isolated from 65.7% of the water samples taken in hospitals. Strains of the Legionella pneum

[Beta-lactamases of Gram negative bacteria: never-ending clockwork!].

The acquired resistance against the wide-spectrum and highly stable beta-lactams including third-generation cephalosporins (3GC) and carbapenems is constinuously increasing and widespead with the discovery of various plasmid-encoded, or genes cassette or integrons coding for a novel beta-lactamase, always a major mechanism of resistance. To explain resistance against 3GC, with the continuing story with TEM and SHV mutated enzymes, several types of ESBL (class A) emerge the CTX-M type, at least CTX-M-40, but also other non predominant types intitled BES, GES, PLA, PER, VEB. The wider resistance

Selection and characterization of cefepime-resistant gram-negative bacteria.

The NCTC type strains and four clinical isolates of Enterobacter cloacae, Citrobacter freundii, Serratia marcescens, Morganella morganii, Providencia stuartii and Pseudomonas aeruginosa were exposed, in agar, to cefepime at 3, 5, and 10 x MIC and a breakpoint concentration of 16 mg/L. Mutants were selected at a frequency of approximately 10(-8) that had decreased susceptibility to cefepime and cefpirome, and species-dependent resistance to other beta-lactams. Any putative mutant with a greater than four-fold increase in the MIC was examined to determine its beta-lactamase expression and outer

IMT-P8 potentiates Gram-positive specific antibiotics in intrinsically resistant Gram-negative bacteria.

Gram-negative bacteria (GNB) pose a major global public health challenge as they exhibit a remarkable level of resistance to antibiotics. One of the factors responsible for promoting resistance against a wide range of antibiotics is the outer membrane (OM) of Gram-negative bacteria. The OM acts as a barrier that prevents the entry of numerous antibiotics by reducing their influx (due to membrane impermeability) and enhancing their efflux (with the help of efflux pumps). Our study focuses on analyzing the effect of IMT-P8, a cell-penetrating peptide (CPP), to enhance the influx of various Gram-

Rational Design of Natural Xanthones Against Gram-negative Bacteria.

Most antibiotics are ineffective against Gram-negative bacteria owing to the existence of the outer membrane (OM) barrier. The rational design of compounds to expand their antibacterial spectra of antibiotics solely targeting Gram-positive pathogens remains challenging. Here, the design of skeletons from natural products to penetrate the OM are deciphered. Structure-activity relationship analysis shows the optimization of the model of natural xanthones α-mangostin endows the broad-spectrum antibacterial activity. Mechanistic studies demonstrate the lead compound A20 penetrates the OM in a self

Cefoperazone/sulbactam: New composites against multiresistant gram negative bacteria?

Sulbactam, a class A β-lactamase inhibitor, added to cefoperazone either at a fixed 8 mg/L level of sulbactam or at a level of fixed cefoperazone: sulbactam ratio (2:1) would constitute a combination form of cefoperazone/sulbactam, which has better activities against Enterobacteriaceae, Pseudomonas aeruginosa and Acinetobacter baumannii than cefoperazone alone. Cefoperazone/sulbactam (1:1 or 1:2) has greater in-vitro activity against most multidrug-resistant organisms (ESBL- and AmpC-producing Enterobacteriaceae and carbapenem-resistant A. baumannii except for carbapenem-resistant P. aeruginos

Lipopolysaccharide modification in Gram-negative bacteria during chronic infection.

The Gram-negative bacterial lipopolysaccharide (LPS) is a major component of the outer membrane that plays a key role in host-pathogen interactions with the innate immune system. During infection, bacteria are exposed to a host environment that is typically dominated by inflammatory cells and soluble factors, including antibiotics, which provide cues about regulation of gene expression. Bacterial adaptive changes including modulation of LPS synthesis and structure are a conserved theme in infections, irrespective of the type or bacteria or the site of infection. In general, these changes resul

[Resistant gram-negative bacteria. Therapeutic approach and risk factors].

The rapid spread of multidrug-resistant bacteria has become a serious threat, especially in critical care units, thereby prolonging the hospital stay. Enterobacteriaceae have a high capacity to adapt to any environment. Plasmids are the reason behind their expansion. The choice of empiric therapy for intra-abdominal or urinary infections requires knowledge of the intrinsic microbiological variability of each hospital or critical care unit, as well as the source of infection, safety or antibiotic toxicity, interaction with other drugs, the dosage regimen and the presence of risk factors. Detection of AmpC β Lactamases in Gram-negative Bacteria.

AmpC β-lactamases are clinically important cephalosporinases encoded on the chromosomes of many Enterobacteriaceae and a few other organisms, where they mediate resistance to cephalothin, cefazolin, cefoxitin, most penicillins, and β-lactamase inhibitor/β-lactam combinations. The increase in antibiotic resistance among Gram-negative bacteria is a notable example of how bacteria can procure, maintain and express new genetic information that can confer resistance to one or several antibiotics. Detection of organisms producing these enzymes can be difficult, because their presence does not always

[Clinically important beta-lactamases of gram-negative bacteria: AmpC].

Beta-lactamases are the most common cause of beta-lactam resistance in Gram-negative bacteria. With third-generation and fourth-generation cephalosporins being introduced into practice, new beta-lactamases have evolved, able to hydrolyze these antibiotics. AmpC-type beta lactamases (cephalosporinases) are serine enzymes with the ability to hydrolyze penicillins, monobactams and cephalosporins of all generations, including cephamycins. Over the last two decades, transferable plasmid-mediated class C beta-lactamases have been reported with increasing frequency. The genes for resistance to other

Bone and joint infections caused by gram-negative bacteria.

Infections of the bones and joints caused by gram-negative bacteria from the family Enterobacteriaceae account for 10%-15%. This family usually cause opportunistic infections, due to a low virulence factor, host and environmental factors. Gram-negative bacteria cause osteomyelitis, infectious arthritis and Reiter's syndrome. The family Enterobacteriaceae belong to the physiological flora of man and cause opportunistic and hospital infections. In order to reach an accurate etiologic diagnosis, it is therefore important for the samples for bacteriologic examination to be properly taken. The qual

Immunity targeting common core antigens of gram-negative bacteria.

Antibodies against common gram-negative core antigens can provide protection against environmental, commensual, and contagious bacteria that cause life-threatening gram-negative sepsis/endotoxemia. Cross-protective immunity may be effective against many common livestock diseases, including neonatal coliform septicemia, coliform mastitis, septic metritis, and severe enteritis. The greatest potential benefit of cross-protective immunity may be realized in the reduction of ill-thrift and mortality of neonates. The use of active and passive immunization against core antigens common to gram-negativ

[Genetic determinants of antibiotic resistance in gram negative bacteria].

Modern data on spreading, structural and functional organization and evolution of the genetic determinants of antibiotic resistance in the gram-negative bacteria are reviewed. Some mechanisms for resistance to trimethoprime, sulphonamides, tetracyclines, chloramphenicol aminoglycosides, beta-lactam antibiotics controlled by the plasmid and chromosomal genes are presented. The problem of using the molecular DNA-probes containing the genetical determinants for antibiotic resistance in the practical work of clinical laboratories is discussed.

Extremely thermophilic gram-negative bacteria from hot tap water.

Two strains of heterotrophic, non-motile, Gram-negative extreme thermophiles have been isolated from hot tap water. These strains (NH and DI) have been characterized and compared with strains of genus Thermus. Few of the single organic compounds tested supported growth in the presence of ammonium salts, and, like Thermus strains, growth on undefined media was restricted to dilute tryptone-yeast extract-mineral salt solutions. Nutrients agar and similar common laboratory media did not support growth. The growth rate was similar to that of Thermus strains, as was the unusual pattern of antibioti

New cationic diketopyrrolopyrrole photosensitizers for the photoinactivation of gram-negative and gram-positive bacteria.

Diketopyrrolopyrroles (DPP) are a versatile group of dyes and pigments with a wide range of applications, namely in the biological field. In this paper, we report the synthesis and the photodynamic activity of new cationic DPP derivatives that effectively inactivate Gram-positive and Gram-negative bacteria. These new compounds were synthetized using an easy synthetic procedure to introduce one or two pyridin-4-ylsulfanyl units followed by cationization. The photodynamic effectiveness of these DPP derivatives against Pseudomonas aeruginosa [Gram-negative] and methicillin-resistant Staphylococcu
